The supercritical fluid extraction (SFE) of Lepidium apetalum seed oil and its anti-oxidant activity were studied. The SFE process was optimized using response surface methodology (RSM) with a central composite design (CCD). Independent variables, namely operating pressure, temperature, time and flow rate were evaluated. The maximum extraction of Lepidium apetalum seed oil by SFE-CO? (about 36.3%) was obtained when SFE-CO? extraction was carried out under the optimal conditions of 30.0 MPa of pressure, 70 °C of temperature, 120 min of extraction time and 25.95 L/h of flow rate. GC-MS analysis showed the presence of four fatty acids in Lepidium apetalum seed oil, with a high content (91.0%) of unsaturated fatty acid. The anti-oxidant activity of the oil was assessed by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging assay and 2,2'-azino- bis(3-ethylbenzthiazoline-6-sulphonic acid) diammonium salt (ABTS) test. Lepidium apetalum seed oil possessed a notable concentration-dependent antioxidant activity, with IC?? values of 1.00 and 3.75 mg/mL, respectively.  相似文献

Rapeseed, soybean and sunflower seeds are used for a comparison of solvent extractions by SFE, ASE, fexIKA and Soxtherm apparatus with the official standard method B-I 5 (87) by the DGF. The optimal extraction parameters for each method are evaluated. The extracts are analysed regarding the composition of tocopherols, as a parameter for mild extraction conditions and the content of free fatty acids and diglycerides, as a parameter for the recovery of more polar lipids. The obtained results for oil content using the optimised methods agree well with the standard method. Differences occur in the results of tocopherol composition and free fatty acid and diglyceride contents. The SFE method shows the highest recovery regarding the tocopherol content. The extraction period by SFE is reduced to about 40 min in contrast to 4 h of the DGF standard method. The other methods also provide considerable time reductions, but showed lower tocopherols and free fatty acid contents.  相似文献

Supercritical fluid extraction (SFE) of daphnoretin, 7-methoxy-daphnoretin and 1,5-diphenyl-1- pentanone from Stellera chamaejasme L. was performed. An orthogonal L9 (3)4 test design was applied to select the optimum extraction parameters including pressure, temperature, modifier and sample particle size on yield using an analytical-scale SFE system. The process was then scaled up by 100 times using a preparative SFE system under the optimized conditions of 25 MPa of pressure, 45 degrees C of temperature, 40-60 mesh of sample particle size and modified CO2 with 20% methanol. The yield of the crude extract from preparative SFE was 2.65%, which contained daphnoretin 25.2%, 7-methoxy-daphnoretin 22.8% and 1,5-diphenyl-1-pentanone 21.1%, respectively. Then the crude extract was successfully isolated and separated by preparative high-speed counter-current chromatography (HSCCC) with a two-phase solvent system composed of n-hexane-ethyl acetate-methanol-water (10:13:13:10, v/v) by increasing the flow-rate of the mobile phase stepwise from 1.0 to 2.0 ml/min after 90 min. The target compounds isolated and purified by HSCCC were analyzed by high-performance liquid chromatography (HPLC). The separation produced total of 69.2mg of daphnoretin at 99.2% purity, 63.4 mg of 7-methoxy-daphnoretin at 98.7% purity and 58.3 mg of 1,5-diphenyl-1-pentanone at 98.1% purity from 300 mg of the crude extract in one-step separation. The recoveries of daphnoretin, 7-methoxy-daphnoretin and 1,5-diphenyl-1-pentanone were 90.8, 91.5 and 90.4%, respectively, in HSCCC isolation step and the chemical structure identification was carried out by MS, 1H NMR and 13C NMR.  相似文献

Supercritical fluid extraction (SFE) of orotinin, orotinin-5-methyl ether and licoagrochalcone B from Patrinia villosa was performed. The optimization of parameters including pressure, temperature, modifier and sample particle size on yield was carried out using an analytical-scale SFE system. The process was then scaled up by 100 times using a preparative SFE system under the optimized conditions of 25 MPa, 45 degrees C, a sample particle size 40-60 mesh and modified CO2 with 20% methanol. The yield of the preparative SFE was 2.82% (crude extract I) and the combined yield of orotinin, orotinin-5-methyl ether and licoagrochalcone B was 0.82 mg/g of dry sample mass. Then the crude extract I was re-dissolved in methanol and methanol soluble fraction (crude extract II, 0.17%) was obtained, which was successfully isolated and separated by a preparative high-speed counter-current chromatography (HSCCC) with a two-phase solvent system composed of n-hexane-ethyl acetate-methanol-water (5:6:6:6, v/v/v/v) by increasing the flow-rate of the mobile phase stepwise from 1.0 to 2.0 ml/min after 3 h. The target compounds isolated and purified by HSCCC were analyzed by high performance liquid chromatography. The separation produced total of 38.2 mg of orotinin at 99.2% purity, 19.8 mg of orotinin-5-methyl ether at 98.5% purity and 21.5 mg of licoagrochalcone B at 97.6% purity from 400 mg of the crude extract in a one-step separation. The recoveries of orotinin, orotinin-5-methyl ether and licoagrochalcone B were 91.1, 91.6 and 90.3%, respectively, and the chemical structure identification was carried out by UV, IR, MS, 1H NMR and 13C NMR.  相似文献

