Glycoforms of glargine expressed in Pichia pastoris were isolated by high‐performance liquid chromatography and analyzed by a series of chemical and mass spectrometric methods for the identification of various glycoforms, glycosylation position, nature and structure of glycans. Reduction and alkylation, peptide mapping techniques were used to decipher the amino acid site at which glycosylation had taken place. Chemical methods were coupled with mass spectrometry techniques such as electrospray ionization and matrix‐assisted laser desorption/ionization for identification of the glycosylation site. This study is concerned with the initiation of localized shear bands in plane-strain tension and compression. A theoretical framework which views the initiation of such shear bands as a bifurcation phenomenon from a homogeneous equilibrium field in an elastic-plastic body is first briefly reviewed, and then the predictions of the theory are compared with some experimental observations on an aged maraging steel. The experiments support the physical relevance of the theory within the framework of continuum mechanics.Specifically, the comparison between theory and experiment is concerned with the critical strains to localization and the orientation of the shear bands relative to the load axis. The theoretical predictions are only in qualitative agreement with the experimental observations. A better agreement is obtained with use of the constitutive equations corresponding to the classical deformation theory (a simple vertex model) than with use of the constitutive equations of the classical flow theory. It is concluded that better constitutive equations for elastic-plastic materials are needed before theoretical predictions can be obtained which might be expected to be in closer quantitative accord with experiment.  相似文献

A series of novel luminescent cyclometalated Au(III) neutral complexes of the type cis-[(N(∧)C)AuL] [N(∧)C = 2-phenylpyridine (ppy), L = 1,1'-biphenyl (1)] and cis-[(N(∧)C)AuL(2)] [N(∧)C = 2-phenylpyridine (ppy), L = C(6)H(5) (2), C(6)F(5) (3), C(6)H(4)-CF(3)-p (4), 2-C(4)H(3)S (5)]; [N(∧)C = 2-(2-thienyl)pyridine (thpy), L = C(6)H(5) (6), C(6)F(5) (7)]; [N(∧)C = 2-(5-methyl-2-thienyl)pyridine (5 m-thpy), L = C(6)F(5) (8)] were successfully synthesized. The X-ray crystal structures of all compounds except 3 have been determined. These complexes were found to show long-lived emission in solution at room temperature. The emission origins of the complexes have been tentatively assigned to be derived from triplet states predominantly bearing intraligand (IL) character with some perturbation from the metal center. Density functional theory (DFT) calculations were performed to evaluate the stability associated with the complexes and TD-DFT calculations to ascertain the nature of the excited state. Variation of the cyclometalated ligands in the complexes readily leads to the tuning of the nature of the lower energy emissive states.  相似文献

Li–Mn–O thin film cathode materials are prepared by high frequency (27.12 MHz) RF magnetron sputtering. The high RF frequency gives higher deposition rates without compromising on the quality of the films. This investigation focuses on the effects of post-annealing on the micro-structural, morphological and electrical properties of Li–Mn–O films. It is observed that with the increase of annealing temperature the crystallinity as well as the electrical conductivity of the films increases. The films annealed at 600–700 °C are found to have high structural perfection and good electrical properties.  相似文献

Miniaturized and tunable optical components, such as the waveguide, lens, and prism, have been of great interest for lab-on-chip systems. This Letter reports an optofluidic aperture stop formed by the liquid-core/liquid-cladding flow. The aperture size can be tuned accordingly by adjusting the flow rates. Manipulation of the aperture size allows control of the amount of light passing through the corresponding optical system as well as the angular aperture on the image side. This optofluidic aperture enables lab-on-chip optical systems to have a greater flexibility and more functionalities.  相似文献

An inexpensive activated carbon (AC) air cathode was developed as an alternative to a platinum-catalyzed electrode for oxygen reduction in a microbial fuel cell (MFC). AC was cold-pressed with a polytetrafluoroethylene (PTFE) binder to form the cathode around a Ni mesh current collector. This cathode construction avoided the need for carbon cloth or a metal catalyst, and produced a cathode with high activity for oxygen reduction at typical MFC current densities. Tests with the AC cathode produced a maximum power density of 1220 mW/m2 (normalized to cathode projected surface area; 36 W/m3 based on liquid volume) compared to 1060 mW/m2 obtained by Pt catalyzed carbon cloth cathode. The Coulombic efficiency ranged from 15% to 55%. These findings show that AC is a cost-effective material for achieving useful rates of oxygen reduction in air cathode MFCs.  相似文献
