Three new heteroscorpionate ligands, (2‐hydroxyphenyl)bis(imidazol‐1‐yl)methane (HL1), (4‐diethylamino‐2‐hydroxyphenyl)bis(imidazol‐1‐yl)methane (HL2) and (5‐bromo‐2‐hydroxyphenyl)bis(imidazol‐1‐yl)methane (HL3), and their heteroleptic copper(II) complexes of the type [Cu(L1–3)diimine]ClO4 ( 1 – 6 ; where diimine =2,2′‐bipyridyl or 1,10‐phenanthroline) have been synthesized and characterized using spectroscopic methods. The molecular structure of ligand HL1 was determined by single‐crystal X‐ray diffraction. UV–visible, electron paramagnetic resonance and theoretical studies suggest a distorted square pyramidal geometry around copper(II) ion. Analyses of highest occupied and lowest unoccupied molecular orbitals have been used to explain the charge transfer taking place within the complexes. The antioxidant activities of the heteroscorpionate ligands and their heteroleptic copper(II) complexes were determined using ABTS, DPPH and H2O2 free radical scavenging assays with respect to standard antioxidant ascorbic acid. In molecular docking studies, the complexes showed π–π, hydrogen bonding, van der Waals and electrostatic interactions with fibroblast growth factor receptor kinase. In vitro cytotoxicity activities of ligands and copper(II) complexes were examined on human breast adenocarcinoma (MCF‐7), cervical (HeLa) and lung (A549) cancer cell lines and normal human dermal fibroblast cell line using MTT assay. Complex 4 exhibited higher anticancer activity than the other complexes against all three cancer lines, being more potent than the standard drug cisplatin.  相似文献

Three ruthenium(II) hydrazone complexes of composition [RuCl(CO)(PPh3)2L] were synthesized from the reactions of [RuHCl(CO)(PPh3)3] with hydrazones derived from 4‐methoxybenzhydrazide and 4‐formylbenzoic acid (HL1), 4‐methylbenzaldehyde (HL2) and 2‐bromobenzaldehyde (HL3). The synthesized hydrazone ligands and their metal complexes were characterized using elemental analysis and infrared, UV–visible, NMR (1H, 13C and 31P) and mass spectral techniques. The hydrazone ligands act as bidentate ones, with O and N as the donor sites, and are predominantly found in the enol form in all the complexes studied. The molecular structures of the ligands HL1, HL2 and HL3 were determined using single‐crystal X‐ray diffraction. The interactions of the ligands and the complexes with calf thymus DNA were studied using absorption spectroscopy and cyclic voltammetry which revealed that the compounds could interact with calf thymus DNA through intercalation. The DNA cleavage activity of the complexes was evaluated using a gel electrophoresis assay which revealed that the complexes act as good DNA cleavage agents. In addition, all the complexes were subjected to antioxidant assay, which showed that they all possess significant scavenging activity against 2,2‐diphenyl‐2‐picrylhydrazyl, OH and NO radicals. The in vitro cytotoxic effect of the complexes examined on cancerous cell lines (HeLa and MCF‐7) showed that the complexes exhibit substantial anticancer activity. A series of six new Zn (II) compounds, viz., [Zn(HLASA)2(Py)2] ( 1 ), [Zn(HLMASA)2(Py)2] ( 2 ), [Zn(HLMASA)2(4‐MePy)2] ( 3 ), [Zn(HLCASA)2(4‐MePy)2] ( 4 ), [Zn(HLBASA)2(Py)2] ( 5 ), [Zn(HLBASA)2(4‐MePy)2] ( 6 ) and representative Cu (II) and Cd (II) complexes, viz., [Cu(HLASA)2(Py)2(H2O)] ( 7 ) and [Cd(HLBASA)2(Py)3] ( 8 ) [(HLXASA)? = para‐substituted 5‐[(E)‐2‐(aryl)‐1‐diazenyl]‐2‐hydroxybenzoate with X = H (ASA), Me (MASA), Cl (CASA) or Br (BASA); Py = pyridine; 4‐MePy = 4‐methylpyridine] have been synthesized and characterized by spectroscopic techniques and single‐crystal X‐ray diffraction analysis. The structural characterization of the compounds revealed distorted tetrahedral ( 1 – 6 ), square‐pyramidal ( 7 ) and pentagonal‐bipyramidal ( 8 ) coordination geometries around the metal atom, in which the aryl‐substituted diazosalicylate ligands are coordinated only through the oxygen atoms of carboxylate groups, either in an anisobidentate or isobidentate mode; meanwhile, the 2‐hydroxy groups of the monoanionic ligand (HLXASA)? are involved only in intramolecular O‐H···O hydrogen bonds with the carboxylate function. In the crystal structures of 1 – 8 , the complex molecules are assembled by π‐stacking interactions giving mostly infinite 1D strands. The intermolecular binding in the solid state structures is accomplished by diverse additional non‐covalent contacts including C‐H···O, C‐H···N, C‐H···π, C‐H···Br, O···Br, Br···π and van der Waals contacts. Although the primary and secondary ligands in the Zn (II) complex series 1 – 6 carry different substituents at the periphery (X = H, Me, Cl, Br for (HLXASA)? and R = H, Me for 4‐Py‐R), five of the crystal structures were isostructural. Additionally, the antimicrobial activity of the pro‐ligands H2LXASA and their Zn (II), Cu (II) and Cd (II) compounds were studied in a comparative manner, showing high sensitivity (IZD ≥ 20) against Bacillus subtilis.  相似文献

