To explore the mechanisms in Surface‐enhanced Raman Scattering (SERS) measurements, silver nanoparticles (AgNPs) were first prepared by a silver mirror reaction to form different particle sizes and different distributions on glass substrates. After the resulting surfaces were probed with molecules of p‐Amino‐thiophenol (pATP), p‐Nitrothiophenol (pNTP), and p‐Mercaptobenzoic acid (pMBA) individually, the substrates were placed into reaction solutions to grow additional AgNPs. In this way, probe molecules could be trapped between two nanoparticles, possibly having the so‐called “hot spot” effect. To examine the variations of morphologies of AgNPs in each of the steps, the substrates were examined by field‐emission scanning electron microscope (FE‐SEM). The morphologies also were correlated with the SERS signals. Two bands in the SERS spectra of probe molecules were selected as indications of the enhancements from electromagnetic (EM) effect and charge‐transfer (CT). Results indicate that the SERS signals from the EM effect were increased ca. 5 times after growing additional AgNPs on the molecule‐modified AgNPs substrates. The SERS signals from CT effect were increased two orders of magnitude after growing additional AgNPs. The increase of enhancement for molecules between AgNPs was caused mostly by CT effect. Based on the effect of particle size and distribution of the AgNPs, the EM effect was strongly influenced by the particle size of the AgNPs, while the CT effect was less sensitive to the variation of the morphologies of the AgNPs.  相似文献

In this work, stacked but isolated silver nanoparticles (AgNPs) on silver chloride were prepared in order to create sensitive substrates that could be used to determine analytes in aqueous solution by performing surface-enhanced infrared absorption (SEIRA) measurements. These substrates have a high loading capacity, which improves sensitivity, thus allowing the detection of biospecies such as weak infrared absorbers (including amino acids) in aqueous solution. AgNPs were obtained by converting the surface layers of the silver chloride crystals into stacked but isolated AgNPs using reducing agents. To optimize the conditions used to prepare the SEIRA substrates, the roles of four common reducing agents—hydrazine, sodium borohydride, glucose, and formaldehyde—were explored by probing the finished substrates with para-nitrobenzoic acid (PNBA). Factors that influence the morphologies of the AgNPs were examined systemically. These factors include the pH of the reducing solution, the reaction time, and the concentration of the reducing agent. Results indicate that the concentration of the reducing agent and the pH of the solution strongly influence the AgNP morphology and hence the SEIRA signals. Under optimized conditions, the resulting substrates showed intense SEIRA spectra from PNBA, with enhancement factors of around two orders of magnitude compared to conventional transmission methods. The new substrates offer a high loading capacity and good heat tolerance, allowing the direct infrared detection of tyrosine in aqueous solution.  相似文献

In this study, a fast, low‐cost, and facile spray method was proposed. This method deposits highly sensitive surface‐enhanced Raman scattering (SERS) silver nanoparticles (AgNPs) on the paper‐microfluidic scheme. The procedures for substrate preparation were studied including different strategies to synthesize AgNPs and the optimization of spray cycles. In addition, the morphologies of the different kinds of paper substrates were characterized by SEM and investigated by their SERS signals. The established method was found to be favorable for obtaining good sensitivity and reproducible results. The RSDs of Raman intensity of randomly analyzing 20 spots on the same paper or different filter papers depositing AgNPs are both below 15%. The SERS enhancement factor is approximately 2 × 107. The whole fabrication is very rapid, robust, and does not require specific instruments. Furthermore, the total cost for 1000 pieces of chip is less than $20. These advantages demonstrated the potential for growing SERS applications in the area of environmental monitoring, food safety, and bioanalysis in the future.  相似文献

In this work, silver nanoparticles (AgNPs) decorated magnetic microspheres (MMs) are prepared as surface-enhanced Raman scattering (SERS) substrate for the analysis of adenine in aqueous solutions. To prepare these substrates, magnetic particles were first synthesized by coprecipitation of Fe(II) and Fe(III) with ammonium hydroxide. A thin layer of cross-linked polymer was formed on these magnetic particles by polymerization through suspension of magnetic particles into a solution of divinyl benzene/methyl methacrylate. The resulted polymer protected magnetic particles are round in shape with a size of 80 μm in diameter. To form AgNPs on these MMs, photochemical reduction method was employed and the factors in photochemical reduction method were studied and optimized for the preparation of highly sensitive and stable AgNPs on MMs substrates (abbreviated as AgMMs substrates). By dispersing the AgMMs in aqueous samples, cylindrical magnet was used to attract the AgMMs for SERS detections. The observed enhancement factor of AgMMs reached 7 orders in magnitude for detection of adenine with a detection limit approaching to few hundreds of nanomolar.  相似文献

