Various [5,6]pyrano[2,3‐c]pyrazol‐4(1H)‐thiones were synthesized in high yields by treatment of the corresponding [5,6]pyrano[2,3‐c]pyrazol‐4(1H)‐ones with Lawesson's reagent. Detailed NMR spectroscopic studies were undertaken of the title compounds. Complete and unambiguous assignment of chemical shifts (1H, 13C, 15N) and coupling constants (1H,1H; 13C,1H) was achieved by the combined application of various one‐ and two‐dimensional (1D and 2D) NMR spectroscopic techniques. Unequivocal mapping of most 13C,1H spin coupling constants is accomplished by 2D (δ, J) long‐range INEPT spectra with selective excitation. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

15N NMR data for a series of 12 para‐substituted benzamidoximes and benzamidinium salts were determined in dimethyl sulfoxide. For the amino group of benzamidoximes 1J(N,H) coupling constants were determined using polarization transfer techniques; the other 15N atoms were not detectable owing to fast exchange processes and, thus, standard proton noise decoupled spectra had to be measured. The 15N NMR chemical shifts of the oxime‐type nitrogen atom and the benzamidinium amino group (with two exceptions) correlate with Hammett σ° values (r2>0.95). 15N NMR shift data are a suitable and sensitive means for characterizing far‐ranging electronic substituent effects in these functional groups. Additionally, 13C NMR data in dimethyl sulfoxide solution are given. All spectroscopic data will be used for investigations into the mechanisms of the enzymes involved in the metabolic cycle of oxidation and reduction of benzamidines and benzamidoximes. Bioactive cage‐like polycyclic compounds have attracted the attention of several research groups because of their unique appearance and their biological activities. Their structures were established on the basis of 1H NMR and 13C NMR spectroscopic data. The 1H and 13C signal assignments and most homonuclear hydrogen coupling constants were assigned by use of techniques such as 1D 1H and 13C NMR and 2D gCOSY, non‐edited gHSQC and gHMBC. Two novel oligosaccharides, mono‐ and difructosyllactosucrose {[O‐β‐D ‐fructofuranosyl‐(2 → 1)]n‐β‐D ‐fructofuranosyl‐O‐[β‐D ‐galactopyranosyl‐(1 → 4)]‐α‐D ‐glucopyranoside, n = 1 and 2} were synthesized using 1F‐fructosyltransferase purified form roots of asparagus (Asparagus officinalis L.). Their 1H and 13C NMR spectra were assigned using several NMR techniques. The spectral analysis was started from two anomeric methines of aldose units, galactose and glucose, since they showed separate characteristic signals in their 1H and 13C NMR spectra. After assignments of all the 1H and 13C signals of two units of aldose, they were discriminated as galactose and glucose using proton–proton coupling constants. The HMBC spectrum revealed the galactose residue attached to C‐4 of glucose, fructose residue attached to the C‐1 of glucose, and further fructosyl fructose linkage extended from the glucosyl fructose residues. The first example in the literature of a compound showing anisochronous 15N atoms resulting from diastereotopicity is described. Racemic 1,3‐dimethyl‐2‐phenyloctahydro‐1H‐benzimidazole was prepared and studied by 1H, 13C and 15N NMR spectroscopy. If convenient conditions were used (monitored by theoretical calculations of 2JN‐H spin–spin coupling constants), two 15N NMR signals were observed and corresponded to the diastereotopic atoms. GIAO/density‐functional calculations of chemical shifts were not only in good agreement with the experimental values but also served as prediction tools. This study suggests that 15N NMR spectroscopy could be used to probe chirality.  相似文献

