Histology, including immunohistochemistry, and magnetic resonance imaging microscopy (microMRI) are complementary techniques for the analysis of brain structure. Therefore, microMRI analysis, often of formalin-fixed tissue, precedes histologic evaluation of the same experimental animal in many studies. However, the application of gadopentetate dimeglumine (Gd-DTPA), while of value for MRI studies, has an unknown effect on subsequent histology. We demonstrate here that for the mouse brain, histology with Nissl staining and immunostaining for microtubule-associated protein 2, using standard techniques for tissue preparation, are unaffected by prior perfusion of the tissue with Gd-DTPA. This conclusion was based on qualitative morphologic comparisons of stained sections, as well as quantification of mean immunofluorescence pixel intensities from Gd-treated (mean+/-S.D.=131.2+/-28.4; n=3) as compared to nontreated specimens (116.2+/-34.7; n=3, P=.7). Therefore, Gd-DTPA may be applied as a microMRI contrast agent in formalin-fixed brain tissue prior to histologic studies.  相似文献

In dynamic susceptibility contrast MRI, the shape of the arterial input function (AIF) is commonly obtained in the near vicinity of the middle cerebral artery (MCA). However, the tissue regions where the AIF is sampled also have significant perfusion, which contributes to T(2)* changes. We investigate whether correction of this effect will introduce significant changes in the measurement of the AIF and, subsequently, the assessment of the mean transit time (MTT). Clinical dynamic susceptibility data from 13 patients with brain tumors were analyzed. Patients received either single or double doses of Magnevist followed by a saline flush through a power injector. In the correction procedure, DeltaR(2)* was sampled in a region of gray matter approximately 1-2 cm away from the MCA and then subtracted from the DeltaR(2)* sampled in the immediate vicinity of the MCA. We demonstrate that in the brain, this correction of DeltaR(2)* due to tissue perfusion leads to a narrower width of the AIF curve obtained with DeltaR(2)* (mean+/-S.D.=7.3+/-2.0 and 6.4+/-1.7 s, before and after correction, respectively, P<.001 using a two-tailed paired t-test). Furthermore, the peak of the AIF also moved to a slightly earlier time relative to the time of arrival (mean+/-S.D.=4.7+/-0.9 and 4.3+/-0.8 s, before and after correction, with P<.001). With the use of the corrected AIF, the measured MTT had increased values in areas of both gray and white matter.  相似文献

The E200K mutation on chromosome 20 can cause familial Creutzfeldt-Jakob disease (CJD). Patients with this mutation are clinically similar to those with sporadic CJD, but their imaging features are not well documented. We report here the quantitative and qualitative evaluation of the magnetic resonance (MR) imaging characteristics of this unique group of patients using three-dimensional spoiled gradient recalled (SPGR) echo images, diffusion-weighted imaging (DWI) with apparent diffusion coefficient (ADC) measurements, MR spectroscopy and a fluid-attenuated inversion recovery (FLAIR) sequence. The SPGR and ADC data were analyzed with SPM99. ANCOVA and regression models were used for a region-of-interest (ROI) analysis of ADC and metabolic ratios. CJD patients had a decreased fraction of gray matter and an increased fraction of cerebrospinal fluid (P=.001) in the cortex and cerebellum and increased ADC values in the cortex (P<.001). Focal decreases of ADC were found in the putamen via ROI analysis (548+/-83 vs. 709+/-9 microm(2)/s, P=.02). N-acetyl aspartate (NAA) was generally reduced, with the NAA/Cho ratio lowest in the cingulate gyrus. Qualitative assessment revealed hyperintensities on FLAIR, DWI or both in the putamen (three out of four patients), caudate (three out of four patients) and thalamus. These results provide a framework for future study of patients with genetically defined familial CJD.  相似文献

Purpose

To prospectively evaluate the feasibility of diffusion-weighted magnetic resonance imaging (DWI) for monitoring early treatment response to chemoradiotherapy (CRT) of nasopharyngeal carcinoma (NPC).