Surface-enhanced Raman scattering (SERS) of 4-mercaptopyridine (4-mpy) adsorbed on HNO3 etched silver foil, chemically deposited silver films (silver mirror) and silver colloids were measured. The SERS study has revealed that 4-mpy was adsorbed onto the three kinds of silver surfaces by a sulfur-silver bond with the plane of pyridine ring being normal to the silver substrates. The structure of 4-mpy adsorbed on the silver surfaces depends largely on the pH values of environment. When the pH values of the environment are changed, the structure of 4-mpy adsorbed on silver surfaces can easily be altered through a protonation or deprotonation reaction occurring on the N atom of the pyridine ring, and the modified structure shows unique characters on the SERS spectrum. Owing to the remarkable enhancement ability of SERS technique and characteristic spectrum of different species, a monolayer of 4-mpy assembled on a silver mirror holds potential as a H+ sensor for highly sensitive detection of the proton concentration in an aqueous solution.  相似文献

Surface-enhanced Raman scattering (SERS) spectra of a series of n-hydroxybenzoic acids (n-HBA, n = P, M and O) adsorbed on the silver nano-particles were studied, respectively, in the silver colloidal solution and on the dried silver-coated filter paper. On the same substrate, the different molecules' SERS spectra were different, while on the different substrates the same molecules' SERS spectra were also different. Significant changes were found in the SERS spectra of PHBA molecules adsorbed on the two substrates, and the changes found in MHBA's spectra on two substrates were next to PHBA's, while almost no changes were found in the spectra of OHBA molecules. Moreover, it was found, on the filter paper, that the SERS spectra of the same molecules would change with the coverage density of the silver nano-particles. The analyses showed that the origins of these changes were the different adsorption behavior of molecules adsorbed on the silver nano-particles. Because in these three molecules the relative positions of the carboxyls and hydroxyls on the benzenes are different, the adsorption behaviors of these three molecules adsorbed on the silver surfaces are also different. The experimental results suggest that the surface characteristic of the substrate and the surface configuration of the adsorbate could exert a great influence on the adsorption behavior of the adsorbates on the substrates.  相似文献

Facile and reproducible SERS signals from Shewanella oneidensis were obtained utilizing silver nanoparticles (AgNPs) and silver nanowires (AgNWs). Additionally, SERS images identify the distribution of SERS hot-spots. One important observation is the synergistically enhanced SERS signal when AgNPs and AgNWs are used in conjunction, due to constructively enhanced electromagnetic field.  相似文献

On dried filter paper coated with silver nanoparticles, surface-enhanced Raman scattering (SERS) spectra of p-hydroxybenzoic acid (PHBA) were studied, and high-quality SERS spectra were obtained, indicating that the silver-coated filter paper is a highly SERS-active substrate. The analysis showed that the adsorption behavior of PHBA molecules on silver nanoparticles coated on filter paper was different from that in silver aqueous colloids. On the filter paper, it was found that the SERS spectra of PHBA changed with the proportion of PHBA molecules and silver nanoparticles, indicating that the adsorption behavior of PHBA molecules changed with the proportion. Surface‐enhanced Raman scattering (SERS) has attracted a great deal of interest during the past four decades and emerged as an ultrasensitive optical technique for chemical and biomedical analysis. It is widely accepted that the facile fabrication of SERS substrates with high activity and good reproducibility is of crucial importance for their applications. Herein, we report on a fast and robust method for the synthesis and immobilization of silver nanoparticles (AgNPs) into poly(oligo(ethylene glycol) methacrylate) (POEGMA) brushes under mild conditions without using any reducing agents. POEGMA brushes of different chain lengths were synthesized directly on silicon wafers by surface‐initiated atom transfer radical polymerization with various reaction time. X‐ray photoelectron spectroscopy and field emission scanning electron microscope measurements indicated that the AgNPs were firmly and homogeneously embedded into POEGMA brushes. The resulting POEGMA–AgNP hybrid films were employed as SERS substrates for the detection of 4‐aminothiophenol, giving rise to an enhancement factor of up to 1.9 × 106. To develop a smart free-standing surface enhanced Raman scattering(SERS) substrate,silver nanoparticles(AgNPs) embedded temperature-sensitive nanofibrous membrane was fabricated by electrospinning their aqueous solution containing the copolymer poly(N-isopropylacrylamide-co-Nhydroxymethylacrylamide),followed by heat treatment to form crosslinking structure within its constituent nanofibers.To avoid negative effect of the additive like stabilizer and the reactant like reductant on their SERS efficiency,the AgNPs were in-situ synthesized through reducing Ag~+ions dissolved in the polymer solution by ultraviolet irradiation.The prepared hybrid nanofibrous membrane with high stability in aqueous medium can reach its swelling or deswelling equilibrium state within 15 seconds with the medium temperature changing between 25℃and 50℃alternately.When it was used as a free-standing SERS substrate,10~(-12) mol/L of 4-nitrothiophenol in aqueous solution can be detected at room temperature,and elevating detection temperature can further lower its low detection limit.Since its generated SERS signal has desirable reproducibility,it can be used as SERS substrate for quantitative analysis.Moreover,the hybrid membrane as SERS substrate is capable of real-time monitoring the reduction of 4-nitrothiophenol into 4-aminothiophenol catalyzed by its embedded AgNPs,and the detected intermediate indicates that the reaction proceeds via a condensation route.  相似文献

