Cytochrome c was immobilized on a mixed-thiol (mercaptoundecanoic acid/mercaptoundecanol) modified gold electrode (MUA:MU/cyt c electrode). Characterization of the cyt c electrode showed a quasi-reversible, electrochemical redox behavior with a formal potential of −13±5 mV (versus Ag/AgCl) for the surface adsorbed protein and 3±5 mV for covalently immobilized cyt c. The heterogeneous electron transfer rate constants were determined to be about 70 and 40 s−1 for both states of the protein, respectively. They were found to be significantly higher than those of pure MUA-modified cyt c electrodes (MUA/cyt c electrodes). The interaction of superoxide radicals (O2) with the (MUA:MU)/cyt c electrode was characterized and used for an amperometric O2 detection. The influence of H2O2 and uric acid on the sensor signal was investigated. The sensitivity of the (MUA:MU)/cyt c electrode to O2 was significantly improved compared with that of the MUA/cyt c electrode. Based on a kinetic model for the superoxide detection system, a new calibration method was established. This simple and fast method used the spontaneous dismutation of KO2 and was compared with the enzymatic superoxide generation system using xanthine oxidase.  相似文献

A robust and effective composite film based on gold nanoparticles (GNPs)/room temperature ionic liquid (RTIL)/multi-wall carbon nanotubes (MWNTs) modified glassy carbon (GC) electrode was prepared by a layer-by-layer self-assembly technique. Cytochrome c (Cyt c) was successfully immobilized on the RTIL-nanohybrid film modified GC electrode by electrostatic adsorption. Direct electrochemistry and electrocatalysis of Cyt c were investigated. The results suggested that Cyt c could be tightly adsorbed on the modified electrode. A pair of well-defined quasi-reversible redox peaks of Cyt c was obtained in 0.10 M, pH 7.0 phosphate buffer solution (PBS). RTIL-nanohybrid film showed an obvious promotion for the direct electron transfer between Cyt c and the underlying electrode. The immobilized Cyt c exhibited an excellent electrocatalytic activity towards the reduction of H2O2. The catalysis currents increased linearly to the H2O2 concentration in a wide range of 5.0 × 10−5– 1.15 × 10−3 M. Based on the multilayer film, the third-generation biosensor could be constructed for the determination of H2O2.  相似文献

A Prussian Blue (PB)/polyaniline (PANI)/multi-walled carbon nanotubes (MWNTs) composite film was fabricated by step-by-step electrodeposition on glassy carbon electrode (GCE). The electrode prepared exhibits enhanced electrocatalytic behavior and good stability for detection of H2O2 at an applied potential of 0.0 V. The effects of MWNTs thickness, electrodeposition time of PANI and rotating rate on the current response of the composite modified electrode toward H2O2 were optimized to obtain the maximal sensitivity. A linear range from 8 × 10−9 to 5 × 10−6 M for H2O2 detection has been observed at the PB/PANI/MWNTs modified GCE with a correlation coefficient of 0.997. The detection limit is 5 × 10−9 M on signal-to-noise ratio of 3. To the best of our knowledge, this is the lowest detection limit for H2O2 detection. The electrode also shows high sensitivity (526.43 μA μM−1 cm−2) for H2O2 detection which is more than three orders of magnitude higher than the reported.  相似文献

A new amperometric biosensor for hydrogen peroxide was developed based on cross-linking horseradish peroxidase (HRP) by glutaraldehyde with multiwall carbon nanotubes/chitosan (MWNTs/chitosan) composite film coated on a glassy carbon electrode. MWNTs were firstly dissolved in a chitosan solution. Then the morphology of MWNTs/chitosan composite film was characterized by field-emission scanning electron microscopy. The results showed that MWNTs were well soluble in chitosan and robust films could be formed on the surface. HRP was cross-linked by glutaraldehyde with MWNTs/chitosan film to prepare a hydrogen peroxide biosensor. The enzyme electrode exhibited excellent electrocatalytic activity and rapid response for H2O2 in the absence of a mediator. The linear range of detection towards H2O2 (applied potential: −0.2 V) was from 1.67 × 10−5 to 7.40 × 10−4 M with correction coefficient of 0.998. The biosensor had good repeatability and stability for the determination of H2O2. There were no interferences from ascorbic acid, glucose, citrate acid and lactic acid.  相似文献

