Synthesis of Steroidal Spiro-1′,2′,4′-triazolidine-3′-thiones

Steroidal ketone thiosemicarbazones (4–6), obtained from the corresponding ketones(1–3), on oxidative cyclization with H₂O₂ at 0°C provide title compounds (7–9), respectively. The structures of these compounds have been established on the basis of their elemental analytical and spectral data.     

Synthesis of 3,4-Biheterylthieno[2,3-b]-thiophenes. Part I: Synthesis of 3,4-Bi(1′,3′,4′-thiadiazolyl-, s-triazolyl-,1′,3′,4′-thiadiazinyl-, 1′,3′,4′-triazinyl-, thiazolyl-, 1′,3′-thiazinyl- and primidinyl)-thieno[2,3-b]thiophenes

3,4-Diamino-2,5-dicarbethoxythieno[2,3-b]thiophene ( 1 ) was allowed to react with NaNO 2 and active methylenes to afford the corresponding azo compounds 2a-c . Hydrazonyl chloride 2a was treated with carbon disulfide, phenyl isothiocyanate, benzonitrile, benzyl cyanide, malononitrile, benzalaniline, ethyl mercaptoacetate, and ethyl glycinate to give 1,3,4-thiadiazolyl-, s-triazolyl-, 1,3,4-thiadiazinyl-, 1,3,4-triazinylthieno[2,3-b]thiophenes 3-6 respectively. The reaction of 2b,c with urea, thiourea, and guanidine afforded pyrimidinyl- and thiazinylazothieno [2,3-b]thiophenes 7-10 respectively. Bithiazolylthieno[2,3-b]thiophenes 11 and 13 were synthesized by treating compound 1 with CS 2 along with halo compounds. The addition of S,S-, N,S-, and N,O-acetals to the Schiff base 14 afforded compounds 15-17 respectively.     

Synthesis of 3-(3′-Acetyl-5′-aroyl-1′,3′,4′-oxadiazolyl-2′)-chromones

A general method has been proposed for synthesizing 3-(3-acetyl-5-aroyl-1,3,4-oxadiazolyl-2)-chromones that has been based on conversion of 3-formylchromones to acylhydrazones and of theacylhydrazones into the heterocyclic chromones.     

Synthesis of 3′,4′-Dimethoxyisoflavone Derivaties

2-(Un)substituted-3,4-dimethoxy-7-hydroxyisoflavones were synthesized. Their derivatives substituted at the phenolic hydroxyl were prepared by alkylation and acylation. Aminomethyl derivatives were also prepared.     

X-ray diffraction study of 1,5-bis-(4′,4′,4′-trifluoro-1′-methyl-3′-oxo-but-1′-enylamino)-3-oxapentane

Specific features of the molecular and crystal structures of 1,5-bis-(4′,4′,4′-trifluoro-1′-methyl-3′-oxo-but-1′-enylamino)-3-oxapentane are determined by single crystal XRD.

An Improved Synthesis of 2′-Hydroxy-3′,4′,6′-trimethoxyacylophenones

A wide variety of 2′-hydroxypolymethoxyacetophenones and propiophenones, e.g. 1a and 1b are used in the synthesis of flavone and ehromone natural products.¹ Repeated attempts to prepare 1a and 1b by reacting 1,2,3,5-tetramethoxybenzene 2 with acetyl or propionyl chlorides and AlCl₃ in ether² gave products which were difficult to purify.³ We traced the problem to ring-ethoxy contaminants which were isolated and tentatively identified as 3a and 3b.     

A Novel Fused Phosphorus Heterocycle: 4-(1′, 2′, 3′, 4′-Tetrahydro- 1, 3, 2-benzodiazaphosphorin-2′-sulfide)-3, 4b, 4a- thiazphosphaphenanthridine Derivative

The first example of fused phosphorus heterocyclic 4-[ l′-(β-bromoethyl)-4'-oxo-3'-prop yl-1′, 2′, 3′, 4-tetrahydro-1, 3, 2-benzodiazaphosphorin-2′-sulfide]-1, 2, 3, 4, 4a, 4b, 5, 6-octahydro -6-oxo-5-propyl-3, 4b, 4a-thiazphosphaphenanthridine 4a, 2′-dioxide was synthesized in excellent yield by refluxing a mixture of 1-(2-bromoethyt)-2, 3-dihydro-3-propyl-l, 3, 2-benzodiazaphos phorin-4(1H)-one 2-oxide with carbon disulfide in benzene in the presence of triethylamine.     

Synthesis and mesomorphic properties of 4-methoxyphenyl 4′- n -alkoxy-2′,3′,5′,6′-tetrafluorobiphenyl-1-carboxylates

A novel type of 4-methoxyphenyl 4′-n-alkoxy-2′,3′,5′,6′-tetrafluorobiphenyl-1-carboxylates have been synthesized. Textural observations by polarizing microscopy and DSC measurements of the phase transitions show that all of these compounds are thermotropic liquid crystals with only a nematic mesophase. The relationship between the properties and chemical structures of these compounds is discussed.     

Formation of 3a′,8′-dimethyl-3a′,5′-dihydro-1′H,3′H-dispiro-[cyclohexane-1,3′-furo[3,4-f][2]benzofuran-5′,1″-cyclohexane]-1′,7′(4′H)-dione in the reaction of 3-acetyl-4-methyl-1-oxaspiro[4.5]dec-3-en-2-one with potassium hydroxide

3a′,8′-Dimethyl-3a′,5′-dihydro-1′H,3′H-dispiro[cyclohexane-1,3′-furo[3,4-f][2]benzofuran-5′,1″-cyclohexane]-1′,7′(4′H)-dione was synthesized by reaction of 3-acetyl-4-methyl-1-oxaspiro[4.5]dec-3-en-2-one with potassium hydroxide in water.     

