Diazo carbonyl derivatives of heterocycles. 5. Intramolecular cyclization of 2-hetarylmercaptoalkyl-diazo ketones. Synthesis of condensed 1,3-thiazin-5-ones

The intramolecular heterocyclization of 2-benzazolylmercaptoalkyl diazomethyl ketones under the influence of acidic agents leads to 1,3-thiazino[2,3-b]benzim-idazole, 1,3-thiazino[2,3-b]benzothiazolium, and 1,3-thiazino[2,3-b]benzoxazolium systems. Ionic Liquid-Mediated Facile Synthesis and Antimicrobial Study of Thiazolo [2,3-b]Benzo[h]Quinazolines and Thiazino[2,3-b] Benzo-[h]Quinazolines

Abstract

8-Methoxy-4-phenyl-3,4,5,6-tetrahydrobenzo[h]quinazoline-2(1H)-thione, obtained by the condensation of 2-benzylidene-6-methoxy-3,4-dihydronapthalene-1(2H)-one with thiourea, on reaction with chloroacetic acid and 3-chloropropanoic acid in the presence of the ionic liquid N-methylpyridinium tosylate furnishes 3-methoxy-7-phenyl-7,10-dihydro-5H- benzo[h]thiazolo[2,3-b]quinazoline-9(6H)-one and 3-methoxy-7-phenyl-5,6,10,11-tetrahydro- benzo[h][1,3]thiazino[2,3-b]quinazoline-9(7H)-one. Further, condensation of the thione with 1,2-dibromoethane and 1,3-dibromopropane yields 3-methoxy-7-phenyl-6,7,9,10-tetrahydro-5 H-benzo[h]thiazolo[2,3-b]quinazoline and 3-methoxy-7-phenyl-5,6,7,9,10,11-hexahydrobenzo [h][1,3]thiazino[2,3-b]quinazoline respectively. Arylidene derivatives have been obtained by two routes. The structures of the cyclized compounds have been established on the basis of elemental analysis and spectroscopic data. The synthesized compounds were screened for antimicrobial activity. Some of the compounds showed promising antimicrobial activities.     

Semiempirical calculation on photochromic process of spirooxazines

Structure effect on photochromic mechanism of 1,3-dimethyl-3,2′-(1,3-propyl-ene)spiro-[indoline-2,3′-[3H]naphtho[2,1-b][1,4]oxazine] (SP1) and 1,2′-(1,3-propylene)-3,3-dimethylspiro[indoline-2,3′[3H]naphtho[2,1-b]-[1,4]oxazine] (SP2) has been studied by AM1 calculation. The colored forms of SP1 and SP2 were fully optimized by AM1 method, and the results support the keto structures. The calculation results on stability of colored forms are agreeable with published experimental results.     

1H and 13C spectral assignment of substituted 5H-[1,3]thiazolo[2,3-b]quinazolin-5-one and 12H-[1,3]benzothiazolo[2,3-b] quinazolin-12-one

(1)H and (13)C spectroscopic data for 5H-[1,3]thiazolo[2,3-b]quinazolin-5-one and 12H-[1,3]benzothiazolo[2,3-b]quinazolin-12-one derivatives were fully assigned by combination of one- and two-dimensional experiments (DEPT, HMBC and HMQC). Both heterocyclic systems show similar spectroscopic properties with some remarkable differences.     

Synthesis of certain pyrrolo[2,3-b]pyridine-5-carboxylic acid derivatives as potential antimicrobial agents

Summary A synthetic approach to new 1-benzyl-7-alkyl-2,3-dimethyl-4-oxopyrrolo[2,3-b]pyridine-5-carboxylic acids using 5,6-dimethyl-2,4-dioxopyrrolo[2,3-d][1,3]oxazine as the starting material is reported. The antimicrobial activity of these compound is reported.

Synthese von Pyrrolo[2,3-b]pyridin-5-carbonsäure-Derivaten als potentielle antimikrobielle Substanzen

Zusammenfassung Die Synthese von neuen 1-Benzyl-7-alkyl-2,3-dimethyl-4-oxopyrrolo[2,3-b]pyridin-5-carbonsäuren durch Verwendung von 5,6-Dimethyl-2,4-dioxopyrrolo[2,3-d][1,3]oxazinen als Ausgangsmaterial wird beschrieben. Die antimikrobielle Aktivität dieser Substanzen wurde geprüft.