Two two‐dimensional supramolecular copper(II) and cobalt(III) complexes, Cu(L1)2 ( 1 ; HL1 = 2‐hydroxy‐3‐methoxybenzaldehyde oxime) and [Co(L2)2]2⋅2CH3COOCH2CH3 ( 2 ; HL2 = 1‐(2‐{[(E )‐3‐methoxy‐2‐hydroxybenzylidene]amino}phenyl)ethanone oxime), have been synthesized via complexation of Cu(II) nitrate trihydrate and Co(II) acetate tetrahydrate with HL. A plausible reaction mechanism for the formation of HL1 is proposed. HL was synthesized and characterized using infrared, 1H NMR and 13C NMR spectra, as well as elemental analysis. Complexes 1 and 2 were investigated using single‐crystal X‐ray diffraction and have a 2:1 ligand‐to‐metal ratio. Different geometric features of both complexes are observed. In their crystal structures, 1 and 2 form infinite two‐dimensional structures and 2 forms a three‐dimensional supramolecular framework. Electron paramagnetic resonance spectra of 1 and 2 were also investigated. Moreover, thermal and electrochemical properties and antimicrobial activity of 2 were also studied. In addition, the calculated HOMO and LUMO energies show the character of complex 1 .  相似文献

A series of diorganotin complexes with Schiff base ligands, (E)‐N′‐(5‐bromo‐2‐hydroxybenzylidene)‐3‐hydroxy‐2‐naphthohydrazide, H2L1, and (E)‐N′‐(5‐chloro‐2‐hydroxybenzylidene)‐3‐hydroxy‐2‐naphthohydrazide, H2L2, were synthesized and characterized by elemental analysis, IR, 1H, 13C and 119Sn NMR spectroscopy. The molecular structures of the complexes, [(5‐bromo‐2‐oxidobenzylidene)‐3‐hydroxy‐2‐naphthohydrazidato]di(o‐chlorobenzyl)tin(IV) 6 and [(5‐chloro‐2‐oxidobenzylidene)‐3‐hydroxy‐2‐naphthohydrazidato]dibutyltin(IV) 9, were determined through single‐crystal X‐ray diffraction and revealed a distorted trigonal‐bipyramidal configuration. The in vitro cytotoxic activity of the Schiff bases and their diorganotin complexes was also evaluated against several human carcinoma cell lines, namely HT29 (human colon carcinoma cell line), SKOV‐3 (human ovarian cancer cell line), MCF7 (hormone‐dependent breast carcinoma cell line) and MRC5 (non‐cancer human fibroblast cell line). [(5‐Bromo‐2‐oxidobenzylidene)‐3‐hydroxy‐2‐naphthohydrazidato]dibutyltin(IV) 2 and [(5‐bromo‐2‐oxidobenzylidene)‐3‐hydroxy‐2‐naphthohydrazidato]dibenzyltin(IV) 5 were the most active diorganotin complexes of H2L1 ligand. Among the diorganotin complexes of H2L2 ligand, [(5‐chloro‐2‐oxidobenzylidene)‐3‐hydroxy‐2‐naphthohydrazidato]dicyclohexyltin(IV) 11 showed good cytotoxic activity against all the tested cell lines. As such, the above compounds can be considered agents with potential anticancer activities, and can therefore be investigated further in in vitro or in vivo anticancer studies. In quest of new metallo‐pharmaceuticals with enhanced anticancer activity, four new phosphine‐ and carbodithioate‐based