Three novel complexes [Nd(L)(NO3)(H2O)2]·NO3·2H2O (HL1 = N-pyrimidine norcantharidin acylamide acid, C12H13N3O4; HL2 = N-pyridine norcantharidin acylamide acid, C13H14N2O4; HL3 = N-phenyl norcantharidin acylamide acid, C14H15NO4) were synthesized. HL1, HL2 and HL3 are the ligand of complex(1), complex(2) and complex(3), respectively. Their structures were characterized by elemental analysis, conductivity measurement, infrared spectra and thermogravimetric analysis. The DNA-binding properties of the complexes have been investigated by fluorescence spectroscopy and viscosity measurements. The results suggest that the complexes can bind to DNA by partial intercalation. The liner Stern-Volmer quenching constant Ksq values are 3.3(±0.21)(1), 1.7(±0.19)(2) and 0.9(±0.04)(3), respectively. Complex (1) and (2) have been found to cleave pBR322 plasmid DNA at physiological pH and temperature. The test of antiproliferation activity indicates that complex(1) has strong antiproliferative ability against the SMMC7721 (IC50 = 131.7 ± 23.4 μmol·L−1) and A549 (IC50 = 128.4 ± 19.9 μmol·L−1) cell lines. The inhibition rates of complex(2) (IC50 = 86.3 ± 11.3 μmol·L−1) are much higher than that of NCTD (IC50 = 115.5 ± 9.5 μmol·L−1) and HL2 (111.0 ± 5.7 μmol·L−1) against SMMC7721 cell lines.  相似文献

The flow injection technique is applied to study the binding to DNA of new platinum complexes—E1: ethylenediaminechlorocholylglycinateplatinum(II): [PtCl(CG)(en)], C54H92O12Pt and E2: ethylenediaminebischolylglycinateplatinum(II): [Pt(CG)2(en)], C28H50ClN3O6Pt—derived from cisplatin in which the exchangeable ligands were replaced by bile acids, such that these anticancer drugs have less toxicity and less resistance is developed towards them. Both compounds are fluorescent and their fluorescence is enhanced when they form adducts with DNA, a property that is extremely useful for monitoring the cytotoxic activity and their mechanisms of action. The binding parameters to DNA of E1 [apparent intrinsic binding constant KE1: (11.2 ± 0.4) × 103 M−1 and maximum number of binding sites per nucleotide, n E1: 0.121 ± 2 × 10−3) and E2 (KE2: 9.2 ± 0.7) × 103 M−1 and n E2 0.098 ± 2 × 10−3] were determined following the Scatchard method and the type of binding was studied experimentally through the modifications introduced by each of the compounds into the ethidium bromide–DNA bond.  相似文献

6-Hydroxy chromone-3-carbaldehyde-(4′-hydroxy) benzoyl hydrazone (L) and its Ln (III) complexes, [Ln = La, Nd, Eu and Tb] have been prepared and characterized on the basis of elemental analyses, molar conductivities, mass spectra, 1H NMR, thermogravimety/differential thermal analysis (TG-DTA), UV-vis spectra, fluorescence spectra and IR spectra. The formula of the complex is [Ln L·(NO3)2]·NO3. Spectrometric titration, ethidium bromide displacement experiments and viscosity measurements indicate that Eu (III) complex bind with calf-thymus DNA, presumably via an intercalation mechanism. The intrinsic binding constant of Eu (III) with DNA was 2.48 × 105 M−1 through fluorescence titration data.  相似文献

