A novel, sensitive electrochemical DNA hybridization detection assay, using silver nanoparticles as the oligonucleotide labeling tag, is described. The assay relies on the hybridization of the target DNA with the silver nanoparticle-oligonucleotide DNA probe, followed by the release of the silver metal atoms anchored on the hybrids by oxidative metal dissolution and the indirect determination of the solubilized Ag(I) ions by anodic stripping voltammetry (ASV) at a carbon fiber ultramicroelectrode. The influence of the relevant experimental variables, including the surface coverage of the target oligonucleotide, the duration of the silver dissolution steps and the parameters of the electrochemical stripping measurement of the silver(I) ions, is examined and optimized. The combination of the remarkable sensitivity of the stripping metal analysis at the microelectrode with the large number of silver(I) ions released from each DNA hybrid allows detection at levels as low as 0.5 pmol L(-1) of the target oligonucleotides. 相似文献

Cell status changes are typically accompanied by the simultaneous changes of multiple microRNA (miRNA) levels. Thus, simultaneous and ultrasensitive detection of multiple miRNA biomarkers shows great promise in early cancer diagnosis. Herein, a facile single-molecule fluorescence imaging assay was proposed for the simultaneous and ultrasensitive detection of multiple miRNAs using only one capture anti-DNA/RNA antibody (S9.6 antibody). Two complementary DNAs (cDNAs) designed to hybridize with miRNA-21 and miRNA-122 were labelled with Cy3 (cDNA1) and Cy5 (cDNA2) dyes at their 5′-ends, respectively. After hybridization, both miRNA-21/cDNA1 and miRNA-122/cDNA2 complexes were captured by S9.6 antibodies pre-modified on a coverslip surface. Subsequently, the Cy3 and Cy5 dyes on the coverslip surface were imaged by the single-molecule fluorescence setup. The amount of miRNA-21 and miRNA-122 was quantified by counting the image spots from the Cy3 and Cy5 dye molecules in the green and red channels, respectively. The proposed assay displayed high specificity and sensitivity for singlet miRNA detection both with a detection limit of 5 fM and for multiple miRNA detection both with a detection limit of 20 fM. Moreover, it was also demonstrated that the assay could be used to detect multiple miRNAs simultaneously in human hepatocellular cancer cells (HepG2 cells). Based on self-assembled biotinylated disulfide derivative monolayer on gold electrode, the sensors immobilized monolayer or multilayer membranes composed of avidin and biotinlabeled glucose oxidase (B.GOD) or of avidin-B.GOD complex (ABC) and B.COD were prepared. The present technique may be useful for controlling the enzyme content of the sensors in molecular level by repeating the deposition of enzyme layers. The sensors have the characteristics of shorter response time, higher sensitivity. The linear range is from 6.0 x 10(-6) - 5.0 x 10(-3) M. The sensor can be used for more than 1 month and can be reactivated. The sensor was used to determine glucose in human blood serum, and the results are satisfactory. 相似文献

Aza-Morita-Baylis-Hillman reactions of N-(benzylidene)polyfluoroanilines 1 with methyl acrylate or acrylonitrile were studied. It was found that Lewis base, solvent and reaction temperature can significantly affect the reaction. Using 3-hydroxyquinuclidine (3-HQD) as a Lewis base in the reactions of 1 with methyl acrylate in DMF, the normal aza-Morita-Baylis-Hillman adducts 3 were formed in moderate to excellent yields. For the reactions of 1 with acrylonitrile, 1,4-diazabicyclo[2.2.2]octane (DABCO) is the best Lewis base giving the corresponding aza-Morita-Baylis-Hillman adducts 4 as the sole product in good to moderate yield. However, upon treatment of 1 with acrolein 2c, the corresponding reaction did not occur even in the presence of a variety of catalysts. 相似文献

Aliphatic aldehydes such as ethanal,propanal,n-butanal,isobutyraldehyde,n-valeraldehyde,isovaleraldehyde,n-hexanal and n-octanal were converted into the corresponding 2,4,6-trialkyl-1,3,5-trioxanes through cyclotrimer-ization in the presence of the ferric chloride based ionic liquids at room temperature without solvent in high selec-tivity.The effects of different ionic liquids,acidity of ionic liquids and temperature on cyclotrimerization were alsostudied.The results showed that the ferric chloride based ionic liquids(apparent molar fraction of FeCl_3(x(FeCl_3)=0.62))were a kind of efficient catalysts for the cyclotrimerization of aliphatic aldehyde which could be separatedconveniently from the reaction mixture and recycled without loss of catalytic activity.The conversion of isobu-tyraldehyde and the selectivity to 2,4,6-triisopropyl-1,3,5-trioxane were 91.1% and 99.8% respectively under opti-mum reaction condition(isobutyraldehyde 25.0 g,[Et_3NH]Cl/FeCl_3(x(FeCl_3)=0.62)1.0 g,25 ℃for 1 h). 相似文献
