葛根多糖具有抗氧化、抗肿瘤等众多生物活性,对葛根多糖进行单糖组成分析对其活性研究具有重要意义。该研究利用响应面分析法考察了超声辅助提取法中液料比、超声温度、超声时间和超声功率对葛根多糖提取率的交互影响,并拟合数据得到多元二次回归方程。同时建立了柱前衍生加压毛细管电色谱检测糖类的方法,对分离8种中性单糖的色谱条件进行了探索与优化,并将此方法应用于两种葛根实际样品的单糖组成测定。响应面分析结果表明,4个试验因素中,超声温度对两种葛根多糖提取率的影响程度最大,其次为液料比,超声时间和超声功率影响程度较小。结合软件预测分析得到的最佳条件及设备实际情况,确定葛根多糖的最佳提取工艺条件为:超声温度90℃,粉葛多糖液料比20 mL/g,柴葛多糖液料比40 mL/g,超声时间30 min,超声功率180 W。优化后的色谱分离条件为:采用Halo-2.7 μm核壳型C18填料毛细管色谱柱,以乙腈-50 mmol/L pH 4.1的醋酸铵水溶液(18:82,v/v)为流动相,在250 nm波长下检测,施加电压-20 kV。在此条件下可以实现24 min内对葡萄糖等8种中性单糖衍生物的快速分离,相比传统液相色谱方法大大提升了分离检测速度和分离柱效。方法学考察表明此方法具有较好的线性关系和良好的重复性。对实际样品分离鉴定表明,粉葛多糖主要由葡萄糖、甘露糖、鼠李糖和岩藻糖组成,4种单糖物质的量之比为1.00:0.16:0.14:0.07;柴葛多糖主要由葡萄糖和甘露糖组成,2种单糖物质的量之比为1.00:0.70。该研究为单糖化合物快速高效分离检测提供了新方法,并为葛根多糖单糖组成分析提供了参考。 相似文献

采用1-苯基-3-甲基-5-吡唑啉酮(PMP)柱前衍生化反相高效液相色谱(HPLC)法,建立了9种常见单糖的分离模式,并成功地用于当归多糖的单糖组成分析,具有良好的重复性。结果表明,当归多糖由甘露糖、鼠李糖、葡萄糖醛酸、半乳糖醛酸、葡萄糖、半乳糖及阿拉伯糖7种单糖组成,其摩尔比为0.57:1.00:0.13:3.06:8.16:7.17:12.69。该HPLC方法是简单、快速、灵敏、方便的分析技术,可用于当归多糖的质量控制。 相似文献

New phosphorylated derivatives of carboxymethylcellulose (CMC) and amidic CMC were realized using trisodium trimetaphosphate (STMP) as the phosphating agent. The new polysaccharides were characterized by infrared spectroscopy and elemental analysis. The characterized polysaccharides were then crosslinked and their rheological and swelling properties determined. The presence of phosphate groups made the three‐dimensional structures more compact and harder than the corresponding non‐phosphated hydrogels. Evaluation of the bioactivity of phosphorylated hydrogels toward osteoblast‐like cells (MG63) showed a significant increase in the osteocalcin production. The thermal pyrolysis of carboxymethyl hydroxypropyl sesbania gum and hydroxypropyl sesbania gum in air and nitrogen atmospheres were studied in order to establish the thermal stability of carboxymethyl hydroxypropyl sesbania gum. The results indicate that the stability of carboxymethyl hydroxypropyl sesbania gum against pyrolysis is higher than that of hydroxypropyl sesbania gum. Natural polymer is a frequently used polymer in various food applications and pharmaceutical formulations due to its benefits and its biocompatibility compared to synthetic polymers. One of the natural polymer groups (i.e., polysaccharide) does not only function as an additive in pharmaceutical preparations, but also as an active ingredient with pharmacological effects. In addition, several natural polymers offer potential distinct applications in gene delivery and genetic engineering. However, some of these polymers have drawbacks, such as their lack of water retention and elasticity. Sacran, one of the high-molecular-weight natural polysaccharides (megamolecular polysaccharides) derived from Aphanothece sacrum (A. sacrum), has good water retention and elasticity. Historically, sacran has been used as a dietary food. Moreover, sacran can be applied in biomedical fields as an active material, excipient, and genetic engineering material. This article discusses the characteristics, extraction, isolation procedures, and the use of sacran in food and biomedical applications. 相似文献

玉米芯用PH=3的HCl煮提得到酸提水溶性粗多糖.该粗多糖组成为Glc,Xyl,Gal,经乙醇分级和Sepharose CL-6B柱层析纯化,得到多糖CCCP.经Sephadex A-25柱层析、比旋光度测定、醋酸纤维薄膜电泳等方法鉴定CCCP为均一多糖.经唾液淀粉酶解、纤维素酶解、部分酸水解、高碘酸氧化、Smith降解、甲基化分析及IR,GC和GC/MS等方法分析表明:CCCP为少分枝结构;主链由吡喃型(1→3)Xyl构成,在O(4)处有分枝;支链主要由(1→4)Glc构成,在O(6)处有分枝,支链还存在1→3,1→6键型连接的Glc,Gal;末端基为Xyl,Glc,Gal. 相似文献
