A novel electrothermal atomic absorption spectrometry (ETAAS) method was developed for the halogenation of refractory sample components (Er, Nd and Nb) of lithium niobate (LiNbO3) and bismuth tellurite (Bi2TeO5) optical single crystals to overcome memory effects and carry-over. For this purpose, the cleaning step of a regular graphite furnace heating program was replaced with a halogenation cycle. In this cycle, after the graphite tube cooled to room temperature, a 20 μL aliquot of liquid carbon tetrachloride (CCl4) was dispensed with a conventional autosampler into the graphite tube. The CCl4 was partially dried at 80 °C under the mini-flow (40 cm3 min−1) condition of the Ar internal furnace gas (IFG), then the residue was decomposed (pyrolyzed) by fast furnace heating at 1900-2100 °C under interrupted flow of the IFG. This step was followed by a clean-out stage at 2100 °C under the maximum flow of the IFG. The advantage of the present method is that it does not require any alteration to the graphite furnace gas supply system in contrast to most of the formerly introduced halogenation techniques.The effectiveness of the halogenation method was verified with the determination of Er and Nd dopants in the optical crystals. In these analyses, a sensitivity decrease was observed, which was likely due to the enhanced deterioration of the graphite tube surface. Therefore, the application of mathematical correction (resloping) of the calibration was also required. The calibration curves were linear up to 1.5 and 10 μmol L−1 for Er and Nd, respectively. Characteristic masses of 18 and 241 pg and the limit of detection (LOD) values of 0.017 and 0.27 μmol L−1 were found for Er and Nd, respectively. These LOD data correspond to 0.68 μmol mol−1 Er and 11 μmol mol−1 Nd in solid bismuth tellurite samples. The analytical results were compared with those obtained by a conventional ETAAS method and validated with X-ray fluorescence spectrometry analysis.  相似文献

A new method for the determination of aflatoxins B1, B2, G1, and G2 (AFB1, AFB2, AFG1, AFG2) in cereal flours based on solid-phase microextraction (SPME) coupled with high performance liquid chromatography with post-column photochemical derivatization and fluorescence detection (SPME–HPLC–PD–FD) has been developed. Aflatoxins were extracted from cereal flour samples by a methanol:phosphate buffer (pH 5.8, I = 0.1) (80:20, v/v) solution, followed by a SPME step. Different SPME and HPLC–PD–FD parameters (fiber polarity, temperature, pH, ionic strength, adsorption and desorption time, mobile phase) have been investigated and optimized. This method, which was assessed for the analysis of different cereal flours, showed interesting results in terms of LOD (from 0.035 to 0.2 ng g−1), LOQ (from 0.1 to 0.63 ng g−1, respectively), within and inter-day repeatability (2.27% and 5.38%, respectively) linear ranges (up to 20 ng g−1 for AFB1 and AFG1 and 6 ng g−1 for AFB2 and AFG2), and total raw extraction efficiency (in the range 55–59% at concentrations in the range 0.3–1 ng g−1 and 49–52% at concentrations in the range 1–10 ng g−1). The results were also compared with the purification step carried out by conventional immunoaffinity columns.  相似文献

This work describes the in situ immobilization of Mn(II) phthalocyanine (MnPc) in a porous SiO2/SnO2 mixed oxide matrix obtained by the sol gel processing method. The chemically modified matrix SiO2/SnO2/MnPc, possessing an estimated amount of 8 × 10−10 mol cm−2 of MnPc on the surface, was used to prepare an electrode to analyze dissolved oxygen in water by an electrochemical technique. The electrode was prepared by mixing the material with ultrapure graphite and evaluated using differential pulse voltammetry. Dissolved O2 was reduced at −0.31 V with a limit of detection (LOD) equal to 7.0 × 10−4 mmol L−1. A mechanism involving four electrons in O2 reduction was determined by the rotating disk electrode technique.  相似文献

