The determination of target molecules in complicated matrices such as biological samples is largely dependent on sample pretreatment. Molecularly imprinted solid-phase extraction (SPE), using molecularly imprinted polymers as the adsorbent, has been demonstrated to be effective for the selective enrichment of target molecules in biological samples. In this study, molecularly imprinted polymeric microspheres were fabricated by two-step swelling polymerization using polystyrene particles as seeds, nicotinamide as the template, methacrylic acid as a functional monomer, and ethylene glycol dimethacrylate as a cross-linker. The molecularly imprinted polymeric microspheres were packed into empty SPE cartridges, and the spiked urine and serum samples were loaded separately. After an initial washing and elution step, the effluents were analyzed by high-performance liquid chromatography (HPLC) using 1:9 methanol/0.05% phosphoric acid. The obtained molecularly imprinted polymeric microspheres were uniform, and the spherical particles were well distributed. The established method was validated, and the results showed that the method was linear from 0.499 to 19.96?µg?mL?1. The limits of detection and quantification for nicotinamide were 0.3 and 0.9?µg?mL?1, respectively. The relative standard deviations were 1.55 and 2.86% in urine and serum, respectively. The spiked recoveries of nicotinamide were 86.0–98.8% and 87.0–96.8% in urine and serum, respectively. The molecularly imprinted SPE and HPLC methods in this study are useful for the pretreatment and determination of the target compounds in these matrices.  相似文献

In this work, performance of a molecularly imprinted polymer (MIP) as a selective solid‐phase microextraction sorbent for the extraction and enrichment of tramadol in aqueous solution and rabbit brain tissue, is described. Binding properties of MIPs were studied in comparison with their nonimprinted polymer (NIP). Ten milligrams of the optimized MIP was then evaluated as a sorbent, for preconcentration, in molecularly imprinted solid‐phase microextraction (MISPME) of tramadol from aqueous solution and rabbit brain tissue. The analytical method was calibrated in the range of 0.004 ppm (4 ng mL−1) and 10 ppm (10 μg mL−1) in aqueous media and in the ranges of 0.01 and 10 ppm in rabbit brain tissue, respectively. The results indicated significantly higher binding affinity of MIPs to tramadol, in comparison with NIP. The MISPME procedure was developed and optimized with a recovery of 81.12–107.54% in aqueous solution and 76.16–91.20% in rabbit brain tissue. The inter‐ and intra‐day variation values were <8.24 and 5.06%, respectively. Finally the calibrated method was applied for determination of tramadol in real rabbit brain tissue samples after administration of a lethal dose. Our data demonstrated the potential of MISPME for rapid, sensitive and cost‐effective sample analysis.  相似文献

Different kinds of deep eutectic solvents based on choline chloride and ionic liquids based on 1‐methylimidazole were used to modify hybrid molecularly imprinted polymers with the monomer γ‐aminopropyltriethoxysilane‐methacrylic and three templates (rutin, scoparone, and quercetin). The materials were adopted as solid‐phase extraction packing agents, and were characterized by FTIR spectroscopy and field emission scanning electron microscopy. The hybrid molecularly imprinted polymers modified by deep eutectic solvents had high recoveries and a strong recognition of rutin, scoparone, and quercetin in Herba Artemisiae Scopariae than those modified by ionic liquids. In the procedure of solid‐phase extraction, deep eutectic solvents‐2‐hybrid molecularly imprinted polymers were obtained with the best recoveries with rutin (92.27%), scoparone (87.51%), and quercetin (80.02%), and the actual extraction yields of rutin (5.6 mg/g), scoparone (2.3 mg/g), and quercetin (3.4 mg/g). Overall, the proposed approach with the high affinity of hybrid molecularly imprinted polymers might offer a novel method for the purification of complex samples.  相似文献