Psoralen and isopsoralen were extracted from Fructus Psoraleae (Psoralea corylitolia L.) by supercritical CO2. The effect of various parameters, i.e., pressure, temperature and sample particle size on yield was investigated with an analytical-scale supercritical fluid extraction (SFE) system to find the optimal conditions. The process was then scaled up by 50 times with a preparative SFE system under the optimized conditions of pressure (26 MPa), temperature (60 degrees C) and a sample particle size of 40-60 mesh. The yield of the preparative SFE was 9.1% and the combined yield of psoralen and isopsoralen was 2.5 mg/g of dry seeds. Psoralen and isopsoralen in the extract were separated and purified by high-speed counter-current chromatography with a two-phase solvent system composed of n-hexane-ethyl acetate-methanol-water (1:0.7:1:0.8, v/v), and the fractions were analyzed by HPLC, MS, 1HNMR and 13C NMR. The structures of the products were further confirmed by comparison with authentic samples (National Institute of the Control of Pharmaceutical and Biological Products, Beijing, China).  相似文献

Supercritical fluid extraction (SFE) of the volatile oil from Thymus vulgaris L. aerial flowering parts was performed under different conditions of pressure, temperature, mean particle size and CO2 flow rate and the correspondent yield and composition were compared with those of the essential oil isolated by hydrodistillation (HD). Both the oils were analyzed by GC and GC‐MS and 52 components were identified. The main volatile components obtained were p‐cymene (10.0–42.6% for SFE and 28.9–34.8% for HD), γ‐terpinene (0.8–6.9% for SFE and 5.1–7.0% for HD), linalool (2.3–5.3% for SFE and 2.8–3.1% for HD), thymol (19.5–40.8% for SFE and 35.4–41.6% for HD), and carvacrol (1.4–3.1% for SFE and 2.6–3.1% for HD). The main difference was found to be the relative percentage of thymoquinone (not found in the essential oil) and carvacryl methyl ether (1.0–1.2% for HD versus t?0.4 for SFE) which can explain the higher antioxidant activity, assessed by Rancimat test, of the SFE volatiles when compared with HD. Thymoquinone is considered a strong antioxidant compound.  相似文献

 利用超临界二氧化碳对秋水仙块根(经粉碎)中的秋水仙碱进行了萃取,采用高效液相色谱法对萃取出的秋水仙碱的质量分数进行了测定。实验选择40℃20~40MPa作为超临界苹取的操作条件,采用体积分数为95%的乙醇在索氏提取器中对样品进行了对比苹取实验。结果表明:不加浸泡剂进行浸泡处理的样品中的秋水仙碱很难被超临界二氧化碳萃取,在40℃,35MPa条件下,消耗1.28mol的二氧化碳只得到3%的萃取率。加入极少量的有机溶剂浸泡处理样品15min后再进行超临界萃取,可以极大程度地提高秋水仙碱的萃取率。  相似文献

Essential fatty acids were extracted from Echium amoenum (Boraginaceae) seed oil via supercritical carbon dioxide and the results were compared with conventional Soxhlet method as the base case of 100% recovery. The response surface methodology was used to optimize the effective extraction parameters. The chemical composition of recovered oil was analyzed by polar and non-polar gas chromatograph-flame ionization detector columns. The experimental results indicated that echium seed contained 25 wt% oil and the maximum extraction oil recovery of 92% was obtained via supercritical CO(2) at optimal operating conditions (43°C, 280 bar, 1.5 mL/min, 25 min static time and 130 min dynamic time). At similar operating conditions, applying 2.5 and 5 mol% ethanol as a modifier enhanced the recovery to 96 and 112%, respectively. The results showed that supercritical fluid extraction is a viable technique for separation of constituents such as γ-linolenic acid (7-8%), palmitic acid (6-7%), stearic acid (3-4%), oleic acid (12-13%), linoleic acid (19-20%), α-linolenic acid (40-41%) and stearidonic acid (8-9%) from Boraginaceae.  相似文献

