The homobifunctional crosslinker 1,4‐phenylene diisothiocyanate (PDC) was coupled to amine‐terminated magnetic particles, and human IgG, streptavidin, protein G and protein A were immobilized on the activated magnetic particles. The coupling of PDC to the amine‐terminated magnetic particles was completed in 10 min, and 1 mg of activated magnetic particles was able to immobilize 95 (g of protein G, 120 µg of protein A, 160 µg of streptavidin and 280 µg of IgG. Ultraviolet‐visible spectroscopy, FTIR spectroscopy and electron micrography were used to characterize the functional particles. The results indicated that PDC was successfully coupled to the surface of the amine‐terminated magnetic particles, and the proteins were effectively immobilized on their surface. The activity of protein G immobilized on the activated magnetic particles was confirmed by its ability to purify IgG from plasma.  相似文献

A magnetic molecularly imprinting polymer for domoic acid was fabricated. Synthesis conditions were optimized. The polymer particles have high magnetization for rapid magnetic separation. The apparent maximum absorption amount and dissociation constant of the polymer were 1,600?µg?g?1 and 20.6?µg?mL?1, respectively. The polymer retained 90% of adsorption amount after 5 times of repeated use. It was used as an adsorbent for purification and HPLC detection of domoic acid in shellfish with a detection limit of 0.050?µg?g?1. Thus, the polymer could be applied to the sample pretreatment of aquatic products.  相似文献

The purification of immunoglobulin G (IgG) from human plasma was performed by using a novel metal-chelated adsorbent with nano size. The non-porous nanoparticles were produced by surfactant free emulsion polymerization of ethylene glycol dimethacrylate (EDMA) and 2-methacryloylamidohistidine (MAH). Then, Cu(II) ions were chelated on the nanoparticles. The nano-poly(EDMA-MAH) nanoparticles were characterized by Fourier transform infrared, scanning electron microscope, atomic force microscope and elemental analysis. The non-porous nanoparticles were spherical form and have 100?C250?nm size distribution. The maximum IgG adsorption capacity of the Cu(II) chelated nanoparticles was found to be 463?mg/g polymer at pH 7.0 in HEPES buffer. Desorption of IgG was performed by 1.0?M NaCl and desorption rate was found to be 97?%. IgG was obtained from human plasma with purity of 94?% (up to 578?mg/g polymer). The non-porous nanoparticles allowed one-step purification of IgG from human plasma.  相似文献

Glycosylated proteins modulate various important functions of organisms. To reveal the functions of glycoproteins, in‐depth characterization studies are necessary. Although mass spectrometry is a very efficient tool for glycoproteomic and glycomic studies, efficient sample preparation methods are required prior to analyses. In the study, poly(amidoamine) dendrimer‐coated magnetic nanoparticles were presented for the specific enrichment and fast purification of glycopeptides and glycans. The enrichment and purification performance of the developed method was evaluated both at the glycopeptide, and the glycan level using several standard glycoprotein digests and released glycan samples. The poly(amidoamine) dendrimer‐coated magnetic nanoparticles not only showed selective affinity (Immunoglobulin G/Bovine Serum Albumin, 1/10 by weight) to glycopeptides and released glycans but also good sensitivity (0.4 ng/µL for Immunoglobulin G) for glycoproteomic and glycomic applications. Thirty‐five glycopeptides of Immunoglobulin G were detected after enrichment with poly(amidoamine) dendrimer‐coated magnetic nanoparticles. In addition, 55 18O tagged deamidated glycopeptides belonging to human plasma glycoproteome were confirmed. Finally, fifty 2‐aminobenzoic acid, and 30 procainamide‐labelled human plasma N‐glycans released from human plasma glycoproteins were determined after purifications. The results indicate that the proposed enrichment and purification method using poly(amidoamine) dendrimer‐coated magnetic nanoparticles could be simply adjusted to sample preparation methods.  相似文献

Abstract

Penicillin G (PG) in milk is unsafe for human and is a big problem for the foods industry. Hence, the better analytical and eliminative techniques are demanded. We investigated a PG molecular imprinted polymer (MIP), which can sensitively detect and extract the PG from milk and other samples. The MIP synthesis involved a β-lactam antibiotics PG, methacrylic acid (MAA) and dimethyl formamide (DMF). Its properties were analyzed with Scatchard plot, which showed that there were two affinity sites of the PG polymer. The first equilibrium dissociation constant of the high-affinity binding sites Kd1 was 1.14 × 10?4 mol/L and the binding capacity Qmax1 was 46.01 µmol/g; the second equilibrium dissociation constant of the low-affinity binding sites Kd2 was 1.35 × 10?3 mol/L and the binding capacity Qmax2 was 72.55 µmol/g. Furthermore, two PG determination methods using the polymer were developed. The first was carried out quantitatively with a spectrophotometer and the detection limit was 1 ppm. The other one was the combination of the MIP particles with milk fermentation for PG analysis and the sensitivity was 10 ppb.  相似文献

