Amphiphilic polyester-co-polyether (PEPE) dendrimers synthesized from poly(ethylene glycol) (PEG) were examined to understand the influence of alterations in the architecture of dendrimers on their conformation at interfaces and distribution of various groups on their surface. Effect of changes in the number of branching points, type of terminal functional groups and generation of dendrimer was primarily evaluated. Dendrimers were deposited on mica by spin coating at 0.1 mg/mL. Tapping mode atomic force microscopy (AFM) was employed for the visualization of dendrimer topographies while, X-ray photoelectron spectroscopy (XPS), AFM phase and force imaging were used as the tools for characterization of their surfaces. Individual dendrimer molecules could be imaged by AFM, which showed that they are round or oval in topography. Dendrimers were also flattened on mica but the extent of flattening differed with the chemical structure; for instance, third generation dendrimers were more flattened than second generation dendrimers whereas, dendrimers with higher number of branches had greater height above the mica surface. Hydrophilic and hydrophobic groups present towards the aerial interface existed in distinct zones rather than being distributed randomly, except in dendrimer with higher number of branches. The percentage of various hydrophobic groups on the surface of dendrimer was enhanced by increase in the number of branches but, was lowered by the presence of hydroxyl groups as the pendant terminal groups. Furthermore, the core of dendrimers was not always located towards the centre, its position was found to be altered by the number of branching points, type of terminal functional groups and the generation of dendrimer.  相似文献

Glycosylphosphatidyl-inositol (GPI)-anchored proteins preferentially localize in the most ordered regions of the cell plasma membrane. Acyl and alkyl chain composition of GPI anchors influence the association with the ordered domains. This suggests that, conversely, changes in the fluid and in the ordered domains lipid composition affect the interaction of GPI-anchored proteins with membrane microdomains. Validity of this hypothesis was examined by investigating the spontaneous insertion of the GPI-anchored intestinal alkaline phophatase (BIAP) into the solid (gel) phase domains of preformed supported membranes made of dioleoylphosphatidylcholine/dipalmitoylphosphatidylcholine (DOPC/DPPC), DOPC/sphingomyelin (DOPC/SM), and palmitoyloleoylphosphatidylcholine/SM (POPC/SM). Atomic force microscopy (AFM) showed that BIAP inserted in the gel phases of the three mixtures. However, changes in the lipid composition of membranes had a marked effect on the protein containing bilayer topography. Moreover, BIAP insertion was associated with a net transfer of phospholipids from the fluid to the gel (DOPC/DPPC) or from the gel to the fluid (POPC/SM) phases. For DOPC/SM bilayers, transfer of lipids was dependent on the homogeneity of the gel SM phase. The data strongly suggest that BIAP interacts with the most ordered lipid species present in the gel phases of phase-separated membranes. They also suggest that GPI-anchored proteins might contribute to the selection of their own microdomain environment.  相似文献

ABSTRACT

The effect of polymer coating on MNR relaxometry of maghemite nanoparticles has been studied. The samples were carefully sorted by size in order to reach narrow size distribution (<0.2) with size ranging from 4.5 to 12.5?nm. Relaxation dispersion profile as well as studies at a fixed Larmor frequency, were recorded for numerous either uncoated or polymer coated samples. The NMR relaxivities r1 and r2 increase with nanoparticle diameter. We have analysed the role of polydispersity for nanoparticles with the same mean size on the dispersion curves. We have compared the role of coating on nanoparticles NMR relaxivity between bare and poly(sodium acrylate-co-maleate) coated nanoparticles. We have investigated the influence of nanoparticle size on the T1 and T2 activation energy Ea. While Ea decreases with nanoparticle diameter when determined from T1, it increases from T2 determination. The influence is more important for small particles (<9?nm) than for big particles (>9?nm). Moreover, the PAAMA coating changes the energy Ea obtained from T2: Ea becomes independent of the nanoparticle diameter. These results highlight the need of a complete characterisation of the role of the coating on the relaxation of magnetic particles.  相似文献

