We report the synthesis and characterization of two new meso‐ferrocenylporphyrins, with the ferrocenyl unit attached at the para position of a C6F4 group by a C? N bond with ? NH(CH2)2NH? ( 2a ) and ? NH(CH2)4NH? ( 2b ) spacers. Compounds 2a and 2b were characterized through elemental analyses, electrospray ionization high‐resolution mass spectrometry, 1H NMR, UV–visible and fluorescence spectroscopies, electrochemistry and spectroelectrochemistry. The free base porphyrin 2 was included for comparison purposes. The binding ability of 2 and the porphyrin derivatives 2a and 2b with calf thymus DNA was determined using UV–visible and fluorescence spectroscopies. The results suggest that the interaction of these systems most probably occurs through π‐stacking by non‐classical modes involving the partial insertion of the C6F5 ring between adjacent base pairs on DNA and possible hydrogen interaction with the aliphatic ? NH(CH2)nNH? (n = 2 or 4) groups with calf thymus DNA. Moreover, we also demonstrate that porphyrins generate singlet oxygen species and show good photostability after irradiation.  相似文献

Two novel five‐coordinate zinc(II) complexes with the tripod ligand tris(N‐methylbenzimidazol‐2‐ylmethyl)amine (Mentb) and two different α,β‐unsaturated carboxylates, with the composition [Zn(Mentb)(acrylate)] (ClO4)·DMF·1.5CH3OH ( 1 ) and [Zn(Mentb)(cinnamate)](ClO4)·2DMF·0.5CH3OH ( 2 ), were synthesized and characterized by means of elemental analyses, electrical conductivity measurements, IR, UV, and 1H NMR spectra. The crystal structure of two complexes have been determined by a single‐crystal X‐ray diffraction method, and show that the ZnII atom is bonded to a Mentb ligand and a α,β‐unsaturated carboxylate molecule through four N atoms and one O atom, resulting in a distorted trigonal‐bipyramidal coordination [τ( 1 ) = 0.853, τ( 2 ) = 0.855], with approximate C3 symmetry.  相似文献

A bimolecular rate constant,kDHO, of (29 ± 9) × 10?12 cm3 molecule?1 s?1 was measured using the relative rate technique for the reaction of the hydroxyl radical (OH) with 3,5‐dimethyl‐1‐hexyn‐3‐ol (DHO, HC?CC(OH)(CH3)CH2CH(CH3)2) at (297 ± 3) K and 1 atm total pressure. To more clearly define DHO's indoor environment degradation mechanism, the products of the DHO + OH reaction were also investigated. The positively identified DHO/OH reaction products were acetone ((CH3)2C?O), 3‐butyne‐2‐one (3B2O, HC?CC(?O)(CH3)), 2‐methyl‐propanal (2MP, H(O?)CCH(CH3)2), 4‐methyl‐2‐pentanone (MIBK, CH3C(?O)CH2CH(CH3)2), ethanedial (GLY, HC(?O)C(?O)H), 2‐oxopropanal (MGLY, CH3C(?O)C(?O)H), and 2,3‐butanedione (23BD, CH3C(?O)C(?O)CH3). The yields of 3B2O and MIBK from the DHO/OH reaction were (8.4 ± 0.3) and (26 ± 2)%, respectively. The use of derivatizing agents O‐(2,3,4,5,6‐pentalfluorobenzyl)hydroxylamine (PFBHA) and N,O‐bis(trimethylsilyl)trifluoroacetamide (BSTFA) clearly indicated that several other reaction products were formed. The elucidation of these other reaction products was facilitated by mass spectrometry of the derivatized reaction products coupled with plausible DHO/OH reaction mechanisms based on previously published volatile organic compound/OH gas‐phase reaction mechanisms. © 2004 Wiley Periodicals, Inc. The NiII‐mediated tautomerization of the N‐heterocyclic hydrosilylcarbene L2Si(H)(CH2)NHC 1 , where L2=CH(C?CH2)(CMe)(NAr)2, Ar=2,6‐iPr2C6H3; NHC=3,4,5‐trimethylimidazol‐2‐yliden‐6‐yl, leads to the first N‐heterocyclic silylene (NHSi)–carbene (NHC) chelate ligand in the dibromo nickel(II) complex [L1Si:(CH2)(NHC)NiBr2] 2 (L1=CH(MeC?NAr)2). Reduction of 2 with KC8 in the presence of PMe3 as an auxiliary ligand afforded, depending on the reaction time, the N‐heterocyclic silyl–NHC bromo NiII complex [L2Si(CH2)NHCNiBr(PMe3)] 3 and the unique Ni0 complex [η2(Si‐H){L2Si(H)(CH2)NHC}Ni(PMe3)2] 4 featuring an agostic Si? H→Ni bonding interaction. When 1,2‐bis(dimethylphosphino)ethane (DMPE) was employed as an exogenous ligand, the first NHSi–NHC chelate‐ligand‐stabilized Ni0 complex [L1Si:(CH2)NHCNi(dmpe)] 5 could be isolated. Moreover, the dicarbonyl Ni0 complex 6 , [L1Si:(CH2)NHCNi(CO)2], is easily accessible by the reduction of 2 with K(BHEt3) under a CO atmosphere. The complexes were spectroscopically and structurally characterized. Furthermore, complex 2 can serve as an efficient precatalyst for Kumada–Corriu‐type cross‐coupling reactions.  相似文献