Molecularly imprinted polymer (MIP) will be modified on the surface of the core-shell structure silica magnetic nanoparticles, during which quercetin is used as a template molecule, acrylamide as a functional monomer, azo-bisisobutyronitrile as an initiator and ethylene glycol dimethacrylate as a cross-linker, to synthesize highly efficient and selective quercetin magnetic molecularly imprinted nanoparticles via Reversible Addition-Fragmentation chain Transfer (RAFT) polymerization. FT-IR and X-ray diffraction (XRD) patterns are used to monitor the reaction and show the characteristic groups of each reaction step. Compared with the conventional bulk polymerization (2.7029 mg/g), the adsorption test showed that the MMIP by RAFT activity controlled polymerization had better absorption capacity for quercetin which the saturated adsorption amount was 4.8471 mg/g. Kinetic studies indicate that pseudo first order model is suitable to describe the adsorption mechanism. Thermodynamics experiment revealed that Langmuir model was more applied for explains the adsorption of quercetin onto magnetic molecularly imprinted polymer.  相似文献

Creatinine imprinted cryogel (MIP) cartridge was prepared with functional monomer N-methacryloyl-(L)-histidinemethylester (MAH) under frozen conditions. Creatinine adsorption studies and selectivity of MIP cryogel were evaluated in aqueous solution and artificial urine sample. Maximum adsorbed amount of creatinine was calculated as 6.83 mg/g polymer for MIP cryogel. Langmuir and Freundlich adsorption isotherm models were used to investigate the adsorption behaviour of creatinine. In the artificial urine sample; recovery amounts of creatinine were found 34.7–46.2%. Creatinine imprinted cryogel (MIP) cartridge recognized creatinine, 4.58 and 4.37 times greater competitive molecules. MIP cryogel catridge was repeatedly used many times for adsorption desorption cycles.  相似文献

In this work, we systematically evaluated the potential of using boronic acid functionalized magnetic particles in the capturing of human immunoglobulin G under typical mammalian cell culture conditions. For comparison, Protein A coated magnetic particles were also used. The binding pH was found to significantly influence the adsorption isotherms of boronic acid particles with the higher capacities (0.216 g IgG/g support) being observed at pH 7.4. Comparatively, this value was 0.109 g IgG/g support, for Protein A particles under the same conditions. Both particles revealed very fast adsorption kinetics with more than 70% of the maximum binding capacity being achieved in a few seconds. The effect of glucose and lactate, which are known to interact with boronic acid, was evaluated. For glucose, the binding capacity was significantly influenced by the pH and decreased as pH increased. At pH 9.5, a 70% lower binding capacity was observed for glucose concentrations as low as 0.5 g/l. The effect of lactate was less pronounced and almost pH independent reaching at most 20% decrease in binding capacity. Nevertheless, the effect of both molecules was always lower at pH 7.4. The optimization of the elution conditions enabled complete recovery of bound IgG from boronic acid particles using 50mM Tris-HCl, 200 mM sorbitol, 200 mM NaCl at pH 8.5.  相似文献

Curcumin widely exists in food, and rapid selective and accurate detection of curcumin have great significance in chemical industry. In this experiment, a new magnetic biocompatibility molecularly imprinted polymer was prepared with nontoxic and biocompatible Zein to adsorb curcumin selectively. The polymer has high biocompatibility, good adsorption capacity, and specific adsorption for curcumin. Combined with portable electrochemical workstations, the polymer can be used to detect curcumin rapidly and cost‐effectively. Using curcumin as a template and Zein as the crosslinking agent, the polymers were synthesized on the surface of Fe3O4 particles for solid phase extraction. The experimental results showed that the polymer reached large adsorption capacity (32.12 mg/g) with fast kinetics (20 min). The adsorption characteristic of the polymer followed the Langmuir isotherm and pseudo‐second‐order kinetic models. Hexacyanoferrate was used as electrochemical probe to generate signals, and the linear range was 5–200 µg/mL for measuring curcumin. The experimental analysis showed that the polymer was an ideal material for selective accumulation of curcumin from complex samples. This approach has been successfully applied to the determination of curcumin in food samples with electrochemical detection, indicating that this is a feasible and practical technique.  相似文献

