Based on the enhancing effects of norepinephrine (NE), epinephrine (EP), dopamine (DA) and 5‐hydroxytryptamine (5‐HT) on the chemiluminescence (CL) reaction between [Ag(HIO6)2]5? and luminol in alkaline solution, a high‐performance liquid chromatography (HPLC) method with CL detection was explored for the sensitive determination of monoamine neurotransmitters for the first time. The UV–visible absorption spectra were recorded to study the enhancement mechanism of monoamine neurotransmitters on the CL of [Ag(HIO6)2]5? and luminol reaction. The HPLC separation of NE, EP, DA and 5‐HT was achieved with isocratic elution using a mixture of aqueous 0.2% phosphoric acid and methanol (5:95, v/v) within 11.0 min. Under the optimized conditions, the detection limits of NE, EP, DA, and 5‐HT were 4.8, 0.9, 1.9 and 2.3 ng/mL, respectively, corresponding to 17.6–96.0 pg for 20 μL sample injection. The recoveries of monoamine neurotransmitters in rat brain were >95.6% with the precisions expressed by RSD <5.0%. The validated HPLC‐CL method was successfully applied for the quantification of NE, EP, DA and 5‐HT in rat brain. A novel aptamer‐based CE with chemiluminescence (CL) assay was developed for highly sensitive detection of human immunoglobulin E (IgE). The IgE aptamer was conjugated with gold nanoparticles (AuNPs) to form AuNPs‐aptamer that could specifically recognize the IgE to produce an AuNPs‐aptamer‐IgE complex. The mixture of the AuNPs‐aptamer‐IgE complex and the unbounded AuNPs‐aptamer could be effectively separated by CE and sensitively detected with luminol‐H2O2 CL system. By taking the advantage of the excellent catalytic behavior of AuNPs on luminol‐H2O2 CL system, the ultrasensitive detection of IgE was achieved. The detection limit of IgE is 7.6 fM (S/N = 3) with a linear range from 0.025 to 250 pM. Successful detection of IgE in human serum samples was demonstrated and the recoveries of 94.9–103.2% were obtained. The excellent assay features of the developed approach are its specificity, sensitivity, adaptability, and very small sample consumption. Our design provides a methodology model for determination of rare proteins in biological samples.  相似文献

A sensitive method based on gold nanoparticle‐enhanced CE‐chemiluminescence (CL) detection was developed for quantifying uric acid (UA) in serum. In this work, gold nanoparticles were added into the running buffer of CE to catalyze the post‐column CL reaction between luminol and hydrogen peroxide, achieving highly efficient CL emission. Negative peaks were produced due to the inhibitory effects on CL emission from UA eluted from the electrophoretic capillary. The decrease in CL intensity was proportional to the concentration of UA in the range of 2.5×10?7–1.0×10?5 M. Detection limit was 4.6×10?8 M UA. Ten human serum samples were analyzed by the presented method. Serum level of UA was found to be in the range from 204 to 324 μM for healthy subjects (n=5), and from 464 to 497 μM for diabetic patients (n=5). The two groups were significantly different (p<0.05). The results suggested a potential application of the proposed assay in rapid primary diagnosis of diseases such as diabetes.  相似文献

