Mono(imidazolyl)-substituted Co(II) porphyrin dimer with a ‘picket fence’ structure was synthesized as a new artificial hemoglobin model containing two binding sites. The dimer was confirmed by UV-vis, resonance Raman and ESR spectral measurements to bind two dioxygen molecules reversibly. The dioxygen binding affinity of the dimer was lower than that of the corresponding monomer. The decrease in this affinity is discussed in terms of steric hindrance and orientational effect of the axial ligand.  相似文献

Submicron microspheres were used directly without ligand coupling for the batch and continuous separations of proteins. In the batch experiments for separating BSA (bovine serum albumin) from BHb (bovine hemoglobin), introducing both hydrophobic effects for BSA and electrostatic repulsion for BHb (and vice versa) was required for high selectivity, and microspheres with low number density of surface groups were advantageous. For the continuous experiments, the utilization of a stirred cell was successful, where the microspheres were in the form of latex with good dispersion of particles. The flow rate without a pump was 0.5–1.3 ml min−1, and the ratio of BSA and BHb was varied. In the experiments for eliminating BHb from BSA, elution curves of BHb corresponded to the single component breakthrough curves, while those for BSA did not. The latter is believed to be due to the interference by BHb in the adsorption of BSA.  相似文献

A novel electrochemical method as a sensitive and convenient technique for the determination of heme proteins based on their interaction with ZnO nanorods was developed. A ZnO nanorod modified glassy carbon electrode (ZnO/GCE) was prepared and the electrochemical behaviors of heme proteins, such as hemoglobin (HB) and cytochrome c (Cyt-c), on this modified electrode have been studied. The results showed that both HB and Cyt-c could be oxidized on the modified electrode and the oxidation currents were linear to the concentrations of the analytes in aqueous solutions. In addition, the results of flow injection analysis (FIA) further suggested the high stability and reproducibility of the ZnO nanorod modified electrode. So this method can be applied to the determination of HB and Cyt-c in biological systems.  相似文献

The redox reaction of poly(ethylene oxide) (PEO)-modified hemoglobin (PEO–Hb) was analyzed in PEO oligomers with cyclic voltammetry. The PEO–Hb was made soluble in PEO with molecular weight of 200 (PEO200) containing 0.5 M KCI. Quasi-reversible redox signals of PEO–Hb were obtained by using an indium tin oxide (ITO) glass working electrode. PEO–Hb, cast on the ITO electrode, also showed the redox response in PEO with molecular weight of 400 (PEO400). The peak current of PEO–Hb on the ITO electrode gradually increased during potential cycling. The effect of the scan rate on the quantity of electricity (Q) was analyzed after the peak current reached a constant value. The constant Q value was observed at the scan rate ranging from 30 to 500 mV/sec. The number of reactive PEO–Hb molecules was estimated from this constant Q-value. It was suggested that the electron transfer was carried out at the first layer of the PEO–Hb which was in direct contact with the ITO electrode. The quantity of electricity of PEO–Hb increased when the ITO electrode was first washed in an aqueous medium with ultrasonicator. This strongly suggested that the more effective surface area of the ITO electrode turned to be covered with PEO–Hb when the microporous region of the ITO particles was more hydrated.  相似文献

We have studied the fluorescent properties of flavonols (quercetin, fisetin, morin, rutin) with the aim of studying possible interaction with hemoglobin and bovine serum albumin (BSA). We observed an increase in the intensity of intrinsic fluorescence for all the flavonols except rutin in the presence of BSA. From the changes in the fluorescence spectra, we concluded that tautomeric forms are formed on interaction with hemoglobin.
