A molecularly imprinted polymer (MIP) has been prepared for the first time with ciprofloxacin (CIPRO) as the template molecule, via a noncovalent synthetic procedure. Prior to its use as a sorbent in SPE, the MIP was evaluated chromatographically to confirm that it was indeed molecularly imprinted. The MIP was then used to extract CIPRO selectively from urine samples by means of a two-step SPE procedure in which a commercial Oasis cartridge and a molecularly imprinted SPE cartridge were combined in series. This approach allowed the matrix compounds present in the samples to be removed effectively. The urine extracts obtained after this two-step SPE procedure was applied were relatively clean compared to the original samples, and this made it possible to inject directly the extracts into a mass spectrometer and thus quantify CIPRO in urine samples at low levels and reduce the time of analysis.  相似文献

A simple, selective, and reproducible molecularly imprinted SPE coupled with HPLC method was developed for monitoring quinoxaline‐1,4‐dioxides in feeds. Molecularly imprinted polymers were synthesized in methanol using mequindox (MEQ) as template molecule and acrylamide as functional monomer by bulk polymerization. Under the optimum SPE conditions, the novel polymer sorbents can selectively extract and enrich carbadox, MEQ, quinocetone, and cyadox from a variety of feeds. The molecularly imprinted SPE cartridge was better than nonimprinted, C18, and HLB cartridges in terms of both recovery and precision. Mean recoveries of four quinoxaline‐1,4‐dioxides from six kinds of feeds spiked at 1.0, 10, and 100 mg/kg ranged between 75.2 and 94.7% with RSDs of less than 10%. The decision limits (CCαs) and the detection capabilities (CCβs) of four analytes were 0.15–0.20 mg/kg and 0.44–0.56 mg/kg, respectively. The class selectivity of the polymers was evaluated by checking three drugs with different molecular structures to that of MEQ.  相似文献

A molecularly imprinted polymer (MIP) able to selectively bind triclosan (TCS) and triclocarban (TCC), commonly used antibacterial agents in many consumer products, was prepared using noncovalent molecular imprinting methods. The prepared MIP was evaluated as a selective sorbent in SPE for sample cleanup before HPLC-UV analysis of TCS and TCC in soil and biosolid samples. The MIP was also compared with commercially available C18 SPE sorbent. The molecularly imprinted SPE (MISPE) developed in this study was more efficient than C18 SPE for the cleanup of extracts of soil and biosolid samples prior to the analysis of TCC and TCS using HPLC-UV. The LOQ values for both TCC and TCS in the soil samples were determined to be 40 microg/kg; in the biosolid samples, the LOQ values were 100 and 300 microg/kg for TCC and TCS, respectively. Compared to C18 SPE, using MISPE for sample cleanup may result in a significant reduction of analytical cost, because one MIP can be reused up to 35 times and HPLC-UV instead of HPLC/MS can be used for instrumental analysis following sample cleanup by MISPE.  相似文献

A highly selective sample cleanup procedure combined with molecularly imprinted SPE was developed for the isolation of crystal violet from seawater and seafood samples. The molecularly imprinted polymer was prepared using crystal violet as the template molecule, methacrylic acid as the functional monomer, and ethylene glycol dimethacrylate as the cross‐linker. The crystal violet‐imprinted polymer was used as the selective sorbent for the SPE of crystal violet. An off‐line molecularly imprinted SPE method followed by HPLC with diode‐array detection for the analysis of crystal violet was also established. Good linearity on the molecularly imprinted SPE columns was obtained from 0 to 200 μg/L (R2 > 0.99). The result demonstrated that the proposed method can be used for the direct determination of crystal violet in seawater and seafood samples. Finally, five samples were analyzed and the following crystal violet concentrations were obtained: 0.92 and 0.52 μg/L in two seawater samples, as well as 0.36 and 0.27 μg/kg in two seafood samples. In the present paper, we describe a new on‐line SPE system where molecular imprinting, fiber‐optic detection and flow injection analysis were combined for the first time. This new system has been applied for the on line detection of 4‐nitrophenol (4‐NP). Initially, molecularly imprinted polymers (MIP) have been prepared for the selective extraction of 4‐NP using 4‐vinylpyridine and ethylene glycol dimethacrylate as functional and cross‐linking monomers, respectively. Selective extraction was achieved using the designed MIP with 97% of recovery on imprinted polymer and 10% on control polymer. The system provided a high degree of accuracy, with RSDs varying between 0.7 and 1.39%. In respect of accuracy, reproducibility, and rapidity, this system is comparable with HPLC. In short, the system allows simple, fast, and accurate analyte determination with the possibility of future automation.  相似文献

