The objective of this work was to investigate the extraction of Ilex paraguariensis leaves by means of three extraction techniques: pressurized liquid extraction (PLE, also called accelerated solvent extraction – ASE), maceration, and sonication. Samples of mate tea leaves were collected from an experiment conducted under agronomic control at Indústria e Comércio de Erva‐Mate Bar?o LTDA, Brazil. Six solvents with increasing polarities (n‐hexane, toluene, dichloromethane, ethyl acetate, acetone, and methanol) were used in this investigation. Chemical analysis of the extracts was performed by GC coupled with a mass spectrometer detector. The identification and quantification were accomplished by coinjections of certified standards. The results showed that no significant differences in the qualities of the extracts were noticed regarding the extraction methods. On the other hand, the PLE technique was found to be more effective for the extractions of caffeine, phytol, palmitic, and stearic acid. The use of PLE led to a significant decrease in the total extraction time, amount of solvent consumption, and manipulation of samples compared to maceration and ultrasound‐assisted extraction methods.  相似文献

The comparison of four extraction techniques for isolation of five native and one labelled steroid oestrogens from sediment was described. The three conventional extraction techniques Soxhlet warm extraction (SWE), accelerated solvent extraction (ASE), microwave-assisted extraction (MAE) and a promising technique QuEChERS were tested for isolation of low environmentally relevant oestrogen concentrations using different extraction conditions. The least expensive and time-consuming method QuEChERS provided the best extraction recoveries (53–84%) from all techniques. MAE achieved the highest recovery from conventional techniques for less polar oestrogens using dichloromethane: acetone 3:1 mixture as an extraction solvent (50–71%), but for extraction of the whole group of oestrogens including more polar estriol acetone or methanol must be used. ASE provided higher extraction recoveries using dichloromethane at 60°C (53–74%) for less polar oestrogens. However, the repeatability of results was unsatisfactory and recoveries using other extraction conditions were lower than for MAE. The most time-consuming SWE achieved the worst extraction recoveries and for isolation of low oestrogen concentrations from sediments, it is completely unsuitable.  相似文献

The total content of polyphenols and flavonoids determined in the same plant and their corresponding antioxidant activities may vary widely, depending on the extraction conditions applied. This study was conducted to optimise the extraction conditions of phenolics and flavonoids from the mistletoe plant. Various extraction methods, i.e. ultrasound-assisted extraction technology, maceration, maceration with stirring, accelerated solvent extraction (ASE), and extraction under reflux were evaluated for their percentage extraction of polyphenols (TPC) and flavonoids (TFC) from Viscum album subsp. abietis. In addition, the anti-radical activity of extracts was analysed using the 2,2-diphenyl-1-picrylhydrazyl method. The effects of temperature, solvent type, and concentration on the phenolic extraction efficiency and antioxidant activity were studied using chemometric and statistical methods. The results showed that the extracts of V. album subsp. abietis contained large amounts of polyphenols and flavonoids (up to 57.673 mg g?1 and 9.955 mg g?1 of dry extract, respectively) and exhibited potent antioxidant activity, hence representing promising sources of powerful antioxidants. Due to its high extraction efficiency and considerable saving of time and solvent, ASE was more effective than the other extraction techniques. Extracts prepared with water-polar solvent mixtures displayed the highest TPC, TFC, and antioxidant activity, while organic polar solvents were the least efficient extractants.  相似文献

A new method to quick extraction of vanillin and p-hydroxybenzaldheyde (PHB) of vanilla beans from vanilla fragans is proposed. Samples were irradiated with microwaves energy to accelerate the extraction process and photometric monitoring was performed at 348 and 329 nm (vanillin and PHB, respectively). The simultaneous determination of vanillin and PHB from extracts was performed using the Vierordt's method, which showed a precision, expressed as relative standard deviation, smaller 2.5% for both analytes. Conditions such as microwaves irradiation power, number of irradiation and non-irradiation cycles, irradiation time and ethanol concentration were optimized by means of multivariate screening that showed that irradiation power and number of irradiation cycles are the most significant condition in the vanilla extraction process. The focused microwave-assisted extraction (FMAE) was applied to commercial (dried vanilla beans from fresh green vanilla beans), lyophilised and dried (commercial vanilla dried at 135 °C in oven) vanilla beans samples. The results showed that the extraction of vanillin and PHB in the commercial vanilla samples were higher than in dried and lyophilised samples. With the proposed FMAE a decrease in the extraction time of 62 times and an increase in the vanillin and PHB concentrations between 40 and 50% with respect to the official Mexican extraction method, were obtained.  相似文献

