This work presents the synthesis and characterization of metalated oligopeptide duplex assemblies composed of artificial oligopeptides bearing tethered phenyl terpyridine ligands that are coordinated to Co(II) and Fe(II) ions. The metals cross-link the oligopeptide strands, forming duplex structures. UV-vis spectrophotometric titrations that monitor the absorbance of the metal complexes' characteristic MLCT bands demonstrate stoichiometric metal chelation. Anodic peaks in the cyclic voltammograms of these molecules are consistent with one-electron oxidative reactions without strong coupling between the metal complexes. In separate chronocoulometry measurements, the diffusion coefficients of the metal complexes decrease with increasing oligopeptide length, suggesting the primary products are metal-linked oligopeptide duplex assemblies. Larger metalated oligopeptides adsorb to electrode surfaces during cyclic voltammetry and yield irreversibly adsorbed electroactive films with thicknesses that depend on the number of voltage cycles. Electrochemical and spectroelectrochemical investigations of the films on Pt and ITO electrodes show that the electron transfers in the adsorbed films are chemically reversible but are kinetically quasi-reversible.  相似文献

Two artificial peptides with pendant pyridine or bipyridine ligands have been synthesized and incorporated into oligomeric strands that are analogous to peptide nucleic acid. Spectrophotometric titrations with Cu(2+) and Fe(2+) show that the oligomers bind stoichiometric quantities of transition metals based on the number of pendant ligands. The identities of the titration products are confirmed by high resolution mass spectrometry. In the case of the bipyridine tripeptides, the titration stoichiometry and mass spectra indicate that the metal ions form interstrand cross-links between two oligopeptides, creating duplex structures linked exclusively by metal ions. Calculated molecular structures of the metalated oligopeptides and duplexes indicate that the peptide backbone acts as a scaffold for the directed assembly of metal ions. Electron paramagnetic resonance spectroscopy of the Cu-containing molecules have varying degrees of electronic interaction based on their charge and supramolecular structure. Cyclic voltammetry of the Fe(2+)- and Cu(2+)-linked bpy oligopeptide duplexes shows that they possess unique electrochemical signatures based on the redox reactivity of the metal complex.  相似文献

We have developed a series of short DNA‐binding peptides containing newly synthesized, unnatural as well as natural amino acid building blocks. By a combinatorial‐library approach, oligopeptides were developed with moderate dsDNA‐binding affinities. Two strategies were used to further enhance the binding affinity of the lead peptides: Ac‐Arg‐Ual‐Sar‐Chi‐Chi‐Chi‐Arg‐NH2 and Ac‐Arg‐Cbg‐Cha‐Chi‐Chi‐Tal‐Arg‐NH2. Site‐selective amino acid substitutions increased the binding affinities up to 2 × 10?5 M . Further enhancement of the binding affinities could be achieved by coupling of an acridine intercalating unit, using linker arms of different length and flexibility. With the introduction of a new lysine‐based acridine unit, different types of oligopeptide–acridine conjugates were designed using known dsDNA‐binding ligands as model compounds. The binding capacities of these new oligopeptide–acridine conjugates have been investigated by a fluorescent intercalator (ethidium bromide) displacement (FID) assay. With the synthesis of the dipeptide–acridine conjugates, binding affinities in the low micromolar range were obtained (6.4 × 10?6 M ), which is similar to the binding strength of the well‐known DNA binder Hoechst 33258.  相似文献

A series of platinum(II) complexes bearing a chromophore-acceptor dyad obtained by reacting 4-(p-bromomethylphenyl)-6-phenyl-2,2'-bipyridine or 4'-(p-bromomethylphenyl)-2,2':6',2'-terpyridine with pyridine, 4-phenylpyridine, 4,4'-bipyridine, 1-methyl-4-(pyridin-4'-yl)pyridinium hexafluorophosphate respectively, were synthesized. Their photophysical properties, emission quenching studies by Pt nanoparticles and methyl viologen, electrochemical properties and photoinduced electron-transfer reactions in a photocatalytic hydrogen-generating system containing triethanolamine and colloidal Pt without an extra electron relay, were investigated. A comparison of the rates of hydrogen production for the two photocatalytic systems, one containing a metal-organic dyad and the other comprising a 1:1 mixture of the parental platinum(II) complexes and the corresponding electron relay, showed that intramolecular electron transfer improves the photocatalytic efficiency. Compared with cyclometalated platinum(II) complexes, the related platinum(II) terpyridyl complexes exhibited poor performance for photocatalytic hydrogen evolution. An investigation into the amount of hydrogen generated by three platinum(II) complexes containing cyclometalated ligands with methyl groups located on different phenyl rings revealed that the efficiency of hydrogen evolution was affected by a subtle change of functional group on ligand, and the hydrogen-generating efficiency in the presence or absence of methyl viologen is comparable, indicating electron transfer from the excited [Pt(C^N^N)] chromophore to colloidal Pt. (1)H NMR spectroscopy of the metal-organic dyads in an aqueous solution in the presence of excess triethanolamine revealed that the dyad with a viologen unit was unstable, and a chemical reaction in the compound occurred prior to irradiation by visible light under basic conditions.  相似文献

