An unmediated hydrogen peroxide (H2O2) biosensor was prepared by co‐immobilizing hemoglobin (Hb) with platinum nanoparticles enhanced poly(chloromethyl thiirane) cross‐linked chitosan (CCCS‐PNs) hybrid film. CCCS could provide a biocompatible microenvironment for Hb and PNs could accelerate the electron transfer between Hb and the electrode. Spectroscopic analysis indicated that the immobilized Hb could maintain its native structure in the CCCS‐PNs hybrid film. Entrapped Hb exhibited direct electrochemistry for its heme Fe(III)/Fe(II) redox couples at ?0.396 V in the CCCS‐PNs hybrid film, as well as peroxidase‐like activity to the reduction of hydrogen peroxide without the aid of an electron mediator.  相似文献

A novel biocompatible composite film containing sodium alginate (SA), room temperature ionic liquid 1-butyl-3-methylimidazolium hexafluorophosphate (BMIMPF6), SiO2 nanoparticle, and hemoglobin (Hb) was fabricated and covered on the surface of a traditional carbon paste elecrode (CPE). The immobilized Hb on the electrode surface showed good direct electrochemical behaviors, and a pair of quasi-reversible redox peaks of Hb was obtained, which indicated that the direct electron transfer of Hb with the electrode surface had been achieved. The SA/nano-SiO2/BMIMPF6/Hb/CPE showed dramatically electrocatalytic activity to the reduction of trichloroacetic acid, hydrogen peroxide (H2O2), and oxygen (O2). The kinetic parameters for the electrocatalytic reactions were evaluated. The composite film showed the potential to the biosensor and biocatalysis.  相似文献

We have studied the trans-membrane electron transfer in human red blood cells (RBCs) immobilized in a chitosan film on a glassy carbon electrode (GCE). Electron transfer results from the presence of hemoglobin (Hb) in the RBCs. The electron transfer rate (k s) of Hb in RBCs is 0.42 s?1, and <1.13 s?1 for Hb directly immobilized in the chitosan film. Only Hb molecules in RBCs that are closest to the plasma membrane and the surface of the electrode can undergo electron transfer to the electrode. The immobilized RBCs displayed sensitive electrocatalytic response to oxygen and hydrogen peroxide. It is believed that this cellular biosensor is of potential significance in studies on the physiological status of RBCs based on observing their electron transfer on the modified electrode.
The transmembrane electron transfer rate of Hb in RBCs is slower than hemoglobin molecules directly immobilized on the chitosan film. Only those hemoglobin in RBCs closest to the plasma membrane and electrode could exchange electrons with the electrode. The immobilized RBCs showed sensitive electrocatalytic response to O2 and H2O2.  相似文献

A new amperometric biosensor for hydrogen peroxide was developed based on cross-linking horseradish peroxidase (HRP) by glutaraldehyde with multiwall carbon nanotubes/chitosan (MWNTs/chitosan) composite film coated on a glassy carbon electrode. MWNTs were firstly dissolved in a chitosan solution. Then the morphology of MWNTs/chitosan composite film was characterized by field-emission scanning electron microscopy. The results showed that MWNTs were well soluble in chitosan and robust films could be formed on the surface. HRP was cross-linked by glutaraldehyde with MWNTs/chitosan film to prepare a hydrogen peroxide biosensor. The enzyme electrode exhibited excellent electrocatalytic activity and rapid response for H2O2 in the absence of a mediator. The linear range of detection towards H2O2 (applied potential: −0.2 V) was from 1.67 × 10−5 to 7.40 × 10−4 M with correction coefficient of 0.998. The biosensor had good repeatability and stability for the determination of H2O2. There were no interferences from ascorbic acid, glucose, citrate acid and lactic acid.  相似文献

A novel method to fabricate a third‐generation hydrogen peroxide biosensor was reported. The electrode was first derivatized by electrochemical reduction of in situ generated 4‐carboxyphenyl diazonium salt (4‐CPDS) in acidic aqueous solution yielded stable 4‐carboxyphenyl (4‐CP) layer. The horseradish peroxidase (HRP) enzyme was then covalently immobilized by amidation between NH2 terminus of enzyme and COOH terminus of 4‐CP film making use of the carbodiimide chemistry. Electrodeposition conditions used to control electrode functionalization density and film electron transfer kinetics were assessed by chronoamperometry and electrochemical impedance spectroscopy. The immobilized HRP displayed excellent electrocatalytic activity towards the reduction of hydrogen peroxide (H2O2) without any mediators. The effect of various operational parameters was explored for optimum analytical performance. The reported biosensor exhibited fast amperometric response (within 5 s) to H2O2. The detection limit of the biosensor was 5 μM, and linear range was from 20 μM to 20 mM. Furthermore, the biosensor exhibited high sensitivity, good reproducibility, and long‐term stability.  相似文献