Pd(II) complexes of the type [(R)CS2Pd(PR3)Cl] (where R = 4‐(2‐hydroxyethyl)piperazine ( 1 , 2 ), dibenzyl ( 3 , 4 ); PR3 = diphenyl(p ‐tolyl)phosphine ( 1 , 3 ), tris(4‐tolyl)phosphine ( 2 , 4 )) were synthesized and characterized using elemental analysis, Fourier transform infrared and NMR (1H, 13C and 31P) spectroscopies and single‐crystal X‐ray diffraction. The X‐ray diffraction data confirmed the pseudo square‐planar geometry ensuring bidentate coordination mode of carbodithioate ligands. Anticancer activity of the synthesized complexes and their ligands was assessed against human lung (A549), breast (MCF‐7) and prostate (PC3) carcinoma cells using MTT assay. All the tested compounds showed activity in micromolar range. In many cases, the cytotoxicity of the synthesized complexes was higher than or comparable to that of the standard drugs cisplatin and doxorubicin. Complex 3 emerged as the most promising compound with the lowest IC50 values of 4.83, 3.72 and 5.11 μM for A549, MCF‐7 and PC3 carcinoma cell lines, respectively. DNA binding studies were also carried out using UV–visible spectroscopy. We extended our investigations to explore the mechanism of anticancer activity using in silico tools. Based on the mechanism of action of standard drugs used, extensive docking studies were carried out on the three different biomolecular targets.  相似文献

Acetato, chloro and nitrato Cu(II) complexes of a novel azo compound, namely 2,4‐dihydroxy‐5‐[(5‐mercapto‐1H‐1,2,4‐triazole‐3‐yl)diazenyl]benzaldehyde, have been prepared. The stoichiometry, stereochemistry and bonding fashion of these copper chelates were deduced via elemental analyses, spectral methods and conductivity and magnetic measurements. Infrared spectral data confirmed the participation of azo N atom and the deprotonated OH group. UV–visible spectral data and magnetic measurements indicated octahedral stereo‐structure for the acetato and nitrato compounds and square planer for the chloro compound. Thermogravimetric analysis was applied to investigate the thermal degradation of the metal chelates. The thermo‐kinetic parameters were computed. The molecular modeling technique was used to support the predicted geometry of the prepared chelates. The interaction between the Cu(II) complexes and calf thymus DNA was studied using two techniques: absorption and viscosity measurements. The values of binding constant obtained from the absorption spectral method were calculated and found to be 4.23 × 104, 26.93 × 104, 13.01 × 104 and 5.36 × 104 M?1 for ligand and acetato, chloro and nitrato complexes, respectively. The antimicrobial activities were evaluated against various bacterial and fungi strains. The in vitro antitumor efficacy of the synthesized compounds was investigated against the HEPG2 cell line.  相似文献