     

Halocyclization of 3-allylthio-5H-[1,2,4]triazino[5,6-<Emphasis Type="Italic">b</Emphasis>]indole

The synthesis of 3-allylthio-5H-[1,2,4]triazino[5,6-b]indole has been carried out by the reaction of 3-mercapto-5H-[1,2,4]triazino[5,6-b]indole with allyl bromide in the NaOH–H₂O–DMSO system and in a one-pot synthesis from isatin-β-thiosemicarbazide. Halocyclization of allylthio-5H-[1,2,4]-triazino[5,6-b]indole synthesized the 3-halomethyl-3,10-dihydro-2H-[1,3]thiazolo[3',2':2,3][1,2,4]-triazino[5,6-b]indolium halides.     

Synthesis of disubstituted imidazo[4,5-b]pyridin-2-ones

Regioselective palladium-catalyzed amination of 2-chloro-3-iodopyridine followed by a subsequent palladium-catalyzed amination leads to 2,3-diaminopyridines. Treatment with triphosgene affords highly functionalized unsymmetrical imidazo[4,5-b]pyridin-2-ones in just three synthetic steps. A two-step synthesis of pseudosymmetrically disubstituted imidazo[4,5-b]pyridin-2-ones, 1,4-disubstituted pyrido[2,3-b]pyrazinediones, and 1,3-disubstituted thiadiazolo[3,4-b]pyridin-2-ones is also described.     

A NOVEL SYNTHESIS OF 2-SUBSTITUTED-4H-THIENO [2,3-d][1,3] OXAZIN-4-ONE AND 2,3-DISUBSTITUTED THIENO [2,3-d]PYRIMIDIN-4(3H)-ONE DERIVATIVES

Abstract

The synthesis of acetic 2-{[1-methyl-2-(4-oxo-5,6,7,8-tetrahydro-4H-benzo [4,5]-thieno [2,3-d] [1,3-oxazin-2-yl)ethylidene]amino}-4,5,6,7-tetrahydrohenzo[b]thiophene ?3-carboxylic acid anhydride 5 and 2-(oxopropyl)-5,6,7,8-tetrahydro-4H-benzo-[4,5] thieno[2,3-d][1,3]oxazin-4-one 7, has been achieved in three steps from ethyl 2-amino-4,5,6,7-tetrahydrobenzo(b]thiophene-3-carboxlate 1 via the reaction with ethyl acetoacetate followed by hydrolysis and acetic anhydride-induced cyclization. The 2-substituent in compound 5 has two functional groups i.e. active methylene and acid anhydride which are suitably located for intramolecular transformation. Thermal and/or base catalyzed intramolecular cyclization of 5 afforded 2-(4-acetoxy-(hydroxyl)-2-methyl-5,6,7,8-tetrahydrobenzo[4,5] thieno[2,3-b]pyridin-3-yl)-5,6,7,8- tetrahydro-4H-benzo[4,5]thieno[2,3-d] [1,3]oxazin-4-one 10 and 9 respectively. Treatment of 5 with hydrazine hydrate, aromatic and/or heterocyclic amines induced the same intramolecular cyclization with a concomitant oxazine-pyrimidine interconversion to give 3-amino(aryl or heteryl)-2-(4-hydroxy-5,6,7,8-tetrdhydrobenzo-[4,5]thieno[2,3-b]pyridin-3-yl)-3,4,5,6,7,8-hexahydrobenzo[4,5] thieno[2,3-d] pyrimid in-4-one 11–14 respectively.     

NOVEL SYNTHESIS OF CONDENSED HETEROCYCLIC SYSTEMS CONTAINING 1,2,4-TRIAZOLE RING

3-Aryl-6,7-dihydro-s-triazolo[3,4-b][1,3]thiazines and 3-aryl-5,6-dihydro-thiazolo[2,3-c]-s-triazoles were synthesized by nucleophilic substitution of 3-aryl-5-mercapto-1,2,4-triazoles with 1,3-dibromopropane and 1,2-dichloroethane. And the bis-Mannich reaction of 3-aryl-5-mercapto-1,2,4-triazoles with formaldehyde has been studied.     

