Quantitative analysis of competitive milk protein adsorption to air/water interfaces in aqueous foam was performed by capillary electrophoresis (CE). Foams were made by whipping protein solutions, in which skim milk powder (SMP) and whey protein isolate (WPI) were mixed at 0.5% protein in different proportions at different pH values and NaCl concentrations. Preferential adsorption of beta-casein into foam phases occurred under most solution conditions, if partial dissociation of the casein micelles had occurred. Preferential adsorption of beta-casein was not observed with added Ca2+, due to the re-association of casein micelles. Enrichment of caseins into the foam phase was more apparent than that of whey proteins. The foamability of SMP demonstrated a continuous improvement due to the gradually increasing dissociation of casein micelles when the concentration of NaCl increased from 0 to 0.8 M. The foamability of WPI increased when NaCl concentration rose from 0 to 0.1 M, and decreased with further increase in NaCl concentration. NaCl at low concentration (I < or = 0.4) did not show a significant effect on the competitive adsorption among milk proteins, indicating that electrostatic interactions do not play a key role in competitive adsorption. NaCl at higher concentration, e.g., 0.6 M, caused less whey protein to be adsorbed to the air/water interfaces. The whippability of WPI was highest at pH 4.5 and lowest at pH 3, and that of SMP was the opposite. The proportions of beta-lactoglobulin and alpha-lactalbumin in the foam phase were lower at acidic pH and higher at basic pH, compared with that at natural pH of WPI. 相似文献

Glycolipid presentation by CD1 proteins has emerged as an important aspect of antigen recognition, and presentation of alpha-glycosylceramides by CD1d to natural killer T cells has become a central focus in understanding how glycolipid presentation can influence immune responses. An alpha-galactosylceramide containing relatively long lipid chains has been the subject of intense study because, when presented by CD1d to natural killer T cells, it stimulates the release of both proinflammatory and immunomodulatory cytokines. Using an efficient synthesis of alpha-galactosylceramides, we have prepared a series of glycolipids in which the lipid chain lengths have been incrementally varied. The responses of natural killer T cells to these glycolipids have been determined, and we have found that truncation of the phytosphingosine lipid chain increases the relative amounts of immunomodulatory cytokines released. In similar fashion, the length of the acyl chain in alpha-galactosylceramides influences cytokine release profiles. 相似文献

Novel polydimethylsiloxane (PDMS) based polyionenes were synthesized by a modified Menschutkin reaction involving reaction between bromo-terminated PDMS oligomers and various ditertiary amino compounds. In this study, the nature of the hard segment was varied by using various ditertiary amino compounds and in some cases by incorporating chain extenders, while the soft segment content was varied by changing the molecular weight of the PDMS oligomers. The mechanical properties of these materials were found to be dependent on both the nature and amount of the hard segments. These materials also showed distinct evidence of a microphase-separated morphology where under normal conditions, the hard segments formed in what are believed to be cylindrical ion-rich microdomains dispersed randomly in the soft PDMS matrix. When subjected to uniaxial deformation, the ionic cylinders were found to orient along their long axes in the stretch direction. 相似文献

Reference materials certified for purity are essential to ensure harmonization of analytical measurements. LGC is currently certifying these materials using an indirect multi-method approach quantifying impurities: Related substances using high-performance liquid chromatography, gas chromatography (GC), differential scanning calorimetry; Residual solvents using headspace GC coupled to mass spectrometry; Inorganic content using ashing, acid digest ion couple plasma mass spectrometry or thermogravimetric analysis; Water using oven coulometric Karl Fischer/direct addition coulometric Karl Fischer. Related substances are not straightforward to quantify without an appropriate standard due to possible difference in response factor for the impurity relative to the main compound. In this article, existing LGC RMs certified for purity were purified further using semi-preparative HPLC. These ultra-purified organic substances were virtually free of related substances making their purity assessment faster and more straightforward, i.e., no need to identify impurities and subsequently quantify them. After characterization, these ultra-purified standards were used as calibrants to determine directly the mass fraction of the analyte in the original CRM using exact matching single-point HPLC calibration. This new approach opens the possibility of certifying the purity of low purity substances with a relative small uncertainty without the need of identifying the impurities present in the sample. 相似文献

The close proximity in melting temperature of the LGC Limited DSC standards indium and diphenylacetic acid, has enabled a direct assessment to be made of any differences resulting from the use of a metal or an organic compound in the calibration of DSC equipment. Following calibration with indium, the equilibrium fusion temperatures for diphenylacetic acid, were determined by both the stepwise heating and extrapolation to zero heating rate methods. The results were in excellent agreement with the certificate values and established that indium may be used as a calibrant when making accurate DSC measurements on organic materials in the same temperature range and that it has the advantage that it is non-volatile and can be used a number of times without significant change. Similar agreement was obtained in the measurement of the enthalpy of fusion, although the larger heat capacity change on fusion of diphenylacetic acid resulted in a greater uncertainty than with indium. 相似文献