Magnetic nanoparticles (Fe3O4) were synthesized by a chemical coprecipitation method. X-ray diffraction (XRD) and transmission electron microscope (TEM) were used to confirm the crystallite structure and the particle's radius. The Fe3O4 nanoparticles and chitosan (CS) were mixed to form a matrix in which haemoglobin (Hb) can be immobilized for the fabrication of H2O2 biosensor. The Fe3O4-CS-Hb film exhibited a pair of well-defined and quasi-reversible cyclic voltammetric peaks due to the redox of Hb-heme Fe (III)/Fe (II) in a pH 7.0 phosphate buffer. The formal potential of Hb-heme Fe(III)/Fe(II) couple varied linearly with the increase of pH in the range of 4.0-10.0 with a slope of 46.5 mV pH−1, indicating that electron transfer was accompanied with single proton transportation in the electrochemical reaction. The surface coverage of Hb immobilized on Fe3O4-CS film glassy carbon electrode was about 1.13 × 10−10 mol cm−2. The heterogeneous electron transfer rate constant (ks) was 1.04 s−1, indicating great facilitation of the electron transfer between Hb and magnetic nanoparticles-chitosan modified electrode. The modified electrode showed excellent electrocatalytic activity toward oxygen and hydrogen peroxide reduction. The apparent Michaelis-Menten constant for H2O2 was estimated to be 38.1 μmol L−1.  相似文献

A novel disposable third-generation hydrogen peroxide (H2O2) biosensor based on horseradish peroxidase (HRP) immobilized on the gold nanoparticles (AuNPs) electrodeposited indium tin oxide (ITO) electrode is investigated. The AuNPs deposited on ITO electrode were characterized by UV-vis, SEM, and electrochemical methods. The AuNPs attached on the ITO electrode surface with quasi-spherical shape and the average size of diameters was about 25 nm with a quite symmetric distribution. The direct electron chemistry of HRP was realized, and the biosensor exhibited excellent performances for the reduction of H2O2. The amperometric response to H2O2 shows a linear relation in the range from 8.0 μmol L−1 to 3.0 mmol L−1 and a detection limit of 2 μmol L−1 (S/N = 3). The value of HRP immobilized on the electrode surface was found to be 0.4 mmol L−1. The biosensor indicates excellent reproducibility, high selectivity and long-term stability.  相似文献

A colloidal suspension of nanostructured poly(N-butyl benzimidazole)-graphene sheets (PBBIns-Gs) was used to modify a gold electrode to form a three-dimensional PBBIns-Gs/Au electrode that was sensitive to hydrogen peroxide (H2O2) in the presence of acetic acid (AcOH). The positively charged nanostructured poly(N-butyl benzimidazole) (PBBIns) separated the graphene sheets (Gs) and kept them suspended in an aqueous solution. Additionally, graphene sheets (Gs) formed “diaphragms” that intercalated Gs, which separated PBBIns to prevent tight packing and enhanced the surface area. The PBBIns-Gs/Au electrode exhibited superior sensitivity toward H2O2 relative to the PBBIns-modified Au (PBBIns/Au) electrode. Furthermore, a high yield of glucose oxidase (GOD) on the PBBIns-Gs of 52.3 mg GOD per 1 mg PBBIns-Gs was obtained from the electrostatic attraction between the positively charged PBBIns-Gs and negatively charged GOD. The non-destructive immobilization of GOD on the surface of the PBBIns-Gs (GOD-PBBIns-Gs) retained 91.5% and 39.2% of bioactivity, respectively, relative to free GOD for the colloidal suspension of the GOD-PBBIns-Gs and its modified Au (GOD-PBBIns-Gs/Au) electrode. Based on advantages including a negative working potential, high sensitivity toward H2O2, and non-destructive immobilization, the proposed glucose biosensor based on an GOD-PBBIns-Gs/Au electrode exhibited a fast response time (5.6 s), broad detection range (10 μM to 10 mM), high sensitivity (143.5 μA mM−1 cm−2) and selectivity, and excellent stability. Finally, a choline biosensor was developed by dipping a PBBIns-Gs/Au electrode into a choline oxidase (ChOx) solution for enzyme loading. The choline biosensor had a linear range of 0.1 μM to 0.83 mM, sensitivity of 494.9 μA mM−1 cm−2, and detection limit of 0.02 μM. The results of glucose and choline measurement indicate that the PBBIns-Gs/Au electrode provides a useful platform for the development of oxidase-based biosensors.  相似文献