The Synthesis of Some New Fused and Substituted Chromenes

Some new chromeno[2,3-b]pyrimidines, chromeno[3,2-c]pyridines, chromeno[2,3-b]pyridines and 3-chromenyl-1,3-thiazines were synthesized via the synthetic studies of the reaction of 2-imino-2H-chromen-3-thiocarboxamide with some aromatic aldehydes, active nitriles, and their ylidene derivatives.     

Condensed pyridine bases. A new approach to the synthesis of 3-phenacylbenzo[b]thiophenes and 3-phenacyl-5-ethylthieno[2,3-b]thiophene. Synthesis of new derivatives of benzothieno[2,3-c]pyrylium and thieno[2′,3′:5,4]thieno-[2,3-c]pyrylium and pyridine bases

A new method is proposed for the production of substituted 3-phenacylbenzo[b]thiophenes by the intramolecular cyclization of 1-phenyl-4-phenylthio-1,3-butanediones in polyphosphoric acid. The analogous cyclization of 1-phenyl-4-(5-ethyl-2-thienylthio)-1,3-butanedione leads to 3-phenacyl-5-ethylthieno[2,3-b]thiophene. The acylation of substituted 3-phenacylbenzo[b]thiophenes and 3-phenacyl-5-ethylthieno[2,3-b]thiophene by the anhydrides of aliphatic acids in the presence of perchloric acid leads to 1-alkyl-3-phenylbenzothieno[2,3-c]pyrylium perchlorates and 2-ethyl-5-phenyl-7-methylthieno[2′,3′:5,4]thieno[2,3-c]pyrylium perchlorate. The action of ammonium acetate on the obtained salts gives the corresponding pyridine bases. L. M. Polymethine dyes derived from heterocyclic bases containing condensed thiophene rings

The synthesis of polymethine dyes of the cyanine and merocyanine series derived from 3-methylthionaphtheno[2,3-b]pyridine, 3-methylthionaphtheno[3,2-b]-pyridine, 6-methylthieno[2,3-b]thieno[2,3-b]-pyridine, and 6-methylthieno[3,2-b]thieno[2,3-b]pyridine is described. The color of the dyes obtained is discussed. It has been shown that the replacement of vinylene groups by sulfur atoms in one of the two benzene rings of the hetero residue in 2-substituted 5,6-benzoquinolines in cyanine dyes leads to a bathochromic shift of the absorption maxima.For part VI, see [8].     

Synthesis of naphtho[2',3':4,5]imidazo[2,1-<Emphasis Type="Italic">b</Emphasis>][1,3]-thiazole-5,10-dione and naphtho[2',3':4,5]imidazo-[2,1-<Emphasis Type="Italic">b</Emphasis>][1,3]benzothiazole-7,12-dione

The reaction of 2,3-dibromo-1,4-naphthoquinone with 2-aminothiazole in MeONa/MeOH at 60oC for 3 h gave naphtho[2',3':4,5]imidazo[2,1-b][1,3]thiazole-5,10-dione in 64% yield. The reaction of 2,3-dibromo-1,4-naphthoquinone with 2-aminobenzothiazole under the above-mentioned conditions gave 2-(benzo[d]thiazol-2-ylamino)-3-bromonaphthalene-1,4-dione in 64% yield, which on treatment with Na/THF or NaN₃/acetone under reflux conditions gave naphtho[2',3':4,5]imidazo[2,1-b][1,3]- benzothiazole-7,12-dione in 69 and 56% yields, respectively.     

Synthesis of 3,4-alkoxythieno[2,3-b]thiophene derivatives. The first block copolymer associating the 3,4-ethylenedioxythieno[2,3-b]thiophene (EDOThT) unit with 3,4-ethylenedioxythiophene (EDOT) moieties

The synthesis of 3,4-alkoxythieno[2,3-b]thiophene derivatives has been developed from the readily accessible dimethyl or diethyl 3,4-dihydroxythieno[2,3-b]thiophenedicarboxylates. The yields for the dialkoxy derivatives were strongly dependent both on the base and the alkylating agent used for the nucleophilic substitutions. A trimer associating the new 3,4-ethylenedioxy-thieno[2,3-b]thiophene unit with 3,4-ethylenedioxythiophene moieties has been synthesized and electropolymerized.     