A novel molecularly imprinted sol-gel material based on polysiloxane nanofiber was introduced as a solid-phase microextraction coating on a stainless steel wire for the extraction of simazine. The nanostructured molecularly imprinted fiber was prepared by a simple single step method at room temperature, using methyltriethoxysilane as the sol-gel precursor and simazine as the template molecule. The fiber was applied for the extraction of simazine in different water samples followed by gas chromatography and mass spectrometry detection. The extraction capacity of the molecularly imprinted fiber was 8 ng, and it had better extraction efficiency than the non-imprinted fiber and commercial fibers (PDMS and PA). The fiber had also a good selectivity for simazine and its analogous compounds. Important parameters affecting the extraction and desorption efficiency, such as salt concentration, stirring rate, pH of sample solution, extraction time and temperature, temperature and time of desorption, were investigated. The intra- and inter-day relative standard deviations were in the range of 4.3–7.6%. The fiber-to-fiber reproducibility was 7.7–8.5%. The method showed a good linearity (r2> 0.9980) in the range of 0.02–20 μg L−1 with the detection limit of 0.005 μg L−1. The relative recoveries were also in the range of 94–97% for different water samples.  相似文献

This research aims to synthesize a specific and efficient sorbent to use in the extraction of apixaban from human plasma samples and its determination by high-performance liquid chromatography-tandem mass spectrometry. High specific surface area of metal-organic framework, magnetic property of iron oxide nanoparticles, selectively of molecular imprinted polymer toward the analyte, and the combination of dispersive solid-phase extraction method with a sensitive analysis system provided an efficient analytical method. In this study, first, a molecularly imprinted polymer combined with magnetic metal organic framework nanocomposite was prepared and then characterized using different techniques. Then the sorbent particles were used for selective extraction of the analyte from plasma samples. The efficiency of the method was improved by optimizing effective parameters. According to the validation results, wide linear range (1.02–200 ng mL−1), acceptable coefficient of determination (0.9938), low limit of detection (0.32 ng mL−1) and limit of quantification (1.02 ng mL−1), high extraction recovery (78%), and good precision (relative standard deviations ≤ 2.9% for intra- (n = 6) and interday (n = 6) precisions) were obtainable using the proposed method. These outcomes showed the high potential of the proposed method for screening apixaban in the human plasma samples.  相似文献

Molecularly imprinted polymers grafted on to the surface of carboxyl-modified multiwalled carbon nanotubes were developed using methacrylic acid as a functional monomer and trihydroxymethylpropyl trimethylacrylate as a crosslinker for application to rhodamine B determination. The synthesis, characteristics, and evaluation of the molecularly imprinted polymer are described. The apparent morphology of the polymers was characterized by scanning electron microscopy. To evaluate the binding ability of the molecularly imprinted polymers, equilibrium binding experiments were conducted and revealed the maximal binding capacity to be 561.54 µg g?1. The introduction of nanomaterials into the polymer composite made important contributions to the affinity enhancement and recognition properties of the molecularly imprinted polymers. Moreover, the polymers were preliminarily applied as an adsorbent for separation and extraction of Rhodamine B from dried chili powder samples, based on solid phase extraction technology. The calibration curve was linear in the range of 0–7 µg mL?1. For all tested samples, recoveries were reliable and in the range of 101.75–109.73%. The relative standard deviation ranged from 6.43–14.32%, which demonstrated that the polymer has potential for preconcentration of Rhodamine B from chili powder samples.  相似文献