The mechanism of surface‐enhanced Raman spectroscopy (SERS) is not very clear in view of the magnitude of the contribution of electromagnetic factor as well as the chemical mechanism. This report presents the extent of adsorption at different temperatures in terms of signal enhancements in SERS employing silver nanoparticles (AgNPs) of various shapes as substrate and dye molecules, crystal violet or Rhodamine 6G, as model Raman probes. Initially, the SERS signal increases with increasing temperature until a maximum intensity is reached, before it gradually decreases with increasing temperature. This trend is independent of the shape of the Raman substrates and probes. However, the temperature at which maximum intensity is obtained may depend upon the nature of the Raman probe. The energetics involved in the chemisorption process between dye molecules and AgNPs were determined through isothermal titration calorimetry and their implications for the observed SERS signals were assessed. The maximum heat change occurred at the temperature at which the maximum signal enhancement in SERS was obtained and the enhanced interaction at optimum temperature was confirmed by absorption spectroscopy.  相似文献

Silver nanoparticles (AgNPs) were deposited onto the monodispersed carboxylic polystyrene (CPS) spheres by an improved in situ reduction method. The size and coverage density of the AgNPs on the surface of CPS spheres could be easily tailored by tuning the concentrations of carboxylic functional groups and silver precursor. The morphologies and structures of the resulting CPS/Ag hybrid particles were studied by scanning electron microscopy (SEM), transmission electron microscopy (TEM), X‐ray diffraction (XRD), UV‐Vis‐NIR spectrometer and X‐ray photoelectron spectroscopy (XPS), etc. The surface enhanced Raman scattering (SERS) performances of the resulting uniform CPS/Ag hybrid particles were investigated using 4‐aminobenzenethiol (4‐ABT) as the probe molecule. The optimized CPS/Ag hybrid particles show high enhancement factor (EF) of 2.71×107, low limit of detection (LOD) of 10?10 m and good reproducibility with relative standard deviation (RSD) of 9.64 %. The good SERS improvement properties demonstrate these hybrid particles could be employed as simple and effective substrates in the SERS spectroscopy.  相似文献

Food safety is a topic of great importance for our society which places high demands on analytical methods. Surface enhanced Raman spectroscopy (SERS) meets the requirements for a rapid, sensitive and specific detection technique. The fact that metallic colloids, one of the most often used SERS substrates, are usually prepared in aqueous solution makes the detection of water-insoluble substances challenging. In this paper we present a SERS based approach for the detection of water-insoluble molecules by applying a hydrophobic surface modification onto the surface of enzymatic generated silver nanoparticles. By this approach the detection of the illegal water-insoluble food dyes, such as Sudan III in presence of riboflavin, as water-soluble competitor, is possible. Moreover, we demonstrate the usability of this kind of SERS substrates for determination of Sudan III out of spiked paprika extracts.  相似文献