A series of six N,N‐di‐substituted acylthiourea ArC(O)NHC(S)NRR′ ligands (denoted as HLn) [Ar = 1‐Naph: NRR′ = NPh2, HL1 ( 1 ); N(iPr)Ph, HL2 ( 2 ). Ar = Mes: NRR′ = NPh2, HL4 ( 3 ); N(iPr)Ph, HL5 ( 4 ); NEt2, HL6 ( 5 ). Ar = Ph: NRR′ = N(iPr)Ph, HL8 ( 6 )] were synthesized and characterized. These ligands were deprotonated to form CuII complexes through metathesis or combined redox reaction with copper halides. The structures of the complexes were investigated with single‐crystal X‐ray diffraction. The reaction of the 1‐naphthalene derivative HL1 ( 1 ) with CuBr in the presence of sodium acetate produced cis‐CuL12 ( 7 ), where the deprotonated ligand is bound to the CuII atom in a bidentate‐(O, S) coordination mode. Similarly treatment of HL2 ( 2 ) with NaOAc and CuCl resulted in the formation of the cis‐arranged product [cis‐CuL22 ( 8 )]. The reaction of mesityl derivative HL4 ( 3 ) and CuBr with and without the addition of NaOAc gave the cis‐CuL42 ( 9 ) and cis‐(HL4)2CuBr ( 10 ), respectively. In contrast, reaction of HL5 ( 4 ) and CuI in the presence of NaOAc resulted in trans‐CuL52 ( 11 ). Alternatively trans‐CuL62 ( 12 ) was obtained by the reaction of diethyl‐substituted HL6 ( 5 ) with CuCl2 in the absence of a base.  相似文献

A novel diazadiphosphetidine ligand derived from the reaction of 2,4‐dichloro‐1,3‐dimethyl‐1,3,2,4‐diazadiphosphetidine‐2,4‐dioxide and 2,2′‐(ethane‐1,2‐diylbis[oxy])bis(ethan‐1‐amine) and its Ni(II), Cu(II), and Co(II) complexes have been synthesized, characterized by spectroscopic, elemental analyses, magnetic susceptibility, and conductivity methods, and screened for antimicrobial, DNA binding, and cleavage properties. Spectroscopic analysis and elemental analyses indicate the formula [M(H2L)Cl2] for the Cu(II), Co(II), Ni(II), and Zn(II) complexes and octahedral geometry for all the complexes. The non‐electrolytic nature of the complexes in dimethyl sulfoxide (DMSO) was confirmed by their molar conductance values, which are in the range 12.32–6.73 Ω?1 cm2 mol?1. Computational studies have been carried out at the density functional theory (DFT)‐B3LYP/6‐31G(d) level of theory on the structural and spectroscopic properties of diazadiphosphetidine H2L and its binuclear Cu(II), Co(II), Ni(II), and Zn(II) complexes. Six tautomers and geometrical isomers of the diazadiphosphetidine ligand were confirmed using semiempirical AM1 and DFT method from DMOL3 calculations. The copper complex had the best antibacterial activity against Staphylococcus aureus (ATCC 29213). DNA cleavage activities of the compounds, evaluated on pBR322 DNA by agarose gel electrophoresis in the presence and absence of an oxidant (H2O2) and a free‐radical scavenger (DMSO), indicated no activity for the ligand and moderate activity for the complexes, with the copper complex cleaving pBR322 DNA more efficiently in the presence of H2O2.  相似文献

Hexacoordinated non‐heme iron complexes [FeII(L1)2](ClO4)2 ( 1 ) and [FeII(L2)2](PF6)2 ( 2 ) have been synthesized using ligands L1 = (E)‐2‐chloro‐6‐(2‐(pyridin‐2ylmethylene) hydrazinyl)pyridine and L2 = (E)‐2‐chloro‐6‐(2‐(1‐(pyridin‐2‐yl)ethylidene)hydrazinyl) pyridine]. These complexes are highly active non‐heme iron catalysts to catalyze the C (sp3)?H bonds of alkanes. These iron complexes have been characterized using ESI?MS analysis and molecular structures were determined by X‐ray crystallography. ESI ? MS analysis also helped to understand the generation of intermediate species like FeIII?OOH and FeIV=O. DFT and TD?DFT calculations revealed that the oxidation reactions were performed through high‐valent iron center and a probable reaction mechanism was proposed. These complexes were also utilized for the degradation of orange II and methylene blue dyes.  相似文献