Bis(N, N′‐dialkyldithiocarbamato)antimony(III) alkylenedithiophosphates of the type [R2NCS2]2 SbS(S)POGO [where NR2 = N(CH3)2, N(C2H5)2 and N(CH2)4; G = ? CH2? C(C2H5)2? CH2? , ? CH2? C(CH3)2? CH2? , ? CH(CH3)? CH(CH3)? and ? C(CH3)2? C(CH3)2? ] were synthesized and characterized by physico‐chemical, spectral [UV, IR and NMR (1H, 13C and 31P)] and thermal (TG, DTA and DSC) analysis. The TG decomposition analysis step of the complex indicated the formation of Sb2S3 as the final product. The first endothermic peak in DSC indicated the melting point of the complexes. These complexes were screened for their antimicrobial activities using the disk diffusion method. All the complexes showed good activity as antibacterial and antifungal agents on some selected bacterial and fungal strains, which increased on increasing the concentration. Chloroamphenicol and terbinafin were used as standards for comparison. Copyright © 2007 John Wiley & Sons, Ltd.  相似文献

A novel hydrazone ligand derived from condensation reaction of 3‐hydroxy‐2‐naphthoic hydrazide with dehydroacetic acid, and its Ni(II), Cu(II) and Co(II) complexes were synthesized, characterized by spectroscopic, elemental analyses, magnetic susceptibility and conductivity methods, and screened for antimicrobial, DNA binding and cleavage properties. Spectroscopic analysis and elemental analyses indicated the formula, [MLCl2], for the complexes; square planar geometry for the nickel, and tetrahedral geometry for copper and cobalt complexes. The non‐electrolytic natures of the complexes in Dimethyl Sulphoxide (DMSO) were confirmed by their molar conductance values in the range of 6.11–14.01 Ω?1cm2mol?1. The copper complex had the best antibacterial activity against Staphylococcus aureus (ATCC 29213). DNA cleavage activities of the compounds, evaluated on pBR322 DNA, by agarose gel electrophoresis, in the presence and absence of oxidant (H2O2) and free radical scavenger (DMSO), indicated no activity for the ligand, and moderate activity for the complexes, with the copper complex cleaving pBR322 DNA more efficiently in the presence of H2O2. When the complexes were evaluated for antibacterial and A‐DNA activity using Molecular docking technique, the copper complex was found to be most effective against Gram‐positive (S. aureus) bacteria. [CuLCl2] showed good hydrogen bonding interaction with the major‐groove (C2.G13 base pair) of A‐DNA. Density functional theory (DFT) calculations of the structural and electronic properties of the complexes revealed that [CuLCl2] had a smaller HOMO‐LUMO gap, suggesting a higher tendency to donate electrons to electron‐accepting species of biological targets.  相似文献

In both title compounds, C18H24N2O2, (Ia), and C18H26N2O22+·2ClO4, (II), respectively, the two aryl rings are strictly parallel, with an inversion centre lying at the mid‐point of each central CH2—CH2 bond. Molecules in (Ia) are linked into two‐dimensional layers by N—H...O hydrogen bonds. The component ions in (II) are joined together by a combination of N/O/C—H...O hydrogen bonds and C—H...π and anion...π interactions, forming a three‐dimensional network. A structural understanding of (Ia) and (II) may provide some useful information about how and why their metal–organic complexes display various biological activities and function in catalytic processes.  相似文献