Lead and cadmium have been determined by subtractive anodic-stripping voltammetry (SASV) in the square-wave mode at a silver electrode without removal of oxygen. The sensitivities and detection limits for the two metals differ considerably. Detection limits of 0.05 nM for lead and 1 nM for cadmium have been achieved following 90 s electrodeposition. The repeatability of consecutive SASV runs is good (for lead 0.5% at 20 nM for 30 s electrolysis, 5% at 0.3 nM for 60 s electrolysis; for cadmium 2.5% at 20 nM for 30 s electrolysis, 5% at 5 nM for 60 s). Hundreds of runs can be carried out without any pretreatment of the electrode. The high stability is attributed to renewal of the electrode surface that takes place during the electrodeposition step in a two-electrode cell: the silver counter/quasi-reference electrode generates silver ions that codeposit with lead and cadmium at the Ag-RDE, thus ensuring a continuity of the latter. Underpotential deposition (UPD) plays a central role in anodic-stripping voltammetry (ASV). During the deposition step, the adatom coverage of trace elements is in the range of 0.01-1% and no bulk deposition is invoked for metals that exhibit UPD. The UPD properties and, as a result, the ASV signals are strongly affected by the type and concentration of the supporting electrolyte. The effects of Cl, Br, SO42− and NO3 are shown. The analysis of lead and cadmium in natural waters has been performed. Surfactants distort the SASV signal. In order to ensure surfactant-free solutions, the samples were pretreated by wet ashing.  相似文献

The electrochemical behavior of terazosin at the hanging mercury drop electrode was studied in Britton-Robinson buffer (pH 2-11), acetate buffer (4.5-5.5), and in 0.1 M solution of each of sodium sulfate, sodium nitrate, sodium perchlorate and potassium chloride as supporting electrolytes. The square-wave adsorptive cathodic stripping voltammogram of terazosin exhibited a single well-defined two-electron irreversible cathodic peak which may be attributed to the reduction of CO double bond of the drug molecule. A fully validated, simple, high sensitive, precise and inexpensive square-wave adsorptive cathodic stripping voltammetric procedure was described for determination of terazosin in bulk form, tablets and human serum. A mean recovery for 1×10−8 M terazosin in bulk form, following preconcentration onto the hanging mercury drop electrode for 60 s at a −1.0 V (versus Ag/AgCl/KCls), of 99±0.7% (n=5) was obtained. Limits of detection (LOD) and quantitation (LOQ) of 1.5×10−11 and 5×10−11 M bulk terazosin were achieved, respectively. The proposed procedure was successfully applied to determination of the drug in its Itrin® tablets and human serum samples. The achieved LOD and LOQ of the drug in human serum samples were 5.3×10−11 and 1.8×10−10 M THD, respectively. The pharmacokinetic parameters of the drug in human plasma were estimated as: Cmax=77.5 ng ml−1, tmax=1.75 h, AUC0-t=602.3 ng h ml−1, Ke=0.088 h−1 and t1/2=11.32 h) which are favorably compared with those reported in literature.  相似文献

A sensitive preconcentration methodology for Cd determination at trace levels in water samples was developed in this work. 1-Butyl-3-methylimidazolium hexafluorophosphate ([C4MIM][PF6]) room temperature ionic liquid (RTIL) was successfully used for Cd preconcentration, as cadmium-2-(5-bromo-2-pyridylazo)-5-diethylaminophenol complex [Cd-5-Br-PADAP]. Subsequently, Cd was back-extracted from the RTIL phase with 500 μL of 0.5 mol L−1 nitric acid and determined by electrothermal atomic absorption spectrometry (ETAAS). A preconcentration factor of 40 was achieved with 20 mL of sample. The limit of detection (LOD) obtained under optimum conditions was 3 ng L−1 and the relative standard deviation (R.S.D.) for 10 replicates at 1 μg L−1 Cd2+ concentration level was 3.5%, calculated at peak heights. The calibration graph was linear from concentration levels near the detection limits up to at least 5 μg L−1. A correlation coefficient of 0.9997 was achieved. Validation of the methodology was performed by standard addition method and analysis of certified reference material (CRM). The method was successfully applied to the determination of Cd in river and tap water samples.  相似文献

