In this Communication, we report water-soluble nano-C(60) in the first use as an effective fluorescent sensing platform for the highly sensitive and selective detection of Ag(+). The general concept used in this approach is based on a fluorescently labeled single-stranded DNA (ssDNA) probe that adsorbs on nano-C(60), leading to substantial dye fluorescence quenching; however, in the presence of Ag(+), C-Ag(+)-C coordination induces the probe to fold into a hairpin structure, which does not adsorb on nano-C(60) and thus retains the dye fluorescence. This sensing system exhibits a detection limit as low as 1 nM and has a high selectivity against other metal ions. Finally and most importantly, we demonstrate its performance in real sample analysis.  相似文献

The design of devices with multiple functions, simple handling procedures and sufficient sensitivity has drawn great interests in the field of analysis. Metal-nucleotide based pairs, such as T-Hg(2+)-T and C-Ag(+)-C complexes accompanied by SYBR Green I (SG), are used to selectively bind duplex-strand DNA by observing a bright fluorescence signal in this work, thus yielding a simple method for the rapid detection of Hg(2+) and Ag(+) without a complex labeling process. Based on this principle, 'OR' and 'AND' logic gates for the multiplexed analysis of Hg(2+) and Ag(+) were developed, and their practical application for the detection of Hg(2+) and Ag(+) in drinking water was reported.  相似文献

The authors describe an oligonucleotide-based lateral flow test for visual detection of Ag(I). The assay is based on cytosine-Ag(I)-cytosine [C-Ag(I)-C] coordination chemistry to capture gold nanoparticle (AuNP) tags in the test zone. A thiolated C-rich oligonucleotide probe was immobilized on the AuNPs via gold-thiol chemistry, and a biotinylated C-rich oligonucleotide probe was immobilized on the test zone. The AuNPs labelled with C-rich oligonucleotides are captured by Ag(I) ions in the test zone through the C-Ag(I)-C coordination. The resulting accumulation of AuNPs produces a readily visible red band in the test zone. Under optimized conditions, the test is capable of visually detecting 1.0 ppb of Ag(I) which is 50 times lower than the maximum allowable concentration as defined by the US Environmental Protection Agency for drinking water. Hence, the test is inexpensive and highly sensitive. It was applied to the detection of Ag(I) in spiked samples of tap water and river water. In our perception, the test is a particularly valuable tool in limited resource settings.

In this article we investigate the effect of monovalent cations (Li(+), Na(+), K(+), Cs(+)) on self-assembly of thiol-modified double-stranded DNA (ds-DNA) and single-stranded DNA (ss-DNA) on gold electrodes. Electrochemical characteristics (surface coverage, ion penetration and charge transfer) of ds-DNA and ss-DNA self-assembled monolayers (SAMs) formed with different monovalent cations are inspected based on six important interfacial parameters including surface coverage (Γ(m)), interfacial capacitance (C), phase angle (Φ(1 Hz)), ion transfer resistance (R(it)*), current density difference (Δj) and charge transfer resistance (R(ct)) from chronocoulometry (CC), cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). Three sections are included: (1) Investigation of the relationships of parameters (Γ(m), C, Φ(1 Hz), R(it)*, Δj and R(ct)) for ds-DNA-SAMs and ss-DNA-SAMs with cation types and concentrations; (2) confirmation and explanation of our experimental results combined with our recently proposed simple DNA model and literature reports; (3) exploration of the mechanism for the orders of monovalent cations (Li(+), Na(+), K(+), Cs(+)) on availing the adsorption of ds-DNA and ss-DNA molecules on gold based on their physicochemical parameters (ion size, solvation free energy and enthalpy, ion-water bond length and water exchange rate) and possible binding modes with DNA molecules. This work might provide a useful reference for understanding interactional mechanism of cations with DNA molecules.  相似文献