Reaction of 1H-azoles with 2-hydroxybenzyl alcohols

Condensation of 1H-azoles with 2-hydroxybenzyl alcohols gave a series 2-(1H-azol-1-ylmethyl)phenols. 12H-Benzimidazo[2,1-b][1,3]benzoxazine was synthesized from 2-methylmercaptobenzimidazole and 2-hydroxybenzyl alcohol. Conversions of polyfunctional 3-amino-1(2H)iso quinolines

The acylation of 3-amino-4-cyano-1(2H)isoquinolines with benzoyl chloride leading to the formation of 1,3 oxazino[4,5-clisoquinoline-6-ones has been studied. Previously undescribed 1-aminopyrimido[4,5-cJisoquinolirt 6-ones have been obtained by the reaction of the appopriate 3-amino-](2H)isoginolones with formamide. Nucleophilic replacement has been carried out with 3-amino-1-chloroisoquinoline by the action of sodium hydroxide, primary alcohols, hydrazine hydrate, and various amines. 1,2,4-Triazino[2,3-b]isoginolone has been synthesized by condensing 2,3-diamino-1 (2H)isoquinolone with mesoxalic acid ethyl ester.     

An efficient synthesis of novel pyridothieno-fused thiazolo[3,2-a]pyrimidinones via Pictet–Spengler reaction

An efficient method for the synthesis of novel pyrido[3',2':4',5']thieno[3',2':2,3]pyrido [4,5:d][1,3]thiazolo[3,2-a]pyrimidine-4-one derivatives (5) has been developed using a Pictet-Spengler reaction between 2-(3-aminothieno[2,3-b]pyridin-2-yl)thiazolo[3,2-a] pyrimidin-5-one (3), which could be obtained from the condensation of 7-(chloromethyl)-5H-thiazolo[3,2-a]pyrimidin-5-one (1) with 3-cyanopyridine-2-thione (2) via Thorpe-Ziegler isomerization, and aromatic aldehydes under NH₂SO₃H as catalysis in good yields.     

Efficient synthesis of fused perhydrofuro[2,3-b]pyrans (and furans) by ring opening of 1,2-cyclopropanated sugar derivatives

[structure: see text]. An efficient method has been developed for the construction of fused perhydrofuro[2,3-b]pyrans by diastereoselective ring opening of 1,2-cyclopropanated sugar derivatives. The methodology has been successfully applied to the synthesis of fused perhydrofuro[2,3-b]pyrano-gamma-butyrolactone derivatives.     

Interaction of benzothieno[2,3-c]pyrylium salts with hydrazine. Derivatives of 5H-[2,3]benzothieno[2,3-e]diazepines

A study has been made of the reaction of 1,3-disubstituted bezothieno[2,3-c]pyrylium salts with hydrazine. It has been shown that 1,3-dialkyl-substituted benzothieno[2,3-c]pyrylium salts interact with hydrazine to give N-amino-1,3-dialkylbenzothieno[2,3-c]pyridines. The presence of a pyrylium ring on one of the positions of the phenyl group leads to a mixture of N-amino derivatives and 5H-[2,3]benzothieno[2,3-e]diazepines. In contrast, 1,3-diphenylbenzothieno[2,3-c]pyrylium perchlorate gives exclusively 5H-[2,3]benzothieno[2,3-e]diazepine.L. M. Fluoro-containing Heterocycles. IV. Synthesis of Benzimidazole Derivatives

2-Mercapto-5,6-difluorobenzimidazole reacts with aliphatic and alicyclic ketones in acetic acid in the presence of catalytic amount of sulfuric acid to afford fluorinated derivatives of 2,3-disubstituted benz[4,5]imidazo[2,1-b][1,3]thiazoles. Reaction with aromatic -haloketones occurs in another way: to furnish 2-phenylacylthio-5,6-difluorobenzimidazoles that in the system acetic anhydride-pyridine undergo cyclization into the corresponding fluorinated derivatives of benz[4,5]imidazo[2,1-b][1,3]thiazoles.