The ring‐opening polymerization of ?‐caprolactone (?‐CL) catalyzed by iodine (I2) was studied. The formation of a charge‐transfer complex (CTC) among triiodide, I, and ?‐CL was confirmed with ultraviolet–visible spectroscopy. The monomer ?‐CL was polymerized in bulk using I2 as a catalyst to form the polyester having apparent weight‐average molecular weights of 35,900 and 45,500 at polymerization temperatures of 25 and 70 °C, respectively. The reactivity of both, ?‐CL monomer and ?‐CL:I2 CTC, was interpreted by means of the potential energy surfaces determined by semiempirical computations (MNDO‐d). The results suggest that the formation of the ?‐CL:I2 CTC leads to the ring opening of the ?‐CL structure with the lactone protonation and the formation of a highly polarized polymerization precursor (?‐CL)+. The band gaps approximated from an extrapolation of the oligomeric polycaprolactone (PCL) structures were computed. With semiempirical quantum chemical calculations, geometries and charge distributions of the protonated polymerization precursor (?‐CL)+ were obtained. The calculated band gap (highest occupied molecular orbit/lowest unoccupied molecular orbit differences) agrees with the experiment. The analysis of the oligomeric PCL isosurfaces indicate the existence of a weakly lone pair character of the C?O and C? For the first time, electrospun composite nanofibers comprising polymeric crown ether with polystyrene (PCE‐PS) have been used for the selective extraction of catecholamines – dopamine (DA), norepinephrine (NE) and epinephrine (E) – prior to their analysis by high‐performance liquid chromatography–electrochemical detection. Using a minicartridge packed with PCE‐PS composite nanofibers, the target compounds were extracted effectively from urine samples to which diphenylborinic acid 2‐aminoethyl ester was added as a complexing reagent. The extracted catecholamines could be liberated from the fiber by the addition of acetic acid. A good linearity was observed for catecholamines in the range of 2.0–200 ng mL?1 (NE, E and DA). The detection limits of catecholamines (signal‐to‐noise ratio = 3) were 0.5 ng mL?1 (NE), 0.2 ng mL?1 (E) and 0.2 ng mL?1 (DA), respectively. Under the optimized conditions, the absolute recoveries of the above three catecholamines were 90.6% (NE), 88.5% (E) and 94.5% (DA). The repeatability of extraction performance was from 5.4 to 9.2% (expressed as relative standard deviation). A sensitive competitive flow injection chemiluminescence (CL-FIA) immunoassay for immunoglobulin G (IgG) was developed using gold nanoparticle as CL label. In the configuration, anti-IgG antibody was immobilized on a glass capillary column surface by 3-(aminopropyl)-triethoxysilane and glutaraldehyde to form immunoaffinity column. Analyte IgG and gold nanoparticle labeled IgG were passed through the immunoaffinity column mounted in a flow system and competed for the surface-confined anti-IgG antibody. CL emission was generated from the reaction between luminol and hydrogen peroxide in the presence of Au (III), generated from chemically oxidative dissolution of gold nanoparticle by an injection of 0.10 mol L−1 HCl–0.10 mol L−1 NaCl solution containing 0.10 mmol L−1 Br2. The concentration of analyte IgG was inversely related to the amount of bound gold nanoparticle labeled IgG and the CL intensity was linear with the concentration of analyte IgG from 1.0 ng mL−1 to 40 ng mL−1 with a detection limit of 5.2 × 10−10 g mL−1. The whole assay time including the injections and washing steps was only 30 min for one sample, which was competitive with CL immunoassays based on a gold nanoparticle label and magnetic separation. This work demonstrates that the CL immunoassay incorporation of nanoparticle label and flow injection is promising for clinical assay with sensitivity and high-speed.  相似文献

Glutathione (GSH), l-cysteine (l-Cys) and 6-mercaptopurine (6-MP) inhibit the CL reaction of luminol–H2O2 catalyzed by gold colloids. In order to explore this, GSH, l-Cys and 6-MP were injected into the chemiluminescence system of luminol and H2O2 catalyzed by gold colloids. The results showed that gold colloids interact with GSH, l-Cys and 6-MP and decrease the CL emission. Based on this phenomenon, a simple, sensitive and convenient flow injection CL method was developed for the determination of GSH, l-Cys and 6-MP. This method provides a novel, and effective CL assay for GSH, l-Cys and 6-MP that has been applied to the determination of GSH in human serum.  相似文献