A colorimetric assay has been developed for the simultaneous selective detection of silver(I) and mercury(II) ions utilizing metal nanoparticles (NPs) as sensing element based on their unique surface plasmon resonance properties. In this method, sulfhydryl group modified cytosine-(C)-rich ssDNA (SH-C-ssDNA) was self-assembled on gold nanoparticles (AuNPs) to produce the AuNPs-C-ssDNA complex, and sulfhydryl group modified thymine-(T)-rich ssDNA (SH-T-ssDNA) was self-assembled on silver nanoparticles (AgNPs) to produce the AgNPs-T-ssDNA complex. Oligonucleotides (SH-C-ssDNA or SH-T-ssDNA) could enhance the AuNPs or AgNPs against salt-induced aggregation. However, the presence of silver(I) ions (Ag(+)) in the complex of ssDNA-AuNPs would reduce the stability of AuNPs due to the formation of Ag(+) mediated C-Ag(+)-C base pairs accompanied with the AuNPs color change from red to purple or even to dark blue. Moreover, the presence of mercury(II) ions (Hg(2+)) would also reduce the stability of AgNPs due to the formation of Hg(2+) mediated T-Hg(2+)-T base pairs accompanied with the AgNPs color change from yellow to brown, then to dark purple. The presence of both Ag(+) and Hg(2+) will reduce the stability of both AuNPs and AgNPs and cause the visible color change. As a result, Ag(+) and Hg(2+) could be detected qualitatively and quantitatively by the naked eye or by UV-vis spectral measurement. The lowest detectable concentration of a 5 nM mixture of Ag(+) and Hg(2+) in the river water was gotten by the UV-vis spectral measurement.  相似文献

Potentiometric sensors based on glassy carbon electrode covered with polyaniline and thiacalix[4]arenes containing amidopyridine, morpholide, pyrrolidide and hydrazide functional groups in cone, partial cone and 1,3-alternate conformations have been developed and applied for determination of Ag(+) ions in the range from 1.0 x 10(-2) to 4.0 x 10(-7)M and limits of detection from 1 x 10(-7) to 3.5 x 10(-8)M. The sensitivity of Ag(+) detection decreases in the following range of thiacalix[4]arene substituents: morpholide>pyrrolidide>amidopyridine>hydrazide. Potentiometric selectivity coefficients predominantly showed binding of Ag(+), Hg(II) and Fe(III) ions over other transient and alkali metals. The influence of functional groups and conformation of receptor on the selectivity of the sensor response was investigated. As shown, selectivity and sensitivity of Ag(+) determination depends on the steric accessibility of the binding site and flexibility of the receptor structure. For Fe(III) ions, changes of the sensor potential are also determined by their implementation in redox reactions of polyaniline.  相似文献

A highly sensitive and selective label-free electrochemical sensor was developed for the determination of kanamycin. To improve the sensitivity of the electrochemical sensor, graphene-gold nanoparticles were prepared by a one-step electrochemical coreduction process and were modified on the surface of a glassy carbon electrode. The double-stranded DNA(ds-DNA) duplex probe was immobilized onto the graphene-gold nanoparticle-modified electrode. The introduction of target kanamycin induced the displacement of aptamer from the ds-DNA duplex into the solution. Methylene blue(MB) as a redox indicator monitored the current change using differential pulse voltammetry. Under optimal conditions, the designed electrochemical aptasensor exhibited a wide linear range from 0.1 pmol/L to 10 pmol/L with a detection limit of 0.03 pmol/L for kanamycin. The experimental strategy enabled the direct analysis of milk samples, and the results showed high sensitivity and good selectivity.  相似文献

An integrated PCR-free DNA sensor, which combines a sequence-specific receptor, an optical polymeric transducer, and an intrinsic fluorescence amplification mechanism, is reported. This sensor is based on the different conformations adopted by a cationic polythiophene when electrostatically bound to ss-DNA or ds-DNA, and on the efficient and fast energy transfer between the resulting fluorescent polythiophene/ds-DNA complex and neighboring fluorophores attached to ss-DNA probes. This molecular system allows the detection of only five molecules in 3 mL of an aqueous solution, or 3 zM, in 5 min. Moreover, this work demonstrates, for the first time, the direct detection of single nucleotide polymorphisms (SNPs) from clinical samples in only a few minutes, without the need for nucleic acid amplification.  相似文献

(133)Cs NMR spectroscopy was used to determine the stoichiometry and stability of the Cs(+) ion complex with dibenzo-21-crown-7 (DB21C7) in acetonitrile-dimethylsulfoxide (96.5:3.5, w/w) and nitromethane-dimethylsulfoxide (96.5:3.5, w/w) mixtures. A competitive (133)Cs NMR technique was also employed to probe the complexation of Na(+), K(+), Rb(+), Ag(+), Tl(+), NH(4)(+), Mg(2+), Ba(2+), Hg(2+), Pb(2+) and UO(2)(2+) ions with DB21C7 in the same solvent systems. All the resulting 1:1 complexes in nitromethane-dimethylsulfoxide were more stable than those in acetonitrile-dimethylsulfoxide solution. In both solvent systems, the stability of the resulting complexes was found to vary in the order Rb(+)>K(+) approximately Ba(2+)>Tl(+)>Cs(+)>NH(4)(+) approximately Pb(2+)>Ag(+)>UO(2)(2+)>Hg(2+)>Mg(2+)>Na(+).  相似文献