The first four‐coordinate methanediide/alkyl lutetium complex (BODDI)Lu2(CH2SiMe3)22‐CHSiMe3)(THF)2 (BODDI=ArNC(Me)CHCOCHC(Me)NAr, Ar=2,6‐iPr2C6H3) ( 1 ) was synthesized by a thermolysis methodology through α‐H abstraction from a Lu–CH2SiMe3 group. Complex 1 reacted with equimolar 2,6‐iPrC6H3NH2 and Ph2C?O to give the corresponding lutetium bridging imido and oxo complexes (BODDI)Lu2(CH2SiMe3)22N‐2,6‐iPr2C6H3)(THF)2 ( 2 ) and (BODDI)Lu2(CH2SiMe3)22‐O)(THF)2 ( 3 ). Treatment of 3 with Ph2C?O (4 equiv) caused a rare insertion of Lu–μ2‐O bond into the C?O group to afford a diphenylmethyl diolate complex 4 . Reaction of 1 with PhN=C?O (2 equiv) led to the migration of SiMe3 to the amido nitrogen atom to give complex (BODDI)Lu2(CH2SiMe3)2‐μ‐{PhNC(O)CHC(O)NPh(SiMe3)‐κ3N,O,O}(THF) ( 5 ). Reaction of 1 with tBuN?C formed an unprecedented product (BODDI)Lu2(CH2SiMe3){μ2‐[η22tBuNC(=CH2)SiMe2CHC?NtBu‐κ1N]}(tBuN?C)2 ( 6 ) through a cascade reaction of N?C bond insertion, sequential cyclometalative γ‐(sp3)‐H activation, C?C bond formation, and rearrangement of the newly formed carbene intermediate. The possible mechanistic pathways between 1 , PhN?C?O, and tBuN?C were elucidated by DFT calculations.  相似文献

Synthesis and characterization of mononuclear transition metal complexes viz., Co(II), Ni(II), Cu(II) and Zn(II) with a newly designed ligand, (E)‐2‐benzamido‐N'‐(1‐(2‐hydroxy‐6‐methyl‐4‐oxo‐4H‐pyran‐3‐yl) ethylidene) benzohydrazide ( H 2 L ) are reported. Molecular structures of H 2 L , Ni(II) and Cu(II) complexes were determined by single‐crystal X‐ray diffraction studies. The structures were stabilized by various intra/inter‐molecular H‐bonding, C‐H···π and π···π stacking interactions. H 2 L exists in zwitterionic form and acts in a monoanionic manner. Ligand/metal ratio was 2:1 for cobalt, nickel and zinc, whereas 1:1 for the copper complex. Co(II), Ni(II) and Zn(II) complexes display distorted octahedral geometry, while the Cu(II) complex shows distorted square pyramidal geometry around the metal ion. Hirshfeld surface analysis and 2D fingerprint plots revealed that H 2 L and its complexes were supported mainly by H?H, O?H and C?H intermolecular interactions. The synthesized compounds were screened for in vitro anti‐inflammatory activity by gelatin zymography and the activity was comparable with tetracycline. Their cleavage behavior towards calf thymus DNA has been studied using agarose gel electrophoresis method. H 2 L and Cu(II) complex were selected by National Cancer Institute (NCI) for in vitro single dose testing in the full NCI 60 cell lines panel assay. Finally, molecular docking simulation effectively proves the binding of all the synthesized compounds at cyclooxygenase‐2 (COX‐2) active sites.  相似文献

Schiff bases of 2‐(phenylthio)aniline, (C6H5)SC6H4N?CR (R = (o‐CH3)(C6H5), (o‐OCH3)(C6H5) or (o‐CF3)(C6H5)), and their palladium complexes (PdLCl2) were synthesized. The compounds were characterized using 1H NMR and 13C NMR spectroscopy and micro analysis. Also, electrochemical properties of the ligands and Pd(II) complexes were investigated in dimethylformamide–LiClO4 solution with cyclic and square wave voltammetry techniques. The Pd(II) complexes showed both reversible and quasi‐reversible processes in the ?1.5 to 0.3 V potential range. The synthesized Pd(II) complexes were evaluated as catalysts in Mizoroki–Heck and Suzuki–Miyaura cross‐coupling reactions. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献

