Direct solid sampling Zeeman atomic absorption spectrometric methods were developed and applied to the determination of mercury, cadmium, manganese, lead and tin in seafood. All elements but mercury were measured by a third generation Zeeman atomic absorption spectrometry combined with an automatic solid sampler. In 3-field- and dynamic mode the calibrations concentration range was substantially extended and high amounts of analyte were detectable without laborious dilution of solid samples. The measurements were based on calibrations using certified reference materials of organic matrices. In case solid certified reference materials were not available calibration by aqueous standard solutions was proved to be an alternative. No matrix effects were observed under the optimized conditions. Results obtained were in good agreement with the certified values. Solid sampling Zeeman atomic absorption spectrometry proved to be a reliable, rapid and low-cost method for the control of trace elements in seafood.  相似文献

A non-oxidative alkaline sample digestion procedure using tetramethylammonium hydroxide and a high pressure, microwave assisted autoclave digestion system was developed. The silicon concentrations of the digested samples were measured by inductively coupled axial plasma optical emission spectrometry (ICP-OES). Details of the digestion conditions as well as the optimised instrumental parameters for ICP-OES are described. The method was developed and tested using silicon-spiked ascorbic acid and applied to samples of animal tissue and organs. The total silicon content of two different reference materials, NIST 1577b Bovine liver and BCR 184 Bovine Muscle having neither certified nor informational values for Si was determined. The results obtained are compared with the results of independent methods such as wavelength dispersive x-ray fluorescence spectrometry (WDXRF) and solid sampling electrothermal atomic absorption spectrometry (ETAAS). The method described achieves a limit of detection of 2 mg kg(-1) using 100 mg of solid biological or organic material and covers a concentration range of up to 500 mg kg(-1).  相似文献

A microwave-digestion system with a closed PTFE vessel was used to improve the leaching of inorganic constituents from biological samples with tetramethylammonium hydroxide (TMAH) and ethylenediaminetetraacetic acid (EDTA). The effects of microwave parameter settings and the quantities of TMAH and EDTA used on leaching efficiency were evaluated. This new digestion method has been applied to the standard reference materials NIST SRM 1577 B Bovine Liver 1515 Apple Leaves and NIES CRM No. 1 Pepperbush, No. 3 Chorella, No. 6 Mussel and No. 7 Tea Leaves. The major and minor elements in the digests were analyzed by flame atomic absorption spectrometry or graphite furnace atomic absorption spectrometry. Good agreement of the analytical results with the certified values was obtained.  相似文献

Two procedures for the determination of Co in biological samples by graphite furnace atomic absorption spectrometry (GF AAS) were compared: solid sampling (SS) and alkaline treatment with tetramethylammonium hydroxide (TMAH) using two different instruments for the investigation: a conventional line-source (LS) atomic absorption spectrometer and a prototype high-resolution continuum source atomic absorption spectrometer. For the direct introduction of the solid samples, certified reference materials (CRM) were ground to a particle size ≤50 μm. Alkaline treatment was carried out by placing about 250 mg of the sample in polypropylene flasks, adding 2 mL of 25% m/v tetramethylammonium hydroxide and de-ionized water. Due to its unique capacity of providing a 3-D spectral plot, a high-resolution continuum source (HR-CS) graphite furnace atomic absorption spectrometry was used as a tool to evaluate potential spectral interferences, including background absorption for both sample introduction procedures, revealing that a continuous background preceded the atomic signal for pyrolysis temperatures lower than 700 °C. Molecular absorption bands with pronounced rotational fine structure appeared for atomization temperatures >1800 °C probably as a consequence of the formation of PO. After optimization had been carried out using high resolution continuum source atomic absorption spectrometry, the optimized conditions were adopted also for line-source atomic absorption spectrometry. Six biological certified reference materials were analyzed, with calibration against aqueous standards, resulting in agreement with the certified values (according to the t-test for a 95% confidence level) and in detection limits as low as 5 ng g−1.  相似文献

The determination of trace elements in food and soil samples by atomic absorption spectrometry was investigated. A coprecipitation procedure with holmium hydroxide was used for separation-preconcentration of trace elements. Trace amounts of copper(II), manganese(II), cobalt(II), nickel(ll), chromium(lll), iron(Ill), cadmium(ll), and lead(ll) ions were coprecipitated with holmium hydroxide in 2.0 M NaOH medium. The optimum conditions for the coprecipitation process were investigated for several commonly tested experimental parameters, such as amount of coprecipitant, effect of standing time, centrifugation rate and time, and sample volume. The precision, based on replicate analysis, was lower than 10% for the analytes. In order to verify the accuracy of the method, the certified reference materials BCR 141 R calcareous loam soil and CRM 025-050 soil were analyzed. The procedure was successfully applied for separation and preconcentration of the investigated ions in various food and soil samples. An amount of the solid samples was decomposed with 15 mL concentrated hydrochloric acid-concentrated nitric acid (3 + 1). The preconcentration procedure was then applied to the final solutions. The concentration of trace elements in samples was determined by atomic absorption spectrometry.  相似文献