We describe a terbium-ligand complex (TbL) for a microtiterplate assay for phosphate (P) in the 0.3-100 μmol L−1 range based on luminescence quenching. As the pH optimum is at neutral pH (7.4) the probe is quenched by both, primary (H2PO4) and secondary phosphate (HPO42−). The LOD is 110 nmol L−1. A Stern-Volmer study revealed that quenching is mostly static. Due to the ms-decay time of TbL, the first luminescence lifetime assay for phosphate could also be developed. The lifetime-based calibration plot is linear between 0.5 and 5 μmol L−1 of P. The effect of various surfactants on assay performance and a study on interferents are presented. The probe was successfully applied to determination of P in commercial plant fertilizers and validated against the molybdenum blue test. The probe is the most sensitive lanthanide-based probe for phosphate.  相似文献

We present a new processing scheme for the deposition of microporous, sol–gel derived silica membranes on inexpensive, commercially available anodic alumina (Anodisk™) supports. In a first step, a surfactant-templated mesoporous silica sublayer (pore size 2–6 nm) is deposited on the Anodisk support by dip-coating, in order to provide a smooth transition from the pore size of the support (20 or 100 nm) to that of the membrane (3–4 Å). Subsequently, the microporous gas separation membrane layer is deposited by spin-coating, resulting in a defect-free dual-layer micro-/mesoporous silica membrane exhibiting high permeance and high selectivity for size selective gas separations. For example, in the case of CO2:N2 separation, the CO2 permeance reached 3.0 MPU (1 MPU = 10−7 mol m−2 s−1 Pa−1) coupled with a CO2:N2 separation factor in excess of 80 at 25 °C. This processing scheme can be utilized for laboratory-scale development of other types of microporous or dense inorganic membranes, taking advantage of the availability, low cost and low permeation resistance of anodic alumina (or other metal oxide) meso- and macroporous supports.  相似文献

A microwave-assisted persulfate oxidation method followed by ion chromatographic determination of nitrate was developed for total nitrogen determination in atmospheric wet and dry deposition samples. Various operating parameters such as oxidation reagent concentrations, microwave power, and extraction time were optimized to maximize the conversion of total nitrogen to nitrate for subsequent chemical analysis. Under optimized conditions, 0.012 M K2S2O8 and 0.024 M NaOH were found to be necessary for complete digestion of wet and dry deposition samples at 400 W for 7 min using microwave. The optimized extraction method was then validated by testing different forms of organic nitrogen loaded to pre-baked filter substrates and NIST SRM 1648 (urban particulate matter), and satisfactory results were obtained. In the case of wet deposition samples, standard addition experiments were performed. The suitability of the method for real-world application was assessed by analyzing a number of wet and dry deposition samples collected in Singapore during the period of March-April 2007. The organic nitrogen content was 15% (wet) and 30% (dry) of the total nitrogen. During the study period, the estimated wet fluxes for nitrate (NO3), ammonium (NH4+), organic nitrogen (ON), and total nitrogen (TN) were 16.1 ± 6.5 kg ha−1 year−1, 11.5 ± 5.7 kg ha−1 year−1, 3.8 ± 1.5 kg ha−1 year−1and 31.5 ± 13.2 kg ha−1 year−1, respectively, while the dry fluxes were 2.5 ± 0.8 kg ha−1 year−1, 1.4 ± 0.9 kg ha−1 year−1, 2.3 ± 1.4 kg ha−1 year−1 and 7.5 ± 2.6 kg ha−1 year−1, respectively.  相似文献

A square wave adsorptive stripping voltammetric (SWAdSV) method for the indirect determination of trace amounts of magnesium with thiopentone sodium (TPS) as an electroactive ligand, at carbon paste mercury film electrode (CP-MFE) is proposed. It is observed that the increase of the square wave voltammetric cathodic peak current of TPS, under alkaline conditions, is linear with the increase of Mg concentration. Under optimum experimental conditions viz.; pH 10.75, 3×10−5 M TPS and 0.05 M phosphate buffer (Na2HPO4-NaH2PO4), a linear relation in the range 6×10−9 to 9×10−8 M Mg2+ (0.14-2.16 ppb), at 60 s deposition time, is obtained. The detection limit of Mg2+ is 0.14 ppb for 60 s deposition time with the relative standard deviation is 0.5% (n=5). The proposed method was successfully applied to the determination of magnesium in urine and tap water samples with satisfactory results. The data obtained are compared with the standard flame atomic absorption spectrophotometric method (FAAS).  相似文献