Layered single crystals of the (BEDO-TTF)6[M(CN)6](H3O,CH3CN)2 (M = Fe, Cr) compounds with alternating conducting layers of BEDO-TTF and [M(CN)6](H3O,CH3CN)2 are studied. The contributions to the magnetic susceptibility from charge carriers in BEDO-TTF layers and from the subsystem of localized magnetic moments of iron (or chromium) transition metal complexes are separated for both compounds under investigation. It is revealed that the crystals with [Fe(CN))6]3− anions at a temperature of ∼80 K and the crystals with [Cr(CN))6]3− anions at ∼30 K undergo magnetic transitions which are accompanied by drastic changes in the parameters of the EPR lines associated with the BEDO-TTF layers and the subsystem of localized spins of transition metal complexes. A new anthracene-based fluorescent PET sensor 1 with a tridentate ionophore of amide/β-amino alcohol displays very good selectivity and sensitivity for Fe3+ (K a = 1.6 × 103 M−1) and Hg2+ (K a = 2.1 × 103 M−1) in CH3CN–H2O (3:7, v/v) with detection limit of 1 μM. More fluorescence enhancement was observed when 1 selectively detected Fe3+ or Hg2+ in CH3CN and its detection limit was up to 0.03 μM.  相似文献

Novel bioactive complexes of Co(II), Cu(II), Ni(II) and Zn(II) metal ions with Schiff base ligand derived from histidine and 1,3-indandione were synthesized and thoroughly characterized by various analytical and spectral techniques. The biological investigations were carried out to examine the efficiency of the binding interaction of all the complexes with calf thymus DNA (CT-DNA). The binding properties were studied and evaluated quantitatively by Kb and Ksq values using UV-visible, fluorescence spectroscopy and voltammetric techniques. The experimental results revealed that the mode of binding of all the complexes with CT-DNA is via intercalation. It is further verified by viscosity measurements and thermal denaturation experiments. From the results of the cleavage study with pUC19 DNA it is inferred that all the complexes possess excellent cleaving ability. The present investigation proved that the binding interaction of all the complexes are significantly strong and the order of binding strength of the complexes is [Ni(L)2] (Kb = 3.11 × 106 M?1) > [Co(L)2] (Kb = 2.89 × 106 M?1) > [Cu(L)2] (Kb = 2.64 × 106 M?1) > [Zn(L)2] (Kb = 2.41 × 105 M?1). The complexes were also screened for antibacterial and anticandidal activity. The in vitro cytotoxicity of the ligand and complexes on the NIH/3 T3 mouse fibroblast cell lines were examined using CellTiter-Blue® (CTB) Cell viability assay, which unveiled that all the complexes exhibit more potent activities against NIH/3 T3 cells. The three Ru(II) complexes of [Ru(phen)2dppca]2+ (1) [Ru(bpy)2dppca]2+ (2) and [Ru(dmb)2dppca]2+ (3) (where phen = 1,10 phenanthroline, bpy = 2,2-bipyridine, dmb = 2 ,2-dimethyl 2′,2′-bipyridine and polypyridyl ligand containing a single carboxylate functionality dppca ligand (dipyridophenazine-11-carboxylic acid) have been synthesized and characterized. These complexes have been shown to act as promising calf thymus DNA intercalators and a new class of DNA light switches, as evidenced by UV-visible and luminescence titrations with Co2+ and EDTA, steady-state emission quenching by [Fe(CN)6]4− and KI, DNA competitive binding with ethidium bromide, viscosity measurements, and DNA melting experiments. The results suggest that 1, 2, and 3 complexes bind to CT-DNA through intercalation and follows the order 1 > 2 > 3. Under irradiation at 365 nm, the three complexes have also been found to promote the photocleavage of plasmid pBR322 DNA.  相似文献

Water-soluble Mn2+-doped ZnS quantum dots (QDs) were prepared using mercaptoacetic acid as the stabilizer. The optical properties and structure features were characterized by X-Ray, absorption spectrum, IR spectrum and fluorescence spectrum. In pH 7.8 Tris-HCl buffer, the QDs emitted strong fluorescence peaked at 590 nm with excitation wavelength at 300 nm. The presence of sulfide anion resulted in the quenching of fluorescence and the intensity decrease was proportional to the S2− concentration. The linear range was from 2.5 × 10−6 to 3.8 × 10−5 mol L−1 with detection limit as 1.5 × 10−7 mol L−1. Most anions such as F, Cl, Br, I, CH3CO2 , ClO4 , CO3 2−, NO2 , NO3 , S2O3 2−, SO3 2− and SO4 2− did not interfere with the determination. Thus a highly selective assay was proposed and applied to the determination of S2− in discharged water with the recovery of ca. 103%.  相似文献

