制备了可用作氨基酸及小分子聚合物的MALDI-FTMS分析基质的β-环糊精包覆多壁碳纳米管.通过引入β-环糊精改善其碳纳米管亲水性.所制备的β-环糊精包覆多壁碳纳米管的扫描电镜图表明,该碳纳米管呈现纳米带状结构.与通常的氧化碳纳米管相比,所制备的β-环糊精改性CNT具有更低的背景信号.进一步采用氨基酸及聚乙二醇等小分子化合物对所制备的碳纳米管进行评价,得到很强的氨基酸及聚乙二醇的碱金属离子加合峰,表明该材料可使小分子化合物解吸离子化,且背景干扰小不会影响到小分子化合物检测.由此可见,制备的β-环糊精包覆多壁碳纳米管适于小分子化合物的MALDI-MS分析.  相似文献

The present article deals with the synthesis and characterization of some sulfonated copolyethersulfones. The synthetic approach differs from the post sulfonation approach traditionally reported in the literature. The synthetic procedure is based on the use of sulfonated monomers which are then reacted with previously synthesized telechelic hydoxy‐ended poly (ether sulpnone)s. Combining the MALDI‐TOF MS and 1H NMR analyses, with SEC‐Viscometry and TGA measurements, we demonstrate a powerful tool for characterizing the chemical composition, end chains, degree of sulfonation (DS) and molecular mass distribution (MMD) of disulfonated poly(arylene ether‐sulfone) copolymers. The characterization techniques allowed to determine the exact nature of the copolymers synthesized and to reveal some interesting features about the reaction. DMA data show that the glass transition temperature of sulfonated copolymers with similar DS increase as raise their MMD. Biobased, unsaturated polyesters derived from isosorbide, maleic anhydride, and succinic acid were synthesized and characterized. The presence of maleic anhydride units in the structure of the polyesters allowed converting them into cured coatings by radical copolymerization with crosslinking agents such as 2‐hydroxyethyl methacrylate, N‐vinyl‐2‐pyrrolidinone, acrylic acid or methacrylamide. The investigated polyesters were obtained via bulk polycondensation, catalyzed by titanium(IV) n‐butoxide. 2D NMR and MALDI‐Tof‐MS spectroscopy proved that this polymerization resulted in isomerization of maleic acid units into fumaric ones and in the formation of slightly branched structures by the reaction of isosorbide (end) groups with main chain unsaturated bonds. Moreover, some double bonds proved to have reacted with the condensation by‐product water. The resulting polyesters displayed the expected correlation between variables such as molecular weight and content of unsaturated bonds and their Tg values. Since the thermal properties of the obtained polyesters were appropriate for coating applications, the polymers were crosslinked with unsaturated monomers by radical copolymerization. The crosslinking process was studied using FTIR spectroscopy and by measurements of the soluble part of the cured coatings. © 2010 Wiley Periodicals, Inc. End‐functionalized poly(phenylacetylene)s were synthesized by the polymerization of phenylacetylene (PA) using the well‐defined palladium catalysts represented as [(dppf)PdBr(R)] {dppf = 1,1′‐bis(diphenylphosphino)ferrocene}. The Pd catalysts having a series of R groups such as o‐tolyl, mesityl, C(Ph)?CPh2, C6H4o‐CH2OH, C6H4p‐CN, and C6H4p‐NO2 in conjunction with silver triflate polymerized PA to give end‐functionalized poly(PA)s bearing the corresponding R groups in high yields. The results of IR and NMR spectroscopies and MALDI‐TOF mass analyses proved the introduction of these R groups at one end of each polymer chain. The poly(PA) bearing a hydroxy end group was applied as a macroinitiator to the synthesis of a block copolymer composed of poly(PA) and poly(β‐propiolactone) moieties. © 2010 Wiley Periodicals, Inc. A Cyclic aryl thioester dimer was prepared by the reaction of o-phthaloyl dichloride and bis(4-mercaptophenyl)sulfide in good yield under pseudo-high dilution conditions via interfacial polycondensation.The structure of the cyclic dimer was confirmed by a conmbination of MALDI-TOF-Ms,FTIR,gel permeation chromatography and NMR analyses.The X-ray diffraction study of the single crystal of cyclic thioester dimer obtained form two sotutions reveals no severe internal strain on the cyclic structure.  相似文献