A novel, simple, sensitive and precise spectrofluorimetric method was developed for measuring the melamine concentration in different milk batch samples. The method was based upon measuring the quenching of the luminescence intensity of the produced yellow colored ruthenium(II) carbonyl complex of the general formula [Ru(CO)2(L)] (where L = anion of tetradentate Schiff base). The Ru(II) complex exhibited characteristic luminescence band in the visible region. The remarkable quenching of the luminescence intensity of [Ru(CO)2(L)] complex by various concentrations of melamine was successfully used as a chemosensor for the assessment of melamine in different milk samples at λex = 400 nm and pH 7.4 in DMSO with a linear dynamic range 1.0 × 10−6 to 3.0 × 10−9 mol L−1 and lower detection limit (LOD) and quantification detection limit (QOD) of 3.3 × 10−10 and 1.0 × 10−9 mol L−1, respectively.  相似文献

Two accurate, reliable, and highly sensitive spectrofluorimetric methods were developed for simultaneous determination of binary mixture gemfibrozil and rosiglitazone in human plasma without prior separation steps. The first method is based on synchronous fluorescence spectrometry using double scans. At Δλ = 27 nm, gemfibrozil yields detectable signal that is independent of the presence of rosiglitazone. Similarly, at Δλ = 120 nm the signal of rosiglitazone is not influenced by the presence of gemfibrozil. Signals at two wavelengths, 301 (Δλ = 27 nm) and 368 nm (Δλ = 120 nm) vary linearly with gemfibrozil and rosiglitazone concentrations over the range 100-700 ng mL−1 (for gemfibrozil) and 20-140 ng mL−1 (for rosiglitazone), respectively. The limits of detection (LOD) were 2.3 and 2.72 ng mL−1 for gemfibrozil and rosiglitazone, respectively. The second method is based on the technique of simultaneous equations (Vierodt's method), in which 258 nm was selected as the excitation wavelength. Two equations are constructed based on the fact that at (λEm2=302 nm of gemfibrozil) and (λEm2=369 nm of rosiglitazone) the fluorescence of the mixture is the sum of the individual fluorescence of gemfibrozil and rosiglitazone. The limits of detection (LOD) were 28.1 and 23.63 ng mL−1 for gemfibrozil and rosiglitazone, respectively. The proposed methods were successfully applied for the determination of the two compounds in synthetic mixtures and in human plasma with a good recovery.  相似文献

We report a simple and rapid procedure that leads to incorporation of mediator and introduction of amine functionality onto the surface of screen-printed carbon electrodes (SPCE). The electrodes were doped with cobalt phthalocyanine (CoPc) by enhanced adsorption in a process that uses minimal amounts of this redox mediator as compared with CoPc loaded inks. The CoPc-doped SPCE showed a substantially increased sensitivity to hydrogen peroxide and thiocholine as compared to unmodified electrodes. This greatly facilitated their use as transducers for the construction of amperometric biosensors based on enzymes producing oxidizable products such as hydrogen peroxide or thiols. Immobilisation of enzymes including glucose oxidase, acetylcholinesterase and choline oxidase was achieved through their multi-contact electrostatic interaction with polyethyleneimine (PEI) which was electrodeposited on the surface of CoPc-doped electrodes in one step from ethanolic solution. The efficiency of enzyme immobilisation was shown to depend on the molecular weight of the PEI used, reaching a maximum for 25 kDa PEI. The biosensors shown sensitivity to glucose at 130 nA mM−1 (LOD 0.15 mM) and to acetylcholine at 70 nA mM−1 (LOD 0.10 mM) under +0.6 V. Detection of glucose has been demonstrated at +0.4 V with the sensitivity of 60 nA mM−1 and LOD of 0.33 mM. Possibility of the inhibition analysis of pesticides has been shown for acetylcholinesterase-based sensors.  相似文献

