The migration of melamine monomers from food contact materials has aroused particular attention since the 2008 melamine-tainted milk scandal in China. However, the determination of melamine monomer’s migratory quantity (MMMQ) has remained an open question because of the complex sample pretreatment and the low sensitivity. Based on the hydrogen bonding interaction between DNA thymine and melamine, this paper described a simple and rapid method focusing on the measurement of MMMQ from melamine tableware by gold nanoparticles (GNPs) and dynamic light scattering (DLS). With the presence of probe DNA (p-DNA), the GNPs were stable in NaCl solution (0.06 M), whereas they became aggregated when the p-DNA hybridized with melamine. The change in the hydrodynamic diameter of GNPs could be detected by DLS technology. Under the optimal conditions, the average diameter increased linearly with the concentration of melamine over the range from 5.0 to 320.0 μg L−1, and showed a detection limit of 2.0 μg  L−1 (3σ/slope). The MMMQ was investigated within a range from 6.00 × 10−4 to 2.58 × 10−1 mg dm−2 (n ≥ 3) in four different food simulants at different temperatures and time points. The results suggest that the DLS method has great potential in the analysis of the migration of melamine monomers.  相似文献

A simple, rapid, field-portable colorimetric method for the detection of melamine based on melamine-induced color change of label-free gold nanoparticles (Au NPs) was developed in this study. Melamine can induced the aggregation of Au NPs and results in the color change from wine-red to purple, which provided a platform for rapid and field-portable colorimetric detection of melamine. The proposed method can be used to detect melamine in liquid milk and infant formula with a detection limit of 1.0 and 4.2 ppm, respectively, within 30 min by naked eyes observation without the aid of any advanced instrument and the need of any complex pretreatment, and detect as low as 0.15 ppm of melamine in liquid milk and 2.5 ppm of melamine in infant formula with UV-vis-spectroscopy. The proposed method is promising for on-site screening of melamine adulterant in milk products.  相似文献

Owing to its high affinity with phosphate, Zr(IV) can induce the aggregation of adenosine 5′-triphosphate (ATP)-stabilized AuNPs, leading to the change of surface plasmon resonance (SPR) absorption spectra and color of ATP-stabilized AuNP solutions. Based on these phenomena, visual and SPR sensors for Zr(IV) have been developed for the first time. The A660 nm/A518 nm values of ATP-stabilized AuNPs in SPR absorption spectra increase linearly with the concentrations of Zr(IV) from 0.5 μM to 100 μM (r = 0.9971) with a detection limit of 95 nM. A visual Zr(IV) detection is achieved with a detection limit of 30 μM. The sensor shows excellent selectivity against other metal ions, such as Cu2+, Fe3+, Cd2+, and Pb2+. The recoveries for the detection of 5 μM, 10 μM, 25 μM and 75 μM Zr(IV) in lake water samples are 96.0%, 97.0%, 95.6% and 102.4%, respectively. The recoveries of the proposed SPR method are comparable with those of ICP-OES method.  相似文献

A novel, simple, sensitive and precise spectrofluorimetric method was developed for measuring the melamine concentration in different milk batch samples. The method was based upon measuring the quenching of the luminescence intensity of the produced yellow colored ruthenium(II) carbonyl complex of the general formula [Ru(CO)2(L)] (where L = anion of tetradentate Schiff base). The Ru(II) complex exhibited characteristic luminescence band in the visible region. The remarkable quenching of the luminescence intensity of [Ru(CO)2(L)] complex by various concentrations of melamine was successfully used as a chemosensor for the assessment of melamine in different milk samples at λex = 400 nm and pH 7.4 in DMSO with a linear dynamic range 1.0 × 10−6 to 3.0 × 10−9 mol L−1 and lower detection limit (LOD) and quantification detection limit (QOD) of 3.3 × 10−10 and 1.0 × 10−9 mol L−1, respectively.  相似文献

