A highly efficient electrode material, three-dimensional reduced graphene oxide with varying wrinkles and folds (WRGO), applicable for electrochemical determination of Hg(II) was obtained by treating graphene oxide (GO) with KOH aqueous solution. After alkaline etching, the relatively flat graphene was altered and its self-aggregation was significantly alleviated, producing more wrinkles and folds, which provided more active adsorption sites for heavy metal ions. WRGO-5 modified electrode system herein offers a highest sensitivity of (31.83 μAμM−1) and a lowest LOD of (16.28 nM). Moreover, the electrode sensor possesses good stability and reproducibility.  相似文献

Parallel solution-phase chemistry has yielded a series of secondary amide analogues of the fungal natural product 3-chloro-4-hydroxyphenylacetamide. 3-Chloro-4-hydroxyphenylacetic acid was coupled to a variety of primary amines using 1-ethyl-3-(3'-dimethylamino- propyl)-carbodiimide hydrochloride. The desired products were obtained in good yield and high purity following rapid silica purification. All analogues were spectroscopically characterised using NMR, UV, IR and MS data. One compound displayed moderate cytotoxicity against the human melanoma and prostate cell lines, MM96L and DU145.  相似文献

Here we exploit the simple, ultra-stable, modular architecture of consensus-designed tetratricopeptide repeat proteins (CTPRs) to create a platform capable of displaying both single as well as multiple functions and with diverse programmable geometrical arrangements by grafting non-helical short linear binding motifs (SLiMs) onto the loops between adjacent repeats. As proof of concept, we built synthetic CTPRs to bind and inhibit the human tankyrase proteins (hTNKS), which play a key role in Wnt signaling and are upregulated in cancer. A series of mono-valent and multi-valent hTNKS binders was assembled. To fully exploit the modular scaffold and to further diversify the multi-valent geometry, we engineered the binding modules with two different formats, one monomeric and the other trimeric. We show that the designed proteins are stable, correctly folded and capable of binding to and inhibiting the cellular activity of hTNKS leading to downregulation of the Wnt pathway. Multivalency in both the CTPR protein arrays and the hTNKS target results in the formation of large macromolecular assemblies, which can be visualized both in vitro and in the cell. When delivered into the cell by nanoparticle encapsulation, the multivalent CTPR proteins displayed exceptional activity. They are able to inhibit Wnt signaling where small molecule inhibitors have failed to date. Our results point to the tremendous potential of the CTPR platform to exploit a range of SLiMs and assemble synthetic binding molecules with built-in multivalent capabilities and precise, pre-programmed geometries.  相似文献

Host cell proteins (HCPs) are widely regarded as a critical quality attribute for a biotherapeutic product. Bottom up MS is the present gold standard for HCP analysis but suffers from incomplete protein identification due to complex nature of the HCP mixture and limited separation efficiency of the preceding LC-based systems. In this paper, we present for the first time an application involving use of LC-CE-MS/MS platform for analysis of HCPs. It has been demonstrated that the proposed platform has been able to successfully identify 397 HCPs from the supernatants of recombinant Chinese hamster ovary cells, twice and thrice the number of proteins identified by the state-of-the-art LC-MS/MS (189 HCPs) and CE-MS/MS (128 HCPs) analyses, respectively. Of these, 225 HCPs were unique to the LC-CE-MS/MS approach and were not identified by either LC-MS/MS or CE-MS/MS. It is observed that the LC-CE-MS/MS platform combines the benefits of LC-MS/MS and CE-MS/MS techniques and identifies peptides in a wider range of size, pI, and hydrophobicity. Additionally, LC-CE-MS/MS also identified more HCPs associated with cellular components, molecular functions, biological processes, peptidases, and secretory proteins. The proposed approach would thus be a useful addition in HCP analysis and secretome studies of mAb-producing Chinese hamster ovary cells.  相似文献

