In this paper,we developed and validated a simple,sensitive,and selective high-performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS) method to identify and measure the following prohibited substances that may be found in cosmetic products:minoxidil,hydrocortisone, spironolactone,estrone,canrenone,triamcinolone acetonide and progesterone.Chromatographic separation was performed on a Waters Symmetry C18(100 mm×2.1 mm,3.5μm particle size) with a gradient elution system composed of 0.2%(v/v) formic acid aqueous solution and methanol containing 0.2%(v/v) formic acid at a flow rate of 0.3 mL/min.The substances were detected using a triple quadrupole mass spectrometer in the multiple reaction monitoring mode with an electrospray ionization source.All of the calibration curves showed good linearity(r > 0.999) within the tested concentration ranges.The limit of detection was <25 pg.The relative standard deviations for intraday precision for each of the prohibited substances were <3.5%at two concentration levels(2μg/g,10μg/g). The relative recovery rate for each of the prohibited substances ranged from 91.8%to 111%at three concentration levels(0.1μg/g,2μg/g,10μg/g),including the limit of quantification.In conclusion,we have developed and validated a method that can identify seven prohibited substances in cosmetic products.  相似文献

1,4-Diazabicyclo[2.2.2]octane (DABCO) has been used as a mild and efficient catalyst for the synthesis of various tetrahydrobenzo[b]pyran derivatives via a one-pot, three component condensation of aromatic aldehydes, dimedone, and active methylene compounds. This method provides several advantages: a simple workup procedure, environmental friendliness, neutral conditions, and good yields. In addition, water or 50% aqueous ethanol was chosen as a green solvent.

A simple, mild, and ecofriendly method has been developed for the synthesis of 3-vinylchromones from 4-oxo-4H-1-benzopyran-3-carboxaldehyde (3-formylchromone) by simple Knoevenagel condensation with various active methylene compounds (AMC) in aqueous micellar media in the presence of catalytic amounts of cetyl trimethylammonium bromide (CTAB) and 1,4-diazabicyclo[2.2.2]octane (DABCO). In the case of malonic acid as AMC, the reaction resulted in formation of only Doebner decarboxylated products under the standard reaction condition. It has been also observed that 3-formylchromone derivatives primarily undergo tandem Knoevenagel and Michael reactions in the presence of > 2 equiv. of ethyl acetoacetate to produce benzophenone derivatives, by opening of pyran ring, as the sole product in good yields.  相似文献

A method has been developed for the determination of trace quantities of 2,4-dichloro-phenoxyacetic acid (2,4-D), 2,4,5-trichlorophenoxyacetic acid (2,4,5-T), 2,4-dichlorophenol (2,4-DCP), and 2,4,5-trichlorophenol (2,4,5-TCP) in human and rat urine. The method involves acid hydrolysis of the phenolic conjugates, extraction of the free phenols and acids, ethylation with diazoethane, silica-gel column chromatography clean-up of the derivatized urine extract, and gas chromatographic determination using the electron-capture detector. The average recoveries of 2,4-D, 2,4,5-T, 2,4-DCP, and 2,4,5-TCP from rat urine spiked with known amounts of the herbicides and their phenols were 94%, 98%, 92%, and 90%, respectively. The limits of detection for 2,4-D, 2,4,5-T, DCP, and TCP in rat urine were: 0.05, 0.01, 0.10, and 0.01 ppm, respectively. The method was used to analyze urine of rats given various levels of 2,4-D and 2,4,5-T by gavage. Results showed that levels of exposure of 3.75 mcg/kg for 2,4-D and 5.0 mcg/kg for 2,4,5-T in rats can be detected in urine within 24 hr from exposure. Urine samples from occupationally exposed people were analyzed and found to contain 0.2 to 1.0 ppm 2,4-D and 0.05 to 3.6 ppm 2,4,5-T.  相似文献

Abstract

A three-column ion chromatographic system for the removal of humic substances from natural waters, and subsequent on-line concentration and determination of nitrate and sulfate using non-suppressed ion chromatography is presented. Humic substances are removed using disposable adsorption columns packed with chemically bonded amine silica material. The sample is directly transfered to an ion exchange column where the anions are concentrated ca 10 times. After reversing the flow, the ions are transferred to a third column where they are separated and quantified. The detection limit is less than 1 mg L?1 of nitrate or sulfate in water containing 45mgL?1 of humic acid.  相似文献