A robust and effective composite film combined the benefits of Nafion, room temperature ionic liquid (RTIL) and multi‐wall carbon nanotubes (MWNTs) was prepared. Hemoglobin (Hb) was successfully immobilized on glassy carbon electrode surface by entrapping in the composite film. Direct electrochemistry and electrocatalysis of immobilized Hb were investigated in detail. A pair of well‐defined and quasi‐reversible redox peaks of Hb was obtained in 0.10 mol·L?1 pH 7.0 phosphate buffer solution (PBS), indicating that the Nafion‐RTIL‐MWNTs film showed an obvious promotion for the direct electron transfer between Hb and the underlying electrode. The immobilized Hb exhibited an excellent electrocatalytic activity towards the reduction of H2O2. The catalysis current was linear to H2O2 concentration in the range of 2.0×10?6 to 2.5×10?4 mol·L?1, with a detection limit of 8.0×10?7 mol·L?1 (S/N=3). The apparent Michaelis‐Menten constant (Kmapp) was calculated to be 0.34 mmol·L?1. Moreover, the modified electrode displayed a good stability and reproducibility. Based on the composite film, a third‐generation reagentless biosensor could be constructed for the determination of H2O2.  相似文献

A novel nanocomposite of colloidal gold (GNPs) and hydroxyapatite nanotubes (Hap) was prepared for immobilization of a redox protein, hemoglobin (Hb), on glassy carbon electrode. The immobilized Hb showed fast direct electron transfer and excellent electrocatalytic behavior toward reduction of hydrogen peroxide. A synergic effect between GNPs and Hap for accelerating the surface electron transfer of Hb was observed, which led to a pair of redox peaks with a formal potential of (?340±2) mV at pH 7.0, and a new biosensor for hydrogen peroxide with a linear range from 0.5 to 25 μM and a limit of detection of 0.2 μM at 3σ. Owing to the good biocompatibility of the nanocomposite, the biosensor exhibited good stability and acceptable reproducibility. The as‐prepared nanocomposite film provided a good matrix for protein immobilization and biosensor preparation.  相似文献

Abstract

Platinum nanowires (PtNW) were prepared by an electrodeposition strategy using nanopore alumina template. The nanowires prepared were dispersed in chitosan (CHIT) solution and stably immobilized onto the surface of glassy carbon electrode (GCE). The electrochemical behavior of PtNW‐modified electrode and its application to the electrocatalytic reduction of hydrogen peroxide (H2O2) are investigated. The modified electrode allows low potential detection of hydrogen peroxide with high sensitivity and fast response time. As an application example, the glucose oxidase was immobilized onto the surface of PtNW‐modified electrode through cross‐linking by glutaric dialdehyde. The detection of glucose was performed in phosphate buffer at –0.2 V. The resulting glucose biosensor exhibited a short response time (<8 s), with a linear range of 10?5?10?2 M and detection limit of 5×10?6 M.  相似文献

A novel biopolymer/room‐temperature ionic liquid composite film based on carrageenan, room temperature ionic liquid (IL) [1‐butyl‐3‐methylimidazolium tetra?uoroborate ([BMIM]BF4)] was explored for immobilization of hemoglobin (Hb) and construction of biosensor. Direct electrochemistry and electrocatalytic behaviors of Hb entrapped in the IL‐carrageenan composite ?lm on the surface of glassy carbon electrode (GCE) were investigated. UV‐vis spectroscopy demonstrated that Hb in the IL‐carrageenan composite ?lm could retain its native secondary structure. A pair of well‐de?ned redox peaks of Hb was obtained at the Hb‐IL‐carrageenan composite ?lm modi?ed electrode through direct electron transfer between the protein and the underlying electrode. The heterogeneous electron transfer rate constant (ks) was 2.02 s?1, indicating great facilitation of the electron transfer between Hb and IL‐carrageenan composite film modi?ed electrode. The modi?ed electrode showed excellent electrocatalytic activity toward reduction of hydrogen peroxide with a linear range of 5.0×10?6 to 1.5×10?4 mol/L and the detection limit was 2.12×10?7 mol/L (S/N=3). The apparent Michaelis‐Menten constant KMapp for hydrogen peroxide was estimated to be 0.02 mmol/L, indicating that the biosensor possessed high af?nity to hydrogen peroxide. A novel protocol for immobilization of horseradish peroxidase (HRP) onto diazonium functionalized screen‐printed gold electrode (SPGE) has been successfully developed. This protocol involved 1) electrochemical reduction of p‐nitrophenyl diazonium salts synthesized in situ in acidic aqueous solution to graft a layer of p‐nitrophenyl on SPGE, 2) electrochemical reduction of the nitro groups to convert to amines, 3) chemical reaction with nitrous acid to transform the amine to diazonium derivative and 4) chemical coupling of the enzyme with the diazonium group to form a covalent diazo bond. The fabricated biosensor showed the direct electrochemistry of HRP and displayed electrocatalytic activity towards the reduction of hydrogen peroxide (H2O2) without any mediator. The biosensor exhibited fast amperometric response to H2O2. The catalytic current increased with increasing H2O2 concentration from 5 μM to 30 μM and the detection limit of the biosensor was 2 μM. The biosensor exhibited acceptable sensitivity, good reproducibility and long‐term stability.  相似文献

