The Friedel–Crafts reaction of electron‐rich phenols with isatins was developed by employing bifunctional thiourea–tertiary amine organocatalysts. Cinchona alkaloid derived thiourea epiCDT‐ 3 a efficiently catalyzed the Friedel–Crafts‐type addition of phenols to isatin derivatives to provide 3‐aryl‐3‐hydroxy‐2‐oxindoles 7 and 9 in good yield (80–95 %) with good enantiomeric excess (83–94 %). Friedel–Crafts adduct 7 t was subjected to a copper(I)‐catalyzed azide–alkyne cycloaddition to obtain biologically important 3‐aryl‐3‐hydroxy‐2‐oxindole 11 in good enantiomeric excess and having a 1,2,3‐triazole moiety.  相似文献

Herein, we present details of our conformationally flexible, 1,3‐diamine‐tethered guanidine/bisthiourea organocatalysts for chemo‐, regio‐, and enantioselective 1,4‐type Friedel–Crafts reactions of phenols. These organocatalysts show a unique stereo‐discrimination governed by the differential activation entropy (ΔΔS), rather than by the differential activation enthalpy (ΔΔH). Extensive kinetic analyses using Eyring plots for a series of guanidine/bisthiourea organocatalysts revealed the key structural motif in the catalysts associated with a large magnitude of differential activation entropy (ΔΔS). A plausible guanidine–thiourea cooperative mechanism for the enantioselective Friedel–Crafts reaction is proposed.  相似文献

Organocatalytic enantioselective aza‐Friedel–Crafts reactions of cyclic ketimines with pyrroles or indoles were catalyzed by imidazoline/phosphoric acid catalysts. The reaction was applied to various 3H‐indol‐3‐ones to afford products in excellent yields and enantioselectivities. The chiral catalysts can be recovered by a single separation step using column chromatography and are reusable without further purification. Based on the experimental investigations, a possible transition state has been proposed to explain the origin of the asymmetric induction.  相似文献

A new chiral reagent has been developed for generalized installation of pharmacophoric (S)‐ or (R)‐2‐(alkoxyphosphono)‐1‐amino‐2,2‐difluoroethyl group into organic compounds. The original synthetic application of this new reagent is exemplified by Friedel–Crafts reactions with indoles, which proceed efficiently with excellent diastereoselectivity to give enantiomerically pure products.  相似文献

N‐Boc ketimines derived from pyrazolin‐5‐ones were explored to develop an unprecedented domino aza‐Friedel–Crafts/N,O‐acetalization reaction with 2‐naphthols. The novel method requires a catalyst loading of only 0.5 mol % of a bifunctional squaramide catalyst, is scalable to gram amounts, and provides a new series of furanonaphthopyrazolidinone derivatives bearing two vicinal tetra‐substituted stereogenic centers in excellent yields (95–98 %) and stereoselectivity (>99:1 d.r. and 97–98 % ee ). A different reactivity was observed in the case of 1‐naphthols and other electron‐rich phenols, which led to the aza‐Friedel–Crafts adducts in 70–98 % yield and 47–98 % ee .  相似文献

The synthesis of 1,1‐dimethyl‐4‐indanols (3a,b) has been achieved by intramolecular Friedel–Crafts cyclization of 2‐(3‐methyl‐2‐butenyl)phenols (5a,b) or 1‐methoxy‐2‐(3‐methyl‐2‐butenyl)benzenes (6a,b) followed by demethylation, respectively.It was found that the solvent was critical for the formation of different products in the intramolecular Friedel–Crafts reactions of 6. The unexpected product 4‐methoxy‐1,1,6,6‐tetramethyl‐as‐hydrindacene (11) was obtained from the Friedel–Crafts reactions of 6a, and its structure was confirmed by X‐ray diffraction analysis. The key intermediates 5a,b were prepared by ortho‐alkenylation of phenols with 1‐bromo‐3‐methyl‐2‐butene, and the reaction temperature exerted an obvious impact on the yield of 2‐(3‐methyl‐2‐butenyl)phenol (5a).  相似文献