Methods have been developed for the preparation of samples containing organic matrices (natural high-color waters, soils, bottom sediments, aquatic organism tissue) to the determination of mercury and other toxic elements (As, Cd, Pb, Se) by different procedures of microwave digestion under elevated pressure (closed systems, vessels with partial removal of the gas phase). It is found that, under optimal oxidative and temperature-time conditions, the partial removal of the gas phase does not lead to losses of volatile elements if sample portions under 2 g are used. The duration of preparation of a series of samples does not exceed 40 min. The detection limits for mercury in the cold vapor atomic absorption spectrometry and for Cd, Pb, As, and Se by electrothermal atomic absorption spectrometry are 5 and 0.13, 6, 13, and 13 μg/kg, respectively. The accuracy of determination is confirmed by the results of analysis of certified reference materials of water and plant materials and also by the standard addition method. The selected conditions of preparation of sludge samples have ensured the determination of mercury by the cold vapor atomic absorption spectrometry in drinking, natural, and sewage waters with a detection limit of 0.07 μg/L.  相似文献

A solid sampling flame furnace atomic absorption spectrometry (SS-FF-AAS) system was developed for Mn and Ni determination in petroleum coke. The proposed system for solid sampling was a quartz cell with two perpendicular tubes (T-shaped tubes), positioned above the burner. Manganese and Ni determination was made using an atomic absorption spectrometer with deuterium background corrector, air-acetylene flame and a single slit burner. Powdered samples of coke were introduced as pellets (up to 62 mg) into the quartz cell with a movable hollow quartz piston. When the sample pellet reached the end of quartz cell (T-connection), in the presence of a constant oxygen flow, it quickly burned and the combustion products were transferred to the upper slit tube where the atomic absorption process occurs. Calibration was possible using aqueous reference solutions applied directly on high purity graphite pellets. Results obtained for Mn and Ni using the proposed SS-FF-AAS system were compared to those obtained by inductively coupled plasma optical emission spectrometry (ICP OES) and inductively coupled plasma mass spectrometry (ICP-MS) after sample decomposition by high pressure microwave assisted acid digestion and also by microwave induced combustion. Agreement better than 96% was obtained for both methods employing a previous step of sample digestion (ICP OES and ICP-MS) and by SS-FF-AAS. Accuracy was evaluated using certified reference materials and also recovery tests. The relative standard deviation was lower than 9% for both analytes. The characteristic mass was 18.3 and 14.7 ng and the limit of detection was 0.6 and 0.8 µg g− 1 for Mn and Ni, respectively. The proposed SS-FF-AAS system can be applied for Mn and Ni determination in petroleum coke, combining a relatively high sample throughput (9 determinations per h), and a suitable precision and accuracy.  相似文献

The literature about direct solid sample analysis of the past 10–15 years using electrothermal atomic absorption spectrometry has been reviewed. It was found that in the vast majority of publications aqueous standards were reported as having been used for calibration after careful program optimization. This means the frequently expressed claim that certified reference materials with a matrix composition and analyte content close to that of the sample have to be used for calibration in solid sample analysis is not confirmed in the more recent literature. There are obviously limitations, and there are examples in the literature where even calibration with certified reference materials did not lead to accurate results. In these cases the problem is typically associated with spectral interferences that cannot be corrected properly by the systems available for conventional line source atomic absorption spectrometry, including Zeeman-effect background correction. Using high-resolution continuum source atomic absorption spectrometry, spectral interferences become visible owing to the display of the spectral environment at both sides of the analytical line at high resolution, which makes program optimization straightforward. Any spectrally continuous background absorption is eliminated automatically, and even rapidly changing background absorption does not cause any artifacts, as measurement and correction of background absorption are truly simultaneous. Any kind of fine-structured background can be eliminated by “subtracting” reference spectra using a least-squares algorithm. Aqueous standards are used for calibration in all published applications of high-resolution continuum source atomic absorption spectrometry to direct solid sample analysis. Two types of sediment reference material (NMIJ 7302-a and 7303-a) for trace elements analysis have been prepared and certified by the National Metrology Institute of Japan in the National Institute of Advanced Industrial Science and Technology (NMIJ/AIST). The original materials were collected from a bay near industrial activity in Kyushu (NMIJ CRM 7302-a; marine sediment) and from Lake Biwa (NMIJ CRM 7303-a; lake sediment). The sediment materials were air-dried, sieved, homogenized, packaged in 1000 glass bottles (60 g each), and radiation sterilized. Certification of these CRM for trace elements was conducted by NMIJ, where each element was determined by at least two independent analytical techniques. Isotope-dilution inductively coupled plasma mass spectrometry (ICP–MS) was applied for certification of all the elements except mono-nuclide elements such as As and Co. Other techniques such as ICP–MS with quadrupole mass spectrometry and sector-field mass spectrometry, inductively coupled plasma atomic emission spectrometry (ICP–AES), and atomic absorption spectrometry (AAS), were also used. Certified values have been provided for 14 elements (Sb, As, Cd, Cr, Co, Cu, Pb, Hg, Mo, Ni, Se, Ag, Sn, and Zn) in both CRM.  相似文献