In this paper, a novel pipette tip micro-solid phase extraction based on molecularly imprinted polymer as a selective sorbent was developed and applied for extraction, pre-concentration and high-performance liquid chromatographic determination of trace amounts of malachite green (MG), rhodamine B (RB), methyl orange (MO) and acid red 18 (AR) dyes in seawater samples. Different parameters affecting the extraction efficiency such as type and volume of eluent solvent, sample volume, number of cycles of extraction and desorption, amount of sorbent and pH of the sample solution were evaluated using one-variable-at-a-time and response surface methodology. In order to optimize dyes extraction, seven factors in three levels were used for Box–Behnken experimental design. Under optimum extraction condition, pH of sample solution was 3.1 for MG, 3.0 for RB, 7.1 for MO and 6.1 for AR; volume of eluent solvent (HCl, 0.5 mol L−1) was 200 µL; volume of the sample solution was 10 mL (for MG) and 4 mL (for RB, MO and AR); the concentration of triton X-114 was 0.085 (MG), 0.10 (RB), 0.08 (MO) and 0.075 (AR) % v/v; the number of extraction cycles was 10 (MG), 6 (RB), 5 (MO) and 7 (AR); the number of elution cycles was 10, 6, 5 and 9, respectively, for MG, RB, MO and AR; NaCl concentration was 0.4 mol L−1; and amount of MIP was 2.0 mg for all dyes. The linear range of calibration curves was 0.5–250.0 µg L−1 for malachite green and methyl orange and 0.5–150.0 µg L−1 for both rhodamine B and acid red 18. The detection limits calculated to be 0.083, 0.10, 0.12 and 0.17 µg L−1 for MG, RB, MO and AR, respectively. The developed protocol was successfully used for determination of dyes in seawater of Chabahar Bay. The mean recoveries were ranged between 76.1 and 97.3% by mean relative standard deviations of 1.2–7.1%.

The quercetin molecularly imprinted polymer microspheres were prepared by reversible addition-fragmentation chain transfer free radical polymerization combined with precipitation method (RAFTPP) with the dibenzyl trithiocarbonate (DBTTC) as RAFT reagent and quercetin as template molecule. The effects of solvents and cross-linking agent on the morphology and size of polymers were investigated, and the polymers were preliminarily screened by scanning electron microscope. The adsorption experiment and adsorption model analysis were carried out on the molecularly imprinted polymers (R-MIP) prepared under optimal condition and on the molecularly imprinted polymers prepared with a traditional precipitation polymerization (T-MIP). Consequently it was determined that, QR-MIP max was 37.71 mg g?1 and QT-MIP max was 29.61 mg g?1. The results showed that the R-MIP had a good adsorptive property and was obviously superior to T-MIP. R-MIP was used as solid-phase extraction filler in combination with the HPLC method to conduct enrichment, separation and measurement of the quercetin in honeysuckle and clove leaves, which effectively removed the matrix interference and the recovery rate of this method was 93.8–102.9%.

As local anesthetics that are illegally added to cosmetics are harmful to consumer health, it is necessary to establish an efficient method for detecting these substances. Herein, a molecularly imprinted polymer (bupivacaine) was prepared by bulk polymerization and packed into a hollow fiber for use as an extraction phase to fabricate a membrane-protected micro-solid-phase extraction device. The optimal values of the influencing parameters for the microextraction process were as follows: a sample solution pH of 9.0, a loading and washing time of 2 h, and an elution time of 32 min. A gas chromatography-mass spectrometry method was established for the determination of local anesthetics and coupled with the microextraction method to successfully detect local anesthetics in cosmetic samples. The calibration curve for the proposed method was linear in the range of 0.4–50 mg/L and showed a good correlation coefficient (r2). The limits of detection for local anesthetics were in the range of 0.01–0.71 mg/L. The molecularly imprinted polymer exhibited good imprinting and selectivity, the micro-solid-phase extraction device was simple and inexpensive and fabrication was reproducible. The combination of molecular imprinting technology, membrane separation, and micro-solid-phase extraction methods used in this study can potentially be applied to pretreat local anesthetics in cosmetic samples.  相似文献

