以靛红酸酐(2)为起始原料,经氮上烷基化、开环、关环以及酯的氨解生成异羟肟酸等多步简单反应合成了目标化合物选择性HDAC6抑制剂4-((2,4-二氧基-3-苯乙基-3,4-二氢喹唑啉-1(2H)-基)-甲基)-N-羟基苯甲酰胺(1)。产物结构经1H NMR、 13C NMR 以及MS 确证。新的合成路线具有反应条件温和、后处理简单等优点,总收率83.8%。   相似文献

A convenient approach for the preparation of (2S,3R,4R,5S,6R)-2-(3-(4-ethylbenzyl)-4-chlorophenyl)-6-(hydroxymethyl)- tetrahydro-2H-pyran-3,4,5-triol I is developed. The target compound via four steps is synthesized from 4-bromo-2-(bromomethyl)- 1-chlorobenzene and the isomers of undesired ortho-products were avoided during the preparation.  相似文献

设计合成了具有降血糖活性的3-甲基-1-苯基4-[4-[[5-甲基-2-(4-取代芳基)-嗯唑4-基]甲氧基]-芳亚甲(苄)基]}-2-吡唑啉-5-酮类化合物.用丁二酮单肟和(取代的)苯甲醛环合、氯化得到氯甲基嗯唑衍生物,与对羟基苯甲醛或香兰醛缩合,再与3-甲基-1-苯基-2-吡唑啉-5-酮进行Knoevenagel反应及催化氢化得到目标化合物(Ⅰ和Ⅱ),共合成了16个未见文献报道的新化合物,并利用元素分析、IR、MS和^1H NMR确证了化合物的结构.初步药理试验结果表明,所合成的化合物有抑制血糖升高的倾向以及能明显加强和延长外源性胰岛素的降血糖作用,其中化合物Ib,Id和If尤为突出,说明这类化合物可能有增强胰岛素敏感性的作用.  相似文献

The title compound 2-(4-fluoro-2-(4-fluorophenoxy)phenyl)-1-(1H-1,2,4-triazol- 1-ylmethyl)-3-methoxy-isopropy alcohol has been synthesized by the treatment of 1-[2-(4-fluoro- 2-(4-fluorophenoxy)phenyl)-2,3-epoxypropyl]-1H-1,2,4-triazole with sodium methoxide. It belongs to orthorhombic, space group P212121, with a = 9.7229(19), b = 11.516(2), c = 16.047(3)A, C18H17F2 N3O3, Mr = 361.35, V = 1796.7(6)A^3, Z = 4, Dc = 1.3359 g/cm^3, F(000) = 752, p = 0.106 mm^-1, the final R = 0.0329 and wR = 0.0803 for 1821 unique reflections. The dihedral angles made by the triazole ring with two benzene rings are 43.56(3) and 54.78(2)°, respectively. The intermolecular hydrogen bond in the crystal lattice plays an important role in stabilizing the structure.  相似文献

The title compound 1-((5-(4-(4-chlorophenoxy)-2-chlorophenyl)-2,2,3-trimethyl-oxazolidin- 5-yl)methyl)-lH-1,2,4-triazole (C21H22Cl2N4O2)has been synthesized and characterized by elemental analysis, IR, 1H NMR, MS and single-crystal X-ray diffraction. The crystal belongs to the triclinic system, space group Pi with a = 6.891(2), b = 9.074(2), c = 18.258(4)AA, α = 99.292(4), β = 95.105(4), γ = 108.068(3)°, C21H22Cl2N4O2, Mr = 433.33, V= 1059.3(4) Aa, Z = 2, Dc = 1.359 g/cm3, F(000) = 452,μ = 0.331 mm-1, the final R = 0.0448 and wR = 0.0994 for 3727 unique reflections. The dihedral angle between the oxazolidine ring taking an envelope conformation with a local pseudo-mirror and the triazole ring is 27.7(9)°. Weak intermolecular C-H...N hydrogen bonds and π-π interactions exist between the triazole rings of neighboring molecules, forming a three-dimensional network, which stabilizes the crystal structure. The primary biological test shows the target compound has certain fungicidal activity.  相似文献

The crystal belongs to triclinic, space group P1, with a = 8.140(3), b = 11.966(4), c = 13.771(5)Aα= 67.366(4), β= 85.165(5), γ= 75.806(4)°, V = 1200.1(7) A3, Z = 2, C26H3008, Mr = 470.50, Dc = 1.302 g/cm^3, F(000) = 500,λ(MoKa) = 0.71073 A, μ= 0.096 mm^-1, R = 0.0659 and wR = 0.1841 for 3080 observed reflections (I 〉 2σ(I)). As a key intermediate of HIV-1 integrase inhibitor, the synthesis and structure confirmation of the title compound are important for further studies.  相似文献