The synthesis and photophysical properties of a rosamine-based fluorescent chemosensor, RosAg, for detecting Ag ion in an aqueous solution are described. This fluorescent sensor has a negligible quantum yield (<0.005) in the absence of Ag(+), whereas a significant increase in fluorescence is observed upon complexation with Ag(+) under physiological conditions. The crystal structure of the silver complex with the chelator moiety of RosAg reveals a trigonal-planar coordination geometry in which three S atoms occupy the metal center. Although a strong coordinative interaction of Ag-N is not observed in the crystal structure, the (1)H NMR experiments suggest that aniline nitrogen is likely to be associated with the Ag(+) center in the solution state. This may inhibit the photoinduced electron transfer process and result in the enhancement of fluorescence.  相似文献

Development of an electrochemical DNA biosensor for the direct detection and discrimination of double-stranded oligonucleotide (dsDNA) corresponding to hepatitis C virus genotype 3a, without its denaturation, using a gold electrode is described. The electrochemical DNA sensor relies on the modification of the gold electrode with 6-mercapto-1-hexanol and a self-assembled monolayer of 14-mer peptide nucleic acid probe, related to the hepatitis C virus genotype 3a core/E1 region. The increase of differential pulse voltammetric responses of methylene blue, upon hybridization of the self-assembled probe with the target ds-DNA to form a triplex is the principle behind the detection and discrimination. Some hybridization experiments with non-complementary oligonucleotides were carried out to assess whether the developed DNA sensor responds selectively to the ds-DNA target. Diagnostic performance of the biosensor is described and the detection limit was found to be 1.8 × 10−12 M in phosphate buffer solution, pH 7.0. The relative standard deviation of measurements of 100 pM of target ds-DNA performed with three independent probe-modified electrodes was 3.1%, indicating a remarkable reproducibility of the detection method.  相似文献

Two novel conjugated polymers containing a 2,2'-biimidazole moiety have been designed, synthesized, and demonstrated to be used as an effective fluorescent sensing platform for detection of Ag(+) and cysteine. Our studies show that electrochemical impedance spectroscopy (EIS) and scanning electrochemical microscopy (SECM) of films of ds-DNA on gold allow us to distinguish between mitochondrial DNA fragments of the cytochrome c(1) oxidase (mt-Cox1) of three related species of the subfamily 'Bovinae' (Bos taurus, Bison bison, and Bison bonasus). In EIS, a perfectly matched DNA gives rise to a considerably larger charge transfer resistance R(ct) compared to mismatched pairings. Differences in charge transfer resistance, ΔR(ct), before and after the addition of Zn(2+) ions provide an additional tool for identification. In addition, all ds-DNA films were studied by SECM and their kinetic parameters were determined. Perfectly matched ds-DNAs are readily distinguished from mismatched duplexes by their lower rate constants. Our system can be used multiple times by dehybridization and rehybridization of capture strands up to the 250 pmole level.  相似文献

A green and simple chemical synthesis of magic sized water soluble blue-emitting ZnS quantum dots (QDs) has been accomplished by reacting anhydrous Zn acetate, sodium sulfide and thiolactic acid (TLA) at room temperature in aqueous solution. Refluxing of this mixture in open air yielded ZnS clusters of about 3.5 nm in diameter showing very strong and narrow photoluminescence properties with long stability. Refluxing did not cause any noticeable size increment of the clusters. As a result, the QDs obtained after different refluxing conditions showed similar absorption and photoluminescence (PL) features. Use of TLA as a capping agent effectively yielded such stable and magic sized QDs. The as-synthesized and 0.5 h refluxed ZnS QDs were used as a fluorescence sensor for Ag(+) ions. It has been observed that after addition of Ag(+) ions of concentration 0.5-1 μM the strong fluorescence of ZnS QDs was almost quenched. The quenched fluorescence can be recovered by adding ethylenediamine to form a complex with Ag(+) ions. The other metal ions (K(+), Ca(2+), Au(3+), Cu(2+), Fe(3+), Mn(2+), Mg(2+), Co(2+)) showed little or no effect on the fluorescence of ZnS QDs when tested individually or as a mixture. In the presence of all these ions, Ag(+) responded well and therefore ZnS QDs reported in this work can be used as a Ag(+) ion fluorescence sensor.  相似文献