Materials and methods

Thirty-one patients with stage III and IV NPC were enrolled in this study from February 2012 to November 2012.T2-weighted and DWI sequences with diffusion factor of 0 and 800mm²/s were performed using a 3.0 T Philips Achieva TX scanner at baseline and 3 days, 20 days (after the first cycle of chemotherapy), 50 days (6 days after radiotherapy initiation) after neoadjuvant chemotherapy (NAC) initiation. The diameter of each primary lesion and target metastatic lymph node before and after the first cycle of NAC was measured and classified into stable disease (SD), partial response (PR) or completed response (CR) based on RECIST 1.1. The apparent diffusion coefficient (ADC) values and changes compared to baseline at each time point were compared between responders (CR and PR) and non-responders (SD). The rates of residual at the end of CRT were compared between these two groups.

Results

A significant increase in ADC was observed at each stage of therapy (P=.001) in lesions of primary and metastatic. The ADC values (ADC), ADC changes (ΔADC) and percentage ADC changes (Δ%ADC) of day 20 in responders were significantly higher than in non-responders for both primary lesions (p=.005, p=.006, p=.008, respectively) and metastatic lymph nodes (p=.002, p=.002, p=.003). Non-responders showed a higher rate of residual for both primary lesions (p=.008) and metastatic lymph nodes (p=.024) than responders.

Conclusions

DW MR imaging allows for detecting early treatment response of NPC. Patients with high ADC values and large ADC increase early after NAC initiation tended to respond better to CRT. Thus, accessing the curative effect of NAC in advanced NPC provides the opportunity to adjust following CRT regimen.  相似文献

BACKGROUND AND PURPOSE: The stage at which normal appearing white matter (NAWM) abnormalities first appear in multiple sclerosis (MS) is not clear. The aim of our study was to monitor water diffusion changes over time in NAWM of patients with early MS.METHODS: Out of a consecutive series of patients enrolled in a MR study on clinically isolated syndrome (CIS), we selected 19 subjects who had completed a one year follow-up. The MR scans obtained at baseline and at 12 months were reviewed according to the new criteria on the diagnosis of MS. Lesion load on T2 and T1 weighted images and the trace of the apparent diffusion coefficient in NAWM were measured both at baseline and at 12 months in patients and in 12 healthy controls.RESULTS: In three patients the diagnosis of MS was done at baseline based on MR. Thirteen patients developed MS during the study and in three patients the diagnosis remained "possible MS." TADC in NAWM in patients was significantly higher than in controls at the 12 months' follow-up but not at baseline (controls mean tADC +/- sd = 0.745 +/- 0.02 mm(2)/sec x 10(-3); patients mean tADC(12) +/- sd = 0.767 +/- 0.02 mm(2)/sec x 10(-3); p < 0.02). TADC and T2 lesion load at 12 months were significantly correlated (p < 0.01). Patients exhibiting tADC(12) above a confidence interval had a significantly greater EDSS score at the same time period (EDSS(12) +/- sd = 1.9 +/- 0.5 and = 1.1 +/- 0.4 respectively; p < 0.01).CONCLUSIONS: This study suggests that diffusion MR cannot detect alterations in NAWM of patients with a CIS suggestive of MS. After one year, when most patients develop MS, diffusion MR abnormalities in NAWM become apparent. These abnormalities are correlated with T2 lesion load and may contribute to neurological impairment.  相似文献