We studied the spectral-luminescent characteristics of the luminescence of mixed-ligand polypyridine-phosphine complexes of ruthenium(II) cis-[Ru(bpy)2(PPh3)X](BF4) n with ligands 2,2′-bipyridyl (bpy) and triphenylphosphine (PPh3) and X = Cl, Br, CN, NO2, NH3, MeCN, pyridine (py), 4-aminopyridine (pyNH2), and 4,4′-bipyridyl (4,4′-bpy) in a 4: 1 EtOH-MeOH alcoholic mixture at 77 K. The radiative and nonradiative deactivation rate constants of the lowest electronically excited state of the complexes are determined. We find that triphenylphosphine has a greater effect on the photophysical characteristics of ruthenium(II) complexes compared to π-acceptor strong-field ligands, such as MeCN, CN, and NO2. At the same time, the characteristics of complexes cis-[Ru(bpy)2(PPh3)X] n+ considerably depend on the nature of the second monodentate ligand X, which is coordinated to ruthenium(II), and correlate with its position in the spectrochemical series of ligands.  相似文献

A sensitive and selective method for the trace determination of 3, 3’, 4, 4’-tetrachlorobiphenyl (PCB77) by using bovine serum albumin (BSA) as a fluorescence probe was introduced. Under optimum conditions, the enhanced fluorescence intensity was proportional to the concentration of polychlorinated biphenyls in the range of 8.9 × 10−8–5.0 × 10−6 mol L−1 for PCB77, and 5.0 × 10−7–5.0 × 10−6 mol L−1 for 2, 2’, 5, 5’-tetrachlorbiphenyl (PCB52). The detection limits (S/N = 3) of PCB77 and PCB52 were 2.6 × 10−8 mol L−1 and 2.9 × 10−7 mol L−1, respectively. Furthermore, the fluorescence enhancement mechanism was discussed in detail. Results indicated that fluorescence enhancement of the system originated from the formation of BSA-PCBs complexes. In addition, PCBs were mainly bound to the tyrosine residues in BSA molecules.  相似文献

In the paper, a chemiluminescence (CL) system was developed based on the catalytical effect of diperiodatocuprate (III) (DPC) on the 1,10-phenanthroline (phen)/hydrogen peroxide (H2O2) in the presence of cetyltrimethylammonium bromide (CTAB). The effects of experimental conditions were investigated. Meanwhile the increase of CL intensity of the DPC/phen/H2O2/CTAB system is proportional to the concentration of phen in the range of low concentration. The linear range of the calibration curve is 5.0 × 10−9–1.0 × 10−6 mol L−1, and the corresponding detection limit is 1.9 × 10−9 mol L−1. The effects of phenolic compounds (PCs) on the system were investigated. Hydroquinone was used as an example to investigate the application of the CL system to the determination of PCs. The quenched CL intensity is linearly related to the logarithm of concentration of hydroquinone. The linear range of the calibration curve is 2.5 × 10−9–1.0 × 10−5 g mL−1, and the corresponding detection limit is 1.8 × 10−9 g mL−1. This phen and hydroquinone can be synchronously determined. The method was applied to the determination of hydroquinone in water samples and the recoveries were from 92% to 106%.  相似文献