In this contribution, surface-enhanced Raman spectroscopy (SERS) based on conical holed glass substrates deposited with silver colloids was reported for the first time. It combines the advantages of both dry SERS assays based on plane films deposited with silver colloids and wet SERS assays utilizing cuvettes or capillary tubes. Compared with plane glass substrates deposited with silver colloids, the conical holed glass substrates deposited with silver colloids exhibited five-to ten-folds of increase in the rate of signal enhancement, due to the internal multiple reflections of both the excitation laser beam and the Raman scattering photons within conical holes. The application of conical holed glass substrates could also yield significantly stronger and more reproducible SERS signals than SERS assays utilizing capillary tubes to sample the mixture of silver colloids and the solution of the analyte of interest. The conical holed glass substrates in combination with the multiplicative effects model for surface-enhanced Raman spectroscopy (MEMSERS) achieved quite sensitive and precise quantification of 6-mercaptopurine in complex plasma samples with an average relative prediction error of about 4% and a limit of detection of about 0.02 μM using a portable i-Raman 785H spectrometer. It is reasonable to expect that SERS technique based on conical holed enhancing substrates in combination with MEMSERS model can be developed and extended to other application areas such as drug detection, environmental monitoring, and clinic analysis, etc.  相似文献

In this study, the adsorption orientation for 2-mercaptobenzothiazole (MBT) and 2-mercaptobenzoxazole (MBO) on to silver mirror and silver sol substrates have been studied by surface enhanced Raman scattering (SERS). The MBT and MBO were chemisorbed on both silver mirror and silver sol after deprotonation with a tilted orientation to the silver surfaces. The surface enhanced properties of MBT and MBO showed that the substrate of silver mirror was superior to the sliver sol. The SERS spectra of MBT and MBO revealed that both of the MBT and MBO were adsorbed on silver surfaces strongly by a common sulfur molecule and a sulfur atom from MBT and an oxygen atom from MBO. Therefore, the adsorption orientation of MBT and MBO was little tilted perpendicularly to the silver surfaces. The adsorption geometry did not undergo any significant changes in acidic and basic solutions. It showed that the adsorption orientation for MBT and MBO were stable in the both solutions.  相似文献

In the present study, Doxorubicin (DOX) drug in healthy blood plasma was the focus of the investigation by surface-enhanced Raman scattering (SERS). In recent years, chemotherapy has been the most popular treatment for various types of cancer; however, its adverse side effects on the patient's health have made a negative aspect regarding the use of this technique. DOX is the most common chemotherapy drug and is used for the treatment of an extensive range of human malignancies. The surface-enhanced Raman scattering (SERS) is a precise technique for the detection of chemicals and biomaterials with significantly low concentrations. The glass fiber substrates coated with silver nanoparticles (AgNPs) have been used to detect DOX. First, the Tollens' method was applied to prepare the AgNPs, and the characteristics of fabricated AgNPs were evaluated using ultraviolet–visible spectroscopy (UV–Vis) and X-ray diffraction (XRD). Then, AgNPs were coated on the glass fiber substrate by a chemical method. Finally, the enhancement of the Raman signal resulted from the molecular vibrations of DOX was evaluated using these SERS-active substrates as plasmonic and Raman spectroscopy sensors. Afterward, for making the sensors practical, the DOX in blood plasma were deposited on the fabricated sensors, and the Raman vibrations were evaluated. The SERS-active substrates, AgNPs deposited on glass fiber substrates, were fabricated for the detection of DOX in and out of the blood plasma; the limit of detection (LOD) for both was 10?10 M, and the mean relative standard deviation at concentrations of 10?10 M of DOX out of blood plasma, and 10?10 M of DOX in blood plasma were obtained to be 3.76% and 3.61%, respectively for ten repeated measurements in which the AgNPs were SERS-active substrates of the biosensors for detecting the DOX. In addition, the enhancement factor was calculated both experimentally and via finite-difference time-domain (FDTD) simulation, which was 29.76 × 103 and 24.95 × 103, respectively. Therefore, these SERS-active substrates can be used to develop microsensors and show positive results for SERS-based investigations.  相似文献

