A short and simple route to the synthesis of pentadec-1-en-4-ol, an important synthetic building block for the aculeatins F and epi-F, insect pheromone 5-hexadecanolide, solenopsin and various other natural products has been developed via proline-catalyzed α-aminoxylation of an aldehyde and hydrolytic kinetic resolution of a terminal epoxide. While the synthesis of aculeatins F and epi-F has been accomplished using a PIFA promoted oxidative spirocyclization/dithiane deprotection reaction sequence and linchpin coupling as key steps, the synthesis of hexadecanolide and a formal synthesis of solenopsin was performed using ring-closing metathesis (RCM) as key step.  相似文献

Motivated by a novel gas embolotherapy technique, a bench top vascular bifurcation model is used to investigate the splitting of long bubbles in a series of liquid-filled bifurcations. The developmental gas embolotherapy technique aims to treat cancer by infarcting tumors with gas emboli that are formed by selective acoustic vaporization of 6 μm, intravascular, perfluorcarbon droplets. The resulting gas bubbles are large enough to extend through several vessel bifurcations. The current bench top experiments examine the effects of gravity and flow on bubble transport through multiple bifurcations. The effect of gravity is varied by changing the roll angle of the bifurcating network about its parent tube. Splitting at each bifurcation is nearly even when the roll angle is zero. It is demonstrated that bubbles can either stick at one of the second bifurcations or in the second generation daughter tubes, even though the flow rate in the parent tube is constant. The findings of this work indicate that both gravity and flow are important in determining the bubble transport, and suggest that a treatment strategy that includes multiple doses may be effective in delivering emboli to vessels not occluded by the initial dose.  相似文献

US FDA released guidelines for bioanalytical method validation in 2001 and it became the basis for guidelines such as ANVISA and EMA. Even though there is a general agreement between these guidelines in terms of evaluation of validation parameters, significant diversity exists with respect to methodology employed. Present review compares and summarizes the regulatory guidelines issued by US FDA, ANVISA and EMA for bioanalytical method validation. This review also discusses evaluation of certain validation parameters such as matrix effect, incurred sample reanalysis, various stability aspects, effect of anticoagulant counter ions, specificity in the presence of concomitant medications, and identification of pharmacokinetic repeats wherein specific guidance and general consensus amongst scientific community does not exist.  相似文献

Fused triazoloquinolines have been prepared starting from (E)-3-(2-nitrophenyl)-1-aryl-prop-2-en-1-ones and sugar or benzyl azides in a sequential [3+2] cycloaddition reaction, followed by one pot Pd–C assisted reduction, cyclization and aromatization. The triazolyl fused quinolines with N1-glycosyl substituents as unnatural nucleosides have inherent potential to generate a library of compounds for bioevaluations.  相似文献

A cinnamyl alcohol dehydrogenase (CAD) from the secondary xylem of Leucaena leucocephala has been purified to homogeneity through successive steps of ammonium sulfate fractionation, DEAE cellulose, Sephadex G-75, and Blue Sepharose CL-6B affinity column chromatographies. CAD was purified to 514.2 folds with overall recovery of 13 % and specific activity of 812. 5 nkat/mg. Native and subunit molecular masses of the purified enzyme were found to be ~76 and ~38 kDa, respectively, suggesting it to be a homodimer. The enzyme exhibited highest catalytic efficiency (Kcat/Km 3.75 μM?1 s?1) with cinnamyl aldehyde among all the substrates investigated. The pH and temperature optima of the purified CAD were pH 8.8 and 40 °C, respectively. The enzyme activity was enhanced in the presence of 2.0 mM Mg2+, while Zn2+ at the same concentration exerted an inhibitory effect. The inclusion of 2.0 mM EDTA in the assay system activated the enzyme. The enzyme was inhibited with caffeic acid and ferulic acid in a concentration-dependent manner, while no inhibition was observed with salicylic acid. Peptide mass analysis of the purified CAD by MALDI-TOF showed a significant homology to alcohol dehydrogenases of MDR superfamily.  相似文献

The current study was undertaken to investigate the antibacterial (against molecular characterized E. coli isolated from poultry faeces) potential of biosynthesized zinc oxide nanoparticles (ZnO-NPs) from Passiflora subpeltata Ortega aqueous leaf extract. The biosynthesized nanoparticles were subjected to physico-chemical characterization to study shape, size and purity by UV–Vis spectroscopy, X-Ray diffraction (XRD), Fourier Transform Infrared Spectroscopy (FT-IR), Scanning Electron Microscopy (SEM), Energy Dispersive Spectroscopy (EDS) and Transmission Electron Microscopy (TEM). The molecular identification of isolated E. coli from faeces samples was carried out by using 16–23s rRNA primers. The results of the physico-chemical characterization revealed that the biosynthesized nanoparticles were of 93.7% purity with an average size between 45 and 50 nm. The ZnO-NPs offered significant inhibition against the isolated Gram-negative E. coli with MIC at 62.5 µg mL?1 concentration. The antibacterial potential of ZnO NPs against E. coli has also been investigated by the cell viability test, and further the effects of ZnO NPs on bacterial morphological structures was analysed by SEM and TEM.

A new approach was proposed for the synthesis of 2‐azido‐1,3‐diols from easily available and inexpensive chiral pool synthon (R)‐2,3‐O‐cyclohexylidene‐D ‐glyceraldehyde, through Mitsunobu azidation of 1,2‐diols. Both C(2) and C(1) azides in variable ratios were obtained in alkyl substituted diols with C(2) as the major one.  相似文献