Hemoglobin was entrapped in composite electrodeposited chitosan‐multiwall carbon nanotubes (MCNTs) film by assembling gold nanoparticles and hemoglobin step by step. In phosphate buffer solution (pH 7), a pair of well‐defined and quasireversible redox peaks appeared with formal potential at ?0.289 V and peak separation of 100 mV. The redox peaks respected for the direct electrochemistry of hemoglobin at the surface of chitosan‐MCNTs‐gold nanoparticles modified electrode. The parameters of experiments have also been optimized. The composite electrode showed excellent electrocatalysis to peroxide hydrogen and oxygen, the peak current was linearly proportional to H2O2 concentration in the range from 1×10?6 mol/L to 4.7×10?4 mol/L with a detection limit of 5.0×10?7 mol/L, and this biosensor exhibited high stability, good reproducibility and better selectivity. The biosensor showed a Michaelis–Menten kinetic response as H2O2 concentration is larger than 5.0×10?4 mol/L, the apparent Michaelis–Menten constant for hydrogen peroxide was calculated to be 1.61 μmol/L.  相似文献

MWCNTs‐nanoNiO composite was used as a glassy carbon electrode modifier for construction of a novel catalase nanobiosensor for hydrogen peroxide. The immobilized catalase exhibited excellent electrocatalytic activity towards the reduction of H2O2. The resulting amperometric biosensor exhibited a linear response over a concentration range of 200 µM to 2.53 mM with a low detection limit of 19.0 µM. Electrochemical impedance measurements revealed that the modified electrode can be used for the sensitive detection of H2O2. The charge transfer resistance found to decrease significantly after enzymatic reaction of nanobiosensor with H2O2. The resulting impedance was highly sensitive to H2O2 over a linear range of 19–170 nM with a detection limit of 2.4 nM.  相似文献

A new amperometric biosensor for hydrogen peroxide (H2O2) has been developed that is based on direct electrochemistry and electrocatalysis of hemoglobin (Hb) in a multilayer inorganic–organic hybrid film. o-Phenylenediamine (PDA) was electropolymerized onto a glassy carbon electrode (GCE), and then negatively charged nanogold particles and positively charged poly(diallyldimethylammonium chloride) (PDDA) were alternately assembled on the PDA/GCE surface. Finally, Hb was electrostatically adsorbed on the surface of gold nanoparticles. The electrochemical behavior of the resulting biosensor (Hb/{nanogold/PDDA}n/PDA/GCE) was assessed and optimized. The performance and factors influencing the biosensor were studied in detail. Under optimal conditions, the immobilized Hb displayed good electrocatalytic response to the H2O2 reduction ranging from 1.3 μM to 1.4 mM with a detection limit of 0.8 μM (at 3δ). In addition, the biosensor exhibited rapid response, good reproducibility, and long-term stability. In this work, a novel hydrogen peroxide biosensor derived from maize tassel (MT) and multiwalled carbon nanotube (MWCNT) composite was used to adsorb horseradish peroxidase (HRP) onto the surface of a glassy carbon electrode through electrostatic interactions. The morphology and structure of the products were characterized by SEM, FTIR and UV‐visible spectroscopy. The electrochemical and electrocatalytic performance of the HRP/MT‐MWCNT/GCE was studied using voltammetric and amperometric methods. The amperometric response of the biosensor varied linearly with concentration of H2O2 from 9 µM to 1 mM with detection limit of 4.0 µM (S/N=3). Furthermore, the biosensor exhibited good reproducibility and stability.  相似文献

A hemoglobin‐titanate composite based biosensor was chosen for determination of H2O2 in an acidic medium. CV results of the Hb‐titanate modified pyrolytic graphite electrode showed a pair of well‐defined, quasi‐reversible redox peaks centered at ?246 mV (vs. Ag/AgCl) in a pH 5.0 HAc‐NaAc buffer solution. The modified electrode exhibited good electrocatalytic response for monitoring H2O2 and had a large linear detection range from 20 μM to 3.2 mM with a detection limit of 8 μM (S/N=3) and a sensitivity of 29.7 mA M?1 cm?2 in the pH 5.0 solution. Horseradish peroxidase, previously modified with 1‐adamantane moieties, was supramolecularly immobilized on gold electrodes coated with perthiolated β‐cyclodextrin. The functionalized electrode was employed for the construction of an amperometric biosensor device for hydrogen peroxide using 1 mM hydroquinone as electrochemical mediator. The biosensor exhibited a fast amperometric response (6 s) and a good linear response toward H2O2 concentration between 12 μM and 450 μM. The biosensor showed a sensitivity of 1.02 mA/M cm2, and a very low detection limit of 5 μM. The electrode retained 97% of its initial electrocatalytic activity after 30 days of storage at 4 0C in 50 mM sodium phosphate buffer, pH 7.0.  相似文献