A simple, fast and sensitive RP-HPTLC method is developed for simultaneous quantitative determination of vanillin and related phenolic compounds in ethanolic extracts of Vanilla planifolia pods. In addition to this, the applicability of accelerated solvent extraction (ASE) as an alternative to microwave-assisted extraction (MAE), ultrasound-assisted extraction (UAE) and Soxhlet extraction was also explored for the rapid extraction of phenolic compounds in vanilla pods. Good separation was achieved on aluminium plates precoated with silica gel RP-18 F(254S) in the mobile phase of methanol/water/isopropanol/acetic acid (30:65:2:3, by volume). The method showed good linearity, high precision and good recovery of compounds of interest. ASE showed good extraction efficiency in less time as compared to other techniques for all the phenolic compounds. The present method would be useful for analytical research and for routine analysis of vanilla extracts for their quality control.  相似文献

In order to determine PAHs in marine sediment samples by GC/MS(SIM) a new extraction approach of ASE-SFE was evaluated using combined accelerated solvent extraction (ASE, dynamic and static mode) and supercritical fluid extraction (SFE, dynamic mode) without further purification of the sample. The solvents used for ASE-SFE were methylene chloride and carbon dioxide. The recovery data, precision and accuracy of the whole method were evaluated statistically. The average recoveries of PAHs, based on deuterated internal standards were 77% for 2-3-ring PAHs, 85% for 4-ring PAHs, 88% for 5-ring PAHs and 97% for 6-ring PAHs. The extraction time required for the ASE-SFE technique was 30 min, which is longer than in the case of independent use of ASE and shorter compared to SFE. ASE-SFE recoveries of PAHs from SRM marine sediment are comparable for (2-3-ring, 4-ring PAHs) or higher (5-ring, 6-ring PAHs) than reported for the conventional extraction methods of ASE and SFE. Method detection limits of (MDL) were statistically estimated. MDL values obtained for 15 PAHs compounds vary between 0.06 ngg(-1) and 3.54 ngg(-1).  相似文献

The extraction and determination of chlorinated biphenyls (CBs) in soils and solid wastes is an ongoing subject of study. This is an overview article that compares the microwave-assisted extraction (MAE) and accelerated solvent extraction (ASE) techniques. The extraction of CBs by ASE has been optimised taking into account the variation of pressure, temperature and extraction time by means of experimental design and the Simplex approach. The recoveries obtained under the optimum conditions are compared and discussed with those obtained from MAE and Soxhlet extractions.  相似文献

Accelerated solvent extraction (ASE) shows higher recoveries for PAHs in comparison with traditional Soxhlet extraction, but in a fraction of time and with less solvent consumption. Better recoveries are especially achieved with PAHs of high reactivity, the latter being expressed by the structure-to-count ratio (SCR). To estimate polar pollutants including phenols/benzenediols, the sample was subjected to a combined in-situ derivatization/extraction approach using 2% v/v acetic anhydride in toluene. The main reason for the better recovery obtained in this way, in comparison with the classical ASE approach, is to overcome strong matrix-analyte interactions. Analogously, fatty acids were analyzed as methyl esters obtained by in-situ derivatization/extraction using boron trifluoride.  相似文献

以烟草中有机氯类农药为研究对象,从检出限、回收率、重复性和色谱图4个方面系统比较了机械振荡萃取法、超声波溶剂萃取法、微波辅助萃取法和加速溶剂萃取法4种常用前处理方法。由于加速溶剂萃取法是在高温高压下进行萃取,在提取出残留农药的同时,也提取出更多的干扰物。机械振荡法、超声波溶剂萃取法和微波辅助萃取法各项结果差别较小,其中机械振荡萃取法回收率和重复性最好,但提取时间较长。考虑溶剂消耗和提取时间等方面的因素,微波辅助萃取是较理想的样品前处理方法。  相似文献