Many efforts have been made toward the achievement of high sensitivity in capillary electrophoresis coupled with chemiluminescence detection (CE‐CL). This work describes a novel dual‐signal amplification strategy for highly specific and ultrasensitive CL detection of human platelet‐derived growth factor–BB (PDGF–BB) using both aptamer and horseradish peroxidase (HRP) modified gold nanoparticles (HRP–AuNPs–aptamer) as nanoprobes in CE. Both AuNPs and HRP in the nanoprobes could amplify the CL signals in the luminol–H2O2 CL system, owing to the excellent catalytic behavior of AuNPs and HRP in the CL system. Meanwhile, the high affinity of aptamer modified on the AuNPs allows detection with high specificity. As proof‐of‐concept, the proposed method was employed to quantify the concentration of PDGF–BB from 0.50 to 250 fm with a detection limit of 0.21 fm. The applicability of the assay was further demonstrated in the analysis of PDGF–BB in human serum samples with acceptable accuracy and reliability. The result of this study exhibits distinct advantages, such as high sensitivity, good specificity, simplicity, and very small sample consumption. The good performances of the proposed strategy provide a powerful avenue for ultrasensitive detection of rare proteins in biological sample, showing great promise in biochemical analysis. This study investigated the synthesis of the biocatalyst, magnesium 2,6‐di‐tert‐butyl‐4‐methylphenoxide (Mg(BHT)2) complex, and the ring‐opening polymerization (ROP) of ε‐caprolactone (CL). The complex demonstrates high catalytic activity and controllable of molecular weight for the ROP of CL in tetrahydrofuran at room temperature, even when polymerization was performed under air. Before this study, the polymerization of CL had never been performed using a magnesium catalyst under air at room temperature. Various forms of alcohols with different purposes were also used as initiators with Mg(BHT)2. The results show that the magnesium complex acts as a perfect catalyst because of its high catalytic activity and control ability without any cytotoxicity in the polymerization of CL, making it suitable for biomedical applications. In addition, nanoparticle formation, cytotoxicity, and phototoxicity of tri‐2‐hydroxyethyl ester [Ce6‐(CH2CH2OPCL)3] were also studied in this article and Ce6‐(CH2CH2OPCL)3 formed nanoparticle can act as a nanophotosensitizer for photodynamic therapy. © 2012 Wiley Periodicals, Inc. In this study, a rapid and sensitive method is described for the catecholamines detection in rat brain. CE with LIF detection for the determination of FITC derivatized catecholamines (dopamine, epinephrine, and norepinephrine) was demonstrated. Conventional water bath and microwave‐assisted derivatization methods were employed and a significant reduction in the derivatization time from 2 h for the conventional water bath at room temperature (ca. 25°C) to 2 min for the microwave‐assisted derivatization was achieved. Online sample concentration of field‐amplified sample stacking (FASS) method was employed to achieve higher sensitivities (the detection limits obtained in the normal injection mode ranged from 2.6 to 4.5 ng L?1 and in the FASS mode ranged from 22 to 34 pg L?1). Furthermore, this microwave‐assisted derivatization CE–LIF method successfully determined catecholamines in rat brain with as low as 100 ng L?1 (FASS mode) to 10 μg L?1 (normal injection mode). This CE–LIF method provided better detection ability when compared to the best reports on catecholamines analyses.  相似文献

A novel core-shell luminol-based SiO2 nanoparticle While these nanoparticles were used as electrogenerated was synthesized by two step micro-emulsion method. chemiluminescence (ECL) reagent, the electrochemical (EC) reaction as well as the subsequent chemiluminescence (CL) reaction not only could be separated spatially, but also presented high efficiency for analytical purpose. In this case, the core-shell luminol-based SiO2 nanoparticles offered more potential to avoid the contradiction between the EC and the CL reaction conditions. A new ECL method based on the nanoparticle was developed, and isoniazid was selected as a model analyte to illustrate the characteristics of this new ECL method. Under the selected conditions, the proposed ECL response to isoniazid concentration was linear in the range of 1.0 ×10^-10 to 1.0 × 10^-6 g/mL with 2 × 10^-11g/mL detection limit.  相似文献

Chemiluminescence (CL) from the cobalt(II)‐catalyzed oxidation of luminol with hydrogen peroxide was dramatically enhanced by the presence of carbonate. The CL signal increases by several orders of magnitude over a wide range of concentrations of Co(II), luminol, or hydrogen peroxide. A limit of detection of 10?12 M for Co(II) and luminol and 10?8 M for hydrogen peroxide can be achieved. The CL emission spectrum exhibits a maximum at 425 nm, indicating that the excited 3‐aminophthalate is the emitting species. ESR spin‐trapping experiments revealed a large increase in the production of hydroxyl and carbonate radicals by the presence of carbonate, which is responsible for the enormous CL enhancement. Uric acid, ascorbic acid, acetaminophen, and p‐hydroxyphenyl acetic acid are capable of scavenging the radicals, thereby inhibiting the CL emission. The inhibition of CL intensity can be used to determine these substances at the sub‐micromolar level.  相似文献

A capillary electrophoresis (CE)/indirect chemiluminescence (CL) detection method is described for monoamines, viz., serotonin (5-HT), dopamine (DA), epinephrine (EP), and norepinephrine (NE) and for catechol (CA). Optimal separation and detection were obtained with an electrophoretic buffer of 10 mM sodium borate (pH 9.5) containing 5 mM luminol and 25 mM H2O2, and a catalyst solution of 30 microM CuSO4 in 30 mM borate buffer (pH 10.0). Complete separation of 5-HT, DA, EP, NE and CA was achieved in less than 5 min. The Cu(II)-catalyzed luminol CL reaction was employed to provide the high and constant background. Since monoamines and catechol can form stable complexes with Cu(II), inverted analyte peaks due to decreased catalytic activity of Cu(II) can be detected. The degree of CL suppression is proportional to the analyte concentrations. Linearity (r> or =20.99) over two orders of magnitude was generally obtained. The concentration limits of detection (CLODs) for the monoamines and catechol studied were between 0.5 and 3.1 uM. The relative standard deviation (RSD) values on peak size and migration time were in the ranges 3.2-4.4% and 0.4-0.5%, respectively. The applicability of the method for the analysis of pharmaceutical and biological samples was examined.  相似文献