Early ischemic change after stroke can be demonstrated with diffusion-weighted imaging (DWI) and quantified by measuring the apparent diffusion coefficient (ADC) and/or lesion volume. We examined the reliability and reproducibility of lesion volume and ADC measurement on DWI images, and discuss the implications for clinical studies. Using 38 DWI scans from 15 stroke patients, two observers (a physicist and a neuroscience graduate) blind to each other, recorded the lesion volume on DWI sequences, measured the ADC values in this volume and calculated the ratio of ischemic: control ADC (ADCr). One observer repeated his measurements blind to his first, and also examined the effect on lesion volume and ADC of deliberately varying by only one pixel, the outline of the visible boundary of the lesion. The inter and intra-rater reliability were worse for lesion volume than ADC or ADCr measurements: lesion volume, inter-rater coefficient of variation (CoV) 85 +/- 130%, intra-rater CoV 20+/-SD80% (p < 0.05); ADC inter-rater CoV 7.7 +/- SD 19%, intra-rater CoV 0.2 +/- SD 12% (p = NS); and ADCr inter-rater CoV 8 +/- SD27%, intra-rater CoV 0.8 +/- SD73% (p = NS). Altering the position of the outline tracing of the lesion boundary by one pixel altered the measured volumes by 22 +/- SD25% (p < 0.05), but ADC values were altered by only 2.9 +/- SD4.9% and ADCr by 2.7 +/- SD4.8% (p = NS). ADC and ADCr values are more reliable and reproducible than DWI lesion size in acute ischemic stroke because altering where the lesion boundary is measured has a much greater impact on lesion volume than on the ADC or ADCr. The main objective of this article was (i) to refocus the residual dipolar and quadrupolar interactions in anisotropic tissues employing magic sandwich echo (MSE) imaging and to compare the results with that of conventional spin-echo (SE) imaging, and (ii) to quantify MSE relaxation and dispersion characteristics in bovine Achilles tendon and compare with spin-lattice relaxation time constant in the rotating frame (T(1rho)). Magic sandwich echo weighted images are approximately 75-100% higher in signal-to-noise ratio than the corresponding T(2)-weighted images. Magic sandwich echo relaxation times varied from 13+/-2 to 19+/-3 ms (mean+/-S.D.), depending upon the structural location of tendon. T(2) relaxation times only varied from 4+/-1 to 10+/-3 ms (mean+/-S.D.) on the same corresponding locations. Magic sandwich echo provides approximately 100% enhancement in relaxation times compared to T(2). Preliminary results based on bovine Achilles tendon and cartilage specimens suggest that the MSE technique has potential for refocusing residual dipolar as well as quadrupolar interactions in anisotropic systems and yields higher intensities than conventional SE imaging as well as T(1rho)-encoded imaging, especially at low-burst pulse amplitudes (250 and 500 Hz).  相似文献

Artificial neuronal network (ANN) in classification of glioblastoma multiforme (GBM) recurrence from treatment effects using advanced magnetic resonance imaging techniques was evaluated. In 56 patients with treated GBM, normalised minimal and mean apparent-diffusion coefficient (ADC) values, vessels number on susceptibility-weighted images (SWI) and Cho/Cr ratio were analysed statistically and by ANN. Significant correlation exists between normalised minimal and mean ADC values, and no correlation between ADC and Cho/Cr values. Cut-off values for tumour presence were: 1.14 for normalised minimal ADC (54% sensitivity, 71% specificity), 1.13 for normalised mean ADC (51% sensitivity, 71% specificity), 1.8 for Cho/Cr ratio (92% sensitivity, 82% specificity), grade 2 for SWI (87% sensitivity, 82% specificity). An accurate prediction of ANN to classify patients into GBM progression or treatment effects group was 99% during the training and 96.8% during the testing phase. Multi-parametric ANN allows distinction between GBM recurrence and treatment effects, and can be used in clinical practice.  相似文献

We present a measurement of time-dependent CP asymmetries and an updated determination of the CP-odd fraction in the decay B0-->D(*+)D(*-) using a data sample of 88x10(6)BB pairs collected by the BABAR detector at the PEP-II B Factory at SLAC. We determine the CP-odd fraction to be 0.063+/-0.055(stat)+/-0.009(syst). The time-dependent CP asymmetry parameters Im(lambda(+)) and /lambda(+)/ are determined to be 0.05+/-0.29(stat)+/-0.10(syst) and 0.75+/-0.19(stat)+/-0.02(syst), respectively. The standard model predicts these parameters to be -sin(2beta and 1, respectively, in the absence of penguin diagram contributions.  相似文献