以两种药材为研究实例,对加速溶剂萃取法(ASE)在中药材有效成分提取研究中的应用进行了简要介绍。采用正交试验法考察了提取丹参中丹酚酸B的提取条件(萃取温度、静态萃取时间、萃取溶剂以及料液比),确定了较好的实验条件。比较了ASE、水蒸气蒸馏法、超声波提取法及索氏提取法对木香挥发油的提取效果,结果表明ASE对木香挥发油的提取效果最好。  相似文献

An extraction technique is described for vitamin K1 in medical foods, using accelerated solvent extraction (ASE) in conjunction with matrix solid-phase dispersion (MSPD). The medical food sample is treated as it would be with MSPD extraction, followed by ASE for a hands-free automated extraction. The vitamin K1 in the ASE extract is then quantitated by reversed-phase liquid chromatography with fluorescence detection. The chromatography specifications are identical to those in previous work that used MSPD only, with a limit of detection of 6.6 pg and a limit of quantitation of 22 pg on column. Recoveries, which were determined for an analyte-fortified zero control reference material for medical foods, averaged 97.6% (n = 25) for vitamin K1. The method provides a rapid, automatic, specific, and easily controlled assay for vitamin K1 in fortified medical foods with minimal solvent usage.  相似文献

Three commonly applied extraction techniques for persistent organic chemicals, Soxhlet extraction (SE), accelerated solvent extraction (ASE) and microwave-assisted extraction (MAE), were applied on soil and fish samples in order to evaluate their performances. For both PCBs and PBDEs, the two more recent developed techniques (ASE and MAE) were in general capable of producing comparable extraction results as the classical SE, and even higher extraction recoveries were obtained for some PCB congeners with large octanol-water partitioning coefficients (Kow). This relatively uniform extraction results from ASE and MAE indicated that elevated temperature and pressure are favorable to the efficient extraction of PCBs from the solid matrices. For PBDEs, difference between the results from MAE and ASE (or SE) suggests that the MAE extraction condition needs to be carefully optimized according to the characteristics of the matrix and analyte to avoid degradation of higher brominated BDE congeners and improve the extraction yields.  相似文献

A selective and sensitive analytical method for determination of ten congeners of polychlorinated biphenyls (PCBs 31, 28, 52, 101, 118, 153, 105, 138, 156, and 180) in mussel samples (Mytilus galloprovincialis) based on accelerated solvent extraction (ASE) and gas chromatography–tandem mass spectrometry (GC–MS–MS) is presented in this work. Extraction conditions were optimised using a Plackett–Burman factorial design. The final extracts were analysed after cleanup on alumina columns. The optimised extraction parameters were solvent percentage, sample amount, extraction temperature, pressure, static extraction time, flush percentage, and purge time. The results suggest that PCBs 118, 105, and 180 extractions appeared affected by only one statistically significant factor, pressure, solvent percentage and static extraction time, respectively. Extraction of PCBs 138 and 156 was affected by amount of sample. PCB 138 extraction was also statistically affected by static extraction time and purge time. Quantitative recoveries (64.8–120.3%) were achieved for all PCBs and method precision (RSD < 19%) was satisfactory.  相似文献

A method was developed to determine eight acetanilide herbicides from cereal crops based on accelerated solvent extraction (ASE) and solid-phase extraction (SPE) followed by gas chromatography-electron capture detector (GC-ECD) analysis. During the ASE process, the effect of four parameters (temperature, static time, static cycles and solvent) on the extraction efficiency was considered and compared with shake-flask extraction method. After extraction with ASE, four SPE tubes (graphitic carbon black/primary secondary amine (GCB/PSA), GCB, Florisil and alumina-N) were assayed for comparison to obtain the best clean-up efficiency. The results show that GCB/PSA cartridge gave the best recoveries and cleanest chromatograms. The analytical process was validated by the analysis of spiked blank samples. Performance characteristics such as linearity, limit of detection (LOD), limit of quantitation (LOQ), precision and recovery were studied. At 0.05 mg/kg spiked level, recoveries and precision values for rice, wheat and maize were 82.3-115.8 and 1.1-13.6%, respectively. For all the herbicides, LOD and LOQ ranged from 0.8 to 1.7 μg/kg and from 2.4 to 5.3 μg/kg, respectively. The proposed analytical methodology was applied for the analysis of the targets in samples; only three herbicides, propyzamid, metolachlor and diflufenican, were detected in two samples.  相似文献

