Chiral, non-racemic 1,2-disubstituted ferrocenes have been prepared from monosubstituted ferrocene derivatives by amine-mediated ortho-directed reactions and subsequent partial reductive removal of the stereogenic ortho-directing group. It was found that the ortho-directing amino group of 2-substituted derivatives of N,N-dimethylaminoethyl-ferrocene and similar compounds can, after quaternisation with methyl iodide, be reductively removed with sodium borohydride to give 2-substituted methyl- or ethylferrocenes. In most cases the substituents I, Br, COOEt, P(O)Ph2 and CN tolerate the reaction conditions used. In addition, a few examples are reported that show how the use of LiTMP allows 2-bromo- and especially 2-cyano-substituted derivatives to be further ortho-lithiated and reacted to give 1,2,3-trisubstituted ferrocenes.  相似文献

We have developed a straightforward approach to diverse synthesis of 2,3-, 2,4-disubstituted pyrimido [1,2-a]benzimidazoles, 2,4,10-trisubstituted 2,10-dihydropyrimido [1,2-a]benzimidazoles and 2,3-disubstituted imidazo [2,1-b]benzothiazoles via multicomponent reactions (MCRs) of heterocyclic azoles, aldehydes with easily storable and handling alkynecarboxylic acids. In the presence of a catalytic amount of CuI and K2CO3, the pyrimido [1,2-a]benzimidazole or imidazo [2,1-b]benzothiazole scaffold could be rapidly constructed through a 6-endo-dig or 5-exo-dig cyclization, respectively. The preliminary mechanistic study suggested that the formation of 2,3- disubstituted pyrimido [1,2-a]benzimidazoles, which completes the assembly of the scaffold and its C-3 position functionalization in one pot, undergoes a novel cascade process involving a decarboxylation, A [3] coupling, 6-endo-dig cyclization, nucleophilic addition and dehydration.  相似文献

Bicyclo[1.1.1]pentane (BCP) motifs are of growing importance to the pharmaceutical industry as sp3-rich bioisosteres of benzene rings and as molecular building blocks in materials science. Herein we explore the behavior of 1,3-disubstituted BCP moieties on metal surfaces by combining low-temperature scanning tunneling microscopy / non-contact atomic force microscopy studies with density functional theory modeling. We examine the configuration of individual BCP-containing precursors on Au(111), their supramolecular assembly and thermally activated dehalogenative coupling reactions, affording polymeric chains with incorporated electronically isolating units. Our studies not only provide the first sub-molecular insights of the BCP scaffold behavior on surfaces, but also extend the potential application of BCP derivatives towards integration in custom-designed surface architectures.  相似文献

Taurine and substituted taurines present a group of important structural elements in many natural products. Various substituted taurines, including 1- and 2-substituted, 1,1-, syn-1,2-, and anti-1,2-disubstituted taurines, were synthesized from the corresponding nitroolefins via Michael addition with thioacetic acid, oxidation with peroxyformic acid, and the catalytic hydrogenation under the catalysis of palladium on carbon or platinum dioxide. It is a general, versatile, and salt-free method for the preparation of substituted taurines, especially for syn- and anti-1,2-disubstituted taurines and some taurines with more bulky substituents. The stereostructures of both syn- and anti-1,2-disubstituted taurines were deduced from the nitroalkyl thioacetates in the Michael addition, which were identified via the Karplus equation analysis and computational analysis, and finally confirmed by the XRD single crystal analysis. The diastereoselectivity in the Michael addition was rationalized with the Cram rule.  相似文献

A “chirality driven self‐sorting” strategy is introduced for the controlled supramolecular organization of donor (D) and acceptor (A) molecules in multicomponent assemblies. The trans‐1,2‐bis(amido)cyclohexane (trans‐BAC) has been identified as a supramolecular motif with strong homochiral recognition to direct this chirality controlled assembly process of enantiomers in solution. Stereoselective supramolecular polymerization of trans‐BAC appended naphthalene diimide monomers (NDIs) has been probed in detail by spectroscopic and mechanistic investigations. This chirality‐driven self‐sorting design of enantiomeric components also offers to realize mixed and segregated D‐A stacks by supramolecular co‐assembly of the NDI acceptors with trans‐BAC appended dialkoxynaphthalene (DAN) donor monomers. Such an unprecedented chirality control on D‐A organization paves the way for the creation of supramolecular p‐n nanostructures with controlled molecular‐level organization.  相似文献