Micro‐Raman spectroscopy (MRS) was utilized for the first time to evaluate the effect of indole‐3‐carbinol (I3C) on acute alcoholic liver injury in vivo. In situ Raman analysis of tissue sections provided distinct spectra that can be used to distinguish alcoholic liver injury as well as ethanol‐induced liver fibrosis from the normal state. Sixteen mice with liver diseases including acute liver injury and chronic liver fibrosis, and eight mice with normal liver tissues, and eight remedial mice were studied employing the Raman spectroscopic technique in conjunction with biomedical assays. The biochemical changes in mouse liver tissue when liver injury/fibrosis occurs such as the loss of reduced glutathione (GSH), and the increase of collagen (α‐helix protein) were observed by MRS. The intensity ratio of two Raman peaks (I1450/I666) and in combination with statistical analysis of the entire Raman spectrum was found capable of classifying liver tissues with different pathological features. Raman spectroscopy therefore is an important candidate for a nondestructive in vivo screening of the effect of drug treatment on liver disease, which potentially decreases the time‐consuming clinical trials. A process for RAFT-controlled radical polymerization in emulsion [36] has been applied to the polymerizations of isoprene and of butadiene in emulsion systems, with the goal of producing latex particles containing block copolymers of acrylic acid (stabilizer and starting polymer), styrene (second polymer) and isoprene or butadiene (third polymer). The microstructure of the polymer chains was examined using dual-detection size-exclusion chromatography, and the nanostructure of the materials was investigated by differential scanning calorimetry and solid-state nuclear magnetic resonance. Reactions were always slow (although faster than the corresponding processes in solution), and exhibited limited reinitiation by isoprene when in emulsion. The materials containing isoprene exhibit a nanostructure with a phase separation into high-Tg polystyrene-rich domains and low-Tg polyisoprene-rich domains, revealed by DSC and NMR. This has the potential to lead to barrier materials with novel physical properties.  相似文献

A new perturbation scheme based on the Barker-Henderson perturbation theory [J. Chem. Phys. 47, 4714 (1967)] is proposed to predict the thermodynamic properties of spherical molecules. Accurate predictions of second virial coefficients and vapor-liquid coexistence properties are obtained for a large variety of potential functions (square well, Yukawa, Sutherland, Lennard-Jones, Buckingham, Girifalco). New Gibbs ensemble Monte Carlo simulations of the generalized exp-m Buckingham potential are reported. An extension of the perturbation approach to mixtures is proposed, and excellent predictions of vapor-liquid equilibria are obtained for Lennard-Jones mixtures. The perturbation scheme can be applied to complex potential functions fitted to ab initio data to predict the properties of real molecules such as neon. The new approach can also be used as an auxiliary tool in molecular simulation studies, to efficiently optimize an intermolecular potential on macroscopic properties or match force fields based on different potential functions.  相似文献

A complete, critical evaluation of all phase diagram and thermodynamic data was performed for all phases of the (Na2SO4 + K2SO4 + Na2S2O7 + K2S2O7) system and optimized model parameters were obtained. The Modified Quasichemical Model in the Quadruplet Approximation was used for modelling the liquid phase. The model evaluates first- and second-nearest-neighbour short-range ordering, where the cations (Na+ and K+) are assumed to mix on a cationic sublattice, while anions were assumed to mix on an anionic sublattice. The Compound Energy Formalism was used for modelling the solid solutions of (Na,K)2SO4 and (Na,K)2S2O7. The models can be used to predict the thermodynamic properties and phase equilibria in multicomponent heterogeneous systems. The experimental data from the literature were reproduced within experimental error limits.  相似文献

Nonaqueous capillary electrophoresis (NACE) was successfully applied to the enantiomeric purity determination of S-timolol maleate using heptakis(2,3-di-O-methyl-6-O-sulfo)-beta-cyclodextrin (HDMS-beta-CD) as chiral selector. With a background electrolyte made up of a methanolic solution of 0.75 M formic acid, 30 mM potassium camphorsulfonate and containing 30 mM HDMS-beta-CD, the determination of 0.1% of R-timolol in S-timolol could be performed with an enantiomeric resolution of 8.5. Pyridoxine was selected as internal standard. The NACE method was then fully validated by applying a novel strategy using accuracy profiles. It is based on beta-expectation tolerance intervals for the total measurement error which includes trueness and intermediate precision. The uncertainty of measurements derived from beta-expectation tolerance intervals was estimated at each concentration level of the validation standards. To confirm the suitability of the developed and validated method, several real samples of S-timolol maleate containing R-timolol maleate at different concentrations were analysed and the results were compared to those obtained by liquid chromatography.  相似文献
