A short ionic liquids (ILs)‐based monolithic cartridge was prepared and used as the selective extraction sorbent. After the material was evaluated by field emission‐scanning electron microscopy (FE‐SEM), a new approach for the extraction and determination of quercetin and myricetin from Chamaecyparis obtusa (C. obtusa) by using ILs‐based, monolithic cartridge system was developed. Chromatographic analysis was conducted on a C18 column with UV detection at 372 nm, an eluting solution consisting of acetonitrile‐water (25/75,V/V) as the mobile phase, and a flow rate of 0.7 mL·min−1. A good linear relationship was demonstrated when the concentrations of quercetin and myricetin were in the range of 0.5–100.0 µg·mL−1. The recoveries ranged from 101.6% to 104.6% and the inter‐ and intra‐day relative standard deviations (RSD) were less than 5.0%. This method effectively removed the impurities and avoided tedious pretreatment. It provided a fast, economic and effective method for assaying trace drugs from natural plants. 相似文献

Azadirachta indica has long been used in traditional medicine. This study focused on isolation and characterisation of active ingredients in the extract, its fractions (NF-EA, NF-AQ, NF-G) and its effect on the cholesterol absorption activity. The NF-EA fraction was identified by marker compounds by LC-ESI-QTOF/MS. Cholesterol absorption activity was performed by measuring the solubility and size of cholesterol micelles. The intestinal motility was also examined by isolated rat’s ileum to test the contraction. The extract and its fractions consist of flavonoids and phenolic compounds, like quercetin, kaempferol and myricetin. We found that A. indica extract and NF-EA increase cholesterol micelles size, while the extract, NF-AQ, myricetin and quercetin, reduced the solubility of cholesterol in micelles. The extract and quercetin inhibited the contraction induced by KCl up to 29 and 18%, respectively, and also decreased CaCl2-induced contraction. This finding is in support to traditional uses of A. indica as cholesterol-lowering agents and regulator of gastrointestinal motility. 相似文献

Abstract In vitro acetylholinesterase (AChE) inhibitory activity of an autochthonous sample of the mushroom Coprinus comatus (encompassing fruiting body FB, mycelia M and filtrate F from the submerged cultivation) was the subject of this study. C. comatus F extract exhibited rather potent anti-AChE activity (73.0?±?1.5%) at in liquid conditions, comparable to those of the conventional drug donepezil (80.6?±?1.4%). Also, the same extract exhibited high anti-AChE activity (1?µg) in solid. While its FTIR spectrum indicated the presence of phenolic compounds, quercetin (28.1?µg g?1 d.w.) was found to affect the observed bioactivity (59.8?±?0.9%). This is the first report of profound anti-AChE activity of any C. comatus extract, a medicinal mushroom that has been successfully cultivated in P.R. Abstract In this present study, different forms of quercetin encapsulated beads were synthesized, namely ionic cross-linked gel beads and cryogel beads. Fourier Transform Infrared (FT-IR) spectra of the beads were used to characterize and prove quercetin encapsulation in alginate beads. Swelling and drying profiles were studied. Besides, release kinetics of quercetin molecules from gel beads and cryogels were carefully investigated in two different solvent/media; dimethyl sulfoxide (DMSO) and Roswell Park Memorial Institute Medium (RPMI-1640). Based upon the release kinetic studies, it is found that quercetin release from alginate cryogel beads fits the first-order release model in DMSO and it depends on the concentration of quercetin in the beads. The release of quercetin from alginate gel beads was described by the Higuchi release model, which highlights the release of quercetin molecules through the pores of the matrix. In RPMI-1640, the release of quercetin from both forms of alginate beads fits zero-order release model and it indicates a constant release of quercetin per unit time. Finally, the radical scavenging activity of the quercetin quantities was tested by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) test, and successful results were obtained compared to reference material. 相似文献

采用柱层析方法对迷迭香抗氧化剂(Rao)进行分离,分离出8个含量较高的单体.用光度法、ESR法、滴定法测定了迷迭香的8个单体、单体组合及Rao的抗氧化活性,并分析了抗氧化活性与其结构的关系.结果表明,在单体中,对超氧阴离子自由基及羟基自由基清除率最高的是槲皮素,分别为82.6%和87.1%.对香烟烟气自由基平均清除率最高的是迷迭香二酚,清除率为48.17%.单体活性最强的槲皮素和迷迭香二酚组合后,其抗氧化活性高于单体.活性强的抗氧化剂对油脂过氧化的抑制效果好,但作用时间较短. 相似文献