Hemochromatosis is a hereditary iron overload syndrome characterized by increased iron storage, followed by liver cirrhosis and is often associated with restrictive cardiomyopathy. The purpose of this study was to detect alterations of cardiac high-energy phosphate metabolism in patients with hereditary hemochromatosis (HHC) prior to the development of structural heart diseases. Therefore cardiac phosphorus-31 two-dimensional chemical shift imaging ((31)P 2D CSI) was employed. Twenty-four male patients (mean age 47.2 +/- 12 years) homozygous for the C282Y mutation in the hemochromatosis associated HFE gene and twenty-four male healthy volunteers (mean age 47 +/- 11 years) as age-matched controls were included in this study. Using a 1.5-Tesla whole-body magnetic resonance scanner, electrocardiograph-triggered transversal 31P 2D CSI was performed. Left ventricle mean phosphocreatine (PCr) to beta-adenosine triphosphate (beta-ATP) ratios of patients with HHC (1.60 +/- 0.41) were significantly decreased in comparison to healthy volunteers (1.93 +/- 0.36; p = 0.004). Furthermore, we detected moderate, negative correlations between left ventricular PCr to beta-ATP ratios and transferrin saturation, cholesterol, low-density lipoprotein as well as triglyceride. This study shows that 31P 2D CSI permits the detection of alterations of cardiac high-energy phosphate metabolism in patients with HHC, but without any evidence for heart disease. The decreased PCr to beta-ATP ratios in HHC might be caused by mitochondrial impairment due to cardiac iron overload.  相似文献

In this paper we first present a CG-type method for inverse eigenvalue problem of constructing real and symmetric matrices $M$, $D$ and $K$ for the quadratic pencil $Q(\lambda)=\lambda^2M+\lambda D+K$, so that $Q(\lambda)$ has a prescribed subset of eigenvalues and eigenvectors. This method can determine the solvability of the inverse eigenvalue problem automatically. We then consider the least squares model for updating a quadratic pencil $Q(\lambda)$. More precisely, we update the model coefficient matrices $M$, $C$ and $K$ so that (i) the updated model reproduces the measured data, (ii) the symmetry of the original model is preserved, and (iii) the difference between the analytical triplet $(M, D, K)$ and the updated triplet $(M_{\text{new}}, D_{\text{new}}, K_{\text{new}})$ is minimized. In this paper a computationally efficient method is provided for such model updating and numerical examples are given to illustrate the effectiveness of the proposed method.  相似文献

Using the CLEO detector at the Cornell Electron Storage Ring, we have studied the distribution of kinematic variables in the decay lambda(+)(c)lambda--> e(+)nu(e). By performing a four-dimensional maximum likelihood fit, we determine the form factor ratio, R= f(2)/f(1) = -0.31 +/- 0.05(stat) +/- 0.04(syst), the pole mass, M(pole) = [2.21 +/- 0.08(stat) +/- 0.14(syst)] GeV/c(2), and the decay asymmetry parameter of the lambda(+)(c), alpha (lambda(c)) = -0.86 +/-0.03(stat) +/- 0.02(syst), for q(2) = 0.67 (GeV/c(2))(2). We compare the angular distributions of the lambda(+)(c) and lambda(-)(c) and find no evidence for CP violation: A(lambda(c)) = (alpha(lambda(c)) + alpha (lambda(c)))/(alpha(lambda(c))-alpha(lambda(c))) = 0.00 +/- 0.03(stat) +/- 0.01(syst) +/- 0.02, where the third error is from the uncertainty in the world average of the CP-violating parameter, A(lambda), for ppi(-).  相似文献

Diffusion tensor imaging (DTI) studies of human ischemic stroke within 24 h of symptom onset have reported variable findings of changes in diffusion anisotropy. Serial DTI within 24 h may clarify these heterogeneous results. We characterized longitudinal changes of diffusion anisotropy by analyzing discrete ischemic white matter (WM) and gray matter (GM) regions during the hyperacute (2.5-7 h) and acute (21.5-29 h) scanning phases of ischemic stroke onset in 13 patients. Mean diffusivity (MD), fractional anisotropy (FA) and T2-weighted signal intensity were measured for deep and subcortical WM and deep and cortical GM areas in lesions outlined by a > or =30% decrease in MD. Average reductions of approximately 40% in relative (r) MD were observed in all four brain regions during both the hyperacute and acute phases post stroke. Overall, 9 of 13 patients within 7 h post symptom onset showed elevated FA in at least one of the four tissues, and within the same cohort, 11 of 13 patients showed reduced FA in at least one of the ischemic WM and GM regions at 21.5-29 h after stroke. The fractional anisotropy in the lesion relative to the contralateral side (rFA, mean+/-S.D.) was significantly elevated in some patients in the deep WM (1.10+/-0.11, n=4), subcortical WM (1.13+/-0.14, n=4), deep GM (1.07+/-0.06, n=1) and cortical GM (1.22+/-0.13, n=5) hyperacutely (< or =7 h); however, reductions of rFA at approximately 24 h post stroke were more consistent (rFA= 0.85+/-0.12).  相似文献