Saponins in plant extracts were indirectly determined by estimation of the content of sapogenins. The first step of determination is extraction with high efficiency. One conventional extraction technique (maceration) and two modern ones (accelerated solvent extraction and supercritical fluid extraction) were compared. Methanol and ethanol were used as solvents or co‐solvents. The results were supported by statistical analysis. Saponins were extracted from leaves, roots, and sprouts of Medicago sativa. Acid hydrolysis, purification, and determination by high‐performance liquid chromatography with evaporative light scattering detector were used. The content of sapogenins was the highest in the roots. Smaller amounts of sapogenins were found in sprouts and the smallest ones in leaves. The main ingredient was medicagenic acid with mean concentration of 621.8 µg/g in roots, 456.7 µg/g in sprouts, and 471.3 µg/g in leaf extract. The highest content of sapogenins in extract was obtained after maceration with methanol; however, this method is nonselective in relation to biologically active compounds. Due to the possibility of using the obtained extracts with sapogenins in the cosmetic or pharmaceutical industry, the selection of extraction techniques and solvents is a very important aspect. Additionally, the chosen technique should be considered eco‐friendly and consistent with the assumptions of “green chemistry.”  相似文献

The primary objective of this study was to simultaneously analyze the residues of the most commonly used pesticides, chlorpyrifos-methyl, endosulfan, EPN, and iprodione in the water dropwort, via accelerated solvent extraction (ASE), supercritical fluid extraction (SFE), and conventional solvent extraction (LLE) techniques. Residue levels were determined using GC with electron-capture detection (GC-ECD). The confirmation of pesticide identity was performed by GC-MS in a selected ion-monitoring (SIM) mode. In none of the ASE and SFE techniques were the extraction conditions optimized. Rather, the experimental variables were predicated on the author's experience. The ECD response for all pesticides was linear in the studied range of concentrations of 0.005-5.0 ppm, with correlation coefficients in excess of 0.9991. At each of the two studied fortification levels, the pesticides yielded recoveries in excess of 72% with RSDs between 1 and 19%. The LODs were achieved at a range of levels from 0.001 to 0.063 ppm, depending on the pesticide utilized. The LOQs, which ranged from 0.003 to 0.188 ppm, were lower than the maximum residue limits (MRLs) authorized by the Korean Food and Drug Administration (KFDA). All of the methods were applied successfully to the determination of pesticide residues in the real samples. It could, therefore, be concluded that any of the techniques utilized in this investigation might prove successful, given that the applied extraction conditions are wisely chosen.  相似文献

In the present work, the use of surfactants and oil-in-water (O/W) microemulsions as alternative extractants in accelerated solvent extraction (ASE) for the extraction of polycyclic aromatic hydrocarbons (pyrene and phenanthrene) from soils was investigated. In particular, the effect of each individual component within the microemulsions, i.e., oil phase, surfactant and co-surfactatnt, and extraction conditions on the percentage recovery was systematically studied. When compared to the water and organic solvent, the important findings were that the common surfactant solutions at the concentrations above their critical micelle concentrations (CMC) were shown to enhance the percentage recovery at the lower extraction temperature. Moreover, the highest percentage recovery can be obtained using microemulsion as the extractant. The chemical component within the microemulsions and relative amounts of the oil phase appeared to play a much more significant role in ensuring high percentage recovery. Finally, an overall comparison between the percentage recoveries obtained with ultrasound-assisted extraction (UAE), microwave-assisted extraction (MAE) and ASE using organic solvents, surfactants and microemulsions as extractants was exhibited.  相似文献