Three fluorescent probes were constructed by incorporating an α,β-unsaturated ketone to a coumarin fluorophore. The selective addition of sulfite to the alkene of TSP assisted by cetyltrimethyl ammonium bromide (CTAB) micelle can be visualized by dramatic color and ratiometric fluorescence changes. In CTAB–PBS system, the fluorescence intensity ratio at 465 nm and 592 nm (I465/I592) and the absorbance ratio at 390 nm and 470 nm (A390/A470) were linearly proportional to sulfite concentration in the range of 0.5–150 μM, and the detection limit was 0.2 μM. Good selectivity and competition of TSP1 towards sulfite over several anions and biological thiols were acquired. Probe TSP1 was used to detect sulfite in three realistic samples (mineral water, sugar and white wine) with good recovery.  相似文献

Melamine is a toxic triazine, illegally used as an additive in milk to apparently increase the amount of protein. A chromatographic procedure using a C18 column and a micellar mobile phase of sodium dodecyl sulphate (0.05 M) and propanol (7.5%), buffered at pH 3, and a detection set by absorbance at 210 nm, was reported for the resolution and quantification of melamine in liquid and powdered milk samples. In this work, samples were diluted with a SDS solution and were directly injected, thus avoiding long extraction and experimental procedures. Melamine was eluted in nearly 9.3 min without overlapping the protein band or other endogeneous compounds. The optimal mobile phase composition was taken using a chemometrical approach that considers the retention factor, efficiency and peak shape. Validation was performed following the European Commission's indications (European Decision 2002/657/EC), and the main analytical parameters studied were: linearity (0.02-100 ppm; r2 = 0.999), limit of detection (5 ppb), intra- and inter-day precision (R.S.D. <7.6% and <9.7%, respectively) and robustness (R.S.D. <7.4% for retention time and <5.0% for area). Sensitivity was adequate to detect melamine under the safety limits proposed by the US FDA. Finally, recoveries for several milk samples were found in the 85-109% range.  相似文献

A novel, quantitative analytical method for measuring C-reactive protein (CRP) levels in human serum has been developed based on the catalytic activity of gold nanoparticles (GNPs) and luminol-H2O2 chemiluminescence (CL). The CL intensity in the presence of CRP and its ligand, O-phosphorylethanolamine (PEA), was greatly enhanced due to the aggregation of GNPs after the addition of 0.5 M NaCl. Any pretreatment steps, such as covalent functionalization of GNPs, addition of antibodies, or labeling of CRP, were not needed for CL detection. The CL enhancement was linearly proportional to CRP concentration in the range of 1.88 fM to 1.925 pM. The detection limit of CRP in serum samples was estimated to be as low as 1.88 fM. The detection sensitivity was increased more than 164 times of magnitude over that of the conventional, enzyme-linked immunosorbent assay (ELISA) method. This proposed GNP-based CL detection method offers the advantages of simplicity, rapidity, and sensitivity.  相似文献

In this paper, a highly selective sample cleanup procedure combining molecular imprinting and solid-phase extraction (MI-SPE) was developed for the isolation of melamine in dairy products. The molecularly imprinted polymer (MIP) was prepared using melamine as the template molecule, methacrylic acid as the functional monomer and ethylene glycol dimethacrylate as the cross-linking monomer. The melamine imprinted polymer was used as selective sorbent for the solid-phase extraction of melamine from dairy products. An off-line MI-SPE method followed by high-performance liquid chromatography with diode-array detection for the detection of melamine was also established. The mean recoveries of melamine from ultra-heat treatment (UHT) milk and milk powders were 92.9-98.0% and 91.6-102.8%, respectively. Good linearity was obtained from 0.5 μM to 10 μM (r > 0.999) with a quantitation limit of 0.5 μmol/L (0.06 ppm) which was sufficient to analyse melamine at the maximum level permitted by U.S. Food and Drug Administration (1 ppm) in dairy products. It was demonstrated that the proposed MI-SPE-HPLC method could be applied to direct determination of melamine in dairy products.  相似文献

