A kinetic method for the determination of nanogram quantities of amino acid glycine (Gly) is described. The catalytic activity of cobalt in the reaction of oxidation of purpurin (1,2,4-trihydroxyantraquinone, PP) by hydrogen peroxide in alkaline buffer solution decreases in the presence of micro quantities of glycine. Operating conditions for the successful determination of glycine were optimized and yielded a theoretical detection limit of 6.5 ng/mL. Kinetic equations are proposed for the investigated process. The interference effects of certain foreign ions and amino acids upon the reaction rate were studied in order to assess the selectivity of the method. The procedure was successfully applied to the determination of glycine in various pharmaceutical samples. The unique features of this procedure are that the determination can be carried out rapidly at room temperature and analysis time is short. The procedure is simple, inexpensive and efficient for use in the analysis of a large number of samples. The article is published in the original.  相似文献

In this preliminary study, we used the Multivariate Curve Resolution- Alternating Least Squares (MCR-ALS) algorithm to analyze the excitation-emission matrix for different samples of maize flour contaminated with aflatoxin B1 (AFB1) - uncontaminated, low-contaminated, high-contaminated and flour from the local market. We intended to see if there are differences in emission spectral parameters that depend on degree of contamination. The analysis used genuine emission of the fluorophores in the flour, in absence and presence of AFB1, which enables fast screening of the samples, without sample pre-processing. As a result of the analysis, two fluorescence components were derived from the emission spectra for all analyzed samples. The components’ positions were the same for the uncontaminated reference sample and the commercial flour sample from the local market, whereas for the samples contaminated with the aflatoxin B1, the emitted peaks’ positions were red-shifted. We found that the ratio of the areas of these two components is proportional to the intensity of contamination: 0.071 for uncontaminated sample, 0.090 for the sample from local market, 0.192 for low-contaminated sample and 1.431 for high-contaminated sample. These results indicate that fluorescence EEM coupled with MCR-ALS could be used for rapid and simple estimation of the degree AFB1 contamination in maize flour.  相似文献

Leukocyte subsets, total leukocyte isolates or full blood samples were subjected to medium-strength square-wave electric impulses (100 V/cm field force, 5 ms duration). On the surface of the leukocytes, the expressions of several markers (CD3, CD4, CD8, CD11a, CD11b and ICAM-1) were determined in order to study the influence of pulsed ionic currents on different aspects of the cellular immune response. Large individual differences were observed among randomly chosen healthy donors, both in the initial expression rate and in the response patterns of different antigens. As a general conclusion, it can be stated that electric impulses with the above parameters activate the state of immune response alertness of human leukocytes. Changes in the activities of several enzymes in the serum in response to electric impulses were also tested in order to examine the feasibility of ex vivo electric treatment of human blood for the establishment of an antiviral and immune activated condition. Slightly elevated lactate dehydrogenase (LDH) levels point to a possibility of enhanced haemolysis, while the lack of an elevation in the membrane-bound peroxidase activity indicates the absence of haemolysis. Significant rises were detected in the serum superoxide dismutase (SOD) activities. Since most ex vivo blood manipulations are characterised by the appearance of superoxide radicals in the serum, a SOD activity enhancement is considered beneficial in these cases. A mild, but significant reduction in the blood clotting time indicates that electric treatment of human blood should be performed with special attention to thrombosis-prone conditions, and adequate precautions and countermeasures should be introduced. Although wider examinations are required before this method can be fully recommended, ex vivo blood treatment with medium-strength electric impulses seems to be a promising adjuvant course for the establishment of acute immune potentiation and an antiviral state in patients undergoing dialysis treatment.  相似文献

The structure of the only structurally yet uncharacterized alkali saccharinate, lithium saccharinate, is reported, together with the infrared spectra of its protonated and partially deuterated analogues. The compound crystallizes as strongly hygroscopic colorless triclinic crystal blocks of a 11/6 hydrate, LiC7H4NO311/6H2O, space group , with unit cell dimensions a = 10.070(1), b = 15.755(2), c = 19.134(2) Å, α = 68.085(2), β = 76.940(2), γ = 84.169(2)°, V = 2743.0(5) Å3 and Z = 12 at T = 200(2) K.  相似文献

To characterise electron-trapping sites on the surface of MgO nanoparticles, surface colour centres were generated using UV light in conjunction with selected hydrogen-based electron sources. Four different colour-centre species, including the characteristic (e-)(H+) or F(S)+(H) centre, were identified due to the distinct shape of the respective electron paramagnetic resonance (EPR) signals. The analysis of the EPR saturation behaviour down to microwave powers of 5 x 10(-3) mW reveals an enhanced spin-relaxation probability of the (e-)(H+) centre compared to all other F(S)+ centres that do not exhibit significant magnetic interactions with hydroxylic protons. Beside the dipolar magnetic interaction in the (e-)(H+) centre observed by EPR, the electronic interaction between the unpaired electron and the proton of a closely spaced OH group produces a redshift of the OH stretching band by about 70 to 170 cm(-1), as observed by infrared spectroscopy. EPR and IR spectroscopic data obtained after the selective address of individual reaction channels for surface colour-centre formation point to the fact that (e-)(H+) centres are formed by trapping electrons from H atoms. Consequently, the underlying surface defect does not belong to the sites of the MgO surface, which chemisorb hydrogen via a heterolytic splitting process.  相似文献

Xanthohumol (XN) is a prenylated chalcone with antimutagenic and anticancer activity from hops. A nonaqueous reverse polarity capillary electrophoretic method for the determination of XN in hop extract was developed and validated. The optimal parameters were a 64.5 cm long fused-silica capillary with 50 microm id at 25 degrees C; 30 kV negative voltage (anode at detector side of the capillary); nonaqueous buffer with 75 mM NaOH and 50 mM boric acid in methanol; hydrodynamical injection with 10 mbar for 40 s; and detection at 440 nm. XN, isoxanthohumol (IX), colupulone, adlupulone, and n-lupulone were well resolved on the electropherogram. The LOD for XN was 0.05 mg/L and RSD for peak area was below 3%. The amount of XN in different samples of hop pellets varied from 0.14 to 0.42%.  相似文献

Preventive care, research, and restoration of cultural heritage objects requires multidisciplinary research and the involvement of experts of different profiles, using high technology equipment. Nondestructive methods dominate in the diagnosis of the situation and protection of cultural heritage objects. The application of lasers has opened many possibilities for research in the field of protection, conservation, restoration, and/or assessment of artifacts. We present the results of the interaction of ruby-laser light with the surfaces of Neolithic ceramics (Obrenovac, Serbia) and samples of copper of unknown age. The investigation was conducted in order to determine the maximum energy density of the laser light that can be applied in nondestructive testing and encrustation cleaning of these ceramic and metal cultural heritage objects. We investigate the laser-light interactions using a scanning electron microscope (SEM) with energy-dispersal unit for the analysis of X-rays (EDX).  相似文献

This paper presents a method for solving nonlinear system with singular Jacobian at the solution. The convergence rate in the case of singularity deteriorates and one way to accelerate convergence is to form bordered system. A local algorithm, with finite-difference approximations, for forming and solving such system is proposed in this paper. To overcome the need that initial approximation has to be very close to the solution, we also propose a method which is a combination of descent method with finite-differences and local algorithm. Some numerical results obtained on relevant examples are presented.  相似文献