We present NMR measurements of the diffusion of hyperpolarized 3He in the human lung performed at fields much lower than those of conventional MRI scanners. The measurements were made on standing subjects using homebuilt apparatus operating at 3mT. O(2)-limited transverse relaxation (T(2) up to 15-35s) could be measured in vivo. Accurate global diffusion measurements have been performed in vivo and in a plastic bag; the average apparent diffusion coefficient (ADC) in vivo was 14.2+/-0.6mm(2)/s, whereas the diffusion coefficient in the bag (3He diluted in N(2)) was 79.5+/-1mm(2)/s. 1D ADC mapping with high SNR ( approximately 200-300) demonstrates the real possibility of performing quality lung imaging at extremely low fields.  相似文献

PURPOSE: To describe the use of 3.0-T magnetic resonance imaging (MRI) for the evaluation of chronic liver diseases. MATERIALS AND METHODS: Two groups of patients who had chronic liver diseases and underwent 3.0-T MRI for evaluation of the liver were included in the study. The first group of patients included 66 consecutive patients (33 male, 33 female; mean age+/-standard deviation, 56+/-11). The second group of patients included 30 consecutive patients (18 males, 12 females; mean age+/-standard deviation, 53+/-10) in whom Variable-Rate Selective Excitation (VERSE) pulses and improved adjustments procedure were used during the acquisitions. Imaging findings of chronic liver diseases, predetermined artifacts and image quality of all individual sequences in the first group and predetermined artifacts and image quality of T2-weighted sequences in the second group were reviewed retrospectively and independently by two reviewers. chi-Square tests were used to compare the findings between two groups of patients and individual sequences. Kappa statistics were used to determine the extent of agreement between the reviewers. RESULTS: Fifteen dysplastic nodules in 6 of 66 (9%) patients and 12 hepatocellular carcinomas in 11 of 66 (17%) patients were detected. Excluding motion artifacts, three-dimensional (3D) T1-weighted gradient-echo (GE) sequence was the least affected sequence by the artifacts. Image quality of T1-weighted 3D-GE sequences was excellent in 43 of 66 (65%) patients. In-phase and out-of-phase T1-weighted spoiled GE (SGE) images were fair in 62 of 66 (94%) and 61 of 66 (92%) patients, respectively. The image quality of short tau inversion recovery (STIR) and half-Fourier rapid acquisition with relaxation enhancement (RARE) sequences were fair in 31 of 66 (47%) and 53 of 66 (80%) patients. STIR and half-Fourier RARE sequences in the second group demonstrated significantly better image quality (P=.03 and P<.0001). CONCLUSION: 3.0-T MRI allows the acquisition of very high quality postgadolinium 3D-GE sequence, which permitted the detection and characterization of lesions in the setting of chronic liver diseases. The use of VERSE pulses and improved adjustments procedure improved the image quality of T2-weighted sequences. In-phase/out-of-phase SGE sequences are at present of fair quality.  相似文献

Diffusion-weighted (DW) MRI at 1.5 T was carried out in two groups of patients. MRI data were correlated with the biopsy and histopathology (where available). The performance of two sequences -- a single-shot FSE (14 patients) and a single-shot EPI (15 patients) -- was compared. Average ADC values from the normal peripheral zone (PZ), central gland (CG) and the tumour [prostate carcinoma (PCa)] were calculated from b values of 0 and 600. Tukey-Kramer test was used for statistical analysis. EPI produced higher values of ADC (10(-3) mm(2)/s) than FSE sequence: 1.992+/-0.208 vs. 1.573+/-0.270 in PZ (P<.001), 1.518+/-0.126 vs. 1.373+/-0.179 in CG and 1.214+/-0.254 vs. 0.993+/-0.158 in PCa (P<.01). In conclusion, both EPI and FSE sequences showed differences in ADC between normal PZ, CG and PCa; however, EPI produced significantly higher ADC values than FSE.  相似文献