BF3?OEt2‐catalyzed reactions of vinylidenecyclopropanes (VDCPs) 1 with bis(aryl)methanols 2 were thoroughly investigated. When VDCPs 1 reacted with electron‐rich bis(aryl)methanols 2 , diastereomeric rotamers of indene derivatives formed in excellent yields by a novel cationic 1,4‐aryl migration between two carbon atoms and the subsequent intramolecular Friedel–Crafts reaction pathways in the presence of BF3?OEt2 under mild conditions. As for electron‐deficient or less‐electron‐rich bis(aryl)methanols 2 , either trialkene products formed in good yields by direct deprotonation, or another type of indene derivative was produced by direct intramolecular Friedel–Crafts reaction, depending on the substituents on the cyclopropane of VDCPs. In addition, DFT calculations were carried out to explain the experimental results. Plausible mechanisms for all these transformations are proposed on the basis of the experimental and computational results.  相似文献

A B(C6F5)3‐catalyzed hydroarylation of a series of 1,3‐dienes with various phenols has been established through a combination of theoretical and experimental investigations, affording structurally diverse ortho‐allyl phenols. DFT calculations show that the reaction proceeds through a borane‐promoted protonation/Friedel–Crafts pathway involving a π‐complex of a carbocation–anion contact ion pair. This protocol features simple and mild reaction conditions, broad functional‐group tolerance, and low catalyst loading. The obtained ortho‐allyl phenols could be further converted into flavan derivatives using B(C6F5)3 with good cis diastereoselectivity. Furthermore, this transformation was applied in the late‐stage modification of pharmaceutical compounds.  相似文献

The first arylation strategy for the synthesis of enantioenriched propargylamines is disclosed. This approach, which is complementary to previous alkynylation and alkylation strategies, involves a C(sp2)?C(sp3) bond formation, and is based on the first asymmetric Friedel–Crafts‐type arylation reaction of C‐alkynyl imines. Asymmetric Friedel–Crafts reactions with electron‐deficient phenols, a longstanding unsolved challenge, have thus been realized for the first time, enabled by the combination of our recently introduced C‐alkynyl N‐Boc‐protected N,O‐acetals as electrophiles and chiral phosphoric acids as catalysts. The synthetic utility of the resulting structurally diverse and polyfunctional chiral propargylamines was demonstrated by a series of selective transformations, including controlled reduction of the alkynyl group and iterative cross‐couplings.  相似文献

The title compounds were prepared by aldol reaction of anisaldehyde and the respective N,N‐dibenzyl glycinates. Deprotection of the nitrogen atom with Pearlman’s catalyst delivered the unprotected β‐hydroxytyrosine esters, which were further N‐protected as N,N‐phthaloyl (Phth) and N‐fluorenylmethylcarbonyloxy (Fmoc) derivatives. The Friedel–Crafts reaction with various arenes was studied employing these alcohols as electrophiles. It turned out that the facial diastereoselectivitiy depends on the nitrogen protecting group and on the ester group. The unprotected substrates (NH2) gave preferentially syn‐products but the anti‐selectivity increased when going from NHFmoc over NPhth to NBn2. If the ester substituent was varied the syn‐preference increased in the order Me <Et <iPr. The reactions were shown to be fully stereoconvergent and proceeded under kinetic product control. A model is suggested to explain the facial diastereoselectivity based on a conformationally locked benzylic cation intermediate. The reactions are preparatively useful for the N‐unprotected isopropyl ester, which gave Friedel–Crafts alkylation products with good syn‐selectivity (anti/syn=21:79 to 7:93), and for the N,N‐dibenzyl‐protected methyl ester, which led preferentially to anti‐products (anti/syn=80:20 to >95:5). Upon acetylation of the latter compound to the respective acetate, Bi(OTf)3‐catalyzed alkylation reactions became possible, in which silyl enol ethers served as nucleophiles. An asymmetric aza‐Friedel–Crafts alkylation reaction between indoles and indolenines that were derived in situ from 3‐indolinone‐2‐carboxylates has been developed by using 3,3′‐bis(triphenylsilyl)‐1,1′‐binaphthyl‐2,2′‐diyl hydrogen phosphate as a catalyst. The reaction proceeded under mild conditions and provided chiral indol‐3‐yl‐3‐indolinone‐2‐carboxylate derivatives in good yields with excellent ee values (up to 98.6 %). Similarly, the Mannich‐type addition of indoline‐3‐ones to indolenines provided heterodimers with vicinal chiral quaternary centers. This method was successfully applied to the construction of the core structure of trigonoliimine C.  相似文献