ABSTRACT

In this paper, we present the results of an analytical method that has been recently developed, validated and successfully applied in a biomonitoring approach. In the environmental pollutant studies it is desirable that the analytical method can determine multiple classes of compounds from a single, small volume sample. The presented analytical method with a simple sample pre-treatment allows the quantitation of 13 perfluoroalkyl acids (PFAAs), 6 parabens and cotinine (used as nicotine biomarker) from a single, small volume of 100 µL serum sample by liquid chromatography-triple quadrupole mass spectrometer (LC-MS/MS). The limits of quantitation (LOQ) for PFAAs, parabens and cotinine were 0.10–0.50, 0.20–0.80 and 0.10 ng/mL, respectively. Besides sensitivity the method has excellent trueness/accuracy and repeatability. The trueness of the method for the determination of PFAAs ranged from 95% to 106% and the repeatability (as RSD %) from 0.6% to 5.6%. The accuracy and RSD for parabens were 73–120% and 1.3–9.7%, respectively, and 100–106% and 1.3–3.5 % for cotinine. Biomonitoring data reveals the presence of several PFAAs and parabens in serum samples of Finnish population. The total concentrations for PFAAs and parabens were from 2.0 to 33 ng/mL and from <LOQ to 1100 ng/mL, respectively. Nearly all non-smokers had the serum cotinine concentration below 1.0 ng/mL, which can be suggested as the cut point for cotinine concentration to identify smoking.  相似文献

合成了邻菲罗啉衍生物联吡啶[3,2-a:2',3'-c]-7-氮杂-吩嗪(dpapz)及其铜(I)配合物[Cu(dpapz)2]PF6, 利用核磁共振氢谱(1H NMR), 傅里叶变换红外(FTIR)光谱, 高分辨质谱(HR ESI-MS)等对合成的化合物进行了表征.采用紫外-可见吸收光谱,荧光光谱, DNA熔解温度实验和循环伏安方法研究了dpapz和[Cu(dpapz)2]PF6与小牛胸腺DNA(CT DNA)的相互作用. 配体dpapz与小牛胸腺DNA(CT DNA)作用时未观察到吸收峰红移并且减色效应较小(<30%), 且DNA熔解温度也上升较小(ΔTm=7.8 ℃), 说明dpapz以沟槽结合的方式与CT DNA相互作用. 而[Cu(dpapz)2]PF6与CT DNA作用时, 可观测到较小的吸收峰红移(2-3 nm)和较大的减色效应(>50%), 同时DNA熔解温度上升较大(ΔTm=11.1 ℃), 表明[Cu(dpapz)2]PF6以静电相互作用和部分扦插的方式与DNA结合. 溴乙锭(EB)荧光竞争实验和循环伏安实验进一步证实了这一结论. 配体dpapz和[Cu(dpapz)2]PF6与DNA的结合常数分别为2.88×105和5.32×105 mol·L-1. 光照条件下, [Cu(dpapz)2]PF6产生单重态氧的能力与dpapz相当, 但产生超氧负离子自由基的能力要弱于dpapz. 活性氧猝灭实验表明, 超氧负离子自由基、单重态氧和羟基自由基均参与了dpapz和[Cu(dpapz)2]PF6对DNA的光损伤作用. [Cu(dpapz)2]PF6对DNA的亲和性要高于对dpapz的, 使得[Cu(dpapz)2]PF6对质粒DNA的光损伤效率明显强于dpapz.  相似文献

A capillary electrophoresis method using CDs for quality control of esomeprazole (ESO) in terms of enantiomeric purity and related substances in raw material and pellets was developed. ESO is the S‐enantiomer of omeprazole (OMZ). Several parameters were evaluated, including type and concentration of buffer and CD, concentration of additives and electrolyte pH. Resolution between the enantiomers of OMZ obtained for each parameter tested was calculated and the presence of the main related substance such as OMZ sulfone was carefully monitored. The optimized system consisted of 100 mM Tris‐phosphate buffer pH 2.5 with 20 mM 2‐hydroxypropyl‐β‐CD, 1 mM sodium dithionite, temperature at 15°C, voltage at 28 kV, and UV detection at 301 nm. Once optimized, the electrophoretic system was validated according to ICH guidelines. The limits of detection and quantification for R‐OMZ were 0.6 μg/mL (0.06% w/w of ESO) and 2.0 μg/mL (0.2% w/w of ESO), respectively. A mean concentration of R‐OMZ <0.2% limit established by the United States Pharmacopeia (USP) was found in the raw material and six‐pellet samples of ESO. No other impurities were found in the samples under these conditions. Therefore, the developed method was found to be appropriate not only for enantiomeric quality control of ESO but also for the analysis of ESO and the main related substance in raw material and pharmaceutical formulations as well as for stability indicating studies.  相似文献
