A series of novel isoxazole, dihydropyrazolone, and tetrahydropyridine derivatives were synthesized by the reaction of corresponding ethyl 1‐substituted aryl‐2‐methyl‐4‐oxo‐1,4,5,6‐tetrahydropyridine‐3‐carboxylates with different hydrazines and hydroxylamine. Reaction of tetrahydropyridone with N ,N‐dimethylformamide dimethyl acetal provided 1‐(5‐chloro‐2‐methylphenyl)‐2‐[2‐(dimethylamino)ethenyl]‐4‐oxo‐1,4,5,6‐tetrahydropyridine‐3‐carboxylate, which was cyclized into a bicyclic compound on treatment with ammonium acetate. The structures of all synthesized compounds were confirmed by IR, 1H NMR, and 13C NMR spectroscopy data. The structure of 5‐(5‐chloro‐2‐methylphenyl)‐4‐methyl‐2‐phenyl‐2,5,6,7‐tetrahydro‐3H‐pyrazolo[4,3‐c]pyridin‐3‐one was unambiguously assigned by means of X‐ray analysis data.  相似文献

The crystal structures of N‐aryl‐1,2,3,4‐tetrahydro‐3,3‐dimethyl‐2,4‐quinolinediones bearing methoxy‐ ( 1 ), methyl‐ ( 2 ), and chloro‐ ( 3 ) substituents in 2′‐position of the phenyl ring have been determined by X‐ray crystal structure analysis. The heterocyclic ring in 1–3 adopts an envelope conformation, with the smallest ring puckering in the ortho‐chloro derivative 3 . The N‐aryl ring is almost perpendicular with respect to the quinoline‐2,4‐dione ring. The corresponding dihedral angle values are 83.2(1)°, 80.0(9)°, and 83.4(2)° in 1, 2 and 3 , respectively. The hydrogen bond of C H⋅⋅⋅O type joins the molecules of the ortho‐methoxy derivative 1 into dimers. The supramolecular structure also contains two C H⋅⋅⋅π interactions that link the hydrogen‐bonded dimers into sheets. In ortho‐methyl derivative 2 , one C H⋅⋅⋅π interaction generates infinite chains, whereas two C H⋅⋅⋅O hydrogen bonds and three C H⋅⋅⋅π interactions in the ortho‐chloro derivative 3 form three‐dimensional framework. © 2008 Wiley Periodicals, Inc. A series of N‐aryl 2‐alkenamides were produced efficiently by treating N‐aryl 3‐(phenylsulfonyl)‐propanamides with potassium tert‐butoxide in THF at 0°C. With out isolation, it was further treated with an additional equivalent of potassium tert‐butoxide and allyl bromide to give N‐allyl N‐aryl 2‐alkenamides in one pot in good yields. Followed by a ring‐closing metathesis reaction, these N‐allyl N‐aryl 2‐alkenamides were respectively converted into corresponding N‐aryl α,β‐unsaturated γ‐lactams in moderate yields.  相似文献

The structures of N‐(2‐chlorophenyl)‐4‐hydroxy‐2‐methyl‐2H‐1,2‐benzothiazine‐3‐carboxamide 1,1‐dioxide and N‐(4‐chlorophenyl)‐4‐hydroxy‐2‐methyl‐2H‐1,2‐benzothiazine‐3‐carboxamide 1,1‐dioxide, both C16H13ClN2O4S, are stabilized by extensive intramolecular hydrogen bonds. The 4‐chloro derivative forms dimeric pairs of molecules lying about inversion centres as a result of intermolecular N—H...O hydrogen bonds, forming 14‐membered rings representing an R22(14) motif; the 2‐chloro derivative is devoid of any such intermolecular hydrogen bonds. The heterocyclic thiazine rings in both structures adopt half‐chair conformations.  相似文献