Recent progress in flexible and lightweight electrochemical sensor systems requires the development of paper-like electrode materials. Here, we report a facile and green synthesis of a new type of MnO2 nanowires–graphene nanohybrid paper by one-step electrochemical method. This strategy demonstrates a collection of unique features including the effective electrochemical reduction of graphene oxide (GO) paper and the high loading of MnO2 nanowires on electrochemical reduced GO (ERGO) paper. When used as flexible electrode for nonenzymatic detection of hydrogen peroxide (H2O2), MnO2–ERGO paper exhibits high electrocatalytic activity toward the redox of H2O2 as well as excellent stability, selectivity and reproducibility. The amperometric responses are linearly proportional to H2O2 concentration in the range 0.1–45.4 mM, with a detection limit of 10 μM (S/N = 3) and detection sensitivity of 59.0 μA cm−2 mM−1. These outstanding sensing performances enable the practical application of MnO2–ERGO paper electrode for the real-time tracking H2O2 secretion by live cells macrophages. Therefore, the proposed graphene-based nanohybrid paper electrode with intrinsic flexibility, tailorable shapes and adjustable properties can contribute to the full realization of high-performance flexible electrode material used in point-of-care testing devices and portable instruments for in-vivo clinical diagnostics and on-site environmental monitoring.  相似文献

We proposed an electrochemical DNA sensor by using peroxidase-like magnetic ZnFe2O4–graphene quantum dots (ZnFe2O4/GQDs) nanohybrid as a mimic enzymatic label. Aminated graphene and Pd nanowires were successively modified on glassy carbon electrode, which improved the electronic transfer rate as well as increased the amount of immobilized capture ssDNA (S1). The nanohybrid ZnFe2O4/GQDs was prepared by assembling the GQDs on the surface of ZnFe2O4 through a photo-Fenton reaction, which was not only used as a mimic enzyme but also as a carrier to label complementary ssDNA (S3). By synergistically integrating highly catalytically activity of nano-sized GQDs and ZnFe2O4, the nanohybrid possessed highly-efficient peroxidase-like catalytic activity which could produce a large current toward the reduction of H2O2 for signal amplification. Thionine was used as an excellent electron mediator. Compared with traditional enzyme labels, the mimic enzyme ZnFe2O4/GQDs exhibited many advantages such as environment friendly and better stability. Under the optimal conditions, the approach provided a wide linear range from 10−16 to 5 × 10−9 M and low detection limit of 6.2 × 10−17 M. The remarkable high catalytic capability could allow the nanohybrid to replace conventional peroxidase-based assay systems. The new, robust and convenient assay systems can be widely utilized for the identification of other target molecules.  相似文献

The nucleophilic substitution reaction of S2O32− with [Ru(HaaiR′)2(OH2)2](ClO4)2 (1) [HaaiR′ = 1-alkyl-2-(phenylazo)imidazole] and [Ru(ClaaiR′)2(OH2)2](ClO4)2 (2) [ClaaiR′ = 1-alkyl-2-(chlorophenylazo)imidazole] [where R′ = Me(a), Et(b) or Bz(c)] in acetonitrile–water (50% v/v) medium to yield Na2[Ru(HaaiR′)2(S2O3)2] (3a, 3b or 3c) and Na2[Ru(ClaaiR′)2(S2O3)2] (4a, 4b or 4c) has been studied. The products were characterized by microanalytical data and spectroscopic techniques (UV–Vis, NMR and mass spectroscopy). The reaction proceeds in two consecutive steps (A → B → C); each step follows first order kinetics with respect to each complex and S2O32−, and the first step second order rate constant (k2) is greater than the second step one (k2). An increase in the π-acidity of the ligand increases the rate. Thermodynamic parameters, the standard enthalpy of activation (ΔH0) and the standard entropy of activation (ΔS0), have been calculated for both steps using the Eyring equation from variable temperature kinetic studies. The low ΔH0 and large negative ΔS0 values indicate an associative mode of activation for both aqua ligand substitution processes.  相似文献