A series of novel cytotoxic compounds, [Mn(cpt)2], [Zn(tpt)(H2O)2]?DMA?2(H2O) and [Cu(tpt)]?DMA (cpt = 4′‐(4‐carboxyphenyl)‐2,2′:6′,2″‐terpyridine, tpt = 4‐(2,4,6‐tricarboxylphenyl)‐2,2′:6′,2″‐terpyridine, DMA = (CH3)2NH), were isolated and characterized. The structures of these complexes were characterized using single‐crystal X‐ray diffraction. The mode and extent of binding between fish sperm DNA and the complexes were investigated using fluorescence spectroscopy and molecular docking. These results indicate the ability of the complexes to bind to DNA with different binding affinities. The binding of the Zn(II) complex with DNA is stronger than that of the corresponding Cu(II) analogue, which is expected due to the z* effect and geometry. The ability of these complexes to cleave pBR322 plasmid DNA was demonstrated using gel electrophoresis assay, showing that the complexes have effective DNA cleavage activity. In addition, the cytotoxic effects of these complexes were examined on HeLa cells (human cervix epithelia carcinoma cells) in vitro. The three complexes exhibit different cytotoxic effects and decent cancer cell inhibitory rate. This means that the structures and type of metal have a great influence on the activity of these novel complexes.  相似文献

Intramolecular H‐atom transfer in model peptide‐type radicals was investigated with high‐level quantum‐chemistry calculations. Examination of 1,2‐, 1,3‐, 1,5‐, and 1,6[C ? N]‐H shifts, 1,4‐ and 1,7[C ? C]‐H shifts, and 1,4[N ? N]‐H shifts (Scheme 1), was carried out with a number of theoretical methods. In the first place, the performance of UB3‐LYP (with the 6‐31G(d), 6‐31G(2df,p), and 6‐311+G(d,p) basis sets) and UMP2 (with the 6‐31G(d) basis set) was assessed for the determination of radical geometries. We found that there is only a small basis‐set dependence for the UB3‐LYP structures, and geometries optimized with UB3‐LYP/6‐31G(d) are generally sufficient for use in conjunction with high‐level composite methods in the determination of improved H‐transfer thermochemistry. Methods assessed in this regard include the high‐level composite methods, G3(MP2)‐RAD, CBS‐QB3, and G3//B3‐LYP, as well as the density‐functional methods B3‐LYP, MPWB1K, and BMK in association with the 6‐31+G(d,p) and 6‐311++G(3df,3pd) basis sets. The high‐level methods give results that are close to one another, while the recently developed functionals MPWB1K and BMK provide cost‐effective alternatives. For the systems considered, the transformation of an N‐centered radical to a C‐centered radical is always exothermic (by 25 kJ ? mol?1 or more), and this can lead to quite modest barrier heights of less than 60 kJ ? mol?1 (specifically for 1,5[C ? N]‐H and 1,6[C ? N]‐H shifts). H‐Migration barriers appear to decrease as the ring size in the transition structure (TS) increases, with a lowering of the barrier being found, for example when moving from a rearrangement proceeding via a four‐membered‐ring TS (e.g., the 1,3[C ? N]‐H shift, CH3? C(O)? NH..CH2? C(O)? NH2) to a rearrangement proceeding via a six‐membered‐ring TS (e.g., the 1,5[C ? N]‐H shift, .NH? CH2? C(O)? NH? CH3 → NH2? CH2? C(O)? NH? CH2.).  相似文献