The title compound, N′‐benzylidene‐N‐[4‐(3‐methyl‐3‐phenyl‐cyclobutyl)‐thiazol‐2‐yl]‐chloro‐acetic acid hydrazide, has been synthesized and characterized by elemental analysis, IR, 1H and 13C NMR, and X‐ray single crystal diffraction. The compound crystallizes in the orthorhombic space group P 21 21 21 with a = 5.8671 (3) Å, b = 17.7182 (9) Å, and c = 20.6373 (8) Å. Moreover, the molecular geometry from X‐ray experiment, the molecular geometry, vibrational frequencies, and gauge‐including atomic orbital 1H and 13C chemical shift values of the title compound in the ground state have been calculated by using the Hartree–Fock and density functional methods (B3LYP) with 6‐31G(d) and 6‐31G(d,p) basis sets. The results of the optimized molecular structure are exhibited and compared with the experimental X‐ray diffraction. Besides, molecular electrostatic potential, Frontier molecular orbitals, and thermodynamic properties of the title compound were determined at B3LYP/6‐31G(d) levels of theory. © 2011 Wiley Periodicals, Inc. Possible approaches to synthesis of 5‐methyl‐4‐oxo‐2‐(coumarin‐3‐yl)‐N‐aryl‐3,4‐dihydrothieno[2,3‐d]pyrimidine‐6‐carboxamides 4 have been discussed. It is shown that the preferable approach is cyclization of 2‐iminocoumarin‐3‐carboxamides 1 , utilizing 5‐amino‐3‐methyl‐N2‐arylthiophene‐2,4‐dicarboxamides 2 as binucleophilic reagents. The proposed procedure allowed us to easily obtain 4 in two stages, using common reagents. © 2007 Wiley Periodicals, Inc. In a search for new insect growth regulators with unusual biological properties and different activity spectrum, we thought that the preservation of the bioactive unit and the introduction of 2‐methyl‐3‐(triphenylgermanyl)propoxycarbonyl in Ntert‐butyl‐N,N′‐dibenzoylhydrazine would enhance their larvicidal activities to a significant degree. Therefore, we designed and synthesized N′‐tert‐butyl‐N′‐[2‐methyl‐3‐(triphenylgermanyl)propoxycarbonyl]‐N‐benzoylhydrazine and analogs by two procedures. These novel compounds were characterized by elemental analyses, IR, and 1H NMR. At the same time, Ntert‐butyl‐N‐substitutedbenzoylhydrazines were prepared by a new method, and some reactions involved were studied. The preliminary results indicate that some compounds have inhibitory effects against plant pathogenetic bacteria such as early blight of tomato. Copyright © 2002 John Wiley & Sons, Ltd.  相似文献

N‐acetyl‐4‐nitrotryptophan methyl ester (2), N‐acetyl‐5‐nitrotryptophan methyl ester (3), N‐acetyl‐6‐nitrotryptophan methyl ester (4) and N‐acetyl‐7‐nitrotryptophan methyl ester (5) were synthesized through a modified malonic ester reaction of the appropriate nitrogramine analogs followed by methylation with BF3‐methanol. Assignments of the 1H and 13C NMR chemical shifts were made using a combination of 1H–1H COSY, 1H–13C HETCOR and 1H–13C selective INEPT experiments. Copyright © 2008 Crown in the right of Canada. A one‐step method was reported for the synthesis of 6‐acetamido‐3‐(N‐(2‐(dimethylamino) ethyl) sulfamoyl) naphthalene‐1‐yl 7‐acetamido‐4‐hydroxynaphthalene‐2‐sulfonate by treating 7‐acetamido‐4‐hydroxy‐2‐naphthalenesulfonyl chloride with equal moles of N, N‐dimethylethylenediamine in acetonitrile in the presence of K2CO3. The chemical structure of the obtained compounds was characterized by MS, FTIR, 1H NMR, 13C NMR, gCOSY, TOCSY, gHSQC, and gHMBC. An exceedingly and highly efficient procedure has been described for the synthesis of substituted N‐3‐diaryl‐1,8‐naphthyridin‐2‐amines by the reaction of 2‐chloro‐3‐aryl‐1,8‐naphthyridines with various anilines in the presence of N‐methyl‐2‐pyrrolidone and K2CO3 under thermal green solvent‐free conditions. The significant features of this green reaction include very good yields in purity, simple experimental, short reaction time, easy workability, and avoidance of toxic solvents. All synthesized compounds have been evaluated for their antibacterial activity.  相似文献