A new method for sensitive and fast screening of melamine (MEL) in milk products was developed with a low-cost disposable microfluidic device coupled with ultraviolet (UV) detection. This method avoided the need of sample pretreatment prior to the separation process, thus was simple and green. Due to the advantages of the device and fracture sampling technique, milk sample could be directly sampled through the fracture to achieve baseline separation from amino acids, and proteins in the sample did not interfere with the detection. Using 20 mM phosphate running buffer (pH 9.0), a sampling time of 3 s at +180 V and a separation voltage of +1800 V (240 V/cm), this method could detect MEL in milk within 75 s. At the detection wavelength of 202 nm, the linear range for MEL was from 1.0 to 100 μg mL−1 with a detection limit of 0.23 μg mL−1 (S/N = 3). The novel protocol had been successfully used to screen MEL in milk samples with recovery more than 82%. The environmentally friendly methodology for pretreatment-free sensitive screening of MEL provided promising applications in monitoring the quality of different foods.  相似文献

A convenient colorimetric approach for visual detection of melamine in raw milk was realized by using gold nanoparticles (AuNPs) stabilized by an unsymmetrical terpyridyl zinc complex with a thymine fragment at one terminal and a quaternary ammonium salt at the other. Even without pre-addition of melamine or relative additives, obvious color change from red to blue was observed by naked eye in the presence of trace amount of melamine, which was attributed to the alternation of aggregation state of AuNPs caused by the selective binding between the thymine fragment and melamine via triple hydrogen-bonding interactions. Remarkably, the detection limit for melamine was as low as 2.4 ppb, providing a highly sensitive and efficient approach for the visual detection of melamine.  相似文献

A graphene, chitosan and Fe3O4 nanoparticles (nano-Fe3O4) modified glassy carbon electrode (graphene-chitosan/nano-Fe3O4/GCE) was fabricated. The modified electrode was characterized by scanning electron microscope and electrochemical impedance spectroscopy. The electrochemical oxidation behavior of guanosine was investigated in pH 7.0 phosphate buffer solution by cyclic voltammetry and differential pulse voltammetry. The experimental results indicated that the modified electrode exhibited an electrocatalytic and adsorptive activities towards the oxidation of guanosine. The transfer electron number (n), transfer proton number (m) and electrochemically effective surface area (A) were calculated. Under the optimized conditions, the oxidation peak current was proportional to guanosine concentration in the range of 2.0 × 10−6 to 3.5 × 10−4 mol L−1 with the correlation coefficient of 0.9939 and the detection limit of 7.5 × 10−7 mol L−1 (S/N = 3). Moreover, the modified electrode showed good ability to discriminate the electrochemical oxidation response of guanosine, guanine and adenosine. The proposed method was further applied to determine guanosine in spiked urine samples and traditional Chinese medicines with satisfactory results.  相似文献

News stories about the contamination of milk with melamine in China emerged on Sept 11, 2008, and the situation has since become an international health scare. In this work, a novel analytical method based on enrichment and pretreatment of analytes in the milk sample, hollow fiber sorptive extraction and gas chromatography–mass spectrometry has been developed for the rapid analysis of melamine in the dairy products. In the proposed method, melamine in the fresh milk was extracted by zirconia hollow fiber, enriching on zirconia coating of the hollow fiber, and then analyzed by GC–MS. The method validations including linearity, limit of detection, limit of qualification, recoveries at three different concentrations, precision, and repeatability were investigated. It was found that the proposed method provided linear range from 0.001 to 1000 μg/mL (r2 = 0.9997), low detection limit of 0.001 μg/mL, and preferable recoveries at three different concentrations. The obtained results demonstrated that zirconia hollow fiber combined with GC–MS is a simple, rapid and solvent-free method for the analysis of melamine in the dairy products.  相似文献