The title compound 3,3'-(2,2'-(ethane-1,2-diylbis(oxy))bis(ethane-2,1-diyl))bis(2- (4-chlorophenylamino)quinazolin-4(3H)-one) 3 (C34H30CI2N604, Mr = 657.54) has been synthesized and its crystal structure was determined by single-crystal X-ray diffraction analysis. It crystallizes in space group P1 with a = 9.9185(8), b = 10.6124(9), c = 15.4064(13) A, α = 92.896(2),β = 103.813(2), γ= 94.635(2)°, V= 1565.5(2) A3, Z = 2, Dc = 1.395 g/cm^3,μ = 0.257 mm^-1, F(000) = 684, the final R = 0.0580 and wR = 0.1284 for 3400 observed reflections (I〉 2σ(I). It remains triclinic in the crystal. Intra- and intermolecular N(3)-H(3A)…O(2), N(6)-H(6A)…O(3) and C(19)-H(19A)…O(4) #1 (#1: 1-x, 1-y, -z) hydrogen bonds are observed in the title compound. A two-dimensional chain network is formed from the dimmers and intermolecular π-π interactions in the crystal structure. This open network has solvent-accessible voids (10A3).  相似文献

以邻羟基萘甲醛和乙酰乙酸乙酯为原料,六氢吡啶为催化剂,通过Knoevenagel反应,利用研磨法制得3-乙酰基-5,6-苯并香豆素(A);A与4-甲氧基苯甲醛(B)和醋酸铵经一锅法合成新化合物4-(4-甲氧基苯基)-2,6-双(5,6-苯并香豆素-3-基)吡啶(C),其结构经1H NMR, IR和元素分析表征。对反应条件进行了优化,并采用MTT法研究了化合物C对人体宫颈癌HeLa 229肿瘤细胞株的抑制作用。结果表明:当C浓度为80 μmol·L-1,作用时间为48 h时,抑制率为66.65%, IC50为45.75 μmol·L-1,与依托泊苷(IC50=42 μmol·L-1)相当。  相似文献

The title compound (Z)-1-(3-fluorobenzyl)-5-((3-fluorobenzylimino)(2-hydroxy-4- methoxyphenyl)methyl)pyridin-2(1H)-one (C27H22F2N2O3,Mr = 460.47) was synthesized and crystallized. The crystal belongs to the triclinic system,space group P1 with a = 9.951(6),b = 10.059(6),c = 12.927(7)A,α = 109.828(7),β = 102.304(7),γ = 104.898(7)°,Z = 2,V = 1110.2(11)A^3,Dc =1.377 Mg/m^3,μ(MoKα) = 0.102 mm^-1,F(000) = 480,the final R = 0.0449 and Rw = 0.1250 for 4595 observed reflections (I 〉 2σ(I)). X-ray analysis reveals that both m-fluorobenzyl groups are diorientationally disordered because of the rotation of C–C single bond which was represented as F(1),F(2) and F(1'),F(2'). The diorientational disorder was refined and gave the occupancies of 0.665(4) and 0.335(4) for F(1) and F(1'),0.631(3) and 0.369(3) for F(2) and F(2'). Hydrogen bonds existing in the cell link different components to stabilize the crystal structure. The active data proved that the title compound has good anti-HBV activity.  相似文献

A novel hydrochloride quaternary ammonium salt (E)-4-(benzyloxy)-2-(cinnamo- yloxy)-N,N,N-trimethyl-4-oxobutan-l-aminium chloride (Cz3HzgNO4Cl2, Mr = 454.37) has been synthesized via the sequence of acetylation and esterification by using L-carnitine (L-4-N-trimethy- lammonium-3-hydroxybutyric acid, LC) and cinnamic acid as the starting materials, and its crystal structure was determined by single-crystal X-ray diffraction method. The crystal belongs to monoclinic, space group P212121 with a = 10.1670(4), b = 10.4488(4), c = 22.9795(11)A, V = 2441.18(18)A3, Z = 4, Dc = 1.236 g/cm3,μ(MoKa) = 0.293 mm-1, F(000) = 960, Flack factor = -0.01(11), the final R = 0.0489 and wR = 0.1550 for 3350 observed reflections (I 〉 2σ(/)) and R = 0.0953 for all 5648 unique reflections. The crystal structure involves a conjugated system which shows a reverse olefin structure.  相似文献