Biologically active triazole Schiff bases ( L 1  L 3 ) derived from the reaction of 3‐amino‐1,2,4‐triazole with chloro‐, bromo‐ and nitro‐ substituted salicylaldehydes and their Zn(II) complexes (1–3) have been synthesized and characterized by their physical, spectral and analytical data. Triazole Schiff bases potentially act as tridentate ligands and coordinate with the Zn(II) metal atom through salicylidene‐O, azomethine‐N and triazole‐N. The complexes have the general formula [M(L‐H)2], where M = zinc(II) and L = ( L 1 – L 3 ), and observe an octahedral geometry. The Schiff bases and their Zn(II) complexes have been screened for in‐vitro antibacterial, antifungal and brine shrimp bioassay. The biological activity data show the Zn(II) complexes to be more potent antibacterial and antifungal than the parent simple Schiff bases. Copyright © 2011 John Wiley & Sons, Ltd.  相似文献

Three novel water‐soluble copper(II) complexes – {[Cu(phen)(trp)]ClO4·3H2O}n ( 1 ), {[Cu(4‐mphen)(trp)]ClO4·3H2O}n ( 2 ) and [[Cu(dmphen)(trp)(MeOH)][Cu(dmphen)(trp)(NO3)]]NO3 ( 3 ) (phen: 1,10‐phenanthroline; 4‐mphen: 4‐methyl‐1,10‐phenanthroline; dmphen: 4,7‐dimethyl‐1,10‐phenanthroline; trp: l ‐tryptophan) – have been synthesized and characterized using various techniques. Complexes 1 and 2 are isostructural, and exist as one‐dimensional coordination polymers. Complex 3 consists of two discrete copper(II) complexes containing [Cu(trp)(dmphen)(MeOH)]+, [Cu(trp)(dmphen)(NO3)] and one nitrate anion. The binding interaction of the complexes with calf thymus DNA (CT‐DNA) was investigated using thermal denaturation, electronic absorption and emission spectroscopic methods, revealing that the complexes could interact with CT‐DNA via a moderate intercalation mode. The binding activity of the complexes to CT‐DNA follows the order: 3  >  2 > 1 . The pUC19 DNA cleavage activity of the complexes was investigated in the absence and presence of external agents using the agarose gel electrophoresis method. Especially, in the presence of H2O2 as an activator, the pUC19 DNA cleavage abilities of the complexes are clearly enhanced at low concentration. Addition of hydroxyl radical scavenger dimethylsulfoxide shows a marked inhibition of the pUC19 DNA cleavage activity of the complexes. In vitro cytotoxic effect of the complexes was examined on human tumor cell lines (Caco‐2, A549 and MCF‐7) and healthy cells (BEAS‐2B). The potent cytotoxic effect of complex 3 , with IC50 values of 1.04, 1.16 and 1.72 μM, respectively, is greater relative to clinically used cisplatin (IC50 = 22.70, 31.1 and 22.2 μM) against the Caco‐2, A549 and MCF‐7 cell lines.  相似文献

New mixed ligand complexes of transition metals were synthesized from a Schiff base (L1) obtained by the condensation reaction of oxamide and furfural as primary ligand and 2,2′‐bipyridine (L2) as secondary ligand. The ligands and their metal complexes were studied using various spectroscopic methods. Also thermal analyses were conducted. The mixed ligand complexes were found to have formulae [M(L1)(L2)]Clm n H2O (M = Cr(III) and Fe(III): m  = 3, n  = 0; M = Cu(II) and Cd(II): m  = 2, n  = 1; M = Mn(II), Co(II), Ni(II) and Zn(II): m  = 2, n  = 0). The resultant data revealed that the metal complexes have octahedral structure. Also, the mixed ligand complexes are electrolytic. The biological and anticancer activities of the new compounds were tested against breast cancer (MCF‐7) and colon cancer (HCT‐116) cell lines. The results showed high activity for the synthesized compounds.  相似文献