A series of spinel-type CoxNi1−xFe2O4 (x = 0, 0.2, 0.4, 0.5, 0.6, 0.8, 1.0) magnetic nanomaterials were solvothermally synthesized as enzyme mimics for the eletroctrocatalytic oxidation of H2O2. X-ray diffraction and scanning electron microscope were employed to characterize the composition, structure and morphology of the material. The electrochemical properties of spinel-type CoxNi1−xFe2O4 with different (Co/Ni) molar ratio toward H2O2 oxidation were investigated, and the results demonstrated that Co0.5Ni0.5Fe2O4 modified carbon paste electrode (Co0.5Ni0.5Fe2O4/CPE) possessed the best electrocatalytic activity for H2O2 oxidation. Under optimum conditions, the calibration curve for H2O2 determination on Co0.5Ni0.5Fe2O4/CPE was linear in a wide range of 1.0 × 10−8–1.0 × 10−3 M with low detection limit of 3.0 × 10−9 M (S/N = 3). The proposed Co0.5Ni0.5Fe2O4/CPE was also applied to the determination of H2O2 in commercial toothpastes with satisfactory results, indicating that CoxNi1−xFe2O4 is a promising hydrogen peroxidase mimics for the detection of H2O2.  相似文献

A simple procedure has been used for preparation of modified glassy carbon electrode with carbon nanotubes and copper complex. Copper complex [Cu(bpy)2]Br2 was immobilized onto glassy carbon (GC) electrode modified with silicomolybdate, α-SiMo12O404− and single walled carbon nanotubes (SWCNTs). Copper complex and silicomolybdate irreversibly and strongly adsorbed onto GC electrode modified with CNTs. Electrostatic interactions between polyoxometalates (POMs) anions and Cu-complex, cations mentioned as an effective method for fabrication of three-dimensional structures. The modified electrode shows three reversible redox couples for polyoxometalate and one redox couple for Cu-complex at wide range of pH values. The electrochemical behavior, stability and electron transfer kinetics of the adsorbed redox couples were investigated using cyclic voltammetry. Due to electrostatic interaction, copper complex immobilized onto GC/CNTs/α-SiMo12O404− electrode shows more stable voltammetric response compared to GC/CNTs/Cu-complex modified electrode. In comparison to GC/CNTs/Cu-complex the GC/CNTs/α-SiMo12O404− modified electrodes shows excellent electrocatalytic activity toward reduction H2O2 and BrO3 at more reduced overpotential. The catalytic rate constants for catalytic reduction hydrogen peroxide and bromate were 4.5(±0.2) × 103 M−1 s−1 and 3.0(±0.10) × 103 M−1 s−1, respectively. The hydrodynamic amperommetry technique at 0.08 V was used for detection of nanomolar concentration of hydrogen peroxide and bromate. Detection limit, sensitivity and linear concentration range proposed sensor for bromate and hydrogen peroxide detection were 1.1 nM and 6.7 nA nM−1, 10 nM-20 μM, 1 nM, 5.5 nA nM−1 and 10 nM-18 μM, respectively.  相似文献

Novel condensation reaction of tropone with N-substituted and N,N′-disubstitued barbituric acids in Ac2O afforded 5-(cyclohepta-2′,4′,6′-trienylidene)pyrimidine-2(1H),4(3H),6(5H)-trione derivatives (8a-f) in moderate to good yields. The 13C NMR spectral study of 8a-f revealed that the contribution of zwitterionic resonance structures is less important as compared with that of 8,8-dicyanoheptafulvene. The rotational barriers (ΔG) around the exocyclic double bond of mono-substituted derivatives 8a-c were obtained to be 14.51-15.03 kcal mol−1 by the variable temperature 1H NMR measurements. The electrochemical properties of 8a-f were also studied by CV measurement. Upon treatment with DDQ, 8a-c underwent oxidative cyclization to give two products, 7 and 9-substituted cyclohepta[b]pyrimido[5,4-d]furan-8(7H),10(9H)-dionylium tetrafluoroborates (11a-c·BF4 and 12a-c·BF4) in various ratios, while that of disubstituted derivatives 8d-f afforded 7,9-disubstituted cyclohepta[b]pyrimido[5,4-d]furan-8(7H),10(9H)-dionylium tetrafluoroborate (11d-f·BF4) in good yields. Similarly, preparation of known 5-(1′-oxocycloheptatrien-2′-yl)-pyrimidine-2(1H),4(3H),6(5H)-trione derivatives (14a-d) and novel derivatives 14e,f was carried out. Treatment of 14a-c with aq. HBF4/Ac2O afforded two kinds of novel products 11a-c·BF4 and 12a,c·BF4 in various ratios, respectively, while that of 14d-f afforded 11d-f. The product ratios of 11a-c·BF4 and 12a-c·BF4 observed in two kinds of cyclization reactions were rationalized on the basis of MO calculations of model compounds 20a and 21a. The spectroscopic and electrochemical properties of 11a-f·BF4 and 12a-c·BF4 were studied, and structural characterization of 11c·BF4 based on the X-ray crystal analysis and MO calculation was also performed.  相似文献