In this article we studied the permeable characteristics of thiol-modified double-stranded DNA (ds-DNA) self-assembled monolayers (SAMs) on a gold substrate assembled under different NaCl concentrations by electrochemical methods. It was based on the inspection of five important parameters including interfacial capacitance (C), phase angle (Φ(1?Hz)), ions transfer resistance (R(it)*), current density difference (Δj) and electron transfer rate (k(et)) through cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). Three sections were included: (1) Investigation of the relationships of C, Φ(1?Hz), R(it)*, Δj and k(et) with NaCl concentrations and comparison with the reports from literature. Experimental results showed that ds-DNA-SAMs were permeable films. (2) Construction of a simple model for exploring the permeable characteristics of ds-DNA-SAMs on gold. (3) Confirmation of the simple model by chronocoulometry (CC) and application of the model to explain the permeable mechanism. This study was significant for exploring the mechanism of electron transfer through the interior of ds-DNA duplex helix.  相似文献

Using a cascade signal amplification strategy, an ultrasensitive electrochemical biosensor for specific detection of DNA based on molecular beacon (MB) mediated circular strand displacement polymerization (CSDP) and hyperbranched rolling circle amplification (HRCA) was proposed. The hybridization of MB probe to target DNA resulted in a conformational change of the MB and triggered the CSDP in the presence of bio-primer and Klenow fragment (KF exo), leading to multiple biotin-tagged DNA duplex. Furthermore, the HRCA was implemented to product amounts of double-stranded DNA (ds-DNA) fragments using phi29 DNA polymerase via biotin-streptavidin interaction. After the product of HRCA binded numerous biotinylated detection probes, an ultrasensitive electrochemical readout by further employing the streptavidin-alkaline phosphatase. The proposed biosensor exhibited excellent detection sensitivity and specificity with a log-linear response to target DNA from 0.01 fM to 10 pM as low as 8.9 aM. The proposed method allowed DNA detection with simplicity, rapidness, low cost and high specificity, which might have the potential for application in clinical molecular diagnostics and environmental monitoring.  相似文献

A new 1,1'-thiobis(2-naphthoxy)-based receptor molecule (L) containing a benzimidazole moiety has been synthesized and characterized by (1)H NMR, ESI-MS, and elemental analysis. The selectivity of L has been explored in aqueous methanol, resulting in selective (7.5 ± 0.5)-fold switch-on fluorescence response toward Ag(+) among 14 different transition, alkali, and alkaline earth metal ions studied. The complexation of Ag(+) by L has been addressed by ESI-MS, (1)H NMR, and UV-vis spectra. Microstructural features of L and its Ag(+) complex have been measured by AFM and TEM. The morphological features of L alone and L in the presence of Ag(+) differ dramatically both in shape and size, and the ion induces the formation of chains owing to its coordinating ability toward benzimidazole. Further, the in situ [Ag(+)-L] complex was titrated against 20 naturally occurring amino acids and found that this complex acts as a secondary recognition ensemble toward Cys, Asp, and Glu by switch-off fluorescence.  相似文献

ArgenCast-1 (1), a photocage for silver utilizing acyclic polythioether 3,6,12,15-tetrathia-9-azaheptadecane receptor and 4,5-dimethoxy-2-nitrobenzyl (DMNB) chromophore has been prepared using trimethylsilyl trifluoromethanesulfonate-assisted electrophilic aromatic substitution. Metal binding studies with ArgenCast-1 reveal interactions with Ag(+), Hg(2+) and Cu(+), but only Ag(+) coordinates in both aqueous and organic solvents. The uncaging mechanism of ArgenCast-1 metal complex involves a photoreaction that converts the nitrobenzydrol into the electron withdrawing nitrosobenzophenone that participates in a resonance interaction with a metal-bound aniline nitrogen atom. The structural change following photolysis decreases availability of the nitrogen lone pair for Ag(+) coordination, but strong interactions between Ag(+) and the thioether ligands mitigates metal ion release. A resonance interaction with a key aci-nitro intermediate reduces the photolysis quantum yield of ArgenCast-1, so several naphthyl-based nitrobenzyl groups were screened as alternatives to DMNB. The naphthyl Ag(+) photocages, ArgenCast-2 and -3, exhibit nearly identical quantum yield to ArgenCast-1 owing to the dominance of resonance between aci-nitro intermediate and the aniline nitrogen atom.  相似文献