15N NMR chemical shifts of 2‐aryl‐1,3,4‐oxadiazoles were assigned on the basis of the 1H–15N HMBC experiment. Chemical shifts of the nitrogen and carbon atoms in the oxadiazole ring correlate with the Hammett σ‐constants of substituents in the aryl ring (r2 ≥ 0.966 for N atoms). 15N NMR data are a suitable and sensitive means for characterizing long‐range electronic substituent effects. Complete assignment of 1H and 13C NMR chemical shifts and J(1H/1H and 1H/19F) coupling constants for 22 1‐phenyl‐1H‐pyrazoles' derivates were performed using the concerted application of 1H 1D and 1H, 13C 2D gs‐HSQC and gs‐HMBC experiments. All 1‐phenyl‐1H‐pyrazoles' derivatives were synthesized as described by Finar and co‐workers. The formylated 1‐phenyl‐1H‐pyrazoles' derivatives were performed under Duff's conditions. In this paper, a complete 1H and 13C NMR data assignment of ent-polyalthic acid, a biologically active labdane-type diterpene, is presented. The assignments were carried on the basis of spectroscopic data from 1H NMR, 13C{1H} NMR, gCOSY, gHMQC, and gHMBC experiments. Furthermore, a software-assisted methodology, using FOMSC3_rm_NB and NMR_MultSim programs, supported the detailed and unequivocal assignment of 1H and 13C signals, allowing all hydrogen coupling constants to be determined and thus clarifying all hydrogen signal multiplicities.  相似文献

1H, 13C, 15N and 195Pt NMR studies of gold(III) and platinum(II) chloride organometallics with N(1),C(2′)‐chelated, deprotonated 2‐phenylpyridine (2ppy*) of the formulae [Au(2ppy*)Cl2], trans(N,N)‐[Pt(2ppy*)(2ppy)Cl] and trans(S,N)‐[Pt(2ppy*)(DMSO‐d6)Cl] (formed in situ upon dissolving [Pt(2ppy*)(µ‐Cl)]2 in DMSO‐d6) were performed. All signals were unambiguously assigned by HMBC/HSQC methods and the respective 1H, 13C and 15N coordination shifts (i.e. differences between chemical shifts of the same atom in the complex and ligand molecules: Δ1Hcoord = δ1Hcomplex ? δ1Hligand, Δ13Ccoord = δ13Ccomplex ? δ13Cligand, Δ15Ncoord = δ15Ncomplex ? δ15Nligand), as well as 195Pt chemical shifts and 1H‐195Pt coupling constants discussed in relation to the known molecular structures. Characteristic deshielding of nitrogen‐adjacent H(6) protons and metallated C(2′) atoms as well as significant shielding of coordinated N(1) nitrogens is discussed in respect to a large set of literature NMR data available for related cyclometallated compounds. In the 13C NMR spectra of methylglyoxal bisdimethylhydrazone, the 13C‐5 signal is shifted to higher frequencies, while the 13C‐6 signal is shifted to lower frequencies on going from the EE to ZE isomer following the trend found previously. Surprisingly, the 1H‐6 chemical shift and 1J(C‐6,H‐6) coupling constant are noticeably larger in the ZE isomer than in the EE isomer, although the configuration around the –CH═N– bond does not change. This paradox can be rationalized by the C–H?N intramolecular hydrogen bond in the ZE isomer, which is found from the quantum‐chemical calculations including Bader's quantum theory of atoms in molecules analysis. This hydrogen bond results in the increase of δ(1H‐6) and 1J(C‐6,H‐6) parameters. The effect of the C–H?N hydrogen bond on the 1H shielding and one‐bond 13C–1H coupling complicates the configurational assignment of the considered compound because of these spectral parameters. The 1H, 13C and 15N chemical shifts of the 2‐ and 8‐(CH3)2N groups attached to the –C(CH3)═N– and –CH═N– moieties, respectively, reveal pronounced difference. The ab initio calculations show that the 8‐(CH3)2N group conjugate effectively with the π‐framework, and the 2‐(CH3)2N group twisted out from the plane of the backbone and loses conjugation. As a result, the degree of charge transfer from the N‐2– and N‐8– nitrogen lone pairs to the π‐framework varies, which affects the 1H, 13C and 15N shieldings. Structure elucidation of compounds in the benzisoxazole series ( 1 – 6 ) and naphtho[1,2‐d][1,3]‐ ( 7 – 10 ) and phenanthro[9,10‐d][1,3]oxazole ( 11 – 14 ) series was accomplished using extensive 2D NMR spectroscopic studies including 1H–1H COSY, long‐ range 1H–1H COSY, 1H–13C COSY, gHMQC, gHMBC and gHMQC‐TOCSY experiments. The distinction between oxazole and isoxazole rings was made on the basis of the magnitude of heteronuclear one‐bond 1JC2, H2 (or 1JC3, H3) coupling constants. Complete analysis of the 1H NMR spectra of 11 – 14 was achieved by iterative calculations. Gradient selected