Polyaniline-iron oxide magnetic nanohybrid was synthesized and characterized using various spectroscopic, microstructural and electrochemical techniques. The smart integration of Fe3O4 nanoparticles within the polyaniline (PANI) matrix yielded a mesoporous nanohybrid (Fe3O4@PANI) with high surface area (94 m2 g−1) and average pore width of 12.8 nm. Catechol is quasi-reversibly oxidized to o-quinone and reduced at the Fe3O4@PANI modified electrodes. The amperometric current response toward catechol was evaluated using the nanohybrid and the sensitivity and detection limit were found to be 312 μA μL−1 and 0.2 nM, respectively. The results from electrochemical impedance spectroscopy (EIS) indicated that the increased solution resistance (Rs) was due to elevated adsorption of catechol on the modified electrodes. Photoluminescence spectra showed ligand-to-metal charge transfer (LMCT) between p-π orbitals of the phenolate oxygen in catechol and the d-σ* metal orbital of Fe3O4@PANI nanohybrid. Potential dependent spectroelectrochemical behavior of Fe3O4@PANI nanohybrid toward catechol was studied using UV/vis/NIR spectroscopy. The binding activity of the biomagnetic particles to catechol through Brownian relaxation was evident from AC susceptibility measurements. The proposed sensor was used for successful recovery of catechol in tap water samples.  相似文献

The mean values of the standard massic energy of combustion of caffeine in phase I (or alpha) and in phase II (or beta) measured by static-bomb combustion calorimetry in oxygen, at T = 298.15 K, are Δcu° (C8H10O2N4, I) = −(21823.27 ± 0.68) J · g−1 and Δcu° (C8H10O2N4, II) = −(21799.96 ± 1.08) J · g−1, respectively.The standard (p° = 0.1 MPa) molar enthalpy of formation in condensed phase for each form was derived from the corresponding standard molar enthalpies of combustion as, and .The difference between the standard enthalpy of formation of the two polymorphs in condensed phase was also evaluated by using reaction-solution calorimetry. The obtained result, 2.04 ± 0.25 kJ · mol−1, is in agreement, within the uncertainty, with the difference between the molar enthalpies of formation obtained from combustion experiments (4.5 ± 3.2) kJ · mol−1, which can be considered as an internal test for consistency of the results.A value for the standard enthalpy of formation of caffeine in the gaseous state was proposed: , estimated from the values of the standard enthalpies of formation of both crystalline forms obtained in this work, and the data on standard enthalpies of sublimation collected from the literature.  相似文献

The vapour pressure of binary mixtures of hydrogen sulphide with ethane, propane, and n-butane was measured at T = 182.33 K covering most of the composition range. The excess Gibbs free energy of these mixtures has been derived from the measurements made. For the equimolar mixtures for (H2S + C2H6), (820.1 ± 2.4) J · mol−1 for (H2S + C3H8), and (818.6 ± 0.9) J · mol−1 for (H2S + n-C4H10). The binary mixtures of H2S with ethane and with propane exhibit azeotropes, but that with n-butane does not.  相似文献