The structures of a series of co-crystals formed by enantiomers of BINOL (2,2′-dihydroxy-1,1′-binaphthalene) with (R,R)-1,2-diaminocyclohexane (DACH) and (R,R)-1,2-diamino-1,2-diphenylethane have been determined by X-ray diffraction to investigate the factors responsible for diastereoselective co-crystallization, the process responsible for the highly effective resolution of rac-BINOL by DACH. The absolute configurations of the molecules and directional preferences of the hydrogen bonds in the supramolecular aggregates have been analyzed. The supramolecular aggregation of hydrogen-bonded BINOL and diamines shows considerable distortions from the helical shape, which indicates that cohesion forces other than hydrogen bonds play a significant role in the diastereoselective resolution.  相似文献

Two complementary protocols for assembly of multisubstituted N-fused heterocycles have been developed. It was demonstrated that 1,3-disubstituted N-fused heterocycles, including indolizines, pyrroloquinoxalines, and pyrrolothiazoles can easily be synthesized via an exceptionally mild and efficient method involving a novel silver-catalyzed cycloizomerization of propargyl-containing heterocycles. Alternatively, 1,2-disubstituted heterocycles can be accessed through the novel cascade transformation involving an alkyne-vinylidene isomerization with concomitant 1,2-shift of hydrogen, silyl, stannyl, or germyl groups. This mild and simple method allows for selective and highly efficient synthesis of indolizines, pyrroloisoquinolines, pyrroloquinoxalines, pyrrolopyrazines, and pyrrolothiazoles.  相似文献

Three types of supramolecular benzoheterocyclic host complexes were successfully prepared using (1R,2S)-2-amino-1,2-diphenylethanol and three types of benzoheterocyclic (benzothiophene, benzofuran, and benzopyrrole) acid derivatives. The host complexes had one-dimensional (1D) channel-like cavities that were formed by the assembly of two-component 21-helical columnar network structures, which included guest alcohol molecules. The release behavior of the guest molecules could be tuned by the type of heterocyclic ring in the supramolecular host complex.  相似文献

Diastereoselective synthesis of cis-2,6-disubstituted piperidines from 1,2-cyclic sulfamidates is described. Regioselective ring-opening reactions of 1,2-cyclic sulfamidates derived from L-phenylalanine, alanine, valine, norvaline with the ketal protected acetylide with a phenyl substituent proceed smoothly to form the N-sulfamate intermediates which on acidic hydrolysis give alkynylated amines with the ketal group intact. Hydrogenation of the alkynylated amines, debenzylation, ketal deprotection, subsequent cyclization (of aminoketones) and stereoselective hydrogenation of the cyclic iminium ion intermediates afford the corresponding cis-2,6-disubstituted piperidines in high diastereoselectivity (98%?≥?d.e.) with good chemical yields (68–86%). The present approach provides a novel route for the stereoselective synthesis of cis-2,6-disubstituted piperidines.  相似文献

An efficient, environmental friendly and substrate controlled method of synthesis of 2-substituted benzimidazole derivatives 3 and 1,2-disubstituted benzimidazole derivatives 4 with high selectivity has been achieved from the reaction of o-phenylenediamine 1 and aldehydes 2 in the presence of water extract of onion and selecting suitable reaction medium. This method is widely applicable for variety of aldehydes such as aromatic/aliphatic/heterocyclic aldehydes and 1,2-diamines to afford 2-substituted benzimidazole derivatives 3 and 1,2-disubstituted benzimidazole derivatives 4 in good to excellent yields (up to 96%). The developed method of water extract of onion catalysis produced 2-substituted benzimidazoles 3 from aromatic aldehydes having electron-withdrawing groups, whereas aromatic aldehydes bearing electron donating groups selectively furnished 1,2-disubstituted benzimidazole 4 derivatives. The process described here has several advantages of cheap, low energy consumption, commercially available starting materials, operational simplicity and nontoxic catalyst. The use of water extract of onion makes this present methodology green and giving a useful contribution to the existing methods available for the preparation of benzimidazole derivatives. In addition, Hammett correlation of substituent constant (σ) vs percentage (%) yield has been established.  相似文献