Direct analysis of solid samples employing a laboratory assembled electrothermal atomic absorption spectrometer is demonstrated to be a feasible approach for determination of trace elements in plant tissue and hair samples for special applications in plant physiology and biomedical research. As an example, the kinetics of Cr uptake by cabbage and its distribution have been measured as a function of chromium speciation in the nutrient solution. Further, longitudinal concentration gradients of Cr, Pb and Cd have been measured in hair of various population groups exposed to different levels of these elements in ambient and/or occupational environments. The techniques are validated for the determination of these trace elements by neutron activation analysis, dissolution atomic absorption spectrometry and by analysis of certified reference materials. Slurry sample introduction is found appropriate for routine trace element determination and in homogeneity testing. Direct sample introduction is indispensable in the analysis of very small (< 1 mg) tissue biopsy samples in the determination of trace element distributions.  相似文献

Trace amounts of germanium can be determined by atomic spectrometry by utilizing the vaporization of germanium tetrachloride at ambient temperature. Using an intermittent or continuous flow reactor, the sample solution was mixed with concentrated hydrochloric acid to form volatile germanium tetrachloride which can subsequently be determined by atomic spectrometry. The conditions for the volatilization of germanium chloride were investigated in detail and rapid method for the determination of trace amounts of germanium in real samples was proposed. A detection limit of 0.5 ng ml−1 (3σn−1) was obtained by using atomic fluorescence spectrometric detection and the precision found was 0.8% for a germanium concentration of l00 ng ml−1. Atomic emission and absorption spectrometric methods were also tested. Owing to the high selectivity of the reaction, no interference was found in the determination. The method was applied to the determination of germanium in several standard and certified reference materials; the results obtained were in good agreement with the certified values.  相似文献

The presence of some essential and toxic metals in fat supplements for swine diet was investigated. Collected samples represented a relevant production of the Italian industry. In particular, some samples were enriched with antioxidants or waste cooking oils. The method for the determination of Ca, Cu, Cd, Fe, Mg, Mn, Ni, Pb, and Zn in fat samples was developed by means of a certified reference material (CRM 186) and a representative fatty sample (RFS). All samples were digested in closed vessels in a microwave oven and then analyzed by flame atomic absorption or graphite furnace atomic absorption spectrometry. The entire analytical method provided a satisfactory repeatability and reproducibility confirmed by agreement between the experimental recovery data obtained for the CRM 186 sample and, with the method of standard additions, for the RFS material. The samples generally showed a small amount of metals compared with the recommended daily intake for the essential elements. On the other hand, some samples contained a significant concentration, from an analytical point of view, of Cd, Ni, and Pb. Principal component analysis (PCA) was applied to inspect the experimental data obtained from samples analysis. Basically no differences were detected in terms of metal concentration among the fat supplements analyzed.  相似文献

Slurry-sampling electrothermal atomic absorption spectrometry has been applied to the determination of lead in several biological samples (fish and marine algae). The slurries were prepared both by magnetic shaking and microwave-heating and the effect of instrument operating conditions and slurry preparation conditions on the signal were examined. In addition, results from slurry sampling were compared with those obtained by microwave-assisted acid digestion of the same samples and no significant differences were found between them when the analysis of variance (ANOVA) was applied. The between-batch precision of the slurry techniques employed was similar to that for the microwave-assisted digestion procedure; values were always below 6.7%, except for the Dicentrarchus labrax sample for which the value obtained was 9.5% when using slurry magnetic shaking and 7.6% when using the slurry microwave heating. The accuracy of the slurry methodology employed was also evaluated by analysis of two biological reference materials (NIST-1577b and IAEA-V10); percentage recoveries obtained were between 95.6 and 98.5% of the values certified for lead.  相似文献

Spectral interferences from phosphorus on the determination of selenium in biological tissue materials were not observed when a Zeeman-effect background correction was used with rhodium as a chemical modifier. A suppression effect on the selenium signal resulted when the concentration of phosphorus present was greater than 1.0 mg ml−1. Rhodium was found to be more effective than palladium in overcoming the phosphate interference. Analytical procedures for the direct determination of trace selenium in standard reference materials by graphite furnace atomic absorption spectrometry following sample dissolution in nitric acid and hydrogen peroxide using a microwave oven has been described. The results obtained agreed favourably with the certified values.  相似文献

The comparative determination of lead in plant samples by two atomic spectrometric techniques is reported. At first, slurry sampling electrothermal atomisation atomic absorption spectrometry (ETAAS) was applied. The results obtained were compared with those found after a wet digestion procedure by flame atomic absorption spectrometry (FAAS) or ETAAS. The accuracy of the studied methods was checked using a certified reference material (CL???1 CRM, Cabbage Leaves). The recovery of lead was 90% for slurry sampling ETAAS, and 86.6% for liquid sampling ETAAS. The advantages of the slurry sampling ETAAS method are the simplicity of sample preparation and very good sensitivity.  相似文献