gHMQC‐TOCSY spectra of phenanthro[9,10‐d][1,3]oxazoles 11 – 14 were obtained at different mixing times (12, 24, 36, 48 and 80 ms) to identify the spin system where the protons of phenanthrene ring at H‐5, H‐6 and at H‐9 and H‐7 and H‐8 were highly overlapping. According to the 1H, 13C and 15N NMR spectroscopic data and DFT calculations, the E‐isomer of 1‐vinylpyrrole‐2‐carbaldehyde adopts preferable conformation with the anti‐orientation of the vinyl group relative to the carbaldehyde oxime group and with the syn‐arrangement of the carbaldehyde oxime group with reference to the pyrrole ring. This conformation is stabilized by the C? H···N intramolecular hydrogen bond between the α‐hydrogen of the vinyl group and the oxime group nitrogen, which causes a pronounced high‐frequency shift of the α‐hydrogen signal in 1H NMR (~0.5 ppm) and an increase in the corresponding one‐bond 13C–1H coupling constant (ca 4 Hz). In the Z‐isomer, the carbaldehyde oxime group turns to the anti‐position with respect to the pyrrole ring. The C? H···O intramolecular hydrogen bond between the H‐3 hydrogen of the pyrrole ring and the oxime group oxygen is realized in this case. Due to such hydrogen bonding, the H‐3 hydrogen resonance is shifted to a higher frequency by about 1 ppm and the one‐bond 13C–1H coupling constant for this proton increases by ~5 Hz. An NMR study of a new stereoisomer of isoreserpiline pseudoindoxyl alkaloid is described. The complete 1H and 13C NMR assignments of the compound were carried out using COSY, HMQC and HMBC. Stereochemistry at carbons C‐2, C‐3, C‐15, C‐20 and C‐19 was established using values of 1H chemical shifts, coupling constants and NOESY experiments. 1H, 13C and two‐dimensional NMR analyses were applied to determine the NMR parameters of 6‐(2′,3′‐dihydro‐1′H‐inden‐1′‐yl)‐1H‐indene. The measurements were accomplished with 0.5 mg of the substance, this quantity being sufficient to determine the chemical shifts of all the H and C atoms, and also the appropriate coupling constants and to give the complete NMR resonance assignments of the molecule. The predicted patterns of the four different H atoms of the methylene groups of the indane structural element coincided completely with the complex patterns in the NMR spectra. Determination of the level and absolute configuration of 2‐hydroxyglutaric acid in a patient's urine is a method of diagnosing two metabolic diseases. Such a determination can be done with the aid of NMR spectroscopic methods. In this paper the careful interpretation of 1H and 13C NMR spectra of this metabolite and its lactone measured under conditions used in biomedical assays is reported. Understanding the complex thermodynamic behavior of confined amphiphilic molecules in biological or mesoporous hosts requires detailed knowledge of the stacking structures. Here, we present detailed solid‐state NMR spectroscopic investigations on 1‐butanol molecules confined in the hydrophilic mesoporous SBA‐15 host. A range of NMR spectroscopic measurements comprising of 1H spin–lattice (T1), spin–spin (T2) relaxation, 13C cross‐polarization (CP), and 1H,1H two‐dimensional nuclear Overhauser enhancement spectroscopy (1H,1H 2D NOESY) with the magic angle spinning (MAS) technique as well as static wide‐line 2H NMR spectra have been used to investigate the dynamics and to observe the stacking structure of confined 1‐butanol in SBA‐15. The results suggest that not only the molecular reorientation but also the exchange motions of confined molecules of 1‐butanol are extremely restricted in the confined space of the SBA‐15 pores. The dynamics of the confined molecules of 1‐butanol imply that the 1H,1H 2D NOESY should be an appropriate technique to observe the stacking structure of confined amphiphilc molecules. This study is the first to observe that a significant part of confined 1‐butanol molecules are orientated as tilted bilayered structures on the surface of the host SBA‐15 pores in a time‐average state by solid‐state NMR spectroscopy with the 1H,1H 2D NOESY technique.  相似文献
