Photocaged antibody fragments, termed photobodies, have been developed that are impaired in their antigen‐binding capacity and can be activated by irradiation with UV light (365 nm). This rational design concept builds on the selective photocaging of a single tyrosine in a nanobody (a single‐domain antibody fragment). Tyrosine is a frequently occurring residue in central positions of the paratope region. o‐Nitrobenzyl‐protected tyrosine variants were incorporated into four nanobodies, including examples directed against EGFR and HER2, and photodeprotection restores the native sequence. An anti‐GFP photobody exhibited an at least 10 000‐fold impaired binding affinity before photodeprotection compared with the parent nanobody. A bispecific nanobody–photobody fusion protein was generated to trigger protein heterodimerization by light. Photoactivatable antibodies are expected to become versatile protein reagents and to enable novel approaches in diagnostic and therapeutic applications.  相似文献

This paper proposes the use of multiagent cooperation for solving global optimization problems through the introduction of a new multiagent environment, MANGO. The strength of the environment lays in its flexible structure based on communicating software agents that attempt to solve a problem cooperatively. This structure allows the execution of a wide range of global optimization algorithms described as a set of interacting operations. At one extreme, MANGO welcomes an individual non-cooperating agent, which is basically the traditional way of solving a global optimization problem. At the other extreme, autonomous agents existing in the environment cooperate as they see fit during run time. We explain the development and communication tools provided in the environment as well as examples of agent realizations and cooperation scenarios. We also show how the multiagent structure is more effective than having a single nonlinear optimization algorithm with randomly selected initial points.  相似文献

We employ QCD sum rules to calculate the coupling constant g by studying the three point -correlation function. The problem of embedding of linear spaces in finite projective planes has been examined by several authors ([1], [2], [3], [4], [5], [6]). In particular, it has been proved in [1] that a linear space which is the complement of a projective or affine subplane of order m is embeddable in a unique way in a projective plane of order n. In this article, we give a generalization of this result by embedding linear spaces in a finite projective plane of order n, which are complements of certain regularA-affine linear spaces with respect to a finite projective plane.  相似文献

The general analysis of the rare Bc→Ds *+- decay is presented by using the most general, model independent effective Hamiltonian. The dependencies of the branching ratios and of the longitudinal, normal and transversal polarization asymmetries for ℓ- and the combined asymmetries for ℓ- and ℓ+ on the new Wilson coefficients are investigated. Our analysis shows that the lepton polarization asymmetries are very sensitive to the scalar and tensor type interactions, which will be very useful in looking for new physics beyond the standard model.  相似文献

OBJECTIVE: The aim of this study was to identify the effects of objective laryngopharyngeal reflux (LPR) on the acoustic parameters of patients by comparing their voice samples with that of control subjects. STUDY DESIGN: Prospective study in two tertiary reference hospitals. METHODS: 48 consecutive patients with symptoms related to LPR and 64 control subjects were included in the study. Suspected LPR patients underwent a 24-hour ambulatory pH monitoring, and 25 (52%) of them were shown to have objective LPR. Acoustical evaluation results of objective LPR patients were compared with that of symptomatic LPR patients and control subjects. RESULTS: All frequency perturbation values obtained from objective and symptomatic LPR patients were higher than the control subjects (P<0.01). Mean fundamental frequency, amplitude perturbation measures, and noise-to-harmonics ratio were not significantly different between groups. CONCLUSION: LPR patients have significantly different frequency perturbation values than control subjects.  相似文献

The (2,2′-dipyridylamine)(pyridine-2,6-dicarboxylato)copper(II) trihydrate complex was synthesized and characterized by spectroscopic (IR, UV-vis, EPR), X-ray diffraction technique and electrochemical methods. The copper(II) center is surrounded by one bidentate 2,2′-dipyridylamine (dpa) and one tridentate dipicolinate (dpc) ligand, and exhibits a distorted square-pyramidal geometry. The crystal packing involves both hydrogen-bonding and π-π interactions. The solvent water molecules link monomers to one another through hydrogen-bonding interactions, forming ladder-type chains in the ab plane. π-π interactions also occur between the dpa rings of neighboring molecules and are responsible for interchain packing. Based on EPR and optical absorption studies, spin-Hamiltonian and bonding parameters have been calculated. The g-values, calculated for title complex in polycrystalline state at 298 K and in frozen DMF (110 K), indicate the presence of the unpaired electron in the dx2-y2 orbital. The evaluated metal-ligand bonding parameters showed strong in-plane σ- and π-bonding. The cyclic voltammogram of the title complex investigated in DMF (dimethylformamide) solution exhibits only metal centered electroactivity in the potential range ±1.25 V versus Ag/AgCl reference electrode.  相似文献

A series of alkyl α‐hydroxymethacrylate derivatives with various secondary functionalities (ether, ester, carbonate, and carbamate) and terminal groups (alkyl, cyano, oxetane, cyclic carbonate, phenyl and morpholine) were synthesized to investigate the effect of intermolecular interactions, H‐bonding, π–π interactions, and dipole moment on monomer reactivity. All of the monomers except one ester and one ether derivative are novel. The polymerization rates, determined by using photo‐DSC, showed the average trend (aromatic carbamate > hydroxyl > ester > carbonate ~ aliphatic carbamate ~ ether), with several exceptions due to the differences in terminal groups. There is a correlation between the chemical shift differences of the double bond carbons, the calculated dipole moments, and the reactivities only for nonhydrogen bonded monomers. © 2011 Wiley Periodicals, Inc. We consider one-way classification model in experimental design when the errors have generalized secant hyperbolic distribution. We obtain efficient and robust estimators for block effects by using the modified maximum likelihood estimation (MML) methodology. A test statistic analogous to the normal-theory F statistic is defined to test block effects. We also define a test statistic for testing linear contrasts. It is shown that test statistics based on MML estimators are efficient and robust. The methodology readily extends to unbalanced designs.  相似文献