We report on a novel electrochemical biosensor that was fabricated by immobilizing hemoglobin (Hb) onto the surface of a gold electrode modified with a chitosan@Fe3O4 nano-composite. The Fe3O4 nanoparticles were prepared by co-precipitation and have an average size of 25 nm. They were dispersed in chitosan solution to obtain the chitosan@Fe3O4 nano-composite particles with an average diameter of 35 nm as verified by transmission electron microscopy. X-ray diffraction patterns and Fourier transform IR spectroscopy confirmed that the crystallite structure of the Fe3O4 particles in the nano-composite has remained unchanged. At pH 7.0, Hb gives a pair of redox peaks with a potential of about ?0.21 V and ?0.36 V. The Hb on the film maintained its biological activity and displays good electrocatalytic reduction activity towards hydrogen peroxide. The linear range for the determination of H2O2 is from 2.3 μM to 9.6 mM, with a detection limit at 1.1 μM concentration (at S/N?=?3). The apparent Michaelis-Menten constant is 3.7 mM and indicates the high affinity of Hb for H2O2. This biosensor also exhibits good reproducibility and long-term stability. Thus, it is expected to possess potential applications in the development of the third-generation electrochemical biosensors.

A nanobiocompatible composite containing hemoglobin (Hb), ZnO nanoparticles (nano‐ZnO) and ionic liquid 1‐butyl‐3‐methylimidazolium hexafluorophosphate (BMIMPF6) was fabricated and further modified on the glassy carbon electrode (GCE). The electrochemical behaviours of Hb in the composite film were carefully studied and a pair of quasi‐reversible redox peaks appeared in pH 7.0 phosphate buffer solution, which was attributed to the electrode reaction of Hb heme Fe(III)/Fe(II) redox couple. The presences of nano‐ZnO and BMIMPF6 in the film can retain the bioactivity of Hb and greatly enhance the direct electron transfer of Hb. The immobilized Hb showed high stability and good electrocatalytic ability to the reduction of hydrogen peroxide and O2.  相似文献

A hydrogen peroxide (H2O2) biosensor based on the combination of Au@Ag core‐shell nanoparticles with a hemoglobin‐chitosan‐1‐butyl‐3‐methyl‐imidazolium tetrafluoroborate (Hb‐CHIT‐BMIM×BF4) composite film was prepared. UV‐vis spectroscopy and transmission electron microscopy confirmed a core‐shell nanostructure of Au@Ag nanoparticle was successfully obtained. Cyclic voltammetric results showed a pair of well‐defined redox peaks appeared with the formal potential (EO′) of ‐0.301 V (versus Ag/AgCl reference electrode) and the peak‐to‐peak separation (ΔEp) was 84 mV in 0.1 M phosphate buffer solutions. Due to the synergetic effect of Au@Ag core‐shell nanoparticles and Hb‐CHIT‐BMIM×BF4, the biosensor exhibited good electrocatalytic activity to the reduction of H2O2 in a linear range from 1.0 × 10?6 to 1.0 × 10?3 M with a detection limit of 4 × 10?7 M (S/N = 3). The apparent Michaelis‐Menten constant (KM) was estimated to be 4.4 × 10?4 M, showing its high affinity. Thus, the study proved that the combination of Au@Ag core‐shell nanoparticles and Hb‐CHIT‐BMIM×BF4 is able to open up new opportunities for the design of enzymatic biosensors.  相似文献

A bioelectrochemical platform has been constructed for the direct electron transfer and biosensing purposes of microperoxidase‐11 (MP‐11) immobilized on the chitosan dispersed multilayer graphene nanocomposite. The immobilized MP‐11 at the modified gold electrode displays a well‐defined and quasireversible redox peaks, with a formal potential of ?0.38 V/SCE in a buffer solution (pH 7.0). MP‐11 absorbed on the electrode surface exhibits high electrocatalytic activity toward the reduction of both oxygen and hydrogen peroxide and also shows good analytical performance for the amperometric detection of H2O2 with a linear range from 2.5 to 135 μM. These results indicate the graphene modified electrode might be used as a third generation biosensor for H2O2 detection.  相似文献