MASS SPECTROMETERY OF THE ISOMERIC OCTAHYDROBENZOPYRIMIDINE-2′-THIONE-4′,1-SPIROCYCLOHEXANE AND 2′-AMINOOCTAHYDROBENZO-3′,1′-THIAZINE-4′,1-SPIROCYCLOHEXANE

Abstract

Some authors have shown that tetrahydropyrimidine-2-thiones are hydrated in acidic solution and then isomerise to the 2-imino-or 2-enamino-1,3-thiazine.     

Synthesis and Crystal Structure of 3′-Ethyl 5′-Methyl 1-Benzyl-4′-methyl-2-oxo-3′,4′-dihydrospiro-[indoline-3,2′-pyrrole]-3′,5′-dicarboxylate

The crystal structure of the title compound(C24H24N2O5, Mr = 420.45) has been determined by single-crystal X-ray diffraction. The crystal is of triclinic, space group P1 with a = 8.991(1), b = 11.166(1), c = 11.169(1) , α = 91.413(2), β = 105.887(2), γ = 90.992(2)°, V = 1077.8(2) 3, Z = 2, Dc = 1.296 g/cm3, F(000) = 444, μ(MoKα) = 0.091 mm-1, the final R = 0.0466 and wR = 0.1507 for 4185 observed reflections(I 2σ(I)). The single-crystal X-ray diffraction data indicated intermolecular C(17)–H(17)O(4), C(5)–H(5)… O(4) hydrogen bonds and C–H…π interaction in the structure.     

Synthesis,structure, and physico-chemical properties of 3-methyl and 3,3-dimethyl derivatives of l-(4′,4′-dimethyl-2′,6′-dioxocyclohex-1′-yl)-3,4-dihydroisoquinoline

3,3-Dimethyl- and 3-methyl-1-(4,4-dimethyl-2,6-dioxocyclohex-1-yl)-3,4-dihydroisoquinolines have been synthesized. Crystal and molecular structures of the 3,3-dimethyl derivative have been determined. In the crystalline state this compound exists as a tautomeric form where a hydrogen atom is located at the N atom of the dihydroisoquinoline fragment of the molecule. The tautomeric equilibrium does not shift noticeably in solutions, as shown by IR, UV, and NMR spectroscopy.Translated fromIzvestiya Akademii Nauk. Diastereoselective and enantioselective synthesis of 4′-aza analogues of 2′,3′-dideoxynucleosides

A diastereo- and enantioselective synthesis of 4′-aza analogues of 2′,3′-dideoxynucleosides has been designed by the strategy of the 1,3-dipolar cycloaddition reaction of a Vasella-type nitrone. The reaction leads to (1′R)- and (1′S)-4′-aza analogues of 2′,3′-dideoxythimidine and fluorouridine, in enantiomerically pure forms.     

Stereoselective synthesis and absolute configuration of (1′R,3R,4R)-4-acetoxy-3-(2′,2′,2′-trifluoro-1′-hydroxyethyl)azetidin-2-one

The title compound 3, an intermediate in the synthesis of fluorocarbapenems, is obtained with high stereocontrol by the condensation of (R)-(+)-ethyl 4,4,4-trifluoro-3-hydroxybutanoate with N-trimethylsilyl cinnamylidenimine. X-Ray diffraction analysis of the condensation product and chemical correlations allowed the unambiguous determination of the absolute configuration.     

Synthesis of 1′-substituted 4′,4′-dimethyl-6′-methoxy-4′H-spiro[cyclohexane-1,3′-isoquinolines]

The reaction of 1-(4-methoxyphenyl)-1-(1-methylcyclohexyl)ethanol with nitriles in concentrated sulfuric acid afforded 1′-substituted 6′-methoxy-4′,4′-dimethyl-4′H-spiro[cyclohexane-1,3′-isoquinolines] as a result of consecutive Wagner-Meerwein rearrangement and Ritter reaction.     

Investigations of 2,3′-biquinolines 23. Synthesis of 1′-R-1′,4′-dihydro-2,3′-biquinolin-4′-ones by Claisen condensation

A method has been developed for the synthesis of 1′-R-1′,4′-dihydro-2,3′-biquinolin-4′-ones based on the interaction of quinaldine with N-alkyl-N-formylanthranilic acid methyl esters under the conditions of the Claisen condensation. __________ Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 3, pp. 409–410, March 2008.     

Structure of Ethanol Coordinated 1,8,15,22-Tetra-（2′,2′,4′-trimethyl-3′-pentoxy）phthalocyaninatocobalt

Crystal structure of ethanol coordinated 1,8,15,22-tetra(2′,2′,4′-trimethyl-3-pentoxy) phthalocyaninatocobalt (C64H80N8O4Co·2C2H5OH, Mr = 1176.43) was determined by X-ray diffrac- tion methods. The crystal is of monoclinic, space group P21/c with a = 16.4294(4), b = 8.0560(2), c = 24.3654(7)(A。), β = 98.3680(8)°, Z = 2, V = 3190.6(1) (A。)3, Dc = 1.225 g/cm3, F(000) = 1262, μ(MoKα) = 0.326 mm-1, the final R = 0.0865 and wR = 0.2737 for 4787 observed reflections with I ≥ 2σ(I). The bulky branched alkyloxy substituents lead to a one structural isomer and space the phthalocyanine molecules where two ethanol molecules coordinate to the cobalt atom from two sides of each molecule.
