Galloping is characterized by large and periodical oscillations which may lead to collapse of slender structures. This study is the first attempt of a comprehensive experimental and theoretical investigation of galloping of transversely inclined prisms. A modified quasi-steady model is proposed with a constant term to estimate the galloping of a transversely inclined prism, which is later experimentally investigated by conducting a static Synchronous Multi-Pressure Sensing System (SMPSS) test and an aeroelastic test in a boundary layer wind tunnel. The galloping responses of the prisms were measured in the aeroelastic test, while the aerodynamic force coefficients were determined from the SMPSS test. These experimental results were subsequently utilized to validate the quasi-steady model. Based on the proposed model, the galloping responses of the prisms were predicted and compared with the experimental results. The experimentally measured and theoretically predicted galloping responses are discussed with respect to aerodynamic damping ratios, onset galloping wind speeds, distributed pressure coefficients, point pressure spectra and vortex shedding frequencies. Based on the mechanical experimental results of methane hydrate (MH), a bond contact model considering the rate-dependency of MH is proposed. A CFD–DEM scheme considering fluid compressibility is used to simulate a series of undrained cyclic shear tests of numerical methane-hydrate-bearing sediment (MHBS) samples. The dynamic behavior, including stress–strain relationship, dynamic shear modulus, and damping ratio, is investigated. In addition, the force chains, contact fabric and averaged pure rotation rate (APR) are examined to investigate the relationships between micromechanical variables and macromechanical responses in the DEM MH samples. The effects of temperature, confining pressure and MH saturation are also analyzed. Due to the micro-structural strengthening by the MH bonds, no obvious change in microscopic quantities is observed, and the samples remain at the elastic stage under the applied low-shear stress level. When confining pressure and MH saturation increase, the dynamic elastic modulus increases, while the damping ratio decreases. An increasing temperature (leading to weakening of MH bonds) can lower the dynamic elastic modulus, but has almost no impact on the damping ratio. On the contrary, an increasing cyclic shear stress level lowers the damping ratio, but has almost no effect on the dynamic elastic modulus. 相似文献

Skin cancer studies depend on questionnaires to estimate exposure to ultraviolet light and subsequent risk but are limited by recall bias. We investigate the feasibility of developing a short checklist of categories comprising outdoor activities that can improve sun exposure questionnaires for use in epidemiologic studies. We recruited 124 working and retired U.S. radiologic technologists (52% women). Each subject was instructed to complete a daily activity diary, listing main indoor and outdoor activities between 9:00 A.M. and 5:00 P.M. during a 7 day period. A total of 4697 entries were associated with 1408 h (21.1%) of the total 6944 h spent outdoors. We were able to classify the activities into seven main activity categories: driving, yard work, home-maintenance, walking or performing errands, water activities, other recreational or sports activities and leisure activities or relaxing outside. These activities accounted for more than 94% of time spent outdoors both for working and retired men and women. Our data document the feasibility and guidance for developing a short checklist of outdoor activities for use in epidemiologic questionnaires for estimating sunlight exposures of working and retired indoor workers. 相似文献

channel and consequently affects the separation quality. We describe a method of reducing the Joule heating effect by incorporating
photonic crystal fiber into a micro capillary electrophoresis chip. The photonic crystal fiber consists of a bundle of extremely
narrow hollow channels, which ideally work as separation columns. Electrophoretic separation of DNA fragments was simultaneously
but independently carried out in 54 narrow capillaries with a diameter of 3.7 μm each. The capillary bundle offers more efficient

Side-chain backbone interactions (or "effects") between nearest neighbours may severely restrict the conformations accessible to a polypeptide chain and thus represent the first step in protein folding. We have quantified nearest-neighbour effects (i to i+1) in peptides through reversed-phase liquid chromatography (RP-HPLC) of model synthetic peptides, where L- and D-amino acids were substituted at the N-terminal end of the peptide sequence, adjacent to a L-Leu residue. These nearest-neighbour effects (expressed as the difference in retention times of L- and D-peptide diastereomers at pHs 2 and 7) were frequently dramatic, depending on the type of side-chain adjacent to the L-Leu residue, albeit such effects were independent of mobile phase conditions. No nearest-neighbour effects were observed when residue i is adjacent to a Gly residue. Calculation of minimum energy conformations of selected peptides supported the view that, whether a L- or D-amino acid is substituted adjacent to L-Leu, its orientation relative to this bulky Leu side-chain represents the most energetically favourable configuration. We believe that such energetically favourable, and different, configurations of L- and D-peptide diastereomers affect their respective interactions with a hydrophobic stationary phase, which are thus quantified by different RP-HPLC retention times. Side-chain hydrophilicity/hydrophobicity coefficients were generated in the presence of these nearest-neighbour effects and, despite the relative difference in such coefficients generated from peptides substituted with L- or D-amino acids, the relative difference in hydrophilicity/hydrophobicity between different amino acids in the L- or D-series is maintained. Overall, our results demonstrate that such nearest-neighbour effects can clearly restrict conformational space of an amino acid side-chain in a polypeptide chain. 相似文献

We report on a novel concept of sample loading for microfluidic devices using a benchtop centrifuge and a magnetically actuated circular closed-loop PCR microchip as a model system. The PCR mixture and the ferrofluid were loaded into a specially designed microchip. The microchip was then placed in an off-the-shelf 50-mL tube and centrifuged. The strong centrifugal force drives the PCR mixture and the ferrofluid into the microchannels of the microchip, and simultaneously expels any trapped microbubbles. PCR was successfully carried out on single and parallel closed-loop PCR microchips. The addition of a few off-chip handling steps allows great simplification of the device design. This new loading concept may be useful for designing robust and low-cost lab-on-a-chip devices because benchtop centrifuges are quite common in most laboratories. Figure