The adsorption processes and electrochemical behavior of 4-nitroaniline (4-NA) adsorbed onto glassy carbon electrodes (GCE) have been investigated in aqueous 0.1 M nitric acid (HNO3) electrolyte solutions using cyclic voltammetry (CV). 4-NA adsorbs onto GCE surfaces, and upon potential cycling past −0.2 V, is transformed into the arylhydroxylamine (ArHA) derivative which exhibits a well-behaved pH dependent redox couple centered at 0.32 V at pH 1.5. It is noted as arylhydroxylamine modified glassy carbon electrodes (HAGCE). This modified electrode can be readily used as an immobilization matrix to entrap proteins and enzymes. In our studies, myoglobin (Mb) was used as a model protein for investigation. A pair of well-defined reversible redox peaks of Mb (Fe(III)-Fe(II)) was obtained at the Mb/arylhydroxylamine modified glassy carbon electrode (Mb/HAGC) by direct electron transfer between the protein and the GCE. The formal potential (E0), the apparent coverage (Γ*) and the electron-transfer rate constant (ks) were calculated as −0.317 V, 8.26 × 10−12 mol/cm2 and 51 ± 5 s−1, respectively. Dramatically enhanced biocatalytic activity was exemplified at the Mb/HAGC electrode by the reduction of hydrogen peroxide (H2O2), trichloroacetic acid (TCA) and oxygen (O2). The Mb/arylhydroxylamine film was also characterized by UV-visible spectroscopy (UV-vis), scanning electron microscope (SEM) indicating excellent stability and good biocompatibility of the protein in the arylhydroxylamine modified electrode. This new Mb/HAGC electrode exhibited rapid electrochemical response (2 s) for H2O2 and had good stability in physiological condition, showing the potential applicability of the films in the preparation of third generation biosensors or bioreactors based on direct electrochemistry of the proteins.  相似文献

A simple and sensitive method for the determination of nanomolar levels of hydrogen peroxide (H2O2) in seawater has been developed and validated. This method is based on the reduction of H2O2 by ferrous iron in acid solution to yield hydroxyl radical (OH) which reacts with benzene to produce phenol. Phenol is separated from the reaction mixture by reversed phase high performance liquid chromatography and its fluorescence intensity signals were measured at excitation and emission of 270 and 298 nm, respectively. Under optimum conditions, the calibration curve exhibited linearity in the range of (0-50) × 103 nmol L−1 H2O2. The relative standard deviations for five replicate measurements of 500 and 50 nmol L−1 H2O2 are 1.9 and 2.4%, respectively. The detection limit for H2O2, defined as three times the standard deviation of the lowest standard solution (5 nmol L−1 H2O2) in seawater is 4 nmol L−1. Interference of nitrite ion (NO2) on the fluorescence intensity of phenol was also investigated. The result indicated that the addition of 10 μmol L−1 NO2 to seawater samples showed no significant interference, although, the addition of 50 μmol L−1 NO2 to the seawater samples decreases the fluorescence intensity signals of phenol by almost 40%. Intercomparison of this method with well-accepted (p-hydroxyphenyl) acetic acid (POHPAA)-FIA method shows excellent agreement. The proposed method has been applied on-board analysis of H2O2 in Seto Inland seawater samples.  相似文献

In this paper, a novel electrochemiluminescence (ECL) sensor was constructed to determine herring sperm (HS) double-stranded (ds) DNA. Tetramethoxysilane and dimethyldimethoxysilane were selected as co-precursors to form an organically modified silicate (ORMOSIL) film for the immobilization of multiwall carbon nanotubes (MWNTs) wrapped by poly(p-styrenesulfonate) (PSS), and then Tris(2,2′-bipyridyl)ruthenium(II) (Ru(bpy)32+) was successfully immobilized on a glassy carbon electrode via ion-association. PSS was employed to increase the conductivity of the ORMOSIL film and disperse the cut MWNTs, which were cut and shortened in a mixture of concentrated sulfuric and nitric acids, in the film. It was found that MWNTs could adsorb Ru(bpy)32+ and acted as conducting pathways to connect Ru(bpy)32+ sites to the electrode. MWNTs also played a key role as materials for the mechanical and thermal properties. The ECL performance of this modified electrode was evaluated in a flow injection analysis (FIA) system, and the detection limit (S/N = 3) for HS ds-DNA was 2.0 × 10−7 g mL−1 with a linear range from 1.34 × 10−6 to 6.67 × 10−4 g mL−1 (R2 = 0.9876). In addition, the ECL sensor presented excellent characteristics in terms of stability, reproducibility and application life.  相似文献