New organotin(IV) derivatives containing the anionic ligands bis(3,5‐dimethylpyrazol‐1‐yl)dithioacetate [LCS2] and bis(3,5‐dimethylpyrazol‐1‐yl)acetate [LCO2] have been synthesized from reaction between (CH3)2SnCl2 and lithium salts of the ligands. Mononuclear complexes of the type {[LCX2](CH3)2SnCl} (X = S or O) have been obtained and fully characterized by elemental analyses and FT‐IR in the solid state and by NMR (1H, 13C and 119Sn) spectroscopy, conductivity measurements and electrospray ionization mass spectrometry in solution. The acute toxicity of new organotin(IV) derivatives on rat was studied, comparing their effect with those of dimethyltin chloride (CH3)2SnCl2. The comparison of LD50 of organotin(IV) complexes and (CH3)2SnCl2 administered intraperitoneally, as a single dose, evaluated in vivo on rats, showed that toxicity decreases as follows: (CH3)2SnCl2 > LCO2 > LCS2. The effect of these organotin(IV) complexes on DNA was evaluated in vitro and in vivo on rats treated with different doses of these compounds (1/20 LD50 and 1/100 LD50). The lymphocyte DNA status was assessed by the comet assay, a rapid and sensitive single‐cell electrophoresis technique, used to detect primary DNA damage in individual cells. After 36 h from the start of treatment the two new organotin(IV) derivatives induced a significant rise in comet assay parameters, indicating an increasing presence of damaged DNA. Mixed sulfur donor ligand complexes of the type bismuth(III) bis(N,N‐dialkyldithiocarbamato) alkylenedithiophosphate, [R2NCS2]2BiS2POGO [where R = CH3 and C2H5; G = ‐CH2‐C(C2H5)2‐CH2‐, ‐CH2‐C(CH3)2‐CH2‐, ‐CH(CH3)‐CH(CH3)‐ and ‐C(CH3)2‐C(CH3)2‐] were synthesized in 1:1 molar ratio of bismuth(III) bis(N,N‐dialkyldithiocarbamate) chloride and ammonium alkylenedithiophosphate in refluxing benzene and characterized by melting point, molecular weight determinations, elemental analysis (C, H, N, Bi and S) and spectral [UV, IR,NMR (1H,13C and 31P) and powder X ray diffraction] studies; all these studies were in good agreement with the synthesized complexes. These newly synthesized derivatives are yellow and brown colored solids and are soluble in common organic solvents like benzene, chloroform, dichloromethane and DMF. Based on the physicochemical and spectral studies, a tentative structure of these newly synthesized complexes was assigned and the average particle size of the synthesized complexes determined by powder XRD, showing that nano range polycrystalline particles were formed with a monoclinic crystal system. These complexes were also screened for their antimicrobial activities using the well diffusion method. The free ligands as well as their mixed metal complexes were tested in vitro against four bacterial strains: two Gram‐positive, Staphylococcus aureus (ATCC 9144) (G+) and Bacillus subtilis (ATCC 6051), (G+) and two Gram‐negative, Escherichia coli (ATCC 9637) (G?) and Pseudomonas aeruginosa (ATCC 25619) (G?) to assess their antimicrobial properties. The results were indeed positive and exhibited good antibacterial effects. Chloroamphenicol used as a standard for comparison and synthesized complexes showed good antibacterial effects over chloroamphenicol. On the basis of these studies, the synthesized complexes help to understand the different structural and biological properties of main group elements with sulfur donor ligands. A new 1,3,4‐oxadiazole‐containing bispyridyl ligand, namely 5‐(pyridin‐4‐yl)‐3‐[2‐(pyridin‐4‐yl)ethyl]‐1,3,4‐oxadiazole‐2(3H)‐thione (L), has been used to create the novel complexes tetranitratobis{μ‐5‐(pyridin‐4‐yl)‐3‐[2‐(pyridin‐4‐yl)ethyl]‐1,3,4‐oxadiazole‐2(3H)‐thione}zinc(II), [Zn2(NO3)4(C14H12N4OS)2], (I), and catena‐poly[[[dinitratocopper(II)]‐bis{μ‐5‐(pyridin‐4‐yl)‐3‐[2‐(pyridin‐4‐yl)ethyl]‐1,3,4‐oxadiazole‐2(3H)‐thione}] nitrate acetonitrile sesquisolvate dichloromethane sesquisolvate], {[Cu(NO3)(C14H12N4OS)2]NO3·1.5CH3CN·1.5CH2Cl2}n, (II). Compound (I) presents a distorted rectangular centrosymmetric Zn2L2 ring (dimensions 9.56 × 7.06 Å), where each ZnII centre lies in a {ZnN2O4} coordination environment. These binuclear zinc metallocycles are linked into a two‐dimensional network through nonclassical C—H...O hydrogen bonds. The resulting sheets lie parallel to the ac plane. Compound (II), which crystallizes as a nonmerohedral twin, is a coordination polymer with double chains of CuII centres linked by bridging L ligands, propagating parallel to the crystallographic a axis. The CuII centres adopt a distorted square‐pyramidal CuN4O coordination environment with apical O atoms. The chains in (II) are interlinked via two kinds of π–π stacking interactions along [01]. In addition, the structure of (II) contains channels parallel to the crystallographic a direction. The guest components in these channels consist of dichloromethane and acetonitrile solvent molecules and uncoordinated nitrate anions.  相似文献