Bisphenol A was polycondensed with diphosgene in a dichloromethane/aqueous NaOH system. Temperature, time, and molar ratios of the reactants were optimized according to a previously elaborated optimization of the hydrolytic polycondensation of bisphenol A bischloroformate. Five of the following catalysts were examined: triethylamine, 4‐(N,N‐dimethylamino)pyridine (DMAP), ethyldiisopropylamine (EDPA), tetrabutylammonium hydrogen sulfate, and triethylbenzylammonium chloride (TEBA‐Cl). Triethylamine and DMAP accelerated the hydrolysis of diphosgene by formation of a hydrophilic acylammonium salt. Therefore, the molecular weights decreased with higher concentration of these tert‐amines. However, the molecular weights increased (weight‐average molecular weight up to 106) with higher concentrations of tetraalkylammonium salts because these catalysts favor chain growth in the organic phase via “naked” phenoxide ions without catalyzing the hydrolysis of diphosgene. EDPA gave poor results under all circumstances. Cyclic polycarbonates were discovered in all samples. Their fraction increased with the average molecular weight of the samples. When samples prepared with triethylamine or TEBA‐Cl were fractionated, cycles having molar masses up to 15,000 Da were detected by matrix‐assisted laser desorption/ionization time‐of‐flight mass spectroscopy. © 2003 Wiley Periodicals, Inc. We report an MS‐based workflow for identification of phosphorylated peptides from trypsinized protein mixtures and cell lysates that is suitable for high‐throughput sample analysis. The workflow is based on an in situ enrichment on matrix‐assisted laser desorption/ionization (MALDI) plates that were functionalized by TiO2 using automated ion landing apparatus that can operate unsupervised. The MALDI plate can be functionalized by TiO2 into any array of predefined geometry (here, 96 positions for samples and 24 for mass calibration standards) made compatible with a standard MALDI spotter and coupled with high‐performance liquid chromatography. The in situ MALDI plate enrichment was compared with a standard precolumn‐based separation and achieved comparable or better results than the standard method. The performance of this new workflow was demonstrated on a model mixture of proteins as well as on Jurkat cells lysates. The method showed improved signal‐to‐noise ratio in a single MS spectrum, which resulted in better identification by MS/MS and a subsequent database search. Using the workflow, we also found specific phosphorylations in Jurkat cells that were nonspecifically activated by phorbol 12‐myristate 13‐acetate. These phosphorylations concerned the mitogen‐activated protein kinase/extracellular signal‐regulated kinase signaling pathway and its targets and were in agreement with the current knowledge of this signaling cascade. Control sample of non‐activated cells was devoid of these phosphorylations. Overall, the presented analytical workflow is able to detect dynamic phosphorylation events in minimally processed mammalian cells while using only a short high‐performance liquid chromatography gradient. This review is devoted to the application of MS using soft ionization methods with a special emphasis on electrospray ionization, atmospheric pressure photoionization and matrix‐assisted laser desorption/ionization MS and tandem MS (MS/MS) for the elucidation of the chemical structure of native and modified lignins. We describe and critically evaluate how these soft ionization methods have contributed to the present‐day knowledge of the structure of lignins. Herein, we will introduce new nomenclature concerning the chemical state of lignins, namely, virgin released lignins (VRLs) and processed modified lignins (PML). VRLs are obtained by liberation of lignins through degradation of vegetable matter by either chemical hydrolysis and/or enzymatic hydrolysis. PMLs are produced by subjecting the VRL to a series of further chemical transformations and purifications that are likely to alter their original chemical structures. We are proposing that native lignin polymers, present in the lignocellulosic biomass, are not made of macromolecules linked to cellulose fibres as has been frequently reported. Instead, we propose that the lignins are composed of vast series of linear related oligomers, having different lengths that are covalently linked in a criss‐cross pattern to cellulose and hemicellulose fibres forming the network of vegetal matter. Consequently, structural elucidation of VRLs, which presumably have not been purified and processed by any other type of additional chemical treatment and purification, may reflect the structure of the native lignin. In this review, we present an introduction to a MS/MS top–down concept of lignin sequencing and how this technique may be used to address the challenge of characterizing the structure of VRLs. Finally, we offer the case that although lignins have been reported to have very high or high molecular weights, they might not exist on the basis that such polymers have never been identified by the mild ionizing techniques used in modern MS. In matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry (MALDI TOF MS), analyte signals can be substantially suppressed by other compounds in the sample. In this technical note, we describe a modified thin‐layer sample preparation method that significantly reduces the analyte suppression effect (ASE). In our method, analytes are deposited on top of the surface of matrix preloaded on the MALDI plate. To prevent embedding of analyte into the matrix crystals, the sample solution were prepared without matrix and efforts were taken not to re‐dissolve the preloaded matrix. The results with model mixtures of peptides, synthetic polymers and lipids show that detection of analyte ions, which were completely suppressed using the conventional dried‐droplet method, could be effectively recovered by using our method. Our findings suggest that the incorporation of analytes in the matrix crystals has an important contributory effect on ASE. By reducing ASE, our method should be useful for the direct MALDI MS analysis of multicomponent mixtures. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献

Matrix assisted laser desorption ionization (MALDI) is a technique widely employed in the analysis of proteins and peptides, and nowadays it has also been applied to small molecules. There is little significant information regarding the in‐source dissociation processes on MALDI for natural products. Twenty‐six flavonoids (flavanones, flavones and flavonols) were analyzed by MALDI using different methods (with different matrices) and without matrix to comprehend the in‐source reactions and establish good analysis methods for these compounds. Depending on the class, structure and the laser intensity applied, methoxylated flavonoid aglycones can eliminate methyl radicals (˙CH3) in the source, such as flavonols, but lithium 2,4‐dihydroxybenzoate matrix suppresses the ˙CH3 eliminations and retro‐Diels–Alder cleavages in the source. All of the flavonoid O‐glycosides evaluated herein eliminated the sugar in source, even in the presence of the matrix, and its product radical ions ([M‐H‐sugar]?˙) were observed in the negative mode. The flavone C‐glycosides suffered intense dissociation, which was reduced by the addition of a matrix and the application of low laser intensity, mainly in the negative mode. Depending on the hydroxyl substituents, the [M‐H‐H]?˙ ion was observed with variable relative intensity in the spectra. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献