Abstract

Butylidenephthalide is one of the major active components isolated from Rhizoma Chuanxiong. This paper describes a simple, rapid, specific and sensitive method for the quantification of butylidenephthalide in rat plasma and tissue distribution using a liquid-liquid extraction procedure followed by capillary gas chromatography-selected ion monitoring mode-mass spectrometry (GC-SIM-MS) analysis. The calibration curves were linear over the concentration ranging from 0.02–10.0 µg/mL (r > 0.99) for plasma samples and 0.18–7.25 µg/g (r > 0.99) for the tissue samples. The limit of quantification (LOQ) was 1.0 ng/mL or 1.0 ng/g (ten times signal/noise ratio). Within- and between-day precisions expressed as the relative standard deviation (RSD) for the method were 2.39–2.98% and 2.97–4.26%, respectively. The methods of recovery for all samples were greater than 80% at the low, medium, and high concentrations. The method has been successfully applied to a pharmacokinetics study in rats after an oral administration of Butylidenephthalide with a dose of 20.0 mg/kg. The main pharmacokinetic parameters obtained were T max  = (0.22 ± 0.06) h, C max = (3 ± 1) µg/mL, AUC = (32 ± 6) h?µg/mL, and K a  = (8.5 ± 0.8)/h. The results showed that the butylidenephthalide was easily absorbed. The concentrations of butylidenephthalide in rat kidney, lung, heart, and cerebellum were higher than those in other organs. To determine free fraction in serum, samples were filtered using ultrafiltration membranes with a molecular weight cut-off of 10,000 Da and extracted using liquid-liquid extraction. The extracts were evaporated and analyzed by GC-MS. The protein binding in rat plasma, human plasma, and human serum albumin were 83 ± 4%, 94 ± 3%, and 89 ± 3%, respectively.  相似文献

The aim of this study is to prepare magnetic beads which can be used for the removal of heavy metal ions from synthetic solutions. Magnetic poly(ethylene glycol dimethacrylate‐vinyl imidazole) [m‐poly(EGDMA‐VIM)] beads were produced by suspension polymerization in the presence of magnetite Fe3O4 nano‐powder. The specific surface area of the m‐poly(EGDMA‐VIM) beads was found to be 63.1 m2/g with a size range of 150–200 µm in diameter and the swelling ratio was 85%. The average Fe3O4 content of the resulting m‐poly(EGDMA‐VIM) beads was 12.4%. The maximum binding capacities of the m‐poly(EGDMA‐VIM) beads were 32.4 mg/g for Cu2+, 45.8 mg/g for Zn2+, 84.2 mg/g for Cd2+and 134.5 mg/g for Pb2+. The affinity order on mass basis is Pb2+>Cd2+>Zn2+>Cu2+. Equilibrium data agreed well with the Langmuir model. pH significantly affected the binding capacity of the magnetic beads. Binding of heavy metal ions from synthetic wastewater was also studied. The binding capacities were 26.2 mg/g for Cu2+, 33.7 mg/g for Zn2+, 54.7 mg/g for Cd2+ and 108.4 mg/g for Pb2+. The magnetic beads could be regenerated up to about 97% by treating with 0.1 M HNO3. These features make m‐poly(EGDMA‐VIM) beads a potential candidate for support of heavy metal removal under magnetic field.  相似文献

In this research, nickel ferrite (NiFe2O4) magnetic nanoparticles were synthesised by a simple method and applied as sorbent for magnetic solid-phase extraction of trace amounts of Au(III) from water samples. Detection in this technique was performed by flame atomic absorption spectrometry. The effects of sample pH, amount of sorbent, extraction time, desorption solvent and its volume on the extraction process were optimised. The effects of interfering ions on the recovery of the analyte were also evaluated in model solutions. The best results were obtained at pH 6.5 with 5 mL of eluent solution (0.1 mol L?1 sodium thiosulphate) and an extraction time of 30 min. Under optimal conditions, the sorption capacity was 34.6 mg g?1. Also, enhancement factor (for 100 mL of sample solution) was found to be 19.3. The calibration graph was linear in the range of 4.4–800.0 µg L?1 gold concentration and the limit of detection was 1.32 µg L?1. The relative standard deviation of the method (for n = 8) was 1.57%. The method was successfully applied to the extraction of Au(III) from water samples.  相似文献