A series of new porphyrin derivatives possessing the side arms of alkyl-substituted thiophene oligomer were synthesized. The effects of solvent polarity on the formation of supramolecular assembly have been studied by UV-vis absorption, fluorescence emission, and TEM measurement. The linear-shape porphyrin derivatives bearing two thiophene pentamers at meso-position showed the significant spectral changes in both blue-shift and band broadening of Soret bands which indicate the formation of a relatively larger porphyrin H-aggregate that occurred in nonpolar solvent such as n-hexane.  相似文献

1,2- and 2,3-disubstituted cyclopentenes were hydrogenated with several transition metal catalysts. The hydrogenation of 1,2-disubstituted cyclopentenes gave preferably the cis products with Raney Ni, whereas 2,3-disubstituted cyclopentenes gave preferably the trans isomers. The trans products were favoured in the hydrogenation of 1,2- and 2,3-dialkylcyclopentenes with Pt or Pd catalysts. A detailed product analysis of the hydrogenation of 1,2-dialkylcyclopentenes indicates that the double bond migration occurred to form 2,3-dialkylcyclopentenes in advance of the hydrogenation. In contrast, 1-methyl-2-phenylcyclopentene gave predominantly the cis product irrespective of the kind of catalyst. These results suggest the formation of a π-benzylic species during the course of the reaction. The mechanism is discussed in terms of the modified Horiuti-Polanyi mechanism.  相似文献

The addition of RMgX or R2Mg to nicotinic acid esters, activated with triisopropylsilyl triflate, was investigated. The regioselectivity of this reaction, where 4-unsubstituted and 4-substituted pyridine derivatives were employed as starting materials, was examined. Depending on the structure of the organomagnesium reagent varying ratios of 1,2-, 1,4-, and 1,6-regioisomers were obtained but in any case the 1,4-addition products were clearly predominating. Desilylation of the isomeric pure addition products with LiOH provided stable 4,4-disubstituted dihydropyridines, which could be easily N-alkylated with alkylhalides in high yield. Furthermore, it was shown that the silyl protecting group of the obtained dihydropyridines can be easily replaced by an acyl group.  相似文献

NaIO4 oxidizes lithium bromide efficiently under acidic conditions to functionalize alkylbenzenes and alkanes and produce the corresponding bromo and acetoxy derivatives in excellent yields. Expanding our studies on the anti-angiogenesis activities of 2,4-disubstituted quinazoline derivatives [8], a series of novel N-(2-(quinazolin-2-yl)phenyl)benzamide (SZ) derivatives were designed and synthesized. Cytotoxicity assays indicated that most of these compounds displayed similar cytotoxicity against tumor cells in comparison with our previously reported, but showed a higher cytotoxicity against HUVECs. The SZ derivatives showed a remarkable inhibitive effect against the migration and adhesion of HUVECs, in addition to demonstrating significant in vivo anti-angiogenesis activities in the chick embryo chorioallantoic membrane (CAM) assay. The results proved that the introduction of an aryl group with a basic amide side chain on the 4′ position linked to the amide of the C-2 substituted quinazoline scaffold is an effective approach to improve the anti-angiogenic activity of quinazoline derivatives.  相似文献

Novel diimidazo[1,2-a:5′,1′-c]quinoxaline derivatives were synthesized by a Pd-catalyzed intramolecular coupling reaction of 1,2-bis(2-iodo-1H-imidazol-1-yl)benzene. This method was applied to the selective introduction of an aryl substituent at the 10 position by the addition of a boronic ester. Moreover, a stepwise introduction of the aryl substituent could be achieved by the further coupling reaction at the 3 position. Focused on the optical properties, a more efficient substituent effect at the 10 position was obtained compared with the analogous structure, diimidazo[1,2-a:2′,1′-c]quinoxaline derivatives. In addition, we revealed that the diimidazo[1,2-a:5′,1′-c]quinoxaline skeleton potentially has an electron-donating character on the optical properties by introducing an electron-withdrawing substituent.  相似文献