This study examines the use of unmodified magnetite nanoparticles (Fe3O4 NPs) for selective extraction and enrichment of the catecholamines dopamine (DA), noradrenaline (NE), and adrenaline (E), prior to analysis using capillary electrophoresis with UV detection. Coordination between Fe3+ on‐the‐surface Fe3O4 NPs and the catechol moiety of catecholamines enables Fe3O4 NPs to capture catecholamines from an aqueous solution. We obtained maximum loading of catecholamines on the NP surface by adjusting the pH of the solution to 7.0. In addition, catecholamine loading on the Fe3O4 NPs increased in conjunction with NP concentrations. H3PO4 was found to be efficient for the removal of adsorbed catecholamines on the NP surface. Adding 1.2% poly(diallyldimethylammonium chloride) to the background electrolyte resulted in a baseline separation of the liberated catecholamines within 20 min. Under optimal extraction and separation conditions, the limit of detections at a S/N ratio of 3 for E, NE, and DA were 9, 8, and 10 nM, respectively. Significantly, the combination of a phenylboronate‐containing spin column and the proposed method was successfully applied to the determination of NE and DA in human urine and NE in Portulaca oleracea L. leaves.  相似文献

Neurotransmitters and their metabolites in central nervous system were known to play a significant role in sedation and hypnosis. A rapid and sensitive UFLC‐MS/MS method for simultaneous determination of serotonin, 5‐hydroxyindole acetic acid (5‐HIAA), tryptophan (Try), dopamine (DA), norepinephrine (NE), γ‐aminobutyric acid (GABA), glutamic acid (Glu) and acetylcholine (Ach) in rat brain without derivatization, ion‐pairing reagent or pre‐concentration was developed. Analytes and IS were separated on a Inertsil ODS‐EP column (150 mm × 4.6 mm, 5 µm particles) and analyzed in a single chromatographic run in less than 9.0 min, using gradient elution with the mobile phase consisting of methanol and 0.01% acetic acid in water at a flow rate of 1.2 ml min?1. The detection of the analytes was performed on 4000Q UFLC‐MS/MS system with turbo ion spray source in positive ion and multiple reaction monitoring mode. The developed method provided excellent linear calibration curves for the assay of analytes (R2 ≥ 0.9915). Limits of quantification were in the range of 1.0 ng ml?1 to 1.0 µg ml?1 for the analytes in rat brain. Intra‐ and inter‐day precision and accuracy of analytes were well within acceptance criteria (15%). Mean extraction recoveries of analytes and IS from rat brain were all more than 80.0%. Furthermore, the validated method was successfully applied to comparing profiles of analytes in normal and insomnic rat brains. Results indicated that there were statistically significant differences for serotonin, 5‐HIAA, DA, NE, Glu and Ach, but no significant difference for Try and GABA between two groups. A simple and fast flow method for the trace level determination of p‐toluidine, 2‐methyl‐5‐nitroaniline, and 2,4‐dinitroaniline in aqueous samples is reported. These amino/nitroaromatics are related to trinitrotoluene (TNT) and appear during the degradation process of the explosive. The chemical principles of ion‐pair formation and liquid‐liquid extraction are applied: In aqueous acidic medium, the protonated analyte [HA]+ makes an ion‐pair with the tetrachloroaurate(III) ion, followed by on‐line ion‐pair extraction into the dichloromethane carrier used. After membrane separation, the CH2Cl2 containing the ion‐pair, [HA]+[AuCl4], is mixed with the reversed micellar luminescent reagent of luminol (in 0.3 M Na2CO3) prepared from cetyl‐trimethylammonium chloride in CH2Cl2‐cyclohexane and the [AuCl4‐luminol chemiluminescence (CL) output is recorded. The detection limits (S/N> 3) are: p‐toluidine, 1.0 × 10−4M; 2‐methyl‐5‐nitroaniline, 1.0 × 10−7 M; 2,4‐dinitroaniline, 1.0 × 10−7 M, while the calibration curves are linear between 1.0 × 10−4 — 1.0 × 10−2 M for all the compounds. Although spectral studies indicated the formation and extraction of a very small amount of the ion‐pair species, the reversed micellar‐mediated CL detection system provides an alternative procedure for the determination of degradation products of the explosive TNT in environmental aqueous samples.  相似文献