The aim of this work was to develop a method for the clean‐up and preconcentration of warfarin from biological sample employing a new molecularly imprinted polymer (MIP) as a selective adsorbent for solid‐phase extraction (SPE). This MIP was synthesized using warfarin as a template, pyrrole as a functional monomer and vinyl triethoxysilane as a cross‐linker. The molar ratio of 1:4:20 (template–functional monomer–cross‐linker) showed the best results. Nonimprinted polymers (NIPs) were prepared and treated with the same method, but in the absence of warfarin. The prepared polymer was characterized by Fourier transmission infrared spectrometry and scanning electron microscopy. An adsorption process (SPE) for the removal of warfarin using the fabricated MIPs and NIPs was evaluated under various conditions. Effective parameters on warfarin extraction, for example, type and volume of elution solvent, pH of sample solution, breakthrough volume and maximum loading capacity, were studied. The limits of detection were in the range of 0.0035–0.0050 µg mL?1. Linearity of the method was determined in the range of 0.0165–10.0000 µg mL?1 for plasma and 0.0115–10.0000 µg mL?1 for urine with coefficients of determination (R2) ranging from 0.9975 to 0.9985. As signal molecules, auxins play an important role in mediating plant growth. Due to serious interfering substances in plants, it is difficult to accurately detect auxins with traditional solid‐phase extraction methods. To improve the selectivity of sample pretreatment, a novel molecularly imprinted polymer ‐coated solid‐phase microextraction fiber, which could be coupled directly to high‐performance liquid chromatography, was prepared with indole acetic acid as template molecule for the selective extraction of auxins. The factors influencing the polymer formation, such as polymerization solvent, cross‐linker, and polymerization time, were investigated in detail to enhance the performance of indole acetic acid‐molecularly imprinted polymer coating. The morphological and chemical stability of this molecularly imprinted polymer‐coated fiber was characterized by scanning electron microscopy, infrared spectrometry, and thermal analysis. The extraction capacity of the molecularly imprinted polymer‐coated solid‐phase microextraction fiber was evaluated for the selective extraction of indole acetic acid and indole‐3‐pyruvic acid followed by high‐performance liquid chromatography analysis. The linear range for indole acetic acid and indole‐3‐pyruvic acid was 1–100 µg/L and their detection limit was 0.5 µg/L. The method was applied to the simultaneous determination of two auxins in two kinds of tobacco (Nicotiana tabacum L and Nicotiana rustica L) samples, with recoveries range from 82.1 to 120.6%.  相似文献

A liquid‐phase microextraction method that uses a hollow‐fiber solvent bar microextraction technique was developed by combining gas chromatography with electron capture detection for the analysis of four trihalomethanes (chloroform, dichlorobromomethane, chlorodibromomethane, and bromoform) in drinking water. In the microextraction process, 1‐octanol was used as the solvent. The technique operates in a two‐phase mode with a 5 min extraction time, a 700 rpm stirring speed, a 30°C extraction temperature, and NaCl concentration of 20%. After microextraction, one edge of the membrane was cut, and 1 μL of solvent was collected from the membrane using a 10 μL syringe. The solvent sample was directly injected into the gas chromatograph. The analytical characteristics of the developed method were as follows: detection limits, 0.017–0.037 ng mL−1; linear working range, 10–900 ng mL−1; recovery, 74 ± 9–91 ± 2; relative standard deviation, 5.7–10.3; and enrichment factor, 330–455. A simple, fast, economic, selective, and efficient method with big possibilities for automation was developed with a potential use to apply with other matrices and analytes.  相似文献

In this study, a highly sensitive and selective sample pretreatment procedure using molecularly imprinted silica nanoparticles was developed for the extraction and determination of quercetin in red wine samples coupled with high‐performance liquid chromatography with ultraviolet detection. The imprinted silica nanoparticles were prepared in the presence of N‐acryoyl‐l ‐aspartic acid (functional monomer), quercetin (template), azobisisobutyronitrile (initiator) and methylene bisacrylamide (cross‐linker) and methanol/water (porogen) via surface‐initiated reversible addition‐fragmentation chain transfer polymerization. Surface characterization was performed and several imprinting parameters were investigated, and the results indicated that adsorption of quercetin on the imprinted silica nanoparticles followed a pseudo‐first‐order adsorption isotherm with a maximum adsorption capacity at 26.4 mg/g within 60 min. The imprinted silica nanoparticles also showed satisfactory selectivity towards quercetin as compared with its structural analogues. Moreover, the imprinted nanoparticles preserved their recognition ability even after five adsorption–desorption cycles. Meanwhile, the nanoparticles were successfully applied to selective extraction of quercetin from red wine with a high recovery (99.7–100.4%). The limit of detection was calculated to be 0.058 μg/mL with a correlation coefficient 0.9996 in the range of 0.2–50 μg/mL. As a result, the developed selective extraction method using molecular imprinting technology simplifies the sample pretreatment procedure before determination of quercetin in real samples.  相似文献