A sensitive and reliable method was developed and validated for detection and confirmation of melamine in egg based on gas chromatography-mass spectrometry (GC-MS) and ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). Trichloroacetic acid solution was used for sample extraction and precipitation of proteins. The aqueous extracts were subjected to solid-phase extraction by mixed-mode reversed-phase/strong cation-exchange cartridges. Using ultra-performance liquid chromatography and electrospray ionization in the positive ion mode, melamine was determined by LC-MS/MS, which was completed in 5 min for each injection. For the GC-MS analysis, extracted melamine was derivatized with N,O-bis(trimethylsilyl)trifluoracetamide prior to selected ion monitoring detection in electron impact mode. The average recovery of melamine from fortified samples ranged from 85.2% to 103.2%, with coefficients of variation lower than 12%. The limit of detection obtained by GC-MS and UPLC-MS/MS was 10 and 5 μg kg−1, respectively. This validated method was successfully applied to the determination of melamine in real samples from market.  相似文献

An electrochemiluminescence (ECL) enhancement method combined with solid-phase extraction has been developed for the determination of melamine in dairy products. It was found that melamine in a strong base solution is able to enhance the ECL of Ru(bpy)32+ at glass carbon electrode. The optimum experimental conditions for the determination of trace melamine by ECL, such as scan mode and scan rate of the applied potential, the type of buffer solutions and their pH conditions, were investigated. Under optimized conditions, the enhanced ECL intensity was linearly proportional to the logarithm of melamine concentration in the range of 0.01-1.0 ppb, and the detection limit was 0.003 ppb. The method has been successfully demonstrated to determine melamine in dairy products including liquid milk, yogurt and milk powder samples. The relative standard deviations ranging from 5.3% to 11.2% and the recoveries from 95.2% to 102.4% were acquired by this method. A possible mechanism for the ECL enhancement effect was also proposed.  相似文献

A high performance liquid chromatographic method with fluorimetric detection for the determination of aflatoxin M1 (AFM1) in milk has been optimized and validated according to Commission Decision 2002/657/EC by using the conventional validation approach. The procedure for determining selectivity, recovery, precision, decision limit (CCα), detection capability (CCβ) and ruggedness of the method has been reported. The results of the validation process demonstrate the agreement of the method with the provisions of Commission Regulation 401/2006/EC. The mean recovery calculated at three levels of fortification (0.5, 1.0, and 1.5-fold the MRL) was 91% and the maximum relative standard deviation value for the within-laboratory reproducibility was 15%. Limit of detection (LOD) and limit of quantitation (LOQ) values were 0.006 μg kg−1 and 0.015 μg kg−1 while the CCα and CCβ values were 0.058 μg kg−1 and 0.065 μg kg−1, respectively. The relative expanded measurement uncertainty of the method was 7%. The method was not affected by slight variations of some critical factors (ruggedness minor changes) as pre-treatment and clean-up of milk samples, thermal treatment and different storage conditions, as well as by major changes valued in terms of milk produced by different species (buffalo, goat and sheep). The method allowed accurate confirmation analyses of milk samples, resulted positive by the screening method. In fact, the Z-score values attained in a proficiency test round were well below the reference value of 1, proving the excellent laboratory performances.  相似文献

An ultrasensitive and simple method for the determination of melamine was developed based on the fluorescence quenching of thioglycolic acid (TGA) capped CdTe quantum dots (QDs) at pH 11.0. In strong alkaline aqueous solution, the selectivity of the method has been greatly improved due to most heavy metal ions show no interference as they are in the precipitation form or in their anion form. Furthermore, CdTe quantum dots have higher quantum yields at higher pH. The method has a wider concentration range and lower detection limit. The influence factors on the determination of melamine were investigated and the optimum conditions were determined. Under optimum conditions, the fluorescence intensity change of TGA coated CdTe quantum dots was linearly proportional to melamine over a concentration range from 1.0 × 10−11 to 1.0 × 10−5 mol L−1 with a correlation coefficient of 0.9943 and a detection limit of 5 × 10−12 mol L−1. The mechanism of fluorescence quenching of the QDs has been proposed based on the infrared spectroscopy information and electrophoresis experiments in presence of melamine under alkaline condition. The proposed method was employed to detect trace melamine in milk powder and pet feeds with satisfactory results.  相似文献