Steroid hormones play significant roles in both worms and mammalians. (25S)‐Δ7‐Dafachronic acid (Δ7‐DA, 1 ) is a member of the dafachronic acid hormonal series that regulates both development and lifespan of C. elegans. Despite its importance, effective tools for the illumination of its mode of action are lacking. Herein, we report an efficient synthesis of trideuterated Δ7‐DA, [5,24,25‐D3]‐(25S)‐Δ7‐dafachronic acid ([D3]‐Δ7‐DA, 2 ), as a useful chemical tool for subsequent biological studies. Key steps for this bioinspired synthesis approach include site‐selective aliphatic C?H oxidation mediated by methyl(trifluoromethyl)dioxirane (TFDO), and the iridium/phosphine‐oxazoline‐catalyzed late‐stage asymmetric deuterium reduction.  相似文献

A novel capillary electrochromatography method was developed for the determination of dopamine (DA), epinephrine (EP), and norepinephrine (NE) by using a graphene oxide (GO) molecularly imprinted polymers (MIPs) coated capillary. In this article, GO was introduced as supporting matrix to synthesize MIPs in the presence of DA as template molecule. Then GO MIPs were used as the stationary phase in electrochromatography for the determination of DA, EP, and NE. The separation of these three analytes was achieved under the optimal conditions with a satisfactory correlation coefficients (R2) > 0.9957 in the range of 5.0–200.0 μg/mL for EP and NE, and 20.0–200.0 μg/mL for DA, respectively. The RSDs for the determination of three analytes were <6.19%, and the detection limits were 1.25 μg/mL for EP and NE, and 10.0 μg/mL for DA, respectively. Finally, this method was used for the determination of DA, EP, and NE in human serum and DA hydrochloride injection.  相似文献

Establishing a simple and accurate method for Hg2+ detection is of great importance for the environment and human health. In this work, platinum nanoparticles (Pt NPs) with different capped agents and morphologies were synthesized. It was found that Pt NPs exhibited peroxidase‐like activity that can catalyze the chemiluminescence (CL) of the luminol system without H2O2. The most intensive CL signals were obtained by using PVP‐capped Pt NPs as catalysis. Based on the fact that Hg2+ could further enhance the CL intensity in the Pt NPs‐luminol CL system, a Pt NPs‐catalyzed CL method based on a flow injection system is developed for the sensitive analysis of Hg2+. When the concentration of Hg2+ in the system increases, the CL intensity would together increase, thereby achieving sensitive Hg2+ detection. The limit of detection (LOD) was calculated to be 8.6 nM. This developed method provides a simple and rapid approach for the sensitive detection of Hg2+ and shows great promise for applications in other complex systems.  相似文献

The treatment of [AuCl(SMe2)] with an equimolar amount of NaO5NCS2 (O5NCS2=(aza‐[18]crown‐6)dithiocarbamate) in CH3CN gave [Au2(O5NCS2)2] ? 2 CH3CN ( 2? 2 CH3CN), and its crystal structure displays a dinuclear gold(I)‐azacrown ether ring and an intermolecular gold(I) ??? gold(I) contact of 2.8355(3) Å in crystal lattices. It is noted that two other single crystals of 2 ?tert‐butylbenzene?H2O and 2? 0.5 m‐xylene can be successfully obtained from a single‐crystal‐to‐single‐crystal (SCSC) transformation process by immersing single crystals of 2? 2 CH3CN in the respective solvents, and both also show intermolecular gold(I) ??? gold(I) contacts of 2.9420(5) and 2.890(2)–2.902(2) Å, respectively. Significantly, the emissions of all three 2 ?solvates are well correlated with their respective intermolecular gold(I) ??? gold(I) contacts, where such contacts increase with 2? 2 CH3CN (2.8355(3) Å)< 2? 0.5 m‐xylene (2.890(2)–2.902(2) Å)< 2? tert‐butylbenzene?H2O (2.9420(5) Å), and their emission energies increase with 2? 2 CH3CN (602 nm)< 2? 0.5 m‐xylene (583 nm)< 2? tert‐butylbenzene?H2O (546 nm) as well. In this regard, we further examine the solvochromic luminescence for some other aromatics, and finally their emissions are within 546–602 nm. Obviously, the above results are mostly ascribed to the occurrence of intermolecular gold(I) ??? gold(I) contacts in 2 ?solvates, which are induced by the presence of various solvates in the solid state, as a key role to be responsible for their solvochromic luminescence.  相似文献
