本文比较了超级电容器、锂离子电池和超级电容电池的结构、原理、研究现状和发展前景。超级电容电池的正极具有超级电容器电极的结构和双电层储能机制,负极具有类似锂离子电池负极的结构和快速电化学储能机制。超级电容器和锂离子电池的发展空间都很有限,而作为两者结合的产物的超级电容电池可兼具高比功率、高比能量、高放电电压和长循环寿命的优点,是未来储能电池的发展方向之一,但还面临缺乏具有高分解电压的电解液和高充电电压下电解液中离子枯竭的问题。  相似文献

Dendritic cell vaccine (DCV) holds great potential in tumor immunotherapy owing to its potent ability in eliciting tumor-specific immune responses. Aiming at engineering enhanced DCV, we report the first effort to construct a glycopolymer-engineered DC vaccine (G-DCV) via metabolicglycoengineering and copper-free click-chemistry. Model G-DCV was prepared by firstly delivering tumor antigens, ovalbumin (OVA) into dendritic cells (DC) with fluoroalkane-grafted polyethyleneimines, followed by conjugating glycopolymers with a terminal group of dibenzocyclooctyne (DBCO) onto dendritic cells. Compared to unmodified DCV, our G-DCV could induce stronger T cell activation due to the enhanced adhesion between DCs and T cells. Notably, such G-DCV could more effectively inhibit the growth of the mouse B16-OVA (expressing OVA antigen) tumor model after adoptive transfer. Moreover, by combination with an immune checkpoint inhibitor, G-DCV showed further increased anti-tumor effects in treating different tumor models. Thus, our work provides a novel strategy to enhance the therapeutic effectiveness of DC vaccines.  相似文献

The authentication of mammalian cell cultures and their subpopulations is of great demand in biotechnology and cell therapy. However, current techniques are either not efficient or can be very complex and expensive. Here we report a simple and straightforward approach for authentication of biological cells and their subpopulations with high speed, high throughput, low sample cost, and high sensitivity. We discovered that cell cultures treated with protease under soft, ‘non‐killing’ conditions release fragments of cell surface proteins, whose composition is a strong characteristic of the cells. Mass spectrometric analysis of the released fragments allows a direct comparison of the produced mass spectrum with the mass spectrum of known cells. As an example, we applied this technique to verify subpopulations of human fibroblasts with different origins and which exhibit different medical characteristics. Integrating 2D culture of adherent mammalian cells with single‐cell western blotting (in situ scWB) uses microfluidic design to eliminate the requirement for trypsin release of cells to suspension, prior to single‐cell isolation and protein analysis. To assay HeLa cells from an attached starting state, we culture adherent cells in fibronectin‐functionalized microwells formed in a thin layer of polyacrylamide gel. To integrate the culture, lysis, and assay workflow, we introduce a one‐step copolymerization process that creates protein‐decorated microwells. After single‐cell culture, we lyse each cell in the microwell and perform western blotting on each resultant lysate. We observe cell spreading after overnight microwell‐based culture. scWB reports increased phosphorylation of MAP kinases (ERK1/2, p38) under hypertonic conditions. We validate the in situ scWB with slab‐gel western blot, while revealing cell‐to‐cell heterogeneity in stress responses.  相似文献

提出了一种以MEMS技术为基础, 可在低电压驱动条件下工作的创新型细胞电融合芯片. 该芯片的设计原理在于通过缩短微电极间的间距, 在低电压条件下获得足够强度的排队和融合电场强度. 原型芯片以SOI硅片为加工材料, 通过刻蚀方式在顶层低阻硅形成微电极和微通道; 在微电极上沉淀2 μm厚的铝膜以降低电阻率, 提高导电性; 通过PECVD方法形成150 nm厚SiO2保障铝膜的抗腐蚀性及芯片生物相容性; 芯片最终采用DIP法进行封装. 在该芯片上进行了低电压(传统电融合设备工作电压的1/20)驱动条件下的基于介电电泳的细胞排队实验及后期的细胞电融合实验, 结果表明, 细胞多以两两结合的方式排列, 与传统的细胞融合电仪器相比较, 降低了多细胞排队概率, 进而减少了传统电融合设备多细胞融合的概率, 为细胞高效率融合奠定了基础. 在加载的低电压短脉冲信号后, 微通道中形成了高压短脉冲电场, 在脉冲作用下, 烟草原生质体细胞在微通道中发生了融合, 融合时间(2 min)远低于传统电融合方法(10~30 min), 融合率远远高于传统的PEG方法(融合率小于1%)和传统电融合方法(利用BTX ECM 2001细胞电融合系统得到, 融合率小于5%).  相似文献

The capability to encapsulate single cells in droplets while retaining high cell viability (>90%) has great impact on tissue engineering, high-throughput screening, as well as clinical diagnostics and therapeutics. We demonstrate a novel method to vitrify a small number of cells using cell-encapsulating droplets. The method allows vitrification at low cryoprotectant concentration (1.5 M propanediol and 0.5 M trehalose), similar to that used in slow freezing protocols. The method was successfully applied to five different mammalian cell types: AML-12 hepatocytes, NIH-3T3 fibroblasts, HL-1 cardiomyocytes, mouse embryonic stem cells, and RAJI cells.  相似文献

Madison Darby canine kidney II (MDCK II) cells were seeded out at two different densities and incubated with 125 micrograms/mL of the photosensitizer meso-tetra(4-sulfonatophenyl)porphine (TPPS4) for 18 h, washed and irradiated with blue light. Four hours later the cells were studied by fluorescence microscopy. Apoptotic cells were detected by virtue of the distinct condensation and fragmentation of chromatin, and necrotic cells were detected by uptake of propidium iodide. In addition apoptosis was measured by the TdT assay. The fraction of apoptotic cells and the fraction of necrotic cells were determined for both cell densities at various levels of survival. With < 55% total cell death the apoptotic fraction was significantly higher for cells in confluent monolayers than for cells growing in microcolonies at equitoxic doses. Confluent cells were 2.9 times more sensitive than cells in microcolonies partly due to a 1.5 times higher uptake of TPPS4 in monolayer cells. The difference in mode of cell death for the different cell densities was not related to any observable difference in subcellular localization pattern of TPPS4 at equitoxic doses of photodynamic treatment.  相似文献

The large‐scale and label‐free molecular characterization of single cells in their natural tissue habitat remains a major challenge in molecular biology. We present a method that integrates morphometric image analysis to delineate and classify individual cells with their single‐cell‐specific molecular profiles. 用凹面反射镜和平面反射镜组成结构简单、光路易调的光多次反射装置,计算了这种光学系统的稳定性条件。考虑实际入射光束的直径和发散角,给出了最大反射次数的求法。制成适用于钛宝石激光光声光谱仪的光56次通过的长程池。理论计算的反射光斑分布与实验观测一致。这个技术有利于提高光声光谱和吸收光谱的探测灵敏度。  相似文献