We present a novel electrochemical sensor based on an electrode modified with molecularly imprinted polymers for the detection of chlorpyrifos. The modified electrode was constructed by the synthesis of molecularly imprinted polymers by a precipitation method then coated on a glassy carbon electrode. The surface morphology of the modified electrode was characterized by using field‐emission scanning electron microscopy and transmission electron microscopy. The performance of the imprinted sensor was thoroughly investigated by using cyclic voltammetry and differential pulse voltammetry. The imprinted electrochemical sensor displayed high repeatability, stability, and selectivity towards the template molecules. Under the optimal experimental conditions, the peak current response of the imprinted electrochemical sensor was linearly related to the concentration of chlorpyrifos over the range 1 × 10−10–1 × 10−5 mol/L with a limit of detection of 4.08 × 10−9 mol/L (signal‐to‐noise ratio = 3). Furthermore, the proposed molecularly imprinted electrochemical sensor was applied to the determination of chlorpyrifos in the complicated matrixes of real samples with satisfactory results. Therefore, the molecularly imprinted polymers based electrochemical sensor might provide a highly selective, rapid, and cost‐effective method for chlorpyrifos determination and related analysis.  相似文献

A simple method based on matrix solid-phase dispersion for selective extraction of anthraquinones from rhubarb samples was developed using a molecularly imprinted polymer as sorbent. The molecularly imprinted polymer was prepared using emodin as the template molecule, methacrylic acid as the functional monomer, and ethylene glycol dimethacrylate as the cross-linking agent. The polymer was characterized by scanning electron microscopy and Fourier-transform infrared spectrometry. Isothermal adsorption and dynamic adsorption experiments were performed. The best extraction conditions for anthraquinones were obtained at a ratio of molecularly imprinted polymer to sample of 1:1, a dispersion time of 5 minutes, with 5% aqueous methanol as the washing solvent, and an elution solvent of methanol-acetic acid (99:1, v/v). Once the matrix solid-phase dispersion process was optimized, the extract was reacted with 8% hydrochloric acid for hydrolysis. The anthraquinones extracted from rhubarb were determined by liquid chromatography. The detection limits of chrysophanol, emodin, physcion, and aloe-emodin were 0.23, 0.24, 0.28, and 0.27 µg mL?1, respectively. The proposed method was compared with the method in Chinese pharmacopoeia, and the results show that the extraction yield of anthraquinones obtained by molecularly imprinted polymer–matrix solid-phase dispersion method was higher. Moreover, the proposed method is faster and simpler and can achieve extraction and purification in the same system.  相似文献

Methyltriphenylphosphonium bromide/chalcone/formic acid, a green ternary deep eutectic solvent, was applied as a functional monomer and dummy template simultaneously in the synthesis of a new molecularly imprinted polymer. Ternary deep eutectic solvent based molecularly imprinted polymers are used as a solid‐phase extraction sorbent in the separation and purification of rutin and quercetin from Herba Artemisiae Scopariae combined with high‐performance liquid chromatography. Fourier transform infrared spectroscopy and field‐emission scanning electron microscopy were applied to characterize the deep eutectic solvent based molecularly imprinted polymers synthesized using different molar ratios of chalcone. The static and competitive adsorption tests were performed to examine the recognition ability of the molecularly imprinted polymers to rutin and quercetin. The ternary deep eutectic solvent consisting of formic acid/chalcone/methyltriphenylphosphonium bromide (1:0.05:0.5) had the best molecular recognition effect. After optimization of the washing solvents (methanol/water, 1:9) and eluting solvents (acetonitrile/acetic acid, 9:1), a reliable analytical method was developed for strong recognition towards rutin and quercetin in Herba Artemisiae Scopariae with satisfactory extraction recoveries (rutin: 92.48%, quercetin: 94.23%). Overall, the chalcone ternary deep eutectic solvent‐based molecularly imprinted polymer coupled with solid‐phase extraction is an effective method for the selective purification of multiple bioactive compounds in complex samples.  相似文献