Supercritical fluid extraction (SFE) of the volatile oil from Satureja montana L. was performed under different conditions of pressure (90 and 100 bar), temperature (40 and 50°C), mean particle sizes (0.4, 0.6 and 0.8 mm) and CO2 flow rate (0.8, 1.1 and 1.3 kg/h) to understand the influence of these parameters on the composition and yield of this oil. The results were compared with those obtained for the essential oil isolated by hydrodistillation (HD). The volatile and the essential oil were analysed by GC and GC‐MS. The main compounds are carvacrol (52.2–62.0% for HD vs. 41.7–64.5% for SFE), thymol (8.6–11.0% for HD vs. 6.0–11.3% for SFE), p‐cymene (6.9–12.8% for HD vs. 6.0–17.8% for SFE), γ‐terpinene (6.4–9.4% for HD vs. 2.3–6.0% for SFE) and β‐bisabolene (2.0–2.7% for HD vs. 2.2–3.5% for SFE). The major difference between SFE and HD was the relative amount of thymoquinone, an oxygenated monoterpene with important biological activities, which can be ten‐fold higher in volatile oil (1.6–3.0 for SFE vs. 0.2% for HD). The morphology of the glandular trichomes of S. montana and the effect of the grinding process on them was also evaluated by SEM.  相似文献

Components of hamster feces ranging from low molecular weight fatty acids through the expected range of triglycerides have been eluted in a single SFC run with simultaneous pressure and temperature programming. Temperature programming from 140°C to 240°C was required to provide optimum conditions for separation of the fatty acids and to move the elution region of the sterol esters away from that of the triglycerides. Data from chemical ionization and electron impact mass spectrometry of compounds separated by SFC were used to confirm identities suggested by retention measurements and to provide tentative identities of unknown compounds. SFC with flame ionization detection was used to compare Soxhlet extraction, off-line supercritical fluid extraction (SFE), and on-line SFE of the feces. Although samples obtained by Soxhlet extraction and SFE produced very similar chromatograms, SFE required far less time and consumed much smaller quantities of organic solvent.  相似文献

Supercritical fluid extraction (SFE) of the volatile oil from Santolina chamaecyparissus L. flower heads was performed under different conditions of pressure, temperature, mean particle size and CO2 flow rate. This oil was compared with the essential oil isolated by hydrodistillation (HD). The SFE volatile and essential oils were analysed by GC and GC‐MS. The range of the main volatile components obtained with HD and SFE were, respectively: 1,8‐cineole (25–30% and 7–48%), camphor (7–9% and 8–14%), borneol (7–8% and 2–11%), terpinen‐4‐ol (6–7% and 1–4%), terpinolene (1–4% and 1–7%) and isobornyl acetate (1–2% and 1–11%). The chemical composition of the extracts was greatly influenced by the conditions of pressure and temperature used. In fact, it was possible to enrich the sesquiterpene fraction by increasing the pressure from 8 to 9 MPa, while changing the temperature from 40 to 50°C at 90 bar enriched of the volatiles in n‐alkanes.  相似文献

Superheated hexane extraction has been tested for obtaining fatty acids from grape seed and compared with conventional Soxhlet and hot hexane extractions. Seeds from grape residues from a winery were dried for 46 h at 105 °C, milled and sieved by particle size (d < 0.42 mm, 0.42 < d < 0.84 mm and d > 0.84 mm). An optimization study of influential variables on superheated hexane extraction (namely extraction time, temperature, pression, particle size and sample amount) was carried out by a multivariate approach. All the extracts were concentrated in a rotary evaporator and dried by adding 1 g of Na2SO4. Then, 2 ml of the dried extract were subjected to reaction with 1 ml of a 0.5 M solution of sodium methylate in methanol to obtain fatty acid methyl esters (FAMEs). After derivatization, FAMEs were quantified by GC-FID. The results show that the optimal conditions for superheated hexane extraction are: time extraction, 10 min; temperature, 80 °C; pressure, 40 bar; particle size, d < 0.42 mm; amount of sample, 0.4 g. Under these conditions, around 84% of the fatty acids (out of the amount obtained by Soxhlet extraction) is extracted. Comparison with Soxhlet and hot hexane extractions showed that the percentages of FAMEs are similar in all the extracts and they agree with the data in the bibliography.  相似文献

The distribution of fatty acids in a triacylglycerol is of great importance from nutritional, biochemical, quality and technological points of view. The ratio of triacylglycerol positional isomers containing two linoleic acid (18:2) and one oleic acid (18:1) moieties--namely, 1(3),2-dilinoleoyl-3(1)-oleoyl glycerol (LLO) and 1,3-dilinoleoyl-2-oleoyl glycerol (LOL)--were quantified in grape seed, olive, pumpkin seed, soybean, sunflower and wheat germ oils by high-performance liquid chromatography/atmospheric pressure chemical ionization mass spectrometry (HPLC/APCI-MS) in selected ion monitoring (SIM) mode. Relative LOL contents (LOL/(LLO+LOL)) of the oils were calculated from the mass abundances of the [LL]+ and [LO]+ diacylglycerol fragment ions ([M+H-RCOOH]+) using a calibration curve. The calibration curve of the relative diacylglycerol mass abundances was measured in SIM mode. The relative LOL contents were found to be relatively consistent for each oil variety. The relative LOL content in grape seed, sunflower, pumpkin seed, soybean and wheat germ oils accounted for 44.2 +/- 2.6, 26.8 +/- 3.2, 16.7 +/- 4.6, 15.9 +/- 2.9 and 13.9 +/- 4.3%, respectively. Only olive oils contained practically 100% of the LLO isomer. These results indicate that the unsaturated fatty acids such as linoleic and oleic acids have 'non-random' distribution patterns in various oils.  相似文献