1-(4-Aminoantipyrine)-3-tosylurea (H2L) and its three lanthanide (III) complexes, M(H2L)3 3NO3 [where M = Nd(III), Sm(III) and Eu(III)], have been synthesized and characterized. In addition, the DNA-binding properties of the three complexes have been investigated by UV–vis (ultraviolet and visible) absorption spectroscopy, fluorescence spectroscopy, circular dichroism (CD) spectroscopy, cyclic voltammetry, and viscosity measurements. Results suggest that the three complexes bind to DNA via a groove binding mode. Furthermore, the antioxidant activity (superoxide and hydroxyl radical) of the metal complexes was determined by using spectrophotometer methods in vitro. These complexes were found to possess potent antioxidant activity and be better than standard antioxidants like vitamin C and mannitol. In this paper we reported a metal complex 1-Zn (2,5-di-[2-(3,5-bis(2-pyridylmethyl)amine-4-hydroxy-phenyl)-ethylene]-pyrazine-Zn) as a fluorescent probe sensing DNA. The result of the competitive experiment of the probe with ethidium bromide (EB) to bind DNA, absorption spectral change and polarization change in the presence and absence of DNA revealed that interaction between the probe and DNA was via intercalation. Ionic strength experiment showed the existence of electrostatic interaction as well. Scatchard plots also confirmed the combined binding modes. The fluorescence enhancement of the probe was ascribed to highly hydrophobic environment when it bound the macromolecules such as DNA, RNA or denatured DNA. The binding constant between the probe and DNA was estimated as 3.13 × 107 mol−1 L. The emission intensity increase was proportional to the concentration of DNA. Based on this, the probe was used to determine the concentration of calf thymus DNA (ct-DNA). The corresponding linear response ranged from 2.50 × 10−7 to 4.75 × 10−6 mol L−1, and detection limit was 1.93 × 10−8 mol L−1 for ct-DNA.  相似文献

A proton-conducting polymer electrolyte based on starch and ammonium nitrate (NH4NO3) has been prepared through solution casting method. Ionic conductivity for the system was conducted over a wide range of frequency between 50 Hz and 1 MHz and at temperatures between 303 K and 373 K. Impedance analysis shows that sample with 25 wt.% NH4NO3 has a smaller bulk resistance (R b) compared to that of the pure sample. The amount of NH4NO3 was found to influence the proton conduction; the highest obtainable room temperature conductivity was 2.83 × 10−5 S cm−1, while at 100 °C, the conductivity in found to be 2.09 × 10−4 S cm−1. The dielectric analysis demonstrates a non-Debye behavior. Transport parameters of the samples were calculated using the Rice and Roth model and thus shows that the increase in conductivity is due to the increase in the number of mobile ions.  相似文献

The saccharide binding and conformational characterization of a hemagglutinin, a low molecular weight protein from the seeds of Moringa oleifera was studied using steady state and time resolved fluorescence. The lectin binds sugars LacNAc (K a = 1380 M−1) and fructose (K a = 975 M−1), as determined by the fluorescence spectroscopy. It has a single tryptophan per monomer which is exposed on the surface and is in a strong electropositive environment as revealed by quenching with iodide. Quenching of the fluorescence by acrylamide involved both static (K s = 0.216 M−1) and collisional (K sv = 8.19 M−1) components. The native protein showed two different lifetimes, τ 1 (1.6 ns) and τ 2 (4.36 ns) which decrease and get converted into a single one, (2.21 ns) after quenching with 0.15 M acrylamide. The bimolecular quenching constant, k q was 7.55 × 1011 M−1 s−1. ANS binding studies showed that the native protein has exposed hydrophobic patches which get further exposed at extreme acidic or alkaline pH. However, they get buried in the interior of the protein in presence of 1 M GdnHCl or urea.  相似文献

A new, simple, sensitive and selective spectrofluorimetric method for the determination of Hydrochlorothiazide was developed in acetonitrile at pH 6.2. The Hydrochlorothiazide can remarkably enhance the luminescence intensity of the Tb3+ ion doped in sol–gel matrix at λex = 370 nm. The intensity of the emission band of Tb3+ ion doped in sol–gel matrix was increased due to the energy transfer from the triplet excited state of Hydrochlorothiazide to (5D4) excited energy state of Tb3 ion. The enhancement of the emission band of Tb3+ ion doped in sol–gel matrix at (5D47 F5) 545 nm was directly proportion to the concentration of Hydrochlorothiazide with a dynamic ranges of 5.0 × 10−10—5.0 × 10−6 mol L−1 and detection limit of 2.2 × 10−11 mol L−1.  相似文献