A novel platinum‐catalyzed cascade cyclization reaction was developed by intramolecular Friedel–Crafts‐type C?H coupling of aniline derivatives with a propargyl carbonate unit‐allylic amination sequence. Treatment of various propargyl carbonates tethered to meta‐aniline derivatives with a Pt(dba)3/DPEphos catalyst system afforded the corresponding 3,4‐fused tricyclic 3‐alkylidene indolines in 42–99 % yield, which were transformed into 3,4‐fused indole derivatives by reaction with trifluoroacetic acid. The reaction products exhibited antiproliferative activities against cancer cells, but not normal cells, revealing the potential usefulness of this reaction for medicinal chemistry.  相似文献

We report a highly efficient Friedel–Crafts reaction of 3‐alkyl or 3‐aryl 3‐hydroxyoxindoles with a variety of aromatic and heteroaromatic compounds to unsymmetrical 3,3‐diaryloxindoles or 3‐alkyl‐3‐aryloxindoles, which are interesting medicinal targets and useful building blocks for the synthesis of natural products. Hg(ClO4)2 ? 3 H2O was identified as a powerful catalyst for this reaction, and is significantly more efficient than other screened metal perchlorate hydrates and Brønsted acids such as HOTf and HClO4. The high catalytic property of Hg(ClO4)2 ? 3 H2O originates from the unprecedented dual activation effects of aromatic mercuration, which could generate a strong protic acid to facilitate the generation of a carbocation at the C3‐position of oxindoles and simultaneously form the more reactive nucleophilic reaction partner.  相似文献

Chiral complexes of BINOL‐based ligands with zirconium tert‐butoxide catalyze the Friedel–Crafts alkylation reaction of indoles with β‐trifluoromethyl‐α,β‐unsaturated ketones to give functionalized indoles with an asymmetric tertiary carbon center attached to a trifluoromethyl group. The reaction can be applied to a large number of substituted α‐trifluoromethyl enones and substituted indoles. The expected products were obtained with good yields and ees of up to 99 %.  相似文献

Biaryl and heterobiaryl compounds are important frameworks across a range of fields including pharmaceutical and functional material chemistries. We have accomplished the efficient synthesis of various naphthalene‐linked arenes and heteroarenes as biaryls and heterobiaryls by the FeCl3‐catalyzed Friedel‐Crafts reactions accompanied by the ring‐opening of the 1,4‐epoxy moiety of 1,4‐epoxy‐1,4‐dihydronaphthalenes. Especially, it is noteworthy that 1‐silylated substrates were regioselectively transformed to the 3‐aryl‐1‐silylnaphthalenes and the double Friedel–Crafts reactions using thiophene derivatives could directly produce the corresponding bis‐naphthlated thiophene derivatives.  相似文献