We study the time-dependent Dalitz plot of D-->K_{S};{0}pi;{+}pi;{-} in B;{0}-->D;{(*)}h;{0} decays, where h;{0} is a pi;{0}, eta, eta;{'}, or omega meson and D;{*}-->Dpi;{0}, using a data sample of 383x10;{6} Upsilon(4S)-->BB[over ] decays collected with the BABAR detector. We determine cos2beta=0.42+/-0.49+/-0.09+/-0.13, sin2beta=0.29+/-0.34+/-0.03+/-0.05, and |lambda|=1.01+/-0.08+/-0.02, where the first error is statistical, the second is the experimental systematic uncertainty, and the third, where given, is the Dalitz model uncertainty. Assuming the world average value for sin2beta and |lambda|=1, cos2beta>0 is preferred over cos2beta<0 at 86% confidence level.  相似文献

PurposeMagnetic resonance imaging is used to stage thyroid tumors. Diffusion weighted imaging (DWI) and apparent diffusion coefficient (ADC) can be used to reflect tumor microstructure. Our aim was to compare ADC values of malignant and benign thyroid lesions based on a large sample.MethodsMEDLINE library, EMBASE and SCOPUS databases were screened for the associations between ADC values and thyroid lesions up to August 2021. The primary endpoint of the systematic review were ADC values of benign and malignant thyroid lesions. In total, 29 studies were suitable for the analysis and were included into the present study.ResultsThe included studies comprised a total of 2137 lesions, 1118 (52.3%) benign and 1019 (47.7%) malignant lesions. The pooled mean ADC value of the benign thyroid lesions was 1.88 × 10−3 mm2/s [95% CI 1.77–2.0] and the pooled mean ADC value of malignant thyroid lesions was 1.15 × 10−3 mm2/s [95% CI 1.04–1.25].ConclusionsADC can well discriminate benign and malignant thyroid tumors. Therefore, DWI should be implemented into the presurgical diagnostic work-up in clinical routine.  相似文献

The conventional MR imaging appearance of gangliogliomas is often variable and nonspecific. Conventional MR images, relative cerebral blood volume (rCBV) and vascular permeability (K(trans)) measurements were reviewed in 20 patients with pathologically proven grade 1 and 2 gangliogliomas (n = 20) and compared to a group of grade 2 low-grade gliomas (n = 30). The conventional MRI findings demonstrated an average lesion size of 4.1 cm, contrast enhancement (n = 19), variable degree of edema, variable mass effect, necrosis/cystic areas (n = 8), well defined (n = 12), signal heterogeneity (n = 9), calcification (n = 4). The mean rCBV was 3.66 +/- 2.20 (mean +/- std) for grade 1 and 2 gangliogliomas. The mean rCBV in a comparative group of low-grade gliomas (n = 30), was 2.14 +/- 1.67. p Value < 0.05 compared with grade 1 and 2 ganglioglioma. The mean K(trans) was 0.0018 +/- 0.0035. The mean K(trans) in a comparative group of low-grade gliomas (n = 30), was 0.0005 +/- 0.001. p Value = 0.14 compared with grade 1 and 2 ganglioglioma. The rCBV measurements of grade 1 and 2 gangliogliomas are elevated compared with other low-grade gliomas. The K(trans), however, did not demonstrate a significant difference. Gangliogliomas demonstrate higher cerebral blood volume compared with other low-grade gliomas, but the degree of vascular permeability in gangliogliomas is similar to other low-grade gliomas. Higher cerebral blood volume measurements can help differentiate gangliogliomas from other low-grade gliomas.  相似文献

We report studies of the Cabibbo-suppressed decay B+/--->D(CP)K+/-, where D(CP) denotes CP eigenstates of the D0-D0; system. The analysis is based on a 29.1 fb(-1) sample collected at the Upsilon(4S) resonance with the Belle detector at the KEKB asymmetric e(+)e(-) storage ring. We measure ratios of branching fractions, relative to Cabibbo-favored B+/--->D(CP)pi(+/-), of B(B--->D1K-)/B(B--->D1pi(-))=0.125+/-0.036+/-0.010 and B(B--->D2K-)/B(B--->D2pi(-))=0.119+/-0.028+/-0.006; the index 1 (2) denotes the CP=+1 (-1) eigenstate.