In situ microwave activation has been applied to the electro-deposition and stripping of palladium metal (which is widely used as a catalyst) at cavitation resistant boron-doped diamond electrodes. Focused microwave radiation leading to heating, boiling, and cavitation is explored as an option to improve the speed and sensitivity of the analytical detection procedure. The deposition and anodic stripping of palladium by linear sweep voltammetry in 0.1 M KCl (pH 2) solution and at boron-doped diamond electrodes is shown to be strongly enhanced by microwave activation due to both (i) the increase in mass transport and (ii) the increase in the kinetic rate of deposition and stripping.The temperature at the electrode surface is calibrated with the reversible redox couple Fe(CN)64−/Fe(CN)63− and found to be reach 380 K. In the presence of microwave radiation, the potential of onset of the deposition of palladium is strongly shifted positive from −0.4 to +0.1 V versus SCE. The optimum potential for deposition in the presence of microwaves is −0.4 V versus SCE and the anodic stripping peak current is shown to increase linearly with deposition time. Under these conditions, the stripping peak current varies linearly with the palladium concentration down to ca. 2 μM. At concentration lower than this a logarithmic variation of the stripping peak current with concentration is observed down to ca. 0.1 μM (for 5 min pre-concentration in presence of microwave radiation).  相似文献

Recently, cobalt (Co) is reported to be taken as a supplement by athletes for improving anaerobic performance. For the diagnosis of abuse, the limit of detection (LOD) of Co in the analysis should be lower than the concentrations of Co in plasma and urine of normal persons. A simple, rapid and sensitive method has been developed for the determination of Co in urine. Co was complexed with diethyldithiocarbamate (DDC) and extracted with isoamyl alcohol in the presence of citric acid. The detection of Co was achieved by injecting a 1-μL aliquot of isoamyl alcohol containing Co-DDC complex directly into an electrospray ionization tandem mass spectrometric (ESI-MS-MS) instrument without chromatographic separation. The quantification was performed using selected reaction monitoring at m/z 291 of the product ion Co(C4H10NCS)2+ which was produced by collision-induced dissociation from the precursor ion Co(DDC)2+ at m/z 355. ESI-MS-MS data were obtained in less than 10 min with an LOD of 0.05 μg L−1 and a linear calibration range of 0.1-100 μg L−1 using 10 μL of urine. The procedure was validated with certified reference materials (SRM 2670a and SRM 1643e). This method is suitable for the analysis of Co in the laboratories already equipped with an ESI-MS-MS instrument.  相似文献

A novel on-line preconcentration method based on liquid-liquid (L-L) extraction with room temperature ionic liquids (RTILs) coupled to flame atomic absorption spectrometry (FAAS) was developed for cadmium determination in plastic food packaging materials. The methodology is based on the complexation of Cd with 2-(5-bromo-2-pyridylazo)-5-diethylaminophenol (5-Br-PADAP) reagent after sample digestion followed by extraction of the complex with the RTIL 1-butyl-3-methylimidazolium hexafluorophosphate ([C4mim][PF6]). The mixture was loaded into a flow injection analysis (FIA) manifold and the RTIL rich-phase was retained in a microcolumn filled with silica gel. The RTIL rich-phase was then eluted directly into FAAS. A enhancement factor of 35 was achieved with 20 mL of sample. The limit of detection (LOD), obtained as IUPAC recommendation, was 6 ng g−1 and the relative standard deviation (R.S.D.) for 10 replicates at 10 μg L−1 Cd concentration level was 3.9%, calculated at the peak heights. The calibration graph was linear and a correlation coefficient of 0.9998 was achieved. The accuracy of the method was evaluated by both a recovery study and comparison of results with direct determination by electrothermal atomic absorption spectrometry (ETAAS). The method was successfully applied for Cd determination in plastic food packaging materials and Cd concentrations found were in the range of 0.04-10.4 μg g−1.  相似文献