Mononuclear transition metal(II) complexes of the type M(L)2?2H2O (where M = Co, Ni, Cu, Zn) have been synthesized from uninegative Schiff base ligands (HL1–HL4) designed by condensation of 4‐fluorobenzylamine with 2‐hydroxy‐1‐naphthaldehyde/3,5‐dichlorosalicylaldehyde/3,5‐dibromosalicylaldehyde/3‐bromo‐5‐chlorosalicylaldehyde. The compounds were successfully characterized using spectroscopic and physiochemical methods together with elemental analysis. Spectroscopic elucidation indicates a monobasic bidentate nature of ligands coordinated via deprotonated phenolic oxygen and azomethine nitrogen atom which suggests an octahedral geometry around the central metal ions. The complexes and ligands were screened for their in vitro antimicrobial activity against bacterial and fungal strains, the zinc(II) complexes being more active against the tested microbial strains. Further, the metal complexes were found to be more active than the uncomplexed ligands due to chelation process and, moreover, the complexes were more active against fungal strains than bacterial strains. Cytotoxic activities of all compounds were evaluated towards human alveolar adenocarcinoma epithelial cell line (A549), human breast adenocarcinoma cell line (MCF7), human prostate cancer cell line (DU145) and one normal human lung cell line (MRC‐5) using MTT colorimetric assay with doxorubicin as a standard. The zinc complexes were most active against the cancer cell lines and also found to be less toxic against MRC‐5 normal cell line than standard doxorubicin.  相似文献

A new series of twelve bidentate Schiff's base ligands (HL1–12) was synthesized via condensation of 5‐(arylazo)salicylaldehydes with aromatic amines. When the new salicylaldimine derivatives were reacted with copper(II) chloride, the neutral complexes Cu(L1–12)2 were obtained. The structure of the copper complexes was established from microanalyses, IR and UV spectra and thermal analyses. The results suggested that the ligands were coordinated to the metal ion in a bidentate manner with ON donor sites of the deprotonated phenolic‐OH and azomethine‐N. The composition of the complexes can be represented as CuL2. Evaluation of antimicrobial activity for the synthesized compounds was carried out to probe their activity. The compounds were found to have weak antimicrobial activity.  相似文献

The synthesis and characterization of two pyrazolate‐bridged dicopper(II) complexes, [Cu2(L1)2(H2O)2](ClO4)2 ( 1 , HL1=3,5‐dipyridyl‐4‐(2‐keto‐pyridyl)pyrazole) and [Cu2(L2)2(H2O)2](ClO4)2 ( 2 , HL2=3,5‐dipyridyl‐4‐benzoylpyrazole), are discussed. These copper(II) complexes are formed from the reactions between pyridine‐2‐aldehyde, 2‐acetylpyridine (for compound 1 ) or acetophenone (for compound 2 ), and hydrazine hydrate with copper(II) perchlorate hydrate under ambient conditions. The single‐crystal X‐ray structure of compound 1? 2 H2O establishes the formation of a pyrazole ring from three different carbon centers through C? C bond‐forming reactions, mediated by copper(II) ions. The free pyrazoles (HL1 and HL2) are isolated from their corresponding copper(II) complexes and are characterized by using various analytical and spectroscopic techniques. A mechanism for the pyrazole‐ring synthesis that proceeds through C? C bond‐forming reactions is proposed and supported by theoretical calculations.  相似文献

Two 3,6‐bis(R‐1H‐1,2,3‐triazol‐4‐yl)pyridazines (R=mesityl, monodisperse (CH2 CH2O)12CH3) were synthesized by the copper(I)‐catalyzed azide–alkyne cycloaddition and self‐assembled with tetrakis(acetonitrile)copper(I) hexafluorophosphate and silver(I) hexafluoroantimonate in dichloromethane. The obtained copper(I) complexes were characterized in detail by time‐dependent 1D [1H, 13C] and 2D [1H‐NOESY] NMR spectroscopy, elemental analysis, high‐resolution ESI‐TOF mass spectrometry, and analytical ultracentrifugation. The latter characterization methods, as well as the comparison to analog 3,6‐di(2‐pyridyl)pyridazine (dppn) systems and their corresponding copper(I) and silver(I) complexes indicated that the herein described 3,6‐bis(1H‐1,2,3‐triazol‐4‐yl)pyridazine ligands form [2×2] supramolecular grids. However, in the case of the 3,6‐bis(1‐mesityl‐1H‐1,2,3‐triazol‐4‐yl)pyridazine ligand, the resultant red‐colored copper(I) complex turned out to be metastable in an acetone solution. This behavior in solution was studied by NMR spectroscopy, and it led to the conclusion that the copper(I) complex transforms irreversibly into at least one different metal complex species.  相似文献