Substituted naphthofurans and benzofurans are easily accessible by treatment of naphthols/substituted phenols with nitroallylic acetates through a substitution–elimination process promoted by cesium carbonate. Reactions between naphthols and aromatic/heteroaromatic‐substituted nitroallylic acetates gave the desired functionalized naphthofurans in high to excellent chemical yields (14–97 %). On the other hand, treatment of phenol derivatives (i.e., 3‐dimethylamino‐, 3‐methoxy‐, and 3,5‐dimethoxyphenol) with various nitroallylic acetates afforded the corresponding benzofurans in moderate to good chemical yields (24–91 %). The reaction proceeded through an interesting Friedel–Crafts SN2′ process followed by intramolecular oxa‐Michael cyclization and subsequent aromatization. A plot of log (k/kH) against Hammett constants σp showed satisfactory linearity with a positive ρ value, indicating that the initial Friedel–Crafts‐type SN2′ process constituted the rate‐determining step. This methodology has been applied to the synthesis of various novel C2 and C3 symmetric bis‐ and trisfurans by using catechol and phloroglucinol as the nucleophilic partners. The reactivity decreased when alkyl‐substituted nitroallylic acetate systems were used. This might be related to the decreased electrophilic character of these substrates.  相似文献

Acid‐catalyzed Friedel–Crafts alkylation of 1,3‐dicarbonyl compounds with electrophilic alcohols, is known to be an effective C? C bond forming reaction. However, until now, this reaction has not been amenable for α‐alkylation of aryl methyl ketones because of the notoriously low nucleophilicities of these compounds. Therefore, α‐alkylation of aryl methyl ketone relies on precious metal catalysts and also, the use of primary alcohols is mandatory. In this study, we found that a system composed of a Fe(OTf)3 catalyst and chlorobenzene solvent is sufficient to promote the title Friedel–Crafts reaction by using benzhydrols as electrophiles. 3,4‐Dihydro‐9‐(2‐hydroxy‐4,4‐dimethyl‐6‐oxo‐1‐cyclohexen‐1‐yl)‐3,3‐dimethyl‐xanthen‐1(2 H)‐one was also applicable as an electrophile in this type of benzylation reaction. On the basis of this result, a three‐component reaction of salicylaldehyde, dimedone, and aryl methyl ketone was also developed, and this provided an efficient way for the synthesis of densely substituted 4H‐chromene derivatives.  相似文献

The highly enantioselective organocatalytic construction of spiroindanes containing an all‐carbon quaternary stereocenter by intramolecular Friedel–Crafts‐type 1,4‐addition is described. The reaction was catalyzed by a cinchonidine‐based primary amine and accelerated by water and p‐bromophenol. A variety of spiro compounds containing quaternary stereocenters were obtained with excellent enantioselectivity (up to 95 % ee). The reaction was applied to the asymmetric formal synthesis of the spirocyclic natural products (?)‐cannabispirenones A and B.  相似文献

A one‐pot synthesis of pyrrolo[1,2‐a]quinolin‐1‐ones has been developed from the reactions of 5‐hydroxy‐1‐arylpyrrolidin‐2‐ones with 1,3‐dicarbonyl compounds under the promotion of H3PO4/P2O5 or HOAc/H2SO4. The pyrrolo[1,2‐a]quinolin‐1‐ones are formed by two‐step reactions, that is, the coupling of N‐acyliminium ion intermediates produced from 5‐hydroxy‐1‐arylpyrrolidin‐2‐ones with 1,3‐dicarbonyls and subsequent Friedel–Crafts reactions of the resulting ketone with the aryl ring.  相似文献

A chiral imidazolidine‐containing NCN/Pd‐OTf catalyst ( C4 ) promoted the nucleophilic addition of unprotected indoles to N‐Boc imines. Using sulfinyl amines as the N‐Boc imine precursors, the combined use of C4 with K2CO3 activated the NH indoles to give chiral 3‐indolyl methanamines with up to 98 % ee. Compared with conventional acid‐catalyzed Friedel–Crafts reactions, this reaction proceeds under mildly basic conditions and is advantageous for the use of acid‐sensitive substrates.  相似文献