A fundamental study on the behaviour of heating coal macerals has been undertaken using two novel thermal analysis techniques. The apparent specific heat was determined during heating using an inverse calorimetric method (computer aided thermal analysis, CATA) and combined with pressure and displacement measurements to correlate endothermic and exothermic behaviour with measurement of swelling. The second technique used a post-oxidation stage to combust the tars and gases into products which were analysed. This method was used to study the elemental character of volatiles release from coal maceral concentrates in terms of carbon and hydrogen. Extents of swelling and exothermicity during primary devolatilisation were found to be correlated with vitrinite content and were associated with tar evolution. For the highest vitrinite fraction (of 86.4% vitrinite) swelling was initiated at the same temperature range for exothermic reactions, and maximum swelling coincided with the peak release of light gases. Tar evolution was found to change in chemical character (as defined by H/C ratio) during progressive heating, initially rising in the early stages of tar formation (<430 °C) to a maximum of 1.24, then gradually decreasing to a minimum of 0.64 at 550 °C.  相似文献

We consider acoustic waves in fluid-saturated periodic media with dual porosity. At the mesoscopic level, the fluid motion is governed by the Darcy flow model extended by inertia terms and by the mass conservation equation. In this study, assuming the porous skeleton is rigid, the aim is to distinguish the effects of the strong heterogeneity in the permeability coefficients. Using the asymptotic homogenization method we derive macroscopic equations and obtain the dispersion relationship for harmonic waves. The double porosity gives rise to an extra homogenized coefficient of dynamic compressibility which is not obtained in the upscaled single porosity model. Both the single and double porosity models are compared using an example illustrating wave propagation in layered media.  相似文献

This work bridges a gap in the cross‐coupling of aliphatic redox‐active esters with aryl zinc reagents. Previously limited to primary, secondary, and specialized tertiary centers, a new protocol has been devised to enable the coupling of general tertiary systems using nickel catalysis. The scope of this operationally simple method is broad, and it can be used to simplify the synthesis of medicinally relevant motifs bearing quaternary centers.  相似文献

Thermally stable, flexible polymer gel electrolytes with high ionic conductivity are prepared by mixing the ionic liquid 1-butyl-1-methylpyrrolidinium bis(trifluoromethylsulfonyl)imide (C4mpyrTFSI), LiTFSI and poly(vinylidene difluoride-co-hexafluoropropylene (PVDF-HFP). FT-IR and Raman spectroscopy show that an amorphous film is obtained for high (60 %) C4mpyrTFSI contents. Thermogravimetric analysis (TGA) confirms that the polymer gels are stable below ∼300 °C in both nitrogen and air environments. Ionic conductivity of 1.9×10−3 S cm−2 at room temperature is achieved for the 60 % ionic liquid loaded gel. Germanium (Ge) anodes maintain a coulombic efficiency above 95 % after 90 cycles in potential cycling tests with the 60 % C4mpyrTFSI polymer gel.  相似文献

Suspensions of solid lipid nanoparticles (SLNs) stabilized with emulsifiers have been extensively investigated (since the 1990s) as drug carriers, although details of their ultrastructure are poorly defined. Previously, a novel microwave‐assisted microemulsion‐based technique to prepare SLNs was reported. To understand the detailed internal structure of these SLNs, ultra‐small angle neutron scattering (USANS) and small angle neutron scattering (SANS) experiments are conducted on suspensions of hydrogenated stearic acid SLNs stabilized with hydrogenated Tween 20 surfactant in D2O. Together, SANS and USANS gives a combined Q range of 0.000047 to 0.6 Å?1 (corresponding to a size range of ≈1 nm–15 µm). This extended Q range allows a comprehensive understanding of the hierarchical structure of SLNs. The data are consistent with the multi‐length scale structure of SLNs having polydispersed large particles with roughened surfaces at the microscale level. At the nanoscale level, the results are consistent with the SLNs having an ellipsoidal shape intermediate between spheres and rods, with a crossover from mass fractals to surface fractals. The elucidation of this structure is particularly important given that the structure influences the stability and drug release properties of the nanoparticles. These results assist in the development of systems with desired shape and properties.  相似文献
