A facile and eco‐friendly approach for synthesizing 9‐allyl‐10‐hydroxycamptothecin ( 2 ), a key intermediate for the preparation of camptothecin analogs, is described. The product was obtained in good yield and high purity (≥99% HPLC) under mild Suzuki reaction condition.  相似文献

A four‐step synthesis of 1‐substituted 5‐(2‐aminophenyl)‐1H‐pyrazoles 5 as a novel type of histamine analogs and versatile building blocks for further transformations was developed. The synthesis starts from commercially available 2‐nitroacetophenone ( 12 ), which is converted into the enamino ketone 13 as the key intermediate. Cyclization of the key intermediate 13 with monosubstituted hydrazines 14a – 14l afforded the 5‐(2‐nitrophenyl)‐1H‐pyrazoles 17a – 17l . Finally, catalytic hydrogenation of the nitro compounds 17a, 17c – 17e , and 17g – 17j furnished the title compounds 5a, 5c – 5e , and 5g – 5j , respectively, in good yields. As demonstrated by some further transformations, additional functionalization of compounds 17 and 5 is feasible, either by electrophilic substitution at C(4) of the pyrazole ring, or at the NH2 group.  相似文献

A new and convenient synthesis of 7‐(3‐chloropropoxy)‐4‐hydroxy‐6‐methoxyquinoline‐3‐carbonitrile, the key intermediate to bosutinib, is described on a hectogram scale. 5‐Bromo‐2‐methoxyphenol is adopted as the starting material via the simple chemical process including Friedel‐Crafts reaction, alkylation, bromination, cyano substitution, and so on to give the 3‐amino‐2‐(2‐bromobenzoyl)acrylonitrile compound 25 , which underwent key intramolecular cyclization at K2CO3/DMF condition; the title product was obtained in 36.9% yield over 7 steps and 98.71% purity (HPLC).  相似文献

Synthesis of 9‐[2‐(2‐hydroxymethyl‐2‐methyl‐, ‐(2‐acetoxymethyl‐2‐methyl‐, ‐(2,2‐di(hydroxymethyl)‐, and ‐(2,2‐di(acetoxymethyl)‐1,3‐dioxan‐5‐yl)ethyl] derivatives of guanine and 2‐aminopurine, 2–9 , has been accomplished in seven to eight step sequences from readily available 1‐(tert‐butyldiphenylsilyloxy)‐acetone, 1,3‐di(tert‐butyldiphenylsilyloxy)acetone, and the diol 10 . Formation of cyclic ketals 11 and 12 was carried out successfully under an acidic condition using a catalytic amount of methanesulfonic acid along with excess anhydrous copper(II) sulfate in toluene. Subsequent reactions of desilylation, acetylation, hydrogenolysis, and bromination afforded the key intermediates 19 and 20 , which were coupled with 2‐amino‐6‐chloropurine to produce the purine compounds 21 and 22 in good yields. Guanine derivatives 2–5 were obtained from 21 and 22 by hydrolysis and acetylation, while the dechlorination and hydrolysis of 21 and 22 yielded the 2‐aminopurine compounds 6–9 .  相似文献

A simple and concise approach for the synthesis of a series of new heterocyclic systems of 2‐(benzofuran‐2‐yl)benzo[h]quinoline‐3‐carboxylic acid derivatives ( 3a–3g ) is described. The synthetic strategy features the one‐pot reaction of ethyl 2‐(chloromethyl)benzo[h]quinoline‐3‐carboxylate ( 2 ) with various substituted salicylaldehydes as well as 2‐hydroxy‐1‐naphthaldehyde as a key step. The substrate 2 was prepared in good yield by a mild, efficient and direct reaction of 1‐naphthylamine ( 1 ) with Vilsmeier‐Haack reagent. The structures of all the new compounds were identified by spectral data and elemental analysis.  相似文献

