We investigated the direct electrochemistry of glucose oxidase (GOx) at gelatin-multiwalled carbon nanotube (GCNT) modified glassy carbon electrode (GCE). GOx was covalently immobilized onto GCNT modified GCE through the well known glutaraldehyde (GAD) chemistry. The immobilized GOx showed a pair of well-defined reversible redox peaks with a formal potential (E0′) of ? 0.40 V and a peak to peak separation (ΔEp) of 47 mV. The surface coverage concentration (Г) of GOx in GCNT/GOx/GAD composite film modified GCE was 3.88 × 10? 9 mol cm? 2 which indicates the high enzyme loading. The electron transfer rate constant (ks) of GOx immobilized onto GCNT was 1.08 s? 1 which validates a rapid electron transfer processes. The composite film shows linear response towards 6.30 to 20.09 mM glucose. We observed a good sensitivity of 2.47 μA mM?1 cm? 2 for glucose at the composite film. The fabricated biosensor displayed two weeks stability. Moreover, it shows no response to 0.5 mM of ascorbic acid (AA), uric acid (UA), acetaminophen (AP), pyruvate (PA) and lactate (LA) which shows its potential application in the determination of glucose from human serum samples. The composite film exhibits excellent recovery for glucose in human serum at physiological pH with good practical applicability.  相似文献

Square-wave adsorptive stripping voltammetry technique was used to determine rosiglitazone (ROS) on the hanging mercury dropping electrode (HMDE) surface, in Britton Robinson buffer, pH = 5. The voltammetric cathodic peak was observed at ?1520 mV vs. Ag/AgCl reference electrode. The voltammetric peak response was characterized with respect to pH, supporting electrolyte, accumulation potential, preconcentration time, scan rate, frequency, pulse amplitude, surface area of the working electrode and the convection rate. Under optimal conditions, the voltammetric current is proportional to the concentration of ROS over the concentration range of 5 × 10?8–8 × 10?7 mol l?1 (r = 0.9899) with a detection limit of 3.2 × 10?11 mol l?1 using 120 s accumulation time. The developed SW-AdSV procedure showed a good reproducibility, the relative standard deviation RSD% (n = 10) at a concentration level of 5 × 10?7 mol l?1 was 0.33%, whereas the accuracy was 101% ± 1.0. The proposed method was successfully applied to assay the drug in the human urine and plasma samples with mean recoveries of 90 ± 0.71% and 86 ± 1.0%, respectively.  相似文献

In this work, a new biosensor was prepared through immobilization of bovine liver catalase in a photoreticulated poly (vinyl alcohol) membrane at the surface of a conductometric transducer. This biosensor was used to study the kinetics of catalase–H202 reaction and its inhibition by cyanide. Immobilized catalase exhibited a Michaelis–Menten behaviour at low H202 concentrations (< 100 mM) with apparent constant KMapp = 84 ± 3 mM and maximal initial velocity VMapp = 13.4 μS min? 1. Inhibition by cyanide was found to be non-competitive and inhibition binding constant Ki was 13.9 ± 0.3 μM. The decrease of the biosensor response by increasing cyanide concentration was linear up to 50 μM, with a cyanide detection limit of 6 μM. In parallel, electrochemical characteristics of the catalase/PVA biomembrane and its interaction with cyanide were studied by cyclic voltammetry and impedance spectroscopy. Addition of the biomembrane onto the gold electrodes induced a significant increase of the interfacial polarization resistance RP. On the contrary, cyanide binding resulted in a decrease of Rp proportional to KCN concentration in the 4 to 50 μM range. Inhibition coefficient I50 calculated by this powerful label-free and substrate-free technique (24.3 μM) was in good agreement with that determined from the substrate-dependent conductometric biosensor (24.9 μM).  相似文献