用ESI-MS研究了一系列结构具有可比性的N-磷酰化肽酯及小肽和溶菌酶的非共价相互作用, 比较了磷酰化肽酯及小肽分子中的不同基团对相互作用的影响. 结果表明—OH对其与溶菌酶的相互作用有较大贡献; 芳香环由于位阻原因, 对相互作用有促进和阻碍双重效应; 当—OH与芳香环相连时会发生协同效应, 可使相互作用显著增强. 磷酰化肽酯及小肽的体积大小、空间位阻对相互作用亦有显著影响. 磷酰化二肽中氨基酸残基的构型、顺序、碳链长短的变化(增加1~2个C)对其与蛋白溶菌酶之间的相互作用在质谱中没有表现出影响. 分子结构较为伸展、分子柔顺性好、空间位阻较小的磷酰化小肽更容易使蛋白在溶液中的构象趋于收缩, 而构象较为收缩的蛋白分子更易结合空间位阻较小的磷酰化小肽分子.  相似文献

The 15N NMR data for 105 complexes of Pd(II), Pt(II), Au(III), Co(III), Rh(III), Ir(III), Pd(IV), and Pt(IV) complexes with simple azines such as pyridine, 2,2'-bipyridine, 1,10-phenanthroline, quinoline, isoquinoline, 2,2'-biquinoline, 2,2':6', 2'-terpyridine and their alkyl or aryl derivatives have been reviewed. The 15N NMR coordination shifts, i.e. the differences between the 15N chemical shifts of the same nitrogen in the molecules of the complex and the ligand (Delta(15N) (coord) = delta(15N) (compl)--delta(15N) (lig)), have been related to some structural features of the reviewed coordination compounds, like the type of the central ion and the character of auxiliary ligands (mainly in trans position). These Delta(15N) (coord) parameters are negative, their absolute magnitudes (ca 30-150 ppm) generally increasing in the metal order Au(III) < Pd(II) < Pt(II) and Rh(III) < Co(III) < Pt(IV) < Ir(III), as well as with the enhanced trans influence of the other donor atoms (H, C < Cl < N).  相似文献

The natural product Gramicidin S is a promising scaffold for novel oligopeptide‐based bisphosphine ligands, combining the advantageous rigid chiral backbone with the close proximity of phosphine substituents. The required unnatural, phosphine‐containing, amino acid building blocks were synthesized by means of a novel protocol that involves the enantioselective alkylation of a chiral nickel Schiff base template. Three Ni complexes were prepared with different alkyl chains between the phosphine group and the α‐carbon atom of the incorporated glycine; the absolute stereochemistry of two of them was determined by single‐crystal X‐ray structure analysis. By detaching the template, enantiopure L ‐phosphine amino acids resulted enabling the solid‐phase, stepwise construction of a linear sequence of the phosphine‐modified oligopeptides. On cyclization three bisphosphine‐substituted Gramicidin S analogues resulted, differing only in the size and shape of the linkage between the phosphine groups and the oligopeptides backbone. Their crystal structures suggest these species to have potential as chelating ligands.  相似文献

The palladium(II)‐catalyzed C(sp3)−H alkynylation of oligopeptides was developed with tetrabutylammonium acetate as a key additive. Through molecular design, the acetylene motif served as a linchpin to introduce a broad range of carbonyl‐containing pharmacophores onto oligopeptides, thus providing a chemical tool for the synthesis and modification of novel oligopeptide–pharmacophore conjugates by C−H functionalization. Dipeptide conjugates with coprostanol and estradiol were synthesized by this method for potential application in targeted drug delivery to tumor cells with overexpressed nuclear hormone receptors.  相似文献

