We have previously used inelastic incoherent neutron scattering spectroscopy to investigate the properties of aqueous suspensions of biomolecules as a function of hydration. These experiments led to the identification of signals corresponding to interfacial (hydration) water at low water content. A prediction from these studies was that in the crowded environment inside living cells, a significant proportion of the water would be interfacial, with profound implications for biological function. Here we describe the first inelastic incoherent neutron scattering spectroscopy studies of living cells and tissues. We find that the interfacial water signal is similar to that observed for water interacting with purified biomolecules and other solutes, i.e., it is strongly perturbed in the librational and translational intermolecular optical regions of the spectrum at 20-150 meV. The ratio of interfacial water compared to total water in cells (approximately 30%) is in line with previous experimental data for hydration water and calculations based on simple assumptions.  相似文献

A direct method for silicon determination in milk samples by Electrothermal Atomic Absorption Spectrometry was developed. Palladium was used as chemical modifier at a concentration of 610 mg L(-1); with this modifier, silicon was stable up to 1800 degrees C. The precision and accuracy of the method were investigated. The detection limit was 16.2, 2.7 and 7.2 micro g L(-1) for cows' milk, human milk and infant formula, respectively. The method was applied to silicon determination in 17 infant formula samples, 13 human milk samples and 12 cows' milk samples.  相似文献

Novel dibenzo[a,c]phenanthridines are prepared regioselectively by the application of a straightforward synthetic pathway, starting from new 3,4-diaryl- and 3,4-dihydro-3,4-diarylisoquinolines prepared via Ritter-type heterocyclization and the more classical two-step reductive amination/Bischler-Napieralski cyclization of triarylethanones, respectively. A comparative study of nonphenolic oxidative coupling methodologies provides a highly efficient procedure, based on the hypervalent iodine reagent phenyliodine(III) bis(trifluoroacetate) (PIFA), to accomplish the final coupling step.  相似文献

 Four inter-comparison exercises on organic elemental analysis were carried out between 1997 and 2001 by the Department of Analytical Chemistry of the University of Barcelona, together with the Microanalysis Service and the Institute of the Marine Sciences, which both belong to the CSIC in Barcelona, and the University of A Coru?a. More than sixty laboratories participated in these exercises. Here we describe the design and characteristics of the trials, the samples and the homogeneity tests applied. We report the results obtained for the analysis of carbon, hydrogen, nitrogen, sulphur and oxygen, their statistical analysis, and the most relevant aspects of the technical discussion meetings. [reaction: see text] A new route to oxcarbazepine (Trileptal), the most widely prescribed antiepileptic drug, starting from commercially available 2'-aminoacetophenone and 1,2-dibromobenzene, is reported. The sequentially accomplished key steps are palladium-catalyzed intermolecular alpha-arylation of ketone enolates and intramolecular N-arylation reactions. After several experiments to establish the best conditions for both arylation processes, the target oxcarbazepine is obtained in a satisfactory overall yield, minimizing the number of steps and employing scalable catalytic procedures developed in partially aqueous media.  相似文献

The preparation of indigo from Indigofera suffruticosa following the procedures attributed to ancient Mayas was electrochemically monitored using the voltammetry-of-microparticles approach. The mechanism formation of indigotin and indirubin from its precursors, indican and isatan, is discussed. Comparison of voltammetric profiles for differently prepared and commercial indigos and genuine Maya Blue samples suggests that the preparation procedure of indigo changed during the Late Classical Maya period. A method is described for neutron activation analysis of thorium by employing thorium-233. Decontamination steps include anion exchange in 12.5 N HCL, precipitation of fluoride, extraction with mesityl oxide and thorium oxalate precipitation. The time spent is less than 1 h; spurious activities in counting samples amount only to 2% and can easily be corrected for. The sensitivity of the method is ca. 0.02 μg of thorium in a neutron flux of 1012n/cm2/sec with an irradiation time of 5 min.  相似文献

Application of a potassium ion sensor based on an ion sensitive field effect transistor (ISFET) for ion control of a dialysis solution in an artificial kidney and in blood plasma of patients treated by hemodialysis is presented. Sensors and their long-term stability were characterised in constant contact with test solutions. Test results are compared to those obtained with conventional ion-selective electrodes and commercial blood ion analyser. Tested ISFET sensors showed high reliability in potassium ion measurements in the physiologically significant concentration range which, along with low cost of their production, makes them promising for cited application.  相似文献

Aluminosilicates can present different structures such as crystalline true zeolite molecular sieves or amorphous silica–aluminas. With a large surface area available, both can be involved as catalysts, adsorbents or catalyst supports, and the determination of their surface acidic properties is an important parameter in the study of such materials.

The number, strength and strength distribution of the acidic sites were determined using microcalorimetry linked to a volumetric line. Ammonia was used as a basic probe molecule. The adsorption temperatures ranged from 353 K up to 473 K. The samples consisted of two amorphous silica–aluminas (Si/Al ≈ 6.5) and three microporous zeolites H-β, H-ZSM-5 and H-MCM-22 with similar Si/Al ratios (Si/Al ≈ 13).

The differential heats of ammonia adsorption versus coverage and the corresponding isotherms are given. The H-ZSM-5, H-MCM-22, H-β samples display a plateau of constant adsorption heats near 150 kJ mol−1, while the silica–alumina samples present continuously decreasing heats from 150 kJ mol−1 at zero coverage to 40 kJ mol−1 at high coverage, due to their surface heterogeneity. For amorphous silica–aluminas, the number of acid sites is dependent of the aluminum distribution at the surface.

The differences observed in the adsorption behavior of ammonia over the three zeolites arise from differences in their morphology, i.e. the total free volumes, pore geometries and electric field gradients at the adsorption sites. The adsorption isosteres have also been calculated from the adsorption isotherms, and the isosteric heats of adsorption have been compared with the heats measured by calorimetry.  相似文献