In this paper, the feasibility of applying electroless displacement to prepare silver nanoparticles (AgNPs) on the surface of germanium (Ge) substrate is demonstrated, and the performances of surfaces prepared in this manner for surface-enhanced infrared absorption (SEIRA) spectroscopy are reported. The process used to produce suitable AgNPs for SEIRA by electroless deposition is simple and effective, requiring only pretreatment of the germanium surface with hot air, immersion of the substrate in a dilute solution of silver nitrate, and washing of the resulting plate. To quantify the behavior of AgNPs on a Ge substrate and to optimize the conditions for the preparation of AgNPs on Ge substrates, a monolayer of p-nitrothiophenol (PNTP) was bonded to the surface of the AgNPs by immersion of the plate in a dilute solution of PNTP and measurement of the transmission spectrum. The factors that influenced the formation of AgNPs, and hence the SEIRA signals, included the concentration of AgNO3, the reaction time and the temperature. Results indicated that stronger absorption bands in the SEIRA spectrum of a monolayer of PNTP were obtained if the reaction rate for the displacement of silver ions by Ge was slow. This condition was achieved by keeping the concentration of AgNO3 and the reaction temperature low. Under the optimal conditions found in this work, an enhancement factor of approximately 100 was achieved for commonly used probe molecules in SEIRA measurements.  相似文献

Surface-enhanced Raman scattering (SERS) enhancement and the reproducibility of the SERS signal strongly reflect the quality and nature of the SERS substrates because of diverse localized surface plasmon resonance (LSPR) excitations excited at interstitials or sharp edges. LSPR excitations are the most important ingredients for achieving huge enhancements in the SERS process. In this report, we introduce several gold and silver nanoparticle-based SERS-active substrates developed solely by us and use these substrates to investigate the influence of LSPR excitations on SERS. SERS-active gold substrates were fabricated by immobilizing colloidal gold nanoparticles on glass slides without using any surfactants or electrolytes, whereas most of the SERS-active substrates that use colloidal gold/silver nanoparticles are not free of surfactant. Isolated aggregates, chain-like elongated aggregates and two-dimensional (2D) nanostructures were found to consist mostly of monolayers rather than agglomerations. With reference to correlated LSPR and SERS, combined experiments were carried out on a single platform at the same spatial position. The isolated aggregates mostly show a broadened and shifted SPR peak, whereas a weak blue-shifted peak is observed near 430 nm in addition to broadened peaks centered at 635 and 720 nm in the red spectral region in the chain-like elongated aggregates. In the case of 2D nanostructures, several SPR peaks are observed in diverse frequency regions. The characteristics of LSPR and SERS for the same gold nanoaggregates lead to a good correlation between SPR and SERS images. The elongated gold nanostructures show a higher enhancement of the Raman signal than the the isolated and 2D samples. In the case of SERS-active silver substrates for protein detection, a new approach has been adopted, in contrast to the conventional fabrication method. Colloidal silver nanoparticles are immobilized on the protein functionalized glass slides, and further SERS measurements are carried out based on LSPR excitations. A new strategy for the detection of biomolecules, particularly glutathione, under aqueous conditions is proposed. Finally, supramolecular J-aggregates of ionic dyes incorporated with silver colloidal aggregates are characterized by SERS measurements and correlated to finite-difference time-domain analysis with reference to LSPR excitations. Colloidal silver (Ag) nanoparticles (AgNP) have been widely used for surface-enhanced Raman spectroscopy (SERS) applications. We report a simple, rapid and effective method to prepare AgNP colloids for SERS using the classic organic chemistry Ag mirror reaction with Tollens’ reagent. The AgNP colloid prepared with this process was characterized using SEM, and the reaction conditions further optimized using SERS measurements. It was found that Ag mirror reaction conditions that included 20 mM AgNO3, 5 min reaction time, and 0.5 M glucose produced AgNP colloids with an average size of 319.1 nm (s.d ± 128.1). These AgNP colloids exhibited a significant SERS response when adenine was used as the reporter molecule. The usefulness of these new AgNP colloids was demonstrated by detecting the nucleotides adenosine 5′-mono-phosphate (AMP), guanosine 5′-monophosphate (GMP), cytidine 5′-monophosphate (CMP), and uridine 5′-monophosphate (UMP). A detection limit of 500 nM for AMP was achieved with the as-prepared AgNP colloid. The bacterium Mycoplasma pneumoniae was also easily detected in laboratory culture with these SERS substrates. These findings attest to the applicability of this AgNP colloid for the sensitive and specific detection of both small biomolecules and microorganisms.  相似文献