A synthetic route to enantiomerically pure (1R,2S)‐1‐phenylphospholane‐2‐carboxylic acid ( 1 ), which is a phosphorus analogue of proline, has been established. A key step is the deprotonation–carboxylation of the 1‐phenylphospholane borane complex 3 by using sBuLi/1,2‐dipiperidinoethane (DPE). Configurational stability of the key intermediate, the amine‐coordinated α‐phosphinoalkyllithium borane complex 4 , was investigated by employing lithiodestannylation–carboxylation of both diastereomers of the 1‐phenyl‐2‐trimethylstannylphospholane borane complex 7 in the presence of several kinds of amines, and as a result, 4 was found to be configurationally labile even at ?100 °C. The key intermediate, the DPE‐coordinated trans‐1‐phenyl‐2‐phospholanyllithium borane complex 9 , was isolated, and the structure was identified by X‐ray crystal structure analysis. This is the first X‐ray crystal structure determined for an α‐monophosphinoalkyllithium borane complex. Remarkably, the alkyllithium complex is monomeric and tricoordinate at the lithium center with a slightly pyramidalized environment, and the existence of a Li? C bond (2.170 Å) has been confirmed. Moreover, 1H–7Li HOESY and 6Li NMR analyses suggested the structure of 9 in solution as well as the existence of an equilibrium between 9 , its cis isomer, and the ion pair 8 at room temperature, which was extremely biased towards 9 at ?100 °C. Finally, 1 was used as a chiral ligand in a palladium‐catalyzed allylic substitution, and the desired product was obtained in high yield with good enantioselectivity.  相似文献

Three 2,3‐bis(5‐hexylthiophen‐2‐yl)‐6,7‐bis(octyloxy)‐5,8‐di(thiophen‐2‐yl)‐quinoxaline ( BTTQ )‐based conjugated polymers, namely, PF‐BTTQ ( P1 ), PP‐BTTQ ( P2 ), and PDCP‐BTTQ ( P3 ), were successfully synthesized for efficient polymer solar cells (PSCs) with electron‐rich units of fluorene and dialkoxybenzene and electron‐deficient unit dicyanobenzene, respectively. All the polymers exhibited good solubility in common organic solvents and good thermal stability. Their deep‐lying HOMO energy levels enabled them good stability in the air and the relatively low HOMO energy level assured a higher open circuit potential when used in PSCs. Bulk‐heterojunction solar cells were fabricated using these copolymers blended with a fullerene derivative as an acceptor. All of them exhibited promising performance, and the best device performance with power conversion efficiency up to 3.30% was achieved under one sun of AM 1.5 solar simulator illumination (100 mW/cm2). © 2012 Wiley Periodicals, Inc. Tumor angiogenesis has become an important target for antitumor therapy, with most current therapies aimed at blocking the vascular endothelial growth factor (VEGF) pathway. The VEGF and its receptors have been implicated as key factors in tumor angiogenesis and are major targets in cancer therapy. A series of aryl‐modified benzoylamino‐N‐(5,6‐dimethoxy‐1H‐benzoimidazol‐2‐yl)‐heteroamides were synthesized from 2‐amino‐5,6‐dimethoxy benzimidazole and aryl‐substituted benzoylamino hetero acids. The new compounds were tested for inhibition of VEGF receptors I and II (VEGFR‐1 and VEGFR‐2). Compound 6e displayed VEGFR‐2 inhibitory activity with a 50% inhibition concentration value as low as 0.020 μM in a homogeneous time‐resolved fluorescence enzymatic assay. VEGFR‐2 active compounds display good activity against VEGFR‐1 as well.  相似文献

A green and efficient one‐pot two‐step synthesis of ethyl 2‐(alkylimino)‐4‐methyl‐3‐(alkanoyl)‐2,3‐dihydrothiazole‐5‐carboxylates from the reaction between acyl chlorides, ammonium thiocyanate, primary alkylamines, and ethyl 2‐chloroacetoacetae under mild, solvent‐ and catalyst‐free conditions at room temperature is presented. This efficient and straightforward technique gave the expected products in good to high yields in 2–4 hr without the creation of any by‐product in all reactions.  相似文献

An efficient method has been developed for the synthesis of N‐alkylated 2′‐oxo‐3‐phenylspiro[cyclopropane‐1,3′‐indoline]‐2,2‐dicarbonitrile from 3‐chloroindolin‐2‐one and 2‐benzylidenemalononitrile by using triethylamine as a base at room temperature and obtained the products in moderate to good yields. In extension, the scope of the reaction has been investigated by stepwise and one‐pot methods. Furthermore, in silico antibacterial activity was carried out in order to understand possible binding modes of novel derivatives with the active site of DNA gyrase A enzyme, and the results were well complemented. Additionally, absorption, distribution, metabolism, and excretion properties of compounds have shown drug likeness with good oral absorption and moderate blood–brain barrier permeability.  相似文献