Glomerular filtration rate (GFR) is usually determined by estimation of iothalamate (IOT) clearance. We have developed and validated an accurate and robust method for the analysis of IOT in human plasma and urine. The mobile phase consisted of methanol and 50 mM sodium phosphate (10:90; v/v). Flow rate was 1.2 mL/min on a C18 reverse phase column, Synergi-hydro (250 × 4.6 mm) 4 µm 80 Å, with an ultraviolet detector set to 254 nm. Acetonitrile was used for the deproteination and extraction of IOT from human plasma and urine. Precision and accuracy were within 15% for IOT in both plasma and urine. The recoveries of IOT in urine and plasma ranged between 93.14% and 114.74 and 96.04–118.38%, respectively. The linear range for urine and plasma assays were 25–1500 and 1–150 µg/mL respectively. The lower limits of detection were 0.5 µg/mL for both urine and plasma, with no interference from plasma and urine matices. This method has been fully validated according to FDA guidelines and the new HPLC assay has been applied to a new formulation of IOT (Conray? 43), to calculate GFR in healthy volunteers. The new method is simple, less expensive and it would be instrumental in future clinical and pharmacokinetic studies of iothalamate in kidney patients.  相似文献

The use of highly purified immunoglobulin became among the most powerful adopted strategies in therapeutic trials nowadays. Their role as immunomodulatory and anti-inflammatory agents has widened their scope of use. A novel continuous supermacroporous monolithic cryogels embedded with histidine-epoxy-activated-sepharose beads were synthetized as a new monolithic adsorbents for the separation of immunoglobulin G from human serum. The histidine-epoxy-activated-sepharose beads were embedded into the 2-hydroxyethyl methacrylate (HEMA) cryogels present in frozen aqueous solution inside a plastic syringe. The microstructure morphology of the cryogels was characterized by swelling measurement and scanning electron microscopy. The adsorption of human IgG on the histidine-epoxy-activated-sepharose beads pHEMA cryogels appeared to follow the Langmuir–Freundlich adsorption isotherm model. The maximum IgG adsorption was observed at 4°C and pH 7.4 and was found to be 26.95 mg/g of cryogel which is close to that obtained experimentally (24.49 mg/g). The cryogels were used for several adsorption-desorption cycles without any negligible decrease in their adsorption capacity.  相似文献

The separation of a molecularly imprinted polymer for cordycepin was investigated. The synthesis employed cordycepin as the molecular template, alpha-methylacrylic acid as the functional monomer, glycol dimethyl acrylate as the cross-linking agent, azobisisobutyronitrile as the initiator, and tetrahydrofuran as the solvent and pore-foaming agent. The interaction between cordycepin and the functional monomer was investigated by ultraviolet-visible and infrared spectroscopy. The properties of the molecularly imprinted polymer were analyzed by scanning electron microscopy, equilibrium adsorption experiments, and the Scatchard equation. Static adsorption, solid phase extraction, and high-performance liquid chromatography experiments were employed to evaluate the adsorption properties and selective recognition characteristics. The results showed that the molecularly imprinted polymer had specific adsorption with cordycepin, and the maximum absorption capacity was 1920 µg/g. Scatchard analysis suggested that high affinity and low affinity binding sites were present. For the high affinity case, the dissociation constant and apparent maximum numbers of the binding sites were 0.0089 mmol/L and 4.78 µmol/g, respectively. The dissociation constant and apparent numbers of binding sites were 0.035 mmol/L and 6.047 µmol/g for the low affinity sites. Compared with the corresponding nonimprinted polymer, the cordycepin molecularly imprinted polymer exhibited higher adsorption and selectivity for cordycepin than structural analogs.  相似文献

In the present study, ionic liquid-modified silica-coated magnetic nanoparticles (MNPs) were synthesised and applied as a new anion-exchange sorbent for extraction and determination of Cr(VI) followed by inductively coupled plasma atomic emission spectrometry. The characterisation of MNPs was carried out by scanning electron microscope, Fourier transform infrared and vibrating sample magnetometer. Experimental design and response surface methodology were used for optimisation of different parameters which affect extraction efficiency of Cr(VI). Under the optimised conditions, extraction recoveries within the range of 25–33% with relative standard deviations (RSD%, n = 4) within the range of 3.0–5.0% were obtained. The limit of detection was found to be 0.1 µg L?1. The linearity was studied in the range of 0.5-200 µg L?1 with the determination coefficient of 0.9958. Also, calculated Errors% for determination of Cr(VI) in the range of 5-15 depict that the method offers acceptable accuracy for analysis of Cr(VI). The method was successfully applied for extraction and determination of Cr(VI) selectively in some tannery waste water samples.  相似文献