Nano-sized phosphine and pyridine ligands having tetraphenylphenyl-, m-terphenyl-, poly(benzylether) moieties were synthesized. These ligands showed a remarkable effect on homogeneous transition metal catalyzed reactions. Pd(II) complexes with tetraphenylphenyl substituted pyridine ligands show high catalytic activities for oxidation of ketones suppressing Pd black formation and maintains the catalytic activity for a long time. Rh(I) complex catalysts with m-terphenyl substituted phosphine ligands showed remarkable rate acceleration in the hydrosilylation of ketones. In addition, several phosphinocalixarene ligands were synthesized and their coordination studies with Pd(II), Pt(II), Ru(II), Ir(I), and Rh(I) metals were documented. Ir(I) and Rh(I) cationic complexes with a 1,3,5-triphosphinocalix[6]arene ligand showed dynamic behavior with size-selective molecular recognition.  相似文献

We investigated the self‐aggregation of 12 short ionic oligopeptides constituted by 4–7 amino acid residues to establish useful structure–property relationships that might be exploited in the biomedical field by using the concept of molecular Lego. We show that the critical aggregation concentration (CAC) of tetrapeptides decreases with increasing hydrophobicity of neutral residues. Additionally, the dependence of the CAC of isomeric oligopeptides on the distribution of amino acid residues confirms the high tendency to self‐organization of molecules with alternating ionic and neutral residues. Indeed, atomic force microscopy (AFM) images recorded on oligopeptide solutions above the CAC show the presence of either fibrillar or spherical aggregates depending on oligopeptide structure and concentration, steric hindrance, solution pH, and time. The potential of the investigated oligopeptides in tissue engineering applications is supported by their in vitro cytocompatibility. © 2010 Wiley Periodicals, Inc. A series of dinuclear Pt(II) complexes of the type [Pt2(N,N,N',N'-tetrakis(2-pyridylmethyl)diamine(H2O)2]4+ were synthesized. Acid-base titrations, and concentration and temperature dependent stopped-flow measurements of the reaction with chloride were performed to study the thermodynamic and kinetic behaviour of the dinuclear bridged complexes. The results indicate that there is a clear interaction between the two Pt(II) centres, which becomes weaker as the aliphatic chain increases in length. From a certain chain length onwards, the Pt(II) centres become independent of each other and exhibit identical thermodynamic and kinetic properties. The experimental results are discussed in reference to structures obtained by DFT (BP86/LACVP*) calculations.  相似文献

The asymmetric terphenyl-2'-carboxylate ligand 3,5-dimethyl-1,1':3',1' '-terphenyl-2'-carboxylate, -O2CArPh,Xyl, was prepared in high yield. This ligand facilitates the assembly of the diiron(II) complexes [Fe2(micro-O2CArTol)2(O2CArPh,Xyl)2(THF)2] [2, -O2CArTol=2,6-di-p-tolylbenzoate], [Fe2(micro-O2CArTol)2(O2CArPh,Xyl)2(pyridine)2] (5), [Fe2(micro-O2CArPh,Xyl)2-(O2CArPh,Xyl)2(THF)2] (3), and [Fe2(micro-O2CArPh,Xyl)2(O2CArPh,Xyl)2(pyridine)2] (6), all of which have a windmill geometry. The iron-iron distance of 3.355[10] A in 6 is approximately 1 A shorter than that in the analogue [Fe2(micro-O2CArTol)2(O2CArTol)2(pyridine)2] (4) and similar to the approximately 3.3 A metal-metal separation at the active site of the reduced diiron(II) form of the soluble methane monooxygenase hydroxylase enzyme (MMOHred). A series of ortho-substituted picolyl-based ligands, 2-picSMe, 2-picSEt, 2-picStBu, 2-picSPh, 2-picSPh(Me3) (Ph(Me3)=mesityl), and 2-picSPh(iPr3) (Ph(iPr3)=2,4,6-triisopropylphenyl), were prepared and allowed to react with [Fe2(micro-O2CAr)2(O2CAr)2(THF)2] to produce [Fe2(micro-O2CAr)3(O2CAr)(picSR)] (7-13, Ar=ArTol or ArPh,Xyl) complexes in 45-87% yields. The substrates tethered to the pyridine N-donor ligands picSR, where R=Me, Et, tBu, or Ph, coordinate to one iron atom of the diiron(II) center by the nitrogen and sulfur atoms to form a five-membered chelate ring. The Fe-S distance be-comes elongated with increasing steric hindrance imparted by the R group. The most sterically hindered ligands, 2-picSPh(Me3) and 2-picSPh(iPr3), bind to the metal only through the pyridine nitrogen atom. The reactions of several of these complexes with dioxygen were investigated, and the oxygenated products were analyzed by 1H NMR spectroscopy and GC/MS measurements following decomposition on a Chelex resin. The amount of sulfoxidation product is correlated with the Fe...S distance. The ratio of oxidized to unoxidized thioether substrate varies from 3.5, obtained upon oxygenation of the weakly coordinated 2-picSPh ligand in 10, to 1.0, obtained for the bulky 2-picSPh(iPr3) ligand in 12, for which the iron-sulfur distance is >4 A. External thioether substrates were not oxidized when present in oxygenated solutions of paddlewheel and windmill diiron(II) complexes containing 1-methylimidazole or pyridine ligands, respectively.  相似文献