A novel molecularly imprinted polymer (MIP) monolith for highly selective extraction of cholecystokinin (CCK) neuropeptides was prepared in a micropipette tip. The MIPs were synthesized by epitope imprinting technique and the polymerization conditions were investigated and optimized. The synthesized MIPs were characterized by infrared spectroscopy, elemental analyzer and scanning electron microscope. A molecularly imprinted solid‐phase microextraction (MI‐μ‐SPE) method was developed for the extraction of CCK neuropeptides in aqueous solutions. The parameters affecting MI‐μ‐SPE were optimized. The results indicated that this MIP monolith exhibited specific recognition capability and high enrichment efficiency for CCK neuropeptides. In addition, it showed excellent reusability. This MIP monolith was used for desalting and enrichment of CCK4, CCK5 and CCK8 from human cerebrospinal fluid prior to matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry analysis, and the results show that this MIP monolith can be a useful tool for effective purification and highly selective enrichment of multiple homologous CCK neuropeptides in cerebrospinal fluid simultaneously. A new molecularly imprinted polymer for extraction of crocin from saffron stigmas was prepared using gentiobiose (a glycoside moiety in crocin structure) as a template. Crocin binding to gentiobiose imprinted polymer (Gent‐MIP) was studied in comparison with a blank nonimprinted polymer in aqueous media. Affinity of the Gent‐MIP for the crocin was more than the nonimprinted polymer at all concentrations. In Scatchard analysis, the number of binding sites in each gram of polymer (maximum binding sites) and dissociation constant of crocin to binding sites were 18.4 μmol/g polymer and 11.2 μM, respectively. The Gent‐MIP was then used as the sorbent in an SPE method for isolation and purification of crocin from methanolic extract of saffron stigmas. The recovery of crocin, safranal and picrocrocin was determined in washing and elution steps. The Gent‐MIP had significantly higher affinity for crocin than other compounds and enabled selective extraction of crocin with a high recovery (84%) from a complex mixture. The results demonstrated the possibility of using a part of a big molecule in preparing a molecularly imprinted polymer with a good selectivity for the main structure.  相似文献

The β‐sitosterol imprinted polymer was prepared for selective extraction and analysis of β‐sitosterol from Oldenlandia diffusa (O. diffusa) followed by HPLC with UV detection. The imprinted polymers show high affinity and selectivity to β‐sitosterol. Using this molecularly imprinted polymer (MIP) cartridge as solid‐phase extraction (SPE) material, the interferences could be quickly washed out and β‐sitosterol was selectively retained and enriched. HPLC analysis method was used to evaluate the characteristics of this MIP material. At the condition of mobile phase composed of MeOH/H2O/H3PO4 (99/1/0.1, V/V/V, pH=6.0) and the flow rate of 1.0 mL·min?1, a good linear relationship was demonstrated when the concentrations of β‐sitosterol were in the range of 0.50–100.0 µg·mL?1. The recoveries ranged from 75.3% to 86.5% and the inter‐day and intra‐day relative standard deviations were less than 5%. This developed HPLC method was proved to be acceptable for extraction of β‐sitosterol from O. diffusa.  相似文献

