We describe an approach to efficiently determine the backbone conformation of solid proteins that utilizes selective and extensive 13C labeling in conjunction with two-dimensional magic-angle-spinning NMR. The selective 13C labeling approach aims to reduce line broadening and other multispin complications encountered in solid-state NMR of uniformly labeled proteins while still enhancing the sensitivity of NMR spectra. It is achieved by using specifically labeled glucose or glycerol as the sole carbon source in the protein expression medium. For amino acids synthesized in the linear part of the biosynthetic pathways, [1-13C]glucose preferentially labels the ends of the side chains, while [2-13C]glycerol labels the Cα of these residues. Amino acids produced from the citric-acid cycle are labeled in a more complex manner. Information on the secondary structure of such a labeled protein was obtained by measuring multiple backbone torsion angles φ simultaneously, using an isotropic–anisotropic 2D correlation technique, the HNCH experiment. Initial experiments for resonance assignment of a selectively 13C labeled protein were performed using 15N–13C 2D correlation spectroscopy. From the time dependence of the 15N–13C dipolar coherence transfer, both intraresidue and interresidue connectivities can be observed, thus yielding partial sequential assignment. We demonstrate the selective 13C labeling and these 2D NMR experiments on a 8.5-kDa model protein, ubiquitin. the notion of viscosity solutions to the eikonal equation, and is not restricted to simply-connected cross-sections. Applications

提出了利用调液柱型阻尼器(Tuned Liquid Column Dampers,简称TLCD)和环形调液阻尼器(Circular Tuned Liquid Column Dampers,简称CTLCD)来控制偏心结构在多维地震作用下扭转耦联振动的方法。采用遗传算法,在双向地震作用下,对调液阻尼器的相关参数进行优化。选取了两种目标函数,一种只考虑最大的楼层反应,另一种考虑结构所有自由度的反应。用一个12层的偏心结构作为算例,进行优化计算,结果表明,采用第二种目标函数得到的阻尼器的参数,能有效降低结构的平动反应和扭转反应。 相似文献

We present results of ab initio and DFT calculations of the structure, potential functions of the methyl group internal rotation and the amino group inversion,
and vibrational frequencies and intensities in IR and Raman spectra of methylcarbamate. The calculations were carried out
using different basis sets in the HF, MP2, and DFT/B3LYP approximations. The influence of both the basis set size and the
allowance for electronic correlation on peculiarities of the structure of the amino group in methylcarbamate has been analyzed.
It is shown that the B3LYP/6-311++G(2d, p) and B3LYP/cc-pVDZ calculations reproduce highly accurately experimental geometric
parameters of methylcarbamate. Parameters of torsional and inversion potentials and characteristics of vibrational spectra
calculated in different approximations show satisfactory agreement with experimental values.

在研制基于微型光机电系统技术的光电器件过程中,需要精确测定器件在给定驱动条件下的运动情况。在对制作完成的光开关进行性能测试时,由于微型光机电系统光开关的几何尺寸的限制,如果要准确测定光开关微反射镜的运动位移。将会有一定的困难。提出了一种采用线阵CCD技术的光学测量方法,实现了对光开关悬臂梁在驱动电压作用下的旋转角度的精确测量。通过理论推导得到了悬臂梁扭转角度与可测量物理量的关系.通过求解方程得到了各个物理量的数值选择对于测量结果的影响,并且给定了测量时各个物理量的参考值。通过实验测量,误差小于10%,表明这是一种具有实际意义的测量方案。 相似文献