Two novel chiral ruthenium(II) complexes, Δ‐[Ru(bpy)2(dmppd)]2+ and Λ‐[Ru(bpy)2(dmppd)]2+ (dmppd = 10,12‐dimethylpteridino[6,7‐f] [1,10]phenanthroline‐11,13(10H,12H)‐dione, bpy = 2,2′‐bipyridine), were synthesized and characterized by elemental analysis, 1H‐NMR and ES‐MS. The DNA‐binding behaviors of both complexes were studied by UV/VIS absorption titration, competitive binding experiments, viscosity measurements, thermal DNA denaturation, and circular‐dichroism spectra. The results indicate that both chiral complexes bind to calf‐thymus DNA in an intercalative mode, and the Δ enantiomer shows larger DNA affinity than the Λ enantiomer does. Theoretical‐calculation studies for the DNA‐binding behaviors of these complexes were carried out by the density‐functional‐theory method. The mechanism involved in the regulating and controlling of the DNA‐binding abilities of the complexes was further explored by the comparative studies of [Ru(bpy)2(dmppd)]2+ and of its parent complex [Ru(bpy)2(ppd)]2+ (ppd = pteridino[6,7‐f] [1,10]phenanthroline‐11,13 (10H,12H)‐dione).  相似文献

The aluminum complexes containing two iminophenolate ligands of the type (p‐XC6H4NCHC6H4O‐o)2AlR' (R′=Me ( 3, 4 ) or R′=O(CH2)4OCH=CH2 ( 5, 6 ), X=H ( 3, 5 ), F( 4, 6 )) were synthesized and characterized by 1H, 13C NMR spectroscopy, and X‐ray crystallography. The reaction of AlMe3 with two equivalents of substituted iminophenols gave five‐coordinated {ONR}2AlMe ( 3, 4 ) complexes. Subsequent reaction of these methyl complexes with unsaturated alcohol, HO(CH2)4OCH=CH2, resulted in target compounds 5 and 6 in a good yield. It was shown that the complexes ( 3 ‐ 6 ) are monomeric in solution (NMR) and in solid state (X‐ray analysis). The catalytic activity of the complexes 5 and 6 towards ring‐opening polymerization (ROP) of ?‐caprolactone and d,l ‐lactide was assessed. Complex 5 showed higher activity as compared with 6 , while both of these catalysts induced controlled homo‐ and copolymerization to afford the macromonomers with high content of vinyl ether end groups (Fn > 80%) in a broad range of molecular weights (Mn = 4000–30,000 g mol?1) with relatively narrow MWD (Mw/Mn = 1.1–1.5). © 2014 Wiley Periodicals, Inc. Four new Ag(I)–N‐heterocyclic carbene (NHC) complexes ( 5 – 8 ) bearing symmetrically substituted NHC ligands have been synthesized starting from the corresponding benzimidazolium bromide salts which are accessible in a single step from N ‐substituted benzimidazoles (N ‐alkyl and N ‐aryl) and subsequently reacted with the basic metal source Ag2O in acetonitrile–methanol. These compounds were characterized using elemental analyses, 1H NMR, 13C NMR, Fourier transform infrared and UV–visible spectroscopic techniques, and molar conductivity. Single‐crystal structural studies for complex 5 show that the Ag(I) centre has a perfectly linear C–Ag–C coordination, with quasi‐parallel pairs of aromatic benzimidazole planes. All the complexes interact with Aedes albopictus DNA via intercalation mode by a large hypochromicity of 22 and 27% and smaller hypochromicity of 16 and 19%. Furthermore, all complexes exhibit efficient DNA cleavage activity via a non‐oxidative mechanistic pathway. The DNase activities of the test compounds revealed a time‐ and concentration‐dependent activity pattern. The Ag(I)–NHC complexes showed considerably higher DNA cleavage activity compared to their respective benzimidazolium salts at a lower concentration. The DNA cleavage of these complexes changed from a moderate effect to a good one, corresponding to the increasing lipophilicity order of the complexes as 5  <  6  <  7  <  8 (1.02, 1.05, 1.78 and 2.06 for 5 – 8 , respectively). This order is further corroborated with the DNA binding study, but with the exception of complex 5 , which shows a better binding ability for DNA (K b = 3.367 × 106) than complexes 6 – 8 (6.982 × 105, 8.376 × 105 and 1.223 × 106, respectively).  相似文献

