A promising liquid chromatography-electrospray ionization-mass spectrometry (LC-ESI-MS) for analysis of the sulfates of 5alpha-androgen, androsterone and epiandrosterone (A-S and EpiA-S) in human serum was developed. The method was used to assess one of the markers of 5alpha-reductase activity of males including patients with prostate cancer (PC). After deproteinization with acetonitrile, the androgen sulfates and the internal standard, [7,7,16,16-2H4]dehydroepiandrosterone-S, were extracted from human serum using a solid-phase extraction cartridge and washed with hexane. The extract was reconstituted and applied to the LC/ESI-MS system operated in the selected ion monitoring mode. The method was validated over the range 0.02-5 microg/mL (A-S) and 0.005-1.5 microg/mL (EpiA-S) using 10 microL of human serum. The method was a concise procedure without chemical or enzymatic hydrolysis of the conjugates, purification by high-performance liquid chromatography and/or derivatization, and proved to be satisfactory in its reproducibility and accuracy. The levels of these androgen sulfates tended to decrease during aging, and the A-S levels in the sera obtained from both healthy males and patients with PC were correlated with their EpiA-S levels.  相似文献

采用人工方法制成人牙釉质浅龋及使用抗龋措施的龋损釉质样本,分别测量各组釉质表面显微硬度,并以微动摩擦磨损试验机测试其抗磨性能,试验结果表明:各组实验组釉质较对照组表面硬度都有不同程度下降,氟化钠前处理会使釉质表达硬度显著下降,人工浅龋牙釉质的抗磨损性能不如正常釉质,使用两种抗龋措施对其抗磨性能没有明显改善。  相似文献

A new optical sensor phase for potassium ions has been developed based on the immobilization of the pH-dependent fluorogenic crown ether 4-acryloylamidobenzo-18-crown-6 on the non-ionic polymeric resin Amberlite XAD-2.Two different optical designs, a flow-through sensor and a fibre optic probetype sensor (optrode), have been constructed and their analytical performance characteristics have been evaluated. The resulting fluorimetric sensors for K+ ions exhibited detection limits of 0.4 or 0.8 M of K+ (16 g/l or 31 g/l), depending on the design, while the linear response occurred from 1 to 25 M of the metal concentrations. The precision, evaluated as the relative standard deviation of measurements of K+ levels at around ten times the detection limit (e.g. 5 M), turned out to be around ±2%.Advantageous features of this fluorimetric sensing phase and optrode include ease of construction, simplicity of use, reversibility, short response times (ca. 1 min full scale deflection) selectivity and operational stability, suitable for sensing potassium at low levels in complex matrices such as biological fluids.The fluorimetric optical sensor has been successfully applied to the direct determination of potassium in clinically important samples (serum and urine) and in natural waters. Very good accuracy has been obtained just using adequate synthetic aqueous potassium standards for calibration.  相似文献

报道了35例食道癌患者血清铜/锌比值及血清硒水平,结果发现食道癌患者血清铜/锌比值增加,血清硒水平下降,具有显著性差异(P〈0.001);同时提示随着食道癌分期增加,铜/锌比值进一步增加,血硒水平进一步下降,但无统计学差异。上述结果对临床诊断及观察病情变化有一定意义。  相似文献

As a novel feeding strategy for aptomizing human epidermal growth factor (hEGF) production with a recombinant Hansenula polymorpha DL-1 using the methanol oxidase (MOX) promoter in H. polymorpha DL-1, independent exponential feeding of two substrates was used. A simple kinetic model considering the cell growth on two substrates was established and used to calculate the respective feeding rates of glycerol and methanol. In the fedbatch culture with methanol-only feeding, the optimal set point of specific growth rate on methanol was found to be 0.10 h−1. When the fed-batch cultures were conducted by the independent feeding of glycerol and methanol, the actual specific growth rate on glycerol and methanol was slightly lower than the set point of specific growth rate. By the uncoupled feeding of glycerol and methanol the volumetric productivity of hEGF increased from 6.4 to 8.0 mg/(L·h), compared with methanol-only feeding.  相似文献