In this article, for the first time, a molecularly imprinted polymer (MIP) for the metolcarb was prepared by bulk polymerization using metolcarb as the template, methacrylic acid as the functional monomer and ethylene glycol dimethacrylate as the cross‐linker. The prepared polymer was characterized by FT‐IR, static and kinetic adsorption experiments, and the results showed that it has been successfully synthesized and had good selective ability for metolcarb. The MIP was applied as a sorbent in molecularly imprinted SPE coupled with HPLC‐UV for separation and determination of trace metolcarb in three kinds of food matrices at three concentration levels. Under the optimal conditions, the LODs (S/N=3) of cabbage, cucumber and pear were 7.622, 6.455 and 13.52 μg/kg, respectively, and recoveries were in the range of 68.80–101.31% with RSD (n=3) below 3.78% in all cases. To demonstrate further the selectivity of the MIP obtained, a comparison with commercially available C18 SPE was performed. The results indicated that molecularly imprinted SPE showed better chromatography, better selectivity and higher recoveries for metolcarb than commercially available C18 SPE.  相似文献

In this paper we describe, for the first time, a molecularly imprinted polymer (MIP) for the antibiotic amoxicillin (AMX), synthesised by a noncovalent molecular imprinting approach and used to extract AMX selectively from urine samples. The MIP was applied as a molecularly selective sorbent in molecularly imprinted SPE (MISPE) in an off-line mode, where it showed useful cross-selectivity for a structurally related antibiotic, cephalexin (CPX). By using a MISPE protocol, the MIP was able to selectively extract both AMX and CFX from 5 mL of water spiked with 10 mg/L with recoveries of 75 and 78% for AMX and CFX, respectively. When applied to real samples (urine) at clinically relevant concentrations, recoveries from 2 mL of human urine spiked with 20 mg/L decreased slightly to 65 and 63% for AMX and CFX, respectively. To demonstrate further the selectivity of the MIP obtained, a comparison with commercially available SPE cartridges was performed. Improvements in the retention of both AMX and CFX on the MIP were obtained relative to the commercially available cartridges, and the MISPE extracts were considerably cleaner, due to molecularly selective analyte binding by the MIP.  相似文献

A molecularly imprinted polymer (MIP) was synthesized and evaluated to selectively extract ephedrine from human plasma. The MIP synthesis was performed in chloroform with methacrylic acid as a functional monomer and the target alkaloid as a template molecule. The resulting MIP was applied to the selective extraction of ephedrine from a pure aqueous medium. A recovery about 74% was obtained using the MIP with only 7% on the nonimprinted polymer (NIP). A very straightforward selective SPE procedure was then successfully applied to the direct extraction of ephedrine from spiked human plasma with a high extraction recovery (68%) on the MIP with no recovery on the NIP. Moreover, the MIP was used for the selective extraction of catecholamine neurotransmitters, i.e. adrenaline and noradrenaline.  相似文献

The aim of this work was to develop a method for the clean-up of a mycotoxin, i.e. Ochratoxin A (OTA), from cereal extracts employing a new molecularly imprinted polymer (MIP) as selective sorbent for solid-phase extraction (SPE) and to compare with an immunoaffinity column. A first series of experiments was carried out in pure solvents to estimate the potential of the imprinted sorbent in terms of selectivity studying the retention of OTA on the MIP and on a non-imprinted polymer using conventional crushed monolith. The selectivity of the MIP was also checked by its application to wheat extracts. Then, after this feasibility study, two different formats of MIP: crushed monolith and micro-beads were evaluated and compared. Therefore an optimization procedure was applied to the selective extraction from wheat using the MIP beads. The whole procedure was validated by applying it to wheat extract spiked by OTA at different concentration levels and then to a certified contaminated wheat sample. Recoveries close to 100% were obtained. The high selectivity brought by the MIP was compared to the selectivity by an immunoaffinity cartridge for the clean-up of the same wheat sample. The study of capacity of both showed a significant higher capacity of the MIP.  相似文献