Four new monomeric Pd (II) complexes with formulas [Pd(C,N)‐(2′‐NH2C6H4)C6H4 (N3)(L)] ( A ), ( B ) and [Pd(C,N)‐C6H4CH2NH(C4H9)(N3)(L)] ( C ), ( D ), [L = isonicotinamide for ( A ) and ( C ), L = 4‐N,N‐dimethylaminopyridine for ( B ) and ( D )] have been synthesized using four initial dimers [Pd2{(C,N)‐(2′‐NH2C6H4)C6H4}2(μ‐OAc)2] ( 1 ), [Pd2{(C,N)‐ (2′‐NH2C6H4)C6H4}2(μ‐N3)2] ( 3 ) for A and C , and [Pd2{(C,N)‐C6H4CH2NH(C4H9)}2(μ‐OAc)2] ( 2 ) and [Pd2{(C,N)‐C6H4CH2NH(C4H9)}2(μ‐N3)2] ( 4 ) for B and D . Then synthesized complexes have been characterized by Fourier transform‐infrared, NMR spectroscopy and thermal gravimetric‐differential thermal analysis. Furthermore, UV–Vis spectroscopy, fluorescence spectroscopy, circular dichroism (CD) and helix melting temperature measurements have been employed to study the binding interaction of them with calf thymus‐deoxyribonucleic acid (DNA). The results reveal that all synthesized complexes can interact with DNA via groove‐binding mode. Bovine serum albumin (BSA)‐binding studies have been carried out using UV–Vis spectroscopy, emission titration and CD. However, competitive binding studies using warfarin, ibuprofen and digoxin on site markers demonstrated that the complexes bind to different sites on BSA. The results also indicated that the binding site was mainly located within site‐III for complex A , and site‐I for complexes B , C and D of BSA. In addition, molecular docking studies have been executed to determine the binding site of the DNA and BSA with complexes. Eventually, in vitro cytotoxicity of synthesized palladium complexes and cisplatin were carried out against human promyelocytic leukemia cancer (Hela) and breast cancer (MCF‐7) cell lines. Pursuant to the IC50 values, the cytotoxicity of complexes against MCF‐7 was more than Hela.  相似文献

Four Ru(II) complexes with tridentate ligands viz. (4-hydroxy-N′-(pyridin-2-yl-ethylene) benzohydrazide [Ru(L1)(PPh3)2(Cl)] (1), N′-(pyridin-2-yl-methylene) nicotinohydrazide [Ru(L2)(PPh3)2(Cl)] (2), N′-(1H-imidazol-2-yl-methylene)-4-hydroxybenzohydrazide [Ru(L3)(PPh3)2(Cl)] (3), and N′-(1H-imidazol-2-yl-methylene) nicotinohydrazide [Ru(L4)(PPh3)2(Cl)] (4) have been synthesized and characterized. The methoxy-derivative of L3H (abbreviated as L3H*) exists in E configuration with torsional angle of 179.4° around C7-N8-N9-C10 linkage. Single crystal structures of acetonitrile coordinated ruthenium complexes of 1 and 3 having compositins as [Ru(L1)(PPh3)2(CH3CN)]Cl (1a) and [Ru(L3)(PPh3)2(CH3CN)]Cl (3a) revealed coordination of tridentate ligands with significantly distorted octahedral geometry constructed by imine nitrogen, heterocyclic nitrogen, and enolate amide oxygen, forming a cis-planar ring with trans-placement of two PPh3 groups and a coordinated acetonitrile. Ligands (L1H-L4H) and their ruthenium complexes (1–4) are characterized by 1H, 13C, 31P NMR, and IR spectral analysis. Ru(II) complexes have reversible to quasi-reversible redox behavior having Ru(II)/Ru(III) oxidation potentials in the range of 0.40–0.71 V. The DNA binding constants determined by absorption spectral titrations with Herring Sperm DNA (HS-DNA) reveal that L4H and 1 interact more strongly than other ligands and Ru(II) complexes. Complexes 1–3 exhibit DNA cleaving activity possibly due to strong electrostatic interactions while 4 displays intercalation.  相似文献