Complexes that bind and stabilize G-quadruplex DNA structures are of significant interest due to their potential to inhibit telomerase and halt tumor cell proliferation. We here report the synthesis of the first Pt(II) G-quadruplex selective molecules, containing pi-extended phenanthroimidazole ligands. Binding studies of these complexes with duplex and quadruplex d(T(4)G(4)T(4))(4) DNA were performed. Intercalation to duplex DNA was established through UV/Vis titration, CD spectroscopy, and thermal denaturation studies. Significantly stronger binding affinity of these phenanthroimidazole Pt(II) complexes to G-quadruplex DNA was observed by UV/Vis spectroscopy and competitive equilibrium dialysis studies. Observed binding constants to quadruplex DNA were nearly two orders of magnitude greater than for duplex DNA. Circular dichroism studies show that an increase in pi-surface leads to a significant increase in the thermal stability of the Pt(II)/quadruplex DNA complex. The match in the pi-surface of these phenanthroimidazole Pt(II) complexes with quadruplex DNA was further substantiated by molecular modeling studies. Numerous favorable pi-stacking interactions with the large aromatic surface of the intermolecular G-quadruplex, and unforeseen hydrogen bonds between the ancillary ethylenediamine ligands and the quadruplex phosphate backbone are predicted. Thus, both biological and computational studies suggest that coupling the square-planar geometry of Pt(II) with pi-extended ligands results in a simple and modular method to create effective G-quadruplex selective binders, which can be readily optimized for use in telomerase-based antitumor therapy.  相似文献

Solid-phase oligopeptide synthesis has been well developed and most short oligopeptides can now be easily synthesized. However, when a desired oligopeptide forms a secondary structure or includes less reactive amino acids such as aminoisobutyric acid, its terminal amino groups become less reactive and synthesis of the desired oligopeptides becomes difficult. To expand the number of synthetic peptide sequences, we have developed efficient coupling conditions using 3-nitro-l,2,4-triazol-l-yl-tris(pyrrolidin-1-yl)phosphonium hexafluorophosphate (PyNTP) as a highly reactive condensing reagent on an unswellable solid support. PyNTP demonstrated higher reactivity than conventional condensing reagents and the optical purity of the synthesized oligopeptides was sufficiently high for application to general oligopeptide synthesis.  相似文献

A series of novel dinuclear platinum(II) complexes were synthesized containing a mixed nitrogen-sulfur donor bidentate chelate system in which the two platinum centers are connected by an aliphatic chain of variable length. The bidentate chelating ligands were selected to stabilize the complex toward decomposition. The pK(a) values and reactivity of the four synthesized complexes, namely, [Pt(2)(S(1),S(4)-bis(2-pyridylmethyl)-1,4-butanedithioether)(OH(2))(4)](4+) (4NSpy), [Pt(2)(S(1),S(6)-bis(2-pyridylmethyl)-1,6-hexanedithioether)(OH(2))(4)](4+) (6NSpy), [Pt(2)(S(1),S(8)-bis(2-pyridylmethyl)-1,8-octanedithioether)(OH(2))(4)](4+) (8NSpy), and [Pt(2)(S(1),S(10)-bis(2-pyridylmethyl)-1,10-decanedithioether)(OH(2))(4)](4+) (10NSpy), were investigated. This system is of special interest because only little is known about the substitution behavior of dinuclear platinum complexes that contain a bidentate chelate that forms part of the aliphatic bridging ligand. Moreover, the ligands as well as the dinuclear complexes were examined in terms of their cytotoxic activity, and the 10NSpy complex was found to be active. Spectrophotometric acid-base titrations were performed to determine the pK(a) values of all the coordinated water molecules. The substitution of coordinated water by thiourea was studied under pseudo-first-order conditions as a function of nucleophile concentration, temperature, and pressure, using stopped-flow techniques and UV-vis spectroscopy. The results for the dinuclear complexes were compared to those for the corresponding mononuclear reference complex [Pt(methylthiomethylpyridine)(OH(2))(2)](2+) (Pt(mtp)), by which the effect of the increasing aliphatic chain length of the bridged complexes could be investigated. The results indicate that there is a clear interaction between the two platinum centers, which becomes weaker as the chain length between the metal centers increases. Furthermore, differences and similarities of the N,S-system were compared to the corresponding dinuclear N,N-system studied previously in our group. In addition, quantum chemical calculations were performed to support the interpretation and discussion of the experimental data.  相似文献