A series of ionic iron(III) complexes of general formula [HLn ][FeX4] (HL1 = 1,3‐dibenzylbenzimidazolium cation, X = Cl, 1 ; HL1, X = Br, 2 ; HL2 = 1,3‐dibutylbenzimidazolium cation, X = Br, 3 ; HL3 = 1,3‐bis(diphenylmethyl)benzimidazolium cation, X = Br, 4 ) were easily prepared in high yields by the direct reaction of FeX3 with 1 equiv. of [HLn ]X under mild conditions. All of them were characterized using elemental analysis, Raman spectroscopy and electrospray ionization mass spectrometry, and X‐ray crystallography for 1 and 4 . In the presence of magnesium turnings and LiCl, these air‐ and moisture‐insensitive complexes showed high catalytic activities in direct cross‐couplings of aryl phosphates with primary and secondary alkyl bromides with broad substrate scope, wherein complex 4 was the most effective.  相似文献

A water soluble chloro bridged binuclear copper(II) complex (3) and mononuclear complex (4) have been synthesized from chloro substituted 2‐oxo‐1,2‐dihydroquinolin‐3‐yl‐methylene‐2 hydroxybenzohydrazide 1 and 2 and CuCl2·2H2O. The structures of the complexes have been determined by single crystal X‐ray diffraction. The binding interactions of the ligands and complexes with CT‐DNA and protein have been evaluated by absorption and emission spectroscopic method. CT‐DNA and ethidium bromide (EB) competitive studies revealed that the compounds could interact with CT‐DNA through intercalation binding mode. Interactions of the compounds with BSA were also studied by UV−visible, fluorescence and synchronous fluorescence spectroscopic methods which showed that the compounds had a strong binding affinity with BSA through static quenching process. The cytotoxic effect of the compounds examined on cancer cell lines, such as A549 (lung cancer) and MCF7 (breast cancer) cell lines showed that all four compounds exhibited substantial cytotoxic activity.  相似文献

A novel hydrazone ligand derived from condensation reaction of 3‐hydroxy‐2‐naphthoic hydrazide with dehydroacetic acid, and its Ni(II), Cu(II) and Co(II) complexes were synthesized, characterized by spectroscopic, elemental analyses, magnetic susceptibility and conductivity methods, and screened for antimicrobial, DNA binding and cleavage properties. Spectroscopic analysis and elemental analyses indicated the formula, [MLCl2], for the complexes; square planar geometry for the nickel, and tetrahedral geometry for copper and cobalt complexes. The non‐electrolytic natures of the complexes in Dimethyl Sulphoxide (DMSO) were confirmed by their molar conductance values in the range of 6.11–14.01 Ω?1cm2mol?1. The copper complex had the best antibacterial activity against Staphylococcus aureus (ATCC 29213). DNA cleavage activities of the compounds, evaluated on pBR322 DNA, by agarose gel electrophoresis, in the presence and absence of oxidant (H2O2) and free radical scavenger (DMSO), indicated no activity for the ligand, and moderate activity for the complexes, with the copper complex cleaving pBR322 DNA more efficiently in the presence of H2O2. When the complexes were evaluated for antibacterial and A‐DNA activity using Molecular docking technique, the copper complex was found to be most effective against Gram‐positive (S. aureus) bacteria. [CuLCl2] showed good hydrogen bonding interaction with the major‐groove (C2.G13 base pair) of A‐DNA. Density functional theory (DFT) calculations of the structural and electronic properties of the complexes revealed that [CuLCl2] had a smaller HOMO‐LUMO gap, suggesting a higher tendency to donate electrons to electron‐accepting species of biological targets.  相似文献