The title compound 3-methyl-l-(4-methylphenyl)-4-(N-4-trifluoromethylphenyl) aminomethyl-5-(4-methoxyphenylthio)-lH-pyrazole has been synthesized via a four-step reaction and characterized by IR, 1H NMR, elemental analysis and X-ray crystallography. The compound crystallizes in monoclinic, space group P21/c with a = 8.7170(15), b = 18.355(3), c = 15.292(3) A, fl = 103.445(3)°, V= 2379.7(7) A3, Dc = 1.350 g/cm3, Z = 4, p = 0.184, F(000) = 1008, and the final R = 0.0491 and wR = 0.1339 for 4160 observed reflections (I 〉 2a(/)). The results demonstrate that there is a face-to-face π-π stacking interaction between one benzene ring (C(19)-C(24)) and another (C(I 3)-C(! 8)) at a plane-plane distance of 3.3539 A. The ring normal and vector between the ring centroids form an angle of 18.2° up to the centroid-to-centroid distance of 3.5273 A. The crystal structure is stabilized by the intermolecular hydrogen bond of N(3)-H(3A)...N(2) (symmetry code: A -x+l, -y+l, -z). The preliminary biological test shows that the title compound has a moderate antifungal activity.  相似文献

报道了1-(4-硝基苯基)-3-(2-苯并咪唑)-三氮烯(NPBMT)的合成及其与铜(Ⅱ)的显色反应。在非离子表面活性剂OP存在下,于pH11.5的Na2B4O7-NaOH缓冲介质中,NPBMT与铜(Ⅱ)形成2:1的桔红色络合物,络合物的最大吸收峰为417nm,表观摩尔吸光系数为2.62×10^4 L·mol^-1·cm^-1,10mL溶液中,铜(Ⅱ)量在0—15μg/mL范围内符合比尔定律。该法用于人发和食品中微量铜的测定,测定结果与AAS法相符。  相似文献

以DMF作溶剂,在四丁基氟化铵 (TBAF)存在下,对甲基苯基三甲硅基醚(1)和4,4'-二氯二苯砜(2)于100 ℃反应1 h,合成了4,4'-二(4-甲基苯氧基)二苯砜(3),产率为92%;加入催化量的N-溴代丁二酰亚胺(NBS)并在光照条件下,与氧气反应得到中间体-4,4'-二(4-羧基苯氧基)二苯砜(4),其产率达90%;将化合物4与二氯亚砜反应合成目标产物4,4'-二(4-氯甲酰基苯氧基)二苯砜(5),总收率为74.5%(以对甲基苯基三甲硅基醚为基准计算).  相似文献

Compound 1 (C18H18N2O4S) belongs to orthorhombic, space group P212121 with a = 9.6348(14), b = 11.1517(17), c = 16.412(3) A, V = 1763.4(5) A^3, Mr = 358.40, Z = 4, De = 1.350 g/cm^3,/t = 0.209 mm^-1, F(000) = 752, R = 0.0348 and wR = 0.0714. Compound 2 (CI8H17ClN2O4S) crystallizes in orthorhombic, space group P212121 with a = 9.3128(14), b = 10.9655(16), c = 17.783(3) A, V = 1815.9(5) A^3, Mr = 392.85, Z = 4, De = 1.437 g/cm^3, p = 0.352 mm^-1, F(000) = 816, R = 0.0389 and wR = 0.0845. The absolute structure Flack parameters X of compounds 1 and 2 are -0.03(8) and -0.06(7), respectively. X-ray analysis reveals that the crystal structures of these two compounds both involve two intermolecular N-H…O hydrogen bond's.  相似文献

以2-氨基-6-甲基苯甲酸为起始原料,通过合环反应生成两种4-氧代-3(4H)-喹唑啉-5-羧酸衍生物(1a, 1b), 1a, 1b分别经高锰酸钾氧化合成了4-氧代-3(4H)-喹唑啉-5-羧酸(2a, 2b),其结构经1H NMR, 13C NMR和MS表征。研究了投料比{r[n(高锰酸钾) :n(5-甲基-3(4H)-喹唑啉-4-酮)]}和反应温度等对收率的影响。结果表明:在最佳反应条件(r=7:1,中性高锰酸钾氧化,于90 ℃反应)下合成2总收率可达66%。  相似文献

采用乙醚蒸汽扩散法,由四氟合硼酸四乙腈合铜(I)、4,4’-联吡啶、二(2-二苯基膦基)苯基醚或者三苯基膦的反应后的二氯甲烷和乙腈混合溶液中,晶化[Cu2(4,4'-bipy)(POP)2(CH3CN)2(C2H5O)2](BF4):(1)和[Cu2(4,4'-bipy)(PPh3)4(CH3CN)2](BF4)2(2)两种配合物。对它们进行了元素分析和X射线衍射单晶结构表征,同时测定了UV—Vis光谱及相应的激发态寿命。配合物1和2展示了较好的光物理特性,在275nm紫外光的激发下,固体粉末样品的最大发射峰位分别位于527和483nm,这归因于金属干扰的配体内部跃迁。  相似文献
