Micro enzymatic assay coupled to capillary electrophoresis via liquid junction

Summary A system for capillary electrophoresis combined with enzymatic assay has been evaluated for the two enzymes glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase. Instrumentation included a post-column reactor coupled to the separation capillary by a liquid junction. A technique for generating a substrate solution flow into the reactor by utilizing two high voltage supplies is proposed. This method offers a high degree of freedom in optimizing the separation and enzymatic reaction conditions individually. Possibilities for improving the enzymatic assay sensitivity were also examined.

Quality assurance for the analytical data of micro elements in food

The micro element content of food is an important quality index due to the action of these elements on human health. In this article, we discuss how to ensure the reliability of analytical data on micro elements in order to truly represent the condition of food. Sampling, treatment of the analytical sample, selection of the analytical method, standard solution, and certified reference material, blank test, calibration of the instrument and equipment, application of the quality control chart, assessment of the final analytical result, and quality assurance system are briefly described. Computational Electrochemistry: A Model to Studying Ohmic Distortion of Voltammetry in Multiple Working Electrode,Microfluidic Devices,an Adaptive FEM Approach

Characteristics of mass transport and potential distribution applicable to microfluidic electrochemical flow cell devices has been modelled using the finite element method. A flexible, automatic grid generation algorithm has been combined with an a‐posteriori error indication technique presented by Nann and Heinze to allow irregular cell geometries to be modelled. The code has been applied to the problem of steady state generator – detector linear sweep voltammetry in a channel flow cell showing the effects of IR drop on the voltammetric response of each electrode.     

Monte Carlo simulation of grain growth in polycrystalline materials

Understanding the kinetics of grain growth, under the influence of second phase (such as impurities, voids and bubbles) is fundamental to advances in the control of microstructural evolution. As a precursor to this objective, we have investigated the grain growth kinetics in a polycrystalline material using a standard Q-state Potts’ model under Monte Carlo settings. Based on physical reasoning, new modifications are suggested to circumvent some of the disadvantages in the basic Potts model. The efficacy of these modifications vis-à-vis the basic model is verified. The influence of second phase particles on the impurity loaded grain boundaries is investigated for the study of grain growth kinetics.

As(Ⅲ)-Cu(Ⅱ)协同抑制鲁米诺-MnO_4~-化学发光反应的研究及其应用

本文报道了As(Ⅲ)-Cu(Ⅲ)两种离子对鲁米诺-MnO_4~-化学发光反应的协同抑制作用,据此拟定了测定微量As(Ⅲ)的化学发光新方法。该法灵敏度较高,检测限为6.8×10~(-9)g/ml As(Ⅲ),选择性也较好,同时线性范围宽(7.0×10~(-9)～3.0×10~(-6)g/ml),测定手续简便。应用本法测定工业废水中的As(Ⅲ),结果满意。     

Application of laser ablation–inductively coupled plasma-mass spectrometry (LA–ICP–MS) to investigate trace metal spatial distributions in human tooth enamel and dentine growth layers and pulp

Human tooth enamel provides a nearly permanent and chronological record of an individuals nutritional status and anthropogenic trace metal exposure during development; it might thus provide an excellent bio archive. We investigated the micro-spatial distribution of trace metals (Cu, Fe, Mg, Sr, Pb, and Zn) in 196×339 m² raster pattern areas (6.6×10⁴ m²) in a deciduous tooth using laser ablation-inductively coupled plasma-mass spectrometry (LA–ICP–MS). Ablated areas include prenatal and postnatal enamel, the neonatal line, the dentine–enamel junction (DEJ), dentine, and the dentine–pulp junction. Topographic variations in the surface elemental distribution of lead, zinc, strontium, and iron intensities in a deciduous tooth revealed heterogeneous distribution within and among regions. ⁴³Ca normalized elemental intensities showed the following order: Sr>Mg>>Zn>Pb>Fe>Cu. Elevated zinc and lead levels were present in the dental pulp region and at the neonatal line. This study demonstrates the ability of LA–ICP–MS to provide unique elemental distribution information in micro spatial areas of dental hard tissues. Elemental distribution plots could be useful in decoding nutrition and pollution information embedded in their bio apatite structure.Presented in part at the 2002 Winter Conference on Plasma Spectrochemistry, Scottsdale, AZ, January 6–12, 2002. Fluorometric detection with a packed flow cell in micro high-performance liquid chromatography

Summary A packed flow cell was used for fluorometric detection in micro high-performance liquid chromatography. The flow cell consisted of fused-silica tubing packed with the same material as the separation column. A focusing effect of the stationary phase on the signal intensity was observed, leading to an improvement of the mass detection limit, as achieved by on-column detection.