An efficient and sensitive analytical method based on molecularly imprinted solid‐phase extraction (MISPE) and reverse‐phase ultrasound‐assisted dispersive liquid–liquid microextraction (USA‐DLLME) coupled with LC–MS/MS detection was developed and validated for the analysis of urinary 4‐(methylnitrosamino)‐1‐(3‐pyridyl)‐1‐butanol (NNAL), a tobacco‐specific nitrosamine metabolite. The extraction performances of NNAL on three different solid‐phase extraction (SPE) sorbents including the hydrophilic‐lipophilic balanced sorbent HLB, the mixed mode cationic MCX sorbent and the molecularly imprinted polymers (MIP) sorbent were evaluated. Experimental results showed that the analyte was well retained with the highest extraction recovery and the optimum purification effect on MIP. Under the optimized conditions of MIP and USA‐DLLME, an enrichment factor of 23 was obtained. Good linearity relationship was obtained in the range of 5‐1200 pg/mL with a correlation coefficient of 0.9953. The limit of detection (LOD) was 0.35 pg/mL. The recoveries at three spiked levels ranged between 88.5% and 93.7%. Intra‐ and inter‐day relative standard deviations varied from 3.6% to 7.4% and from 5.4% to 9.7%, respectively. The developed method combing the advantages of MISPE and DLLME significantly improves the purification and enrichment of the analyte and can be used as an effective approach for the determination of ultra‐trace NNAL in complex biological matrices.  相似文献

A molecularly imprinted polymer (MIP) was synthesized in order to specifically extract vinflunine, an anticancer agent, and its metabolite (4‐O‐deacetylvinflunine) from bovine plasma and artificial urine by solid‐phase extraction (SPE). Vinorelbine, a non‐fluorinated analogue of vinflunine, was selected as a template for MIP synthesis. The selectivity of MIP versus the template (vinorelbine) and other alkaloids (catharanthine, vinblastine, vincristine, vinflunine and 4‐O‐deacetylvinflunine) was shown by a SPE protocol carried out with non‐aqueous samples. A second protocol was developed for aqueous samples with two consecutive washing steps (AcOH–NH2OH buffer (pH 7, I=10 mM)–MeOH mixture 95:5 v/v and ACN–AcOH mixture 99:1 v/v) and an elution step (MeOH–AcOH mixture 90:10 v/v). Thus, MIP‐SPE of bovine plasma brought high recoveries, 81 and 89% for vinflunine and its metabolite, respectively. This protocol was slightly modified for artificial urine samples in order to obtain a good MIP/NIP selectivity; furthermore, elution recoveries were 73 and 81% for vinflunine and its metabolite, respectively. Repeatability was assessed in both biological matrices and RSD (%) were inferior to 4%. The MIP also showed a suitable linearity (r2 superior to 0.99), between 0.25 and 10 μg/mL for plasma, and between 1 and 5 μg/mL for artificial urine.  相似文献

Method of molecularly imprinted solid phase extraction (MISPE) of (-)-ephedrine from Chinese Ephedra has been developed in the research. The molecularly imprinted polymer (MIP) with good selectivity and affinity for (-)-ephedrine was synthesized with (-)-ephedrine as the template, methacrylic acid as the functional monomer. The washing and elution conditions in MISPE were selected and optimized for efficient analyte extraction and sample clean-up. A clean analytical HPLC base line of ephedra extract was obtained after MISPE, which indicated that the sample pre-treatment was efficient. Good recovery and precision were obtained in the assessment for the MISPE-HPLC procedure, which demonstrated it is a reliable method and can be used for the determination of (-)-ephedrine in herbal ephedra.  相似文献

A simple, reliable, and low‐cost method based on molecularly imprinted polymer as a selective sorbent of SPE was proposed for the determination of ochratoxin A (OTA) in beer, red wine, and grape juice by HPLC coupled with fluorescence detection (HPLC‐FLD). Samples were diluted with water and cleaned up with an AFFINIMIP® SPE OTA column. After washing and eluting, the analyte was analyzed by HPLC‐FLD. Under the optimized conditions, LOD and LOQ for OTA were 0.025 and 0.08 ng/mL, respectively. The recoveries of OTA from beer, red wine, and grape spiked at 0.1, 2, and 5 ng/mL ranged from 91.6 to 101.7%. Furthermore, after a simple regenerated procedure, the molecularly imprinted polymer based SPE column could be reused at least 14 times to achieve more than 80% recoveries of OTA in real samples. The developed method was applied to the detection of 30 beer, red wine, and grape juice samples and only four samples were contaminated by OTA with levels below the legal limits.  相似文献