A complex Fe(phen)2·PHPIP·3ClO4·2H2O, where phen = 1,10-phenanthroline and PHPIP = p-hydroxyphenylimidazo[f]1,10-phenanthroline, was synthesized and acted as a good fluorescence indicator based on its interaction with double-duplex DNA. Then a fiber-optic DNA biosensor of fluorimetric detection was developed based on the recognition of target DNA in DNA hybridization assays. A probe ssDNA was covalently immobilized onto the surface of quartz optical fibers and then the probe ssDNA hybridized with complementary ssDNA introduced into the local environment of the sensor. The hybridization with complementary strands was monitored in real time by fluorimetric detection. Several factors affecting the probe immobilization, target DNA hybridization, and indicator binding reactions were optimized to maximize the sensitivity and shorten the assay time. Using this method, a sequence of the 16-mer oligonucleotides could be quantified over the range from 4.98 × 10−7 to 4.88 × 10−6 M and a detection limit of 1.08 × 10−7 M. And the designed optic-fiber biosensor could be conveniently regenerated by thermal denature. The utility of the novel hybridization indicator could provide a simple, rapid, low toxicity and reusable detection.  相似文献

In the present work, five systems of samples have been prepared by the solution casting technique. These are the plasticized poly(methyl methacrylate) (PMMA-EC) system, the LiCF3SO3 salted-poly(methyl methacrylate) (PMMA-LiCF3SO3) system, the LiBF4 salted-poly(methyl methacrylate) (PMMA-LiBF4) system, the LiCF3SO3 salted-poly(methyl methacrylate) containing a fixed amount of plasticizer ([PMMA-EC]-LiCF3SO3) system, and the LiBF4 salted-poly(methyl methacrylate) containing a fixed amount of plasticizer ([PMMA-EC]-LiBF4) system. The conductivities of the films from each system are characterized by impedance spectroscopy. The room temperature conductivity in the pure PMMA sample and (PMMA-EC) system is 8.57 × 10−13 and 2.71 × 10−11 S cm−1, respectively. The room conductivity for the highest conducting sample in the (PMMA-LiCF3SO3), (PMMA-LiBF4), ([PMMA-EC]-LiCF3SO3), and ([PMMA-EC]-LiBF4) systems is 3.97 × 10−6, 3.66 × 10−7, 3.40 × 10−5, and 4.07 × 10−7 S cm−1, respectively. The increase in conductivity is due to the increase in number of mobile ions, and decrease in conductivity is attributed to ion association. The increase and decrease in the number of ions can be implied from the dielectric constant, ɛr-frequency plots. The conductivity–temperature studies are carried out in the temperature range between 303 and 373 K. The results show that the conductivity is increased when the temperature is increased and obeys Arrhenius rule. The plots of loss tangent against temperature at a fixed frequency have showed a peak at 333 K for the ([PMMA-EC]-LiBF4) system and a peak at 363 K for the ([PMM-EC]-LiCF3SO3) system. This peak could be attributed to β-relaxation, as the measurements were not carried out up to glass transition temperature, T g. It may be inferred that the plasticizer EC has dissociated more LiCF3SO3 than LiBF4 and shifted the loss tangent peak to a higher temperature. 81Br NQR frequencies and differential scanning calorimetry (DSC) were measured as a function of temperature. [NH3(CH2)4 NH3]CdBr4 (1) and [NH3(CH2)5NH3]CdBr4 (2) showed a doublet and quartet 81Br NQR spectrum, respectively. [NH3(CH2)5NH3]ZnBr4 (3) and [NH3(CH2)6NH3]ZnBr4 (4) exhibited a four-line 81Br NQR spectrum. From the NQR results, it is inferred that (1) and (2) consist of infinite two-dimensional sheets of corner-sharing CdBr6 octahedra, whereas (3) and (4) have isolated [ZnBr4]2− tetrahedra. All of the crystals except (1) showed at least one structural phase transition above 380 K.  相似文献