A graphene (GR) and multi-walled carbon nanotubes (MWCNT) hybrid was prepared and modified on a 1-hexylpyridinium hexafluorophosphate based carbon ionic liquid electrode (CILE). Hemoglobin (Hb) was immobilized on GR-MWCNT/CILE surface with Nafion as the film forming material and the modified electrode was denoted as Nafion/Hb-GR-MWCNT/CILE. Spectroscopic results revealed that Hb molecules retained its native structure in the GR-MWCNT hybird. Electrochemical behaviors of Hb were carefully investigated by cyclic voltammetry with a pair of well-defined redox peaks obtained, which indicated that direct electron transfer of Hb was realized in the hybrid modified electrode. The result could be attributed to the synergistic effects of GR-MWCNT hybrid with enlarged surface area and improved conductivity through the formation of a three-dimensional network. Electrochemical parameters of the immobilized Hb on the electrode surface were further calculated with the results of the electron transfer number (n) as 1.03, the charge transfer coefficient (a) as 0.58 and the electron-transfer rate constant (ks) as 0.97 s−1. The Hb modified electrode showed good electrocatalytic ability toward the reduction of different substrates such as trichloroacetic acid in the concentration range from 0.05 to 38.0 mmol L−1 with a detection limit of 0.0153 mmol L−1 (3σ), H2O2 in the concentration range from 0.1 to 516.0 mmol L−1 with a detection limit of 34.9 nmol/L (3σ) and NaNO2 in the concentration range from 0.5 to 650.0 mmol L−1 with a detection limit of 0.282 μmol L−1 (3σ). So the proposed electrode had the potential application in the third-generation electrochemical biosensors without mediator.  相似文献

In this work, we develop a simple and rapid sensing method for the visual and fluorescent detection of acetamiprid (AC) based on the inner-filter effect (IFE) of gold nanoparticles (AuNPs) on ratiometric fluorescent quantum dots (RF-QDs). The RF-QDs based dual-emission nanosensor was fabricated by assembling green emissive QDs (QDs539 nm, λem = 539 nm) on the surface of red emissive QDs (QDs661 nm, λem = 661 nm)-doped silica microspheres. The photoluminescence (PL) intensity of RF-QDs could be quenched by AuNPs based on IFE. Acetamiprid can adsorb on the surface of AuNPs due to its cyano group that has good affinity with gold, which could induce the aggregation of AuNPs accompanying color change from red to blue. Thus, the IFE of AuNPs on RF-QDs was weakened and the PL intensity of RF-QDs was recovered accordingly. Under the optimized conditions, the PL intensity of the RF-QDs/AuNPs system was proportional to the concentration of AC in the range of 0.025–5.0 μg mL−1, with a detection limit of 16.8 μg L−1. The established method had been used for AC detection in environmental and agricultural samples with satisfactory results.  相似文献

A capillary electrophoretic method was explored to assay aromatic amines in food samples. With an inline-coupled transient isotachophoretic stacking approach, the method has yielded about 200-fold improvement of sensitivity in UV detection of three primary aromatic amines and melamine. By using K+ as a leading ion and Tris+ as a terminating ion, a plug of 10 cm (equivalent to 0.44 μL) sample solution was allowed to introduce into a 60 cm (50 cm effective) capillary for separation, giving limits of detection down to 2.0 × 10−8 M. Baseline separation was achieved within 10 min, with relative standard deviation of 0.41–0.75% (intra-day) or 1.2–1.5% (inter-day) for migration time and 3.8–4.3% (intra-day) or 5.2–6.7% (inter-day) for peak area. The method was directly applicable to assaying the melamine in powder milk samples, with recovery in between 92.0% and 107.1%. The method could also be applied to the analysis of trace primary aromatic amines migrating from composite food packaging bags after combination of a 10-fold off-line concentration step, with limit of detection down to less than 1 μg/L. By this method, 4,4′-diaminophenylmethane and 2,4-diaminotoluene were thus found in three types of composite food packaging bags.  相似文献