Abstract

A simple, rapid, specific, and reliable high performance liquid chromatographic assay of zaleplon in human plasma has been developed. Reversed‐phase chromatography was conducted using a mobile phase of methanol∶ammonium acetate buffer (50∶50) v/v, pH 3.2 adjusted with orthophosphoric acid, UV detection at 232 nm. After extraction from plasma by precipitation the drug was chromatographed using a C18 reversed‐phase analytical column. The average recoveries of zaleplon from spiked plasma in the concentration range from 0.005–0.2 µg/ml were 93.29%, and their respective CV% was 2.557%. Regression analysis for the calibration plot for plasma standards obtained on three different days for the drug concentrations between 0.005–0.2 µg/ml indicated excellent linearity (r>0.999) and the coefficient of variation of the slopes of the three lines was less than 2%. The limit of detection was 5 ng/ml. Analysis of variance of the data showed no detectable difference in the slopes of the three standard plots (F=3.1, P>0.01). The high correlation coefficients and the similarities in the slopes are good indications of the excellent reproducibility and linearity of the proposed method. The proposed method was applied to study the bioequivalence of a commercial product of zaleplon, using as reference standard the innovator drug product. The study was conducted by using one capsule (1×10 mg) of each of the commercial product and the reference standard in a two‐way open randomized crossover design involving 24 volunteers. The criteria used to assess bioequivalence of the products were AUC (0?∞), Cmax, tmax, t1/2, and K. The obtained values for these parameters were 0.246±0.03 µg h/ml, 0.150±0.013 µg/ml, 1 h, 1.26±0.36 h, and 0.5928±0.1732 h?1 for product A whereas, for product B they were 0.256±0.044 µgh/ml, 0.142±0.014 µg/ml, 1 h, 1.18±0.33 h, and 0.63±0.1747 h?1, respectively.  相似文献

Ligand location within particles, detected by immunogold labelling, was shown to influence the biochemical productivity of a silica-based solid phase, Sorbsil C-500, using a model ligand-biomolecule system (immobilised human immunoglobulin G-anti-human immunoglobulin G monoclonal antibody). The distribution of the ligand was in turn affected by the initial ligand challenge used to prepare the immunoadsorbents. Maximal productivity was achieved with adsorbents prepared with an initial challenge of about 3 mg human immunoglobulin G per ml: the ligand in these cases was shown to be more uniformally distributed within the adsorbent particles than adsorbents, exhibiting low productivity, prepared with either low (1 mg/ml) or high (9 mg/ml) concentrations of human immunoglobulin G. The ligand in the latter was restricted to the periphery of the particles.  相似文献

The new ionic liquid-modified polymer sorbent was developed by surface chemical modification of the synthesized polymer particles using ionic liquids. The obtained ionic liquid-modified polymer was successfully used as a special sorbent in a solid-phase extraction (SPE) process to isolate caffeine and theophylline from green tea. A comparison of different SPE cartridges using the blank polymer, C18, and ionic liquid-modified polymer revealed that the highest recovery was obtained using ionic liquid-modified polymer sorbent. Quantitative analysis was carried out by using a C18 column (5 µm, 150 × 4.6 mm) as an analytical column. Good linearity was obtained from 5 × 10?4 to 0.5 mg/mL (r2>0.999) for the two analytes with relative standard deviations <4.5%.  相似文献

A novel and efficient sulfadiazine imprinted polymer was synthesized via co-precipitation method and successfully grafted on magnetic multi-walled carbon nanotubes. The synthesized magnetic imprinted polymer was characterized by scanning electron microscopy, Fourier transform infrared spectroscopy, X-ray powder diffraction analysis, thermal analysis and applied as a sorbent for selective magnetic solid-phase extraction of sulfadiazine. The retained sulfadiazine was eluted by 150.0 µL methanol/acetic acid (6:4) solution and quantified by fiber optic linear array spectrophotometry via formation of a detectable azo dye. All parameters affecting the extraction of sulfadiazine were investigated and optimized. Under the optimized conditions, the method exhibited a linear dynamic range of 2.0–50.0 µg L?1 with a detection limit of 0.56 µg L?1 and enrichment factor of 300.0. The relative standard deviation at 30.0 µg L?1 of sulfadiazine (N = 6) was 2.8 and 4.6% for intra-day and inter-day, respectively. The method was successfully applied to determine sulfadiazine in human urine, honey, milk and environmental water samples.  相似文献
