以甲氧基和卤素取代的苯甲酸为起始原料,经过羧酸酰基化、傅克酰基化、芳香环卤代等反应合成了12个卤代甲氧基二苯甲酮类化合物,其结构经ESI-MS、1H NMR、13C NMR进行了表征和确认。通过考察其对H2O2诱导的人脐静脉内皮细胞(HUVECs)损伤的保护活性,讨论了此类化合物的构效关系。体外活性实验表明,浓度为12.5μmol/L时,化合物7a和9a对HUVECs有一定的保护活性,保护率超过50%。  相似文献

Four novel steroidal alkaloids named cortistatins E (1), F (2), G (3), and H (4) have been isolated from the marine sponge Corticium simplex. The chemical structures of these four cortistatins, which are unique abeo-9(10-19)-stigmastane-type steroidal alkaloids having oxabicyclo[3.2.1]octene and N-methyl piperidine or 3-methylpyridine units in the side chain, were elucidated by the detailed 2D-NMR analysis. These four compounds showed only weak anti-proliferative activity against human umbilical vein endothelial cells (HUVECs) at 0.35-1.9 μM concentrations in contrast to cortistatin A (5), which was isolated as a highly selective inhibitor of proliferation of HUVECs from the same marine sponge.  相似文献

Endothelial cell dysfunction is considered to be one of the major causes of vascular complications in diabetes. Polyphenols are known as potent antioxidants that can contribute to the prevention of diabetes. Corn silk has been reported to contain polyphenols and has been used in folk medicine in China for the treatment of diabetes. The present study aims to investigate the potential protective role of the phenolic-rich fraction of corn silk (PRF) against injuries to vascular endothelial cells under high glucose conditions in vitro and in vivo. The protective effect of PRF from high glucose toxicity was investigated using human umbilical vein endothelial cells (HUVECs). The protective effect of PRF was subsequently evaluated by using in vivo methods in streptozotocin (STZ)-induced diabetic rats. Results showed that the PRF significantly reduced the cytotoxicity of glucose by restoring cell viability in a dose-dependent manner. PRF was also able to prevent the histological changes in the aorta of STZ-induced diabetic rats. Results suggested that PRF might have a beneficial effect on diabetic patients and may help to prevent the development and progression of diabetic complications such as diabetic nephropathy and atherosclerosis.  相似文献

One new pyrrole alkaloid,N-(2E)-3-(3,4-dihydrophenyl)prop-N1-(4-aminoburyl)-3-pyrrole formaldehyde (1),was isolated from the whole herbs of Selaginella moellendorfii Hieron.The structure was elucidated by spectroscopic analyses including UV,1R,1D NMR,2D NMR and MS methods.Additionally, compound 1 exhibited potent protective effect against the injury of human umbilical vein endothelial cell(HUVECs) induced by high concentrations of glucose in vitro.  相似文献

用丝素蛋白(SF)对微生物合成的高分子聚合物聚(3-羟基丁酸酯-co-3-羟基己酸酯)(PHBHHx)进行亲水改性,以提高材料的生物相容性.水接触角测定和表面自由能分析表明,丝素蛋白在支架表面吸附,使PHBHHx材料表面的水接触角从90°降至51°,表面自由能从37.9 mJ/m2增至57.4 mJ/m2,因而增加了材料的亲水性.进一步对亲水性改性前后PHBHHx多孔支架与人脐静脉内皮细胞(HUVECs)的相容性进行了比较.MTT法细胞活力分析表明,细胞在支架上培养3,5,7天后,其在SF改性PHBHHx多孔支架上的活力显著高于在未改性的PHBHHx支架上的活力;扫描电镜观察细胞生长形貌表明,细胞在改性后多孔支架上黏附及生长5天后,形成了连续细胞单层,其生长状态优于在未改性的PHBHHx支架上的生长状态;胶原含量测定表明细胞在改性后支架上比在未改性支架上有更好的胶原分泌能力,即改性后支架更利于诱导HUVECs分泌细胞外基质(ECM)从而构建类似体内的生长环境.  相似文献

Sepsis and septic shock, which are conditions triggered by infection, occur with high incidence in emergency departments and are among the most common causes of death in hospitalized patients worldwide. Therefore, the identification of sepsis biomarkers for the rapid diagnosis is a major goal for researchers in the field of critical care. Endothelial cells play a pivotal role in orchestrating the inflammatory response triggered by sepsis. In this study, we used proteomics to investigate the secretome of EA.hy926 endothelial cells following lipopolysaccharide (LPS) stimulation with 1 μg/mL LPS for 12 or 24 h. SILAC in cell cultures and an online 2D‐LC‐MS/MS system were used to analyze the secretome dynamics in response to LPS. We found that 22 of the 77 secreted proteins identified in both the presence and absence of LPS and that 19 of the secreted proteins were quantified more strongly after LPS treatment for 24 h than after treatment for 12 h. By Gene Ontology and KEGG pathway analyses, we found that proteins related to the regulation of the actin cytoskeleton showed the highest secretion response to LPS stimulation. Out of the 19 candidate proteins, we focused on moesin, which is involved in the function of endothelial cells, and confirmed its amount in cellular lysates and media taken from primary human umbilical vein endothelial cells treated with LPS. To our knowledge, this study provides the first in‐depth analysis of the LPS‐induced secretome in human endothelial cells, and we propose 19 new biomarker candidates for sepsis, including moesin.  相似文献

Three novel anti-angiogenic steroidal alkaloids, cortistatins J (1), K (2), L (3), have been isolated from the Indonesian marine sponge Corticium simplex. The chemical structures of cortistatins J (1), K (2), and L (3) were determined by 2D-NMR analysis to be unique abeo-9(10-19)-androstane-type steroidal alkaloids having isoquinoline unit instead of the side chain part, respectively. Cortistatin J (1) showed cytostatic anti-proliferative activity against human umbilical vein endothelial cells (HUVECs) at 8 nM, in which the selective index was 300-1000-fold in comparison with other cell lines.  相似文献