A palladium‐catalyzed intramolecular α‐arylation of an amide in the presence of a bulky chiral N‐heterocyclic carbene ligand is the key step in the first catalytic synthesis of (3R)‐6‐chloro‐3‐(3‐chlorobenzyl)‐1,3‐dihydro‐3‐(3‐methoxyphenyl)‐2H‐indol‐2‐one ((R)‐ 5 ). This oxindole, in racemic form, had been shown previously to be an anticancer agent. (R)‐ 5 was obtained with an overall yield of 45% and with 96% enantioselectivity.  相似文献

A novel phosphine‐catalyzed [4+1] annulation of maleimides with 4,4‐dicyano‐2‐methylenebut‐3‐enoates has been developed to afford spirocyclic products, and the maleimides serves as C1 synthons. Moreover, a phosphine‐catalyzed formal [3+2] annulation between 4,4‐dicyano‐2‐methylenebut‐3‐enoates and maleic anhydride has been also achieved, and maleic anhydride behaved as a C3 synthon in the reaction, thus efficiently affording the functionalized cyclopentenones. A stable phosphinium‐containing zwitterionic compound is the key reactive intermediate in both annulations and was successfully isolated. Plausible mechanisms have been proposed on the basis of control experiments and deuterium‐labeling experiments.  相似文献

We report a practical and one‐pot synthesis of novel series of ω‐(oxathiolan‐2‐thione‐5‐yl)‐α‐ oxazolidin‐2‐ones ( 3a‐h ). The obtained compounds have been designed, synthesized via reaction of oligoethylene glycols diglycidyl ethers, isocyanate and carbon disulfide in the presence of catalytic amount of lithium bromide. A variety of important oxazolidinone derivatives can be obtained from simple starting materials in good yields and the biological activity of these new products will be investigated in complementary study.  相似文献

An easy, highly efficient and a new convenient one‐pot, two‐step approach to the synthesis of 3‐(3‐benzyl‐2‐(phenylimino)‐2,3‐dihydrothiazol‐4‐yl)‐6‐methyl‐4‐(2‐oxo‐2‐phenylethoxy)‐3,4‐dihydro‐2H‐pyran‐2‐one is described. These compounds were synthesized from 3‐(3‐benzyl‐2‐(phenylimino)‐2,3‐dihydrothiazol‐4‐yl)‐4‐hydroxy‐6‐methyl‐3,4‐dihydro‐2H‐pyran‐2‐one and α‐bromoketones in good yields. The compounds 4 were synthesized by a multi‐component reaction between 1 , 2 , and 3 and the prominent features of this protocol are mild reaction conditions, operation simplicity, and good to high yields of products.  相似文献

A highly enantiospecific, azide‐free synthesis of (?)‐(R)‐ and (+)‐(S)‐piperidin‐3‐ol in excellent yield was developed. The key step of the synthesis involves the enantiospecific ring openings of enantiomerically pure (R)‐ and (S)‐2‐(oxiran‐2‐ylmethyl)‐1H‐isoindole‐1,3(2H)‐diones with the diethyl malonate anion and subsequent decarboxylation.  相似文献

A series of 5‐aryl‐1‐arylaminomethyl‐1,3,5‐triazinane‐2‐thiones has been prepared in a one‐step procedure from condensation of readily available aromatic amines with thiourea and formaldehyde. A mechanism is presented to account for the formation of the products. A simple and efficient approach to synthesize a novel pyrrolocoumarin 9‐hydroxy‐8H‐pyrano[3,2‐f]indol‐2‐one ( 7 ) has been described. Starting from vanillin, the key intermediate 7‐methoxy‐1H‐indol‐6‐yl propiolate ( 6 ) was synthesized in six steps. Then, the target compound was obtained by forming pyrone‐ring and demethylation simultaneously in one step. A plausible mechanism invoking PtCl4 catalyzed one‐step reaction of cyclization and demethylation was also presented.  相似文献

An enantioselective iridium‐catalyzed allylic substitution with a set of highly unstabilized nucleophiles generated in situ from 2‐methylpyridines is described. Enantioenriched 2‐substituted pyridines, which are frequently encountered in natural products and pharmaceuticals, could be easily constructed by this simple method in good yields and excellent enantioselectivity. The synthetic utility of the pyridine products is demonstrated through the synthesis of a key intermediate of a reported Na+/H+ exchanger inhibitor and the total synthesis of (−)‐lycopladine A.  相似文献