A mild and efficient synthesis of N‐substituted‐3‐aryl‐3‐(4‐hydroxy‐6‐methyl‐2‐oxo‐2H‐pyran‐3‐yl)propanamides via four‐component reaction of an aldehyde, amine, Meldrum's acid, and 4‐hydroxy‐6‐methyl‐2H‐pyran‐2‐one in the presence of benzyltriethylammonium chloride (TEBAC) in aqueous medium is described. The Hantzsch synthesis of novel aryl imidazo[1,2‐b]isoxazolyl‐N‐aryl thiazol amines 5 analogues were described. Reaction of 3‐aminoisoxazole 1 with substituted phenacyl bromides 2 in dry ethanol afforded the corresponding 6‐methyl‐3‐arylimidazo[1,2‐b]isoxazoles 3 in good yields. Compounds 3 on reaction with chloroacetyl chloride in 1,4‐dioxane furnished the corresponding 2‐chloro‐1‐(6‐methyl‐3‐arylimidazo[1,2‐b]isoxazol‐2‐yl)ethanones 4 . Compounds 4 on heating with N‐aryl thioureas in an oil bath underwent cyclization to afford the title compounds viz., imidazo[1,2‐b]isoxazolyl‐N‐aryl thiazol amines 5 in moderate to good yields by Hantzsch synthesis.  相似文献

A convenient diastereoselective synthesis of diisopropyl (2R,3R)‐3‐{{{(R/S)‐aryl[(diethoxyphosphinyl)amino]methyl}hydroxyphosphinyl}oxy}‐2‐hydroxybutanedioate through Mannich‐type reactions is reported. The reactions take place under mild conditions in good yields, and this makes it possible to introduce various substituents at the α‐position to the P‐atom of α‐aminophosphonates. The chiral diisopropyl (4R,5R)‐2‐chloro‐1,3,2‐dioxaphospholane‐4,5‐dicarboxylate ( 3 ) was found to be a good phosphonylating agent in this stereoselective reaction.  相似文献

A study on the synthesis of the novel N‐(cyclic phosphonate)‐substituted phosphoramidothioates, i.e., O,O‐diethyl N‐[(trans‐4‐aryl‐5,5‐dimethyl‐2‐oxido‐2λ5‐1,3,2‐dioxaphosphorinan‐2‐yl)methyl]phosphoramidothioates 4a – l , from O,O‐diethyl phosphoramidothioate ( 1 ), a benzaldehyde or ketone 2 , and a 1,3,2‐dioxaphosphorinane 2‐oxide 3 was carried out (Scheme 1 and Table 1). Some of their stereoisomers were isolated, and their structure was established. The presence of acetyl chloride was essential for this reaction and accelerated the process of intramolecular dehydration of intermediate 5 forming the corresponding Schiff base 7 (Scheme 2).  相似文献

The synthesis of a new series of 4‐aryl‐3‐chloro‐2‐oxo‐N‐[3‐(10H‐phenothiazin‐10‐yl)propyl]azetidine‐1‐carboxamides, 4a – 4m , is described. Phenothiazine on reaction with Cl(CH2)3Br at room temperature gave 10‐(3‐chloropropyl)‐10H‐phenothiazine ( 1 ), and the latter reacted with urea to yield 1‐[3‐(10H‐phenothiazin‐10‐yl)propyl]urea ( 2 ). Further reaction of 2 with several substituted aromatic aldehydes led to N‐(arylmethylidene)‐N′‐[3‐(phenothiazin‐10‐yl)propyl]ureas 3a – 3m , which, on treatment with ClCH2COCl in the presence of Et3N, furnished the desired racemic trans‐2‐oxoazetidin‐1‐carboxamide derivatives 4a – 4m . The structures of all new compounds were confirmed by IR, and 1H‐ and 13C‐NMR spectroscopy, FAB mass spectrometry, and chemical methods.  相似文献

Synthesis ofN1‐3‐{(4‐substitute daryl‐3‐chloro‐2‐oxo‐azetidine)‐iminocarbamyl}‐propyl‐6‐nitroindazole 4a – 4s was conducted by a conventional method. All the compounds were synthesized and characterized by IR, 1H NMR, 13C NMR, FAB‐Mass techniques and chemical methods. All the final synthesized compounds were evaluated for their antimicrobial activity and antitubercular activity with MIC values against some selected microorganisms.  相似文献

Acylation of 2‐amino‐N‐methyl‐thiobenzamide with substituted benzoyl chlorides has been used to synthesize the corresponding 2‐benzoylamino‐N‐methylthiobenzamides. Subsequent sodium methoxide‐catalyzed ring closure gives the corresponding 3‐methyl‐2‐phenylquinazoline‐4‐thiones. These compounds were characterized by means of their 1H‐ and 12C‐NMR spectra. The kinetics of the cyclization reaction has been followed with UV‐VIS spectroscopy at 100 °C in methanolic solutions of sodium methoxide.  相似文献