Colombian mango production, which exceeded 261,000 t in 2020, generates about 40% of the whole fruit as solid waste, of which more than 50% are seed kernels (over 52,000 t solid by-product); though none is currently used for commercial purposes. This study reports the results of the supercritical carbon dioxide (scCO2) extraction of an oil rich in essential fatty acids (EFAs) from revalorized mango seed kernels and the optimization of the process by the Response Surface Methodology (RSM). In pilot-scale scCO2 experiments, pressure (23–37 MPa) and temperature (52–73 °C) were varied, using 4.5 kg of CO2. The highest experimental oil extraction yield was 83 g/kg (37 MPa and 63 °C); while RSM predicted that 84 g/kg would be extracted at 35 MPa and 65 °C. Moreover, by fine-tuning pressure and temperature it was possible to obtain an EFA-rich lipid fraction in linoleic (37 g/kg) and α-linolenic (4 g/kg) acids, along with a high oleic acid content (155 g/kg), by using a relatively low extraction pressure (23 MPa), which makes the process a promising approach for the extraction of oil from mango waste on an industrial scale, based on a circular economy model.  相似文献

The lipid fraction of Opuntia ficus indica seeds was extracted and analyzed for its chemical and physical properties such as acid value, free fatty acid percentage (% FFA), iodine index, peroxide value, and saponification value as well as refractive index and density. The yield of seed oil was calculated as 11.75%. The acid and free fatty acid values indicated that the oil has a fairly low acidity. The triacylglycerols, fatty acids, sterols, and tocopherols were identified and their concentrations determined. The main TAGs were LLL (25.60%), OLL (21.53%), PLL (15.53%), and POL + SLL (12.73%). Linoleic acid (60.69%) was the dominant fatty acid, followed by oleic (21.42%) and palmitic (12.76%) acids, respectively. A high level of sterols making up 16.06 g/kg seed oil was present. The sterol marker, β-sitosterol, accounted for 71.60% of the total sterol content in the seed oil. Among the tocopherols present in the oil, γ-tocopherol (421.08 mg/kg) was the main constituent.  相似文献

This study examines the supercritical carbon dioxide (SC-CO(2)) extraction of oryzanols contained rice bran oil from powdered rice bran. The extraction efficiencies and concentration factors of oryzanols, free fatty acids and triglycerides in the SC-CO(2) extracts were determined. With top-flow type SC-CO(2) extraction the total oil yield was 18.1% and the extraction efficiencies of oryzanols and triglycerides were 88.5 and 91.3% respectively, when 2750 g CO(2 )was consumed during the extraction of 35 g rice bran powder. The concentration factors of oryzanols and triglycerides in SC-CO(2)-extracted oil were higher than in the Soxhlet n-hexane extracted oil. SC-CO(2) extractions indicated that pressure can be used more effectively than temperature to enhance the extraction efficiency and concentration factor of oryzanols. A two-factor central composite scheme of response surface methodology was employed to determine the optimal pressure (300 bar) and temperature (313 K) for increasing the concentration of oryzanols in the SC-CO(2) extracted oil.  相似文献

The oil yields, fatty acid compositions, and total protein contents of three varieties of Turkish poppy were investigated. Solvent extraction of yellow seed gave the highest oil yield (49.2%), while white seed (36.8%) and blue seed (33.6) showed considerably lower yields. Yellow seed was also subjected to mechanical extraction, yielding 32.3% oil. Solvent extraction of the oilcake from the press gave 17.6% additional oil. Fatty acid compositions of the oils were determined by GC/MS and major components were found to be linoleic (56.4-69.2%), oleic (16.1-19.4%), and palmitic (10.6-16.3%) depending on the color of the seeds. The values of proximate analysis, nutritional elements and heavy metals content were also reported for the most widely cultivated yellow seed.  相似文献

 研究了用超临界流体萃取法直接从脂肪基质的固体样品(大豆粉)中选择性地萃取有机锡化合物的方法。模拟试样萃取结果表明:用较低压力和较高温度的超临界态CO2作流动相时,有机锡达到最大萃取率,而脂肪类物质仅被少量萃取,从而消除了脂肪类物质对超临界流体色谱法测定有机锡的干扰。  相似文献