The aim of this work was to develop a method for the clean‐up and preconcentration of warfarin from biological sample employing a new molecularly imprinted polymer (MIP) as a selective adsorbent for solid‐phase extraction (SPE). This MIP was synthesized using warfarin as a template, pyrrole as a functional monomer and vinyl triethoxysilane as a cross‐linker. The molar ratio of 1:4:20 (template–functional monomer–cross‐linker) showed the best results. Nonimprinted polymers (NIPs) were prepared and treated with the same method, but in the absence of warfarin. The prepared polymer was characterized by Fourier transmission infrared spectrometry and scanning electron microscopy. An adsorption process (SPE) for the removal of warfarin using the fabricated MIPs and NIPs was evaluated under various conditions. Effective parameters on warfarin extraction, for example, type and volume of elution solvent, pH of sample solution, breakthrough volume and maximum loading capacity, were studied. The limits of detection were in the range of 0.0035–0.0050 µg mL?1. Linearity of the method was determined in the range of 0.0165–10.0000 µg mL?1 for plasma and 0.0115–10.0000 µg mL?1 for urine with coefficients of determination (R2) ranging from 0.9975 to 0.9985. A new LC method to detect fusaric acid (FA) in maize is reported based on a molecularly imprinted SPE clean‐up using mimic‐templated molecularly imprinted polymers. Picolinic acid was used as a toxin analog for imprinting polymers during a thermolytic synthesis. Both acidic and basic functional monomers were predicted to have favorable binding interactions by MP2 ab initio calculations. Imprinted polymers synthesized with methacrylic acid or 2‐dimethylaminoethyl methacrylate exhibited imprinting effects in SPE analysis. FA levels were determined using RP ion‐pairing chromatography with diode‐array UV detection and tetrabutylammonium hydrogen sulfate in the mobile phase. A method was developed to detect FA in maize using molecularly imprinted SPE analysis within the range of 1–100 μg/g with recoveries between 83.9 and 92.1%.  相似文献

A novel molecularly imprinted polymer (MIP) was synthesized by precipitation polymerization with baicalein (BAI) as the template and used as solid‐phase extraction (SPE) adsorbent, aiming at the affinity isolation and selective knockout of BAI from Scutellaria baicalensis Georgi (SB). We used computational simulation to predict the optimal functional monomer, polymerization solvent and molar ratio of template to functional monomer. Characterization and performance tests revealed that MIP exhibited uniform spherical morphology, rapid binding kinetics, and higher adsorption capacity for BAI compared with nonimprinted polymer (NIP). The application of MIP in SPE coupled with high‐performance liquid chromatography to extract BAI from SB showed excellent recovery (94.3%) and purity (97.0%). Not only the single BAI compound, but also the BAI‐removed SB extract was obtained by one‐step process. In this paper, a novel monolithic stir bar based on molecularly imprinted polymer (MIP) was firstly developed by filling modified neodymium magnet (Nd2Fe14B) powders into a glass tube (60 × 4 mm), followed by the imprinted grafting with bisphenol A (BPA) as the template molecule by thermal polymerization. It has been successfully used for the stir bar sorptive extraction (SBSE) and its extraction performance illustrated that the MIP‐encapsulated stir bar had stronger affinity to the template molecule, compared with the stir bar based on the non‐imprinted molecularly polymer (NIP). Under the optimal extraction conditions, a simple method based on the coupling of MIP‐SBSE with high performance liquid chromatography (HPLC) was used for the selective determination of the model mixtures of BPA, 4‐phenylphenol (PP) and phenol (P) in bottled water. The recoveries of BPA, PP and P were in the range of 88.5‐96.1%, 78.2‐89.7%, 81.3‐89.5% at three spiked levels, respectively, demonstrating that higher extraction and the specific absorption occurred between the template molecule and the prepared MIP stir bar.  相似文献