A simple and sensitive method for the simultaneous determination of six fluoroquinolones from serum samples was developed by selective molecularly imprinted matrix solid-phase dispersion (MI-MSPD) coupled with chromatographic separation. By using ethylene glycol dimethacrylate as crosslinker and reformative methanol–water system as reaction medium, the improved water-compatible imprinted polymers were synthesized which show higher affinity to template and its analogues in aqueous environment. The molecularly imprinted polymers (MIPs) were applied as the selective dispersant of matrix solid-phase dispersion (MSPD) could selectively extracted the six fluoroquinolones from serum, while interferences originated from serum matrices were eliminated simultaneously. Good linearity was obtained in a range of 0.05–100 μg mL−1 with the correlation coefficients >0.999. The average recoveries of the six fluoroquinolones at four different spiked levels (0.25–10 μg mL−1) were ranged from 72.2% to 114.1% with the relative standard deviations less than 6.6%. This method is simple and sensitive, and can be used as an alternative tool to the existing HPLC methods for analyzing the residues of fluoroquinolones in biological samples.  相似文献

Ordered macroporous molecularly imprinted polymers were prepared by a combination of the colloidal crystal templating method and the molecular imprinting technique by using SiO2 colloidal crystal as the macroporogen, quercetin as the imprinting template, acrylamide as the functional monomer, ethylene glycol dimethacrylate as the cross‐linker and tetrahydrofuran as the solvent. Scanning electron microscopy and Brunauer–Emmett–Teller measurements show that the ordered macroporous molecularly imprinted polymers have a more regular macroporous structure, a narrower pore distribution and a greater porosity compared with the traditional bulk molecularly imprinted polymers. The kinetic and isothermal adsorption behaviors of the polymers were investigated. The results indicate that the ordered macroporous molecularly imprinted polymers have a faster intraparticle mass transfer process and a higher adsorption capacity than the traditional bulk molecularly imprinted polymers. The ordered macroporous molecularly imprinted polymers were further employed as a sorbent for a solid‐phase extraction. The results show that the ordered macroporous molecularly imprinted polymers can effectively separate quercetin from the Gingko hydrolysate.  相似文献

An environmentally benign method of sample preparation based on dispersive liquid–liquid microextraction and solidification of floating organic droplets (DLLME-SFO) coupled with high-performance liquid chromatography with ultraviolet detection has been developed for analysis of non-steroidal anti-inflammatory drugs (NSAIDs) in biological fluids. A low-toxicity solvent was used to replace the chlorinated solvents commonly used in conventional DLLME. Seven conditions were investigated and optimized: type and volume of extraction solvent and dispersive solvent, extraction time, effect of addition of salt, and sample pH. Under the optimum conditions, good linearity was obtained in the range 0.01–10 µg mL−1, with coefficients of determination (r 2) >0.9949. Detection limits were in the range 0.0034–0.0052 µg mL−1 with good reproducibility (RSD) and satisfactory inter-day and intra-day recovery (95.7–115.6 %). The method was successfully used for analysis of diclofenac, mefenamic acid, and ketoprofen in human urine. Analysis of urine samples from a patient 2 and 4 h after administration of diclofenac revealed concentrations of 1.20 and 0.34 µg mL−1, respectively.