A novel high-performance liquid chromatographic (HPLC) method using pre-column derivatization and UV detection at 275 nm for the determination of clarithromycin in rat plasma has been validated. Clarithromycin was extracted from plasma sample spiked with internal standard (erythromycin) under alkaline condition with ethyl ether and derivatizated with trimethylbromosilane. The analyses were run on a C18 column, maintained at 40 °C during elution, using a mobile phase comprised of potassium dihydrogen phosphate (50 mM, pH 6.8, contained 0.7% triethylamine), acetonitrile, and methanol (30:45:25, v/v/v). The standard calibration curve for clarithromycin was linear (r2 = 0.9998) over the concentration range of 0.1-10 μg ml−1 in rat plasma. The limit of detection (LOD) and limit of quantitation (LOQ) was 30 ng ml−1 and 0.1 μg ml−1 respectively. The intra- and inter-day assay variability range was 2.6-7.4% and 3.3-8.5%, respectively. This method has been successfully applied to a pharmacokinetic study of clarithromycin in rats.  相似文献

An amperometric immunoassay for the determination of tumor necrosis factor alpha (TNFα) protein biomarker in human serum based on the use of magnetic microbeads (MBs) and disposable screen-printed carbon electrodes (SPCEs) has been developed. The specifically modified microbeads were magnetically captured on the working electrode surface and the amperometric responses were measured at −0.20 V (vs. Ag pseudo-reference electrode), upon addition of hydroquinone (HQ) as electron transfer mediator and H2O2 as the enzyme substrate. After a thorough optimization of the assay, extremely low limits of detection were achieved: 2.0 pg mL−1 (36 fM) and 5.8 pg mL−1 (105 fM) for standard solutions and spiked human serum, respectively. The simplicity, robustness and this clinically interesting LOD proved the developed TNFα immunoassay as a good contender for real clinical application.  相似文献

A simple, precise, and accurate hydrophilic interaction liquid chromatographic (HILIC) method has been developed for the determination of five aromatic amines in environmental water samples. Chromatography was carried out on a bare silica column, using a mixture of acetonitrile and a buffer of NaH2PO4–H3PO4 (pH 1.5, containing 10 mM NaH2PO4) (85:15, v/v) as a mobile phase at a flow rate of 1 mL min−1. Aromatic amines were detected by UV absorbance at 254 nm. The linear range of amines was good (r2 > 0.998) and limit of detection (LOD) within 0.02–0.2 mg L−1 (S/N = 3). The retention mechanism for the analytes under the optimum conditions was determined to be a combination of adsorption, partition and ionic interactions. The proposed method was applied to the environmental water samples. Aromatic amines were isolated from aqueous samples using solid-phase extraction (SPE) with Oasis HLB cartridges. Recoveries of greater than 75% with precision (RSD) less than 12% were obtained at amine concentrations of 5–50 μg L−1 from 100 mL river water and influents from a wastewater treatment plant (WWTP). The present HILIC technique proved to be a viable method for the analysis of aromatic amines in the environmental water samples.  相似文献

Determination of Se(IV) and Se(VI) in high saline media was investigated by cathodic stripping voltammetry (CSV). The voltammetric method was applied to assay selenium in seawater, hydrothermal and hemodialysis fluids. The influence of ionic strength on selenium determination is discussed. The CSV method was based on the co-electrodeposition of Se(IV) with Cu(II) ions and Se(VI) determined by difference after sample UV-irradiation for photolytic selenium reduction. UV-irradiation was also used as sample pre-treatment for organic matter decomposition. Detection limit of 0.030 μg L−1 (240 s deposition time) and relative standard deviation (RSD) of 6.19% (n = 5) for 5.0 μg L−1 of Se(IV) were calculated. Linear calibration range for selenium was observed from 1.0 to 100.0 μg L−1. Concerning the pre-treatment step, best results were obtained by using 60 min UV-irradiation interval in H2O2/HCl medium. Se(VI) was reduced to the Se(IV) electroactive species with recoveries between 91.7% and 112.9%. Interferents were also investigated.  相似文献
