Solid-phase synthesis of quinoxaline, thiazine, and oxazine analogs through a benzyne intermediate

A solid-phase synthetic route to quinoxaline, thiazine, and oxazine analogs is described. N-Alloc-3-amino-3-(2,4-difluoro-5-nitrophenyl)propanoic acid was tethered to Rink resin via its carboxylic acid group. The 4-arylfluorine was displaced with a primary amine, alcohol, or thiol to create, respectively, a resin bound aniline, phenol, or thiophenol derivative with one diversity element and one single atom (e.g., N, S, or O) diversity point. A fused heterocyclic system was subsequently created via a benzyne heterocyclization initiated by dehydrofluorination with strong base. Acid treatment released the desired products in high yield and moderate purity.     

Plasmid DNA isolation using amino-silica coated magnetic nanoparticles (ASMNPs)

A simple and efficient approach for the rapid isolation of plasmid DNA from crude cell lysates has been described. The approach took advantage of the amino-modified silica coated magnetic nanoparticles (ASMNPs) with positive zeta potential at neutral pH and superparamagnetism under the external magnetic fields. As a demonstration, the pEGFP-N3 plasmid has been concentrated and isolated from the E. coli DH5α transformed with pEGFP-N3 plasmid through electrostatic binding between the positive charge of the amino group of ASMNPs and the negative charge of the phosphate groups of the plasmid DNA. Then the pEGFP-N3 plasmid has been released easily and quickly from the pEGFP-N3 plasmid-ASMNPs complexes with 3 M NaCl. The entire procedure could be carried out by the aid of external magnetic fields in 15 min and eliminate the need of phenol, cesium chloride gradients or other noxious reagents and complexes operation. Moreover, the pEGFP-N3 plasmid obtained by this approach retains biological activity that can be suitable for restriction enzyme digestion and cells transfection with expression of green fluorescence protein.     

Molecular basis for antifreeze activity difference of two insect antifreeze protein isoforms

The insect spruce budworm(Choristoneura fumiferana) produces antifreeze protein(AFP) to assist in the protection of the over-wintering larval stage and contains multiple isoforms. Structures for two isoforms,known as CfAFP-501 and CfAFP-337,show that both possess similar left-handed β-helical structure,although thermal hysteresis activity of the longer isoform CfAFP-501 is three times that of CfAFP-337. The markedly enhanced activity of CfAFP-501 is not proportional to,and cannot be simply accounted for,by the increased ice-binding site resulting from the two extra coils in CfAFP-501. In or-der to investigate the molecular basis for the activity difference and gain better understanding of AFPs in general,we have employed several different computational methods to systematically study the structural properties and ice interactions of the AFPs and their deletion models. In the context of intact AFPs,a majority of the coils in CfAFP-501 has better ice interaction and causes stronger ice lattice disruption than CfAFP-337,strongly suggesting a cooperative or synergistic effect among β-helical coils. The synergistic effect would play a critical role and make significant contributions to the anti-freeze activity β-helical antifreeze proteins. This is the first time that synergistic effect and its implica-tion for antifreeze activity are reported for β-helical antifreeze proteins.     

Analysis of selected pesticides and alkylphenols in human cord blood by gas chromatograph-mass spectrometer

A total of seven pesticides and eight alkylphenols were monitored using this method for the determination of their trace levels in human cord blood. The pesticides are lindane, diazinon, α-endosulfan, β-endosulfan, endosulfan sulfate, chlorpyrifos and endrin; while the alkylphenols are 4-n-butylphenol, 4-n-pentylphenol, 4-n-hexylphenol, 4-t-octylphenol, 4-n-heptylphenol, nonylphenol, 4-n-octylphenol and bisphenol A. The pesticides and alkylphenols in the cord blood samples were extracted with solid phase extraction IST C18 cartridges and analyzed by selected ion monitoring mode using quadrapole detector in Shimadzu QP-5000 gas chromatograph-mass spectrometer. Trace levels of pesticide and alkylphenols in the range of non-detectable to 15.17 ng ml⁻¹, were detected in the human cord blood samples. This technique of monitoring the levels of endocrine-disruptors in blood samples is consistent, reliable and cost effective while reducing wastage of time and solvents.     

Partial oscillator strengths for the photoionization of N2O and CO2 (20–60 eV)

The photoelectron branching ratios and the partial oscillator strengths (cross sections) for photoionisation of the valence orbitals of N₂O and CO₂ have been obtained in the energy range 20–60 eV using the magic angle dipole (e, 2e) method. In addition to single electron ionization processes there is a large contribution from multiple electron transitions at higher energies in agreement with recent theoretical predictions. The photoionization efficiency and the dipole oscillator strenght for total photoabsorption have also been measured.     

Synthesis of C60-diphenylaminofluorene dyad with large 2PA cross-sections and efficient intramolecular two-photon energy transfer

The first, highly two-photon active C60 derivative comprised of a A-sp3-D conjugate structure was synthesized showing effective two-photon absorption cross-sections (sigma 2' = 196 x 10(-48) cm4 sec-1 molecule-1) in the nanosecond regime among the best values for diphenylaminofluorene-based AFX chromophores.

制备了4-乙烯基吡啶-镍(钴)和丙烯酰胺-镍(钴)络合物并研究了它们的结构。在自由基引发剂的作用下,4-乙烯基吡啶-镍可进行溶液聚合,也可与苯乙烯共聚合,测定了单体的竞聚率。所得的聚(4-乙烯基吡啶-镍)及聚(4-乙烯基吡啶-钴)以NaBH_4还原后可作为一些有机化合物的加氢催化剂。由IR及XPS分析说明在丙烯酰胺-镍络合物中,与镍离子发生了络合作用的是羧基上的氧原子。     

Reduced Overlap Population Analysis for Triosmium Clusters

The use of reduced overlap population (ROP) analysis from EHMO calculations as a means to gauge the presence of metal–metal bonds in triosmium clusters is examined. A number of triosmium clusters containing bridging ligands have been used as test cases, including the bis-ethoxy bridged cluster Os₃(μ-OEt)₂(CO)₁₀, 1b, and some of its group 15 derivatives Os₃(μ-OEt)₂(CO)₉(EPh₃), 2. These latter clusters are newly synthesized and have been characterized completely, including by single-crystal X-ray crystallographic studies. Measurement of pH by flow injection over a wide dynamic range with PVC/neutral carrier electrodes

Two pH-sensitive neutral carrier/PVC electrodes are used simultaneously for the measurement of pH by flow injection. One of these is based on the neutral carrier tridodecylamine and the other on octadecyl isonicotinate, and together they allow the pH range 1–13 to be covered. These electrodes have been used in a very low dispersion miniature potentiometric flow cell designed specifically for use in flow injection in conjunction with a multi-channel data acquisition system. The effects of the solution ionic strength and buffer capacity on the pH measurement are discussed. A flow-injection manifold is proposed which can be used for high-accuracy pH measurements without ionic strength adjustment of the sample and for simultaneous pH and ion concentration measurements with ionic strength adjustment. This has been tested on some synthetic samples for the simultaneous determination of potassium and calcium and the measurement of pH.