Platinum(II) complexes of types PtLX2, PtL2X2, PtLX″ and the Pt(IV) complexes PtLXY (where L = mono- or bidentate organic ligand containing nitrogen donor atoms; X = Cl or Br; X′ = oxalate or malonate and Y = Br) have been synthesized and characterized from their elemental analysis, IR and X-ray photoelectron spectral data. The Pt 4f7/2 binding energies indicate that 1,8-naphthalene-diamine ligand is a better donor of electron density to the metal than other ligands studied here. The Cl 2p3/2 binding energies in the square planar Pt(II) complexes are observed in the range 198.8 ± 0.8 eV. The ν (PtCl) vibrations (ca 335 and 320 cm?1) corresponding to two cis-Cl ligands were observed in the IR spectra.The extent of the interaction between cis-dichloro-bis-(theophylline)platinum(II) with calf thymus DNA has beenstudied. The UV difference spectra resulting from aquated PtII(theoph)2-DNA interaction exhibit bands at 282 and 292 nm attributable to the change in the electron distribution of the base moieties induced by binding with platinum and due to the loss of base stacking. Melting profiles for the DNA samples treated with Pt-complex showed decrease in the melting temperature. Binding of the guanine residues of the DNA, involving probably (N7)-0(6) positions to the metal is implied.  相似文献

Capillary electrophoresis-electrospray ionization-mass spectrometry (CE-ESI-MS) has been used for investigating the influence of the sulfur containing amino acid L-methionine (L-Met) on the binding behavior of oxaliplatin (trans-R,R-diaminocyclohexane-(oxalato)platinum(II)) to 5'-GMP. L-Methionine competes with 5'-GMP for the platinum binding site and forms as well as 5'-GMP adducts with oxaliplatin. The formation of the prognosed complexes [Pt(DACH)(L-Met-S,N)]+ and [Pt(DACH)(5'-GMP)2]2- (DACH = 1,2-diaminocyclohexane) could be proved directly by using CE-ESI-MS. Furthermore, we could now bring forward proofs, that the coordination of 5'-GMP with oxaliplatin is inhibited by L-methionine and could show, that the 5'-GMP ligands of the [Pt(DACH) (5'-GMP)2]2- complex can be replaced slowly by L-methionine whereas methionine can not be replaced by GMP.  相似文献

Potentiometric, bipotentiometric and biamperometric titrations with an iron(II) salt in alkaline solutions containing sorbitol are reported. Suitable conditions are discussed for determinations based on the Bi(III) → Bi°, Cu(II) → Cu(I) → Cu°, Pt(IV) → Pt(II) → Pt° and Te(VI) → Te(IV) → Te° → telluride systems.  相似文献

为清除尿毒症患者血清中分离出的八肽VVRGCTWW(V8), 制备了不同间隔臂长含苯环的吸附剂Phenc. 吸附实验结果显示, 吸附剂Phe3c具有非常好的吸附能力. 采用NMR和分子模拟技术对吸附剂的模型体系的吸附机理进行了研究. 结果显示, 配体中的苯环可与八肽形成π-π堆积, 而且间隔臂的增长可以克服空间位阻效应, 有效增加配体与V8的作用几率, 进而增强吸附剂与八肽的相互作用. 研究结果表明, 采用合理的分子模型及分子对接方法, 不仅可以合理解释吸附剂的吸附机理, 而且可用于吸附剂的虚拟筛选.  相似文献