The objective of this study was to investigate the influence of cyclodextrins on the cutaneous availability of the sunscreen oxybenzone. The interaction between oxybenzone and hydrophilic α-, β- and γ-cyclodextrin derivatives was studied in water by phase-solubility analysis. Among the available cyclodextrins, hydroxypropyl-β-cyclodextrin (HP-β-CD) and especially sulfobutylether-β-cyclodextrin (SBE-β-CD) had the greatest solubilizing activity. Ethanol–water solutions containing oxybenzone free or complexed with HP-β-CD or SBE-β-CD were applied to human skin in Franz diffusion cells and the amount of sunscreen permeated into the different cutaneous compartments was determined by HPLC. As much as 20.5% of the oxybenzone applied dose diffused within the skin tissue after 6 h application. Between 39.4% and 54.9% of the penetrated UV filter was localized in the stratum corneum, with no significant difference between uncomplexed oxybenzone or its complex with HP-β-CD. Conversely, the amount retained in the stratum corneum was markedly decreased (ca. 50%) by complexation with SBE-β-CD. Considerable quantities of oxybenzone accumulated into the viable epidermis (5.7% of the applied dose) and dermis (6.2% of the applied dose) from the preparation containing the free UV filter. The sunscreen penetration to the deeper living layers of the skin was remarkably lower (1.0% and 2.0% of applied dose for epidermis and dermis, respectively) upon application of the sunscreen complexed with SBE-β-CD, whereas HP-β-CD had no effect. In addition, photostability experiments demonstrated that SBE-β-CD complexation did not alter the sunscreen photochemical properties.  相似文献

建立了同时检测人体血浆中芹菜素等5种黄酮类化合物含量的反相高效液相色谱法。血浆样品经固相萃取柱提取后,用Hypersil C18色谱柱分离样品,流动相为甲醇/磷酸水溶液(V/V,2/3,pH2.25),等度洗脱,流量为1 mL/min,柱温为40℃,用电化学检测器在直接模式下,1100 mV分析检测待测物,内标法定量。对于全部待测组分r均大于0.99;槲皮素、毛地黄黄酮、山奈酚、芹菜素和异鼠李黄素线性范围分别为3~7000、3~5900、3~7000、5~7000和3~7400 nmol/L;检出限为1.4~4.8 nmol/L。方法的实际样品加标回收率为86.8%~102.9%;相对标准偏差(RSD)低于7.4%(n=3)。方法简便、快速、准确,可用于芹菜素等黄酮类化合物在人体内的生物利用度及膳食干预研究。  相似文献

A sensitive and specific method for the determination of memantine in human plasma is presented. Memantine was extracted from plasma and derivatized to the pentafluorobenzoyl derivative in a one-step procedure avoiding any sample concentration steps. Amantadine was used as an internal standard. The compounds were measured by gas chromatography/negative ion chemical ionization mass spectrometry without any further processing. Using this detection mode, the fragment ions at m/z 353 and 325 were obtained at high relative abundance. Calibration graphs were linear over the range 0.117-30 ng ml(-1). At the limit of quantification (LOQ), the inter-assay precision was 2.00% and the intra-assay variability was 3.22%. The accuracy at the LOQ showed deviations of -1.42% (intra-assay) and -2.47% (inter-assay). The method is rugged, rapid and robust and was applied to the batch determination of memantine during pharmacokinetic profiling of the drug.  相似文献

A sensitive and specific procedure for simultaneous quantitation of chlorpheniramine and pseudoephedrine in human plasma has been developed and validated. Analytes were extracted from plasma samples by liquid-liquid extraction, separated on a Diamonsil C18 column (250 x 4.6 mm i.d.) and detected by tandem mass spectrometry with an atmospheric pressure chemical ionization interface. Diphenhydramine was used as the internal standard. The method has a lower limit of quantitation of 0.2 and 2.0 ng/mL for chlorpheniramine and pseudoephedrine, respectively. The intra- and inter-day relative standard deviation, calculated from quality control (QC) samples were below 4.3% for chlorpheniramine and below 9.5% for pseudoephedrine. The inter-day relative error as determined from QC samples was within 4.7% for each analyte. The overall extraction recoveries of chlorpheniramine and pseudoephedrine were 77 and 61% on average, respectively. The method was successfully applied to pharmaockinetic study of chlorpheniramine and pseudoephedrine in volunteers receiving formulations containing 4 mg of chlorpheniramine maleate and 60 mg of pseudoephedrine hydrochloride.  相似文献

Interfacial layers have been widely applied to study the formation and stability of emulsion-based systems. However, the application of isolated interfaces to address digestibility of emulsions is often limited because of the complexity of experimental methods and results. This review summarizes the latest developments in analytical methods and literature data on effects of digestion on interfacial layers. Particular emphasis is given to understand the changes on interfacial magnitudes during oral, gastric, and duodenal digestion, either applied separately or sequentially. Limitations of interfacial aspects and key factors that influence emulsion microstructure in bulk and lipid digestion are identified. Understanding the behavior of interfacial layers upon gastrointestinal digestion promotes an accurate tracking of the physiological fate of emulsions.  相似文献
