From two-day etiolated shoots of the cotton plant of variety 108-F we have isolated the total histone and purified it on a column of CM-cellulose. It has been shown by gel electrophoresis and amino acid analysis that the total histone of the cotton plant, like the total histone of wheat, consists of five main fractions, the amount of the lysine-rich fraction A1 being greater in the cotton plant than in wheat (23% and 19%, respectively), and the amount of arginine-rich fraction A4 being less (19.2 and 24.3%, respectively). The total histone of the cotton plant is less basic (ratio of basic amino acids to acidic amino acids in the cotton-plant histone 1.24, in wheat histone 1.9, and in calf thymus histone 1.78) but richer in lysine as compared with arginine than the histones of wheat and calf thymus.V. The results are given of a study of the quantitative content of protein in the seeds of some pea varieties and mutants, the activity of the total inhibitor proteins, and correlations of their activity with the protein content of the seeds and the amino acid compositions of the proteinase inhibitors. In order to isolate individual fractions of the histones of the cotton plant of variety 108-F and to investigate them further, the total histone has been chromatographed on Acrylex P-60 and Bio-Gel P-30. It has been shown that the most complete fractionation is achieved on a Acrylex P-60. By electrophoresis of the fractions in 15% PAAG in the presence of 6.25 M urea and on the basis of amino acid compositions, the following order of elution of the histones has been established: H1, H2B + H2A, H3, and H4. In a comparison of the amino acid composition of the H3 and H4 histones of the cotton plant with histones enriched with arginine from other species, a somewhat higher lysine : arginine ratio in these fractions has been found. On comparing the electrophoretic mobilities of the histone fractions from the cotton plant with the histones of calf thymus, some difference is observed which is apparently connected with a difference in their molecular weights.V. A characterization of the histones of two varieties of the cotton plant,Gossypium hirsutum andG. barbadense, has been given with the aid of various electrophoretic systems. Their molecular weights have been determined. An analysis has been made of the cleavage of histone H1 at tyrosine residues. The 11S globulins of two varieties of the cotton plant — 108-F and Tashkent-1 — have been studied. It has been shown that the 11S globulins do not differ in quaternary structure and each consists of three subunits A, B, and C. Phospholipids of the seeds of kenaf of the variety Opytnyi-1931 are similar with respect to their set of components and the qualitative composition of the fatty acids of homogeneous classes to the phospholipids of the variety Opytnyi-1972, but differ with respect to the amounts of individual acids. The interactions with DNA of a synthetic peptide fragment of histone H1 33 amino acid residues long the sequence of which corresponds to fragment 152–184 and of a synthetic polypeptide (Lys-Ala-Ala)n modeling the properties of the C-terminal fragments of histone H1 have been studied by the CD method. The use of the CD method for recording the spectra of the polypeptides under investigation in the free state and of the artificial complexes of DNA formed with them has permitted the characterization of different structural possibilities of the polypeptides in solution and of their complexes with DNA, and the conformational changes of the components of the complex under various ionic conditions.V. The yields and fatty acid compositions of the free lipids and lipids weakly and strongly bound to proteins from seeds of cottonplant varieties resistant and susceptible to verticillium wilt have been determined and the changes in these indices in the seeds of the infected plants have been measured. It has been established that the seeds of healthy plants of the two varieties are similar in the qualitative compositions of the lipids and fatty acids but differ with respect the amounts of lipids in them bound with protein in different ways and with respect to the amounts of the individual fatty acids. The nature of the relative affinity of histones for DNA has been studied with the aid of the electrophoretic analysis of the DNP complexes obtained by the addition of the total histone to a nuclear histone with a DNA:histone ratio of 1:1. It has been established that, on the addition of histone, complexes are formed the composition of which changes according to the degree of competition of the histone fractions. Some differences have been detected in the nature of the formation of nuclear histones of plant and animal origin due to features of the primary structure of the histones. The results obtained confirm a hypothesis put forward previously about the different strengths of the bonds of histones with DNA and the role of dissociation in the functioning of the genetic apparatus of the cells of higher organisms.V. Conclusions A protamine, which has been named stellin, has been isolated from the mass of sperm of the sevruga(Acipenser stellatus).Stellin is heterogeneous and consists of a mixture of peptides differing in amino acid composition and in chain length. Information is given on the total amino acid composition and the free amino acids of pears (Pyrus communis L), squashes (Cucurbita pepo L), grapefruits (Citrus paradisi L) and apples (Malus sylvestris Mill.). It has been established that during the dispersion of the fruits in the manufacture of sedimentation-stabilized products that the total qualitative and quantitative amino acid compositions scarcely change but the concentration of free amino acids rises by 14.5–45.1% on the total amount of free amino acids in the initial material. The nature of the interaction of high- and low-molecular-weight DNAs (6×106 and 3×105 daltons) with synthetic oligopeptides of the N-end of histone H2B having the sequences 1–21, 1–10, 1–13, 11–21, and 14–21, differing in molecular weight and amino acid composition, as a function of the amount of peptide component in the complex and the ionic strength of the solution has been studied by the methods of UV and CD spectroscopy and the spectrophotometric analysis of melting curves. It has been shown that of all the peptides studied only the 1–21 peptide possesses the capacity of condensing DNA. This capacity depends on the amount of peptide component in the complex, the molecular weight of the DNA, and the ionic strength of the solution. The interaction with peptides under all the conditions studied, without changing the conformational parameters of the DNA, stabilizes its secondary structure in relation to the action of the temperature, which depends on the number of lysine residues in the peptides.N. Oxidation of -amino acids by pyridinium bromochromate (PBC) was studied in acetic acid–water mixture containing perchloric acid. The reaction rate is first order in [PBC] and inverse first order in [H+] and has aldehyde as a product. The results are contrary to that of Karim and Mahanti, who observed first order with [H+] and cyanide as the product in the oxidation of amino acids by quinolinium dichromate. Michaelis–Menten type kinetics has been observed with respect to -amino acids. The rate of reaction increases with a decrease in the polarity of solvent indicating an ion–dipole interaction in the slow step. The reactions exhibit no primary kinetic isotope effect. The activation parameters have been evaluated. The reaction mechanism involving the formation of chromate-ester between unprotonated PBC and unprotonated amino acid followed by C–C bond fission in the slow step has been suggested. The value of the Michaelis constant (substrate–oxidant complex formation constant) increases as the number of carbon atoms increases in the amino acid.  相似文献