Square-wave voltammetry was used to explore the adsorption property of ofloxacine complex with iron ion on the hanging mercury drop electrode (HMDE). By employing the adsorptive stripping voltammetric approach, a sensitive electroanalytical method for the quantitative analysis of ofloxacine antibiotic was achieved. A well-developed voltammetric peak was obtained in pH 7.5 Britton–Robinson buffer (B–R buffer) at ?1400 mV. The cyclic voltammetric studies indicated that the reduction process was irreversible and primarily controlled by adsorption. An investigations of the variation of adsorptive voltammetric peak current with supporting electrolyte, pH, accumulation time, accumulation potential, ion concentration, scan rate, pulse amplitude, SW frequency, working electrode area and convection rate has resulted in the recognition of optimal experimental conditions for ofloxacine analysis. The studied electroanalytical signal showed a linear response for ofloxacine in the concentration range 5 × 10?7 to 1.7 × 10?6 mol l?1 (r = 0.999). A limit of detection of 1.1 × 10?8 mol l?1 (3.98 ppb) with relative standard deviation of 1.21 RSD% and mean recovery of 99.6% were obtained. Possible interferences by several substances usually present in pharmaceutical formulation were also evaluated. The analytical quantification of ofloxacine in commercially available pharmaceutical formulation was performed and compared with data from HPLC technique.  相似文献

The adsorption of uranium (VI) from aqueous solutions onto natural sepiolite has been studied using a batch adsorber. The parameters that affect the uranium (VI) sorption, such as contact time, solution pH, initial uranium(VI) concentration, and temperature, have been investigated and optimized conditions determined. Equilibrium isotherm studies were used to evaluate the maximum sorption capacity of sepiolite and experimental results showed this to be 34.61 mg · g?1. The experimental results were correlated reasonably well by the Langmuir adsorption isotherm and the isotherm parameters (Qo and b) were calculated. Thermodynamic parameters (ΔH° = ?126.64 kJ · mol?1, ΔS° = ?353.84 J · mol?1 · K?1, ΔG° = ?21.14 kJ · mol?1) showed the exothermic heat of adsorption and the feasibility of the process. The results suggested that sepiolite was suitable as sorbent material for recovery and adsorption of uranium (VI) ions from aqueous solutions.  相似文献

In this paper, a novel biosensor was prepared by immobilizing glucose oxidase (GOx) on carbon nanotube-gold-titania nanocomposites (CNT/Au/TiO2) modified glassy carbon electrode (GCE). SEM was initially used to investigate the surface morphology of CNT/Au/TiO2 nanocomposites modified GCE, indicating the formation of the nano-porous structure which could readily facilitate the attachment of GOx on the electrode surface. Cyclic voltammogram (CV) and electrochemical impedance spectrum (EIS) were further utilized to explore relevant electrochemical activity on CNT]Au/TiO2 nanocomposites modified GCE. The observations demonstrated that the immobilized GOx could efficiently execute its bioelectrocatalytic activity for the oxidation of glucose. The biosensor exhibited a wider linearity range from 0.1 mmol L-1 to 8 mmol L^-1 glucose with a detection limit of 0.077 mmol L^- 1.  相似文献

The adsorptive collection of zinc(II) complex with alizarin ligand, coupled with the square-wave voltammetric technique at the hanging mercury drop electrode, yields a very sensitive electroanalytical procedure for the determination of zinc. The optimized experimental conditions include: supporting electrolyte (carbonate buffer), pH (11), alizarin concentration (1 × 10?6 mol l?1), accumulation time (60 s), accumulation potential (?0.1 V), scan rate (700 mV s?1), pulse amplitude (0.06 V) and SW frequency (80 Hz). The monitored stripping voltammetric current was linear over the range of 5 × 10?8 – 4 × 10?7 mol l?1 and the detection limit was 1 × 10?8 mol l?1. The relative standard deviation was calculated as 1.3% (n = 10) for 1 × 10?8 mol l?1 Zn(II) and the obtained electrochemical signal was stabile for up to 60 min. Possible interferences by either co-existing metal ions or other chelating agents were also investigated. The applicability of the proposed SW-AdSV method to the analysis of foodstuff was assessed by the determination of zinc content in instant coffee samples. The accuracy of the obtained voltammetric analytical results was validated by comparing with that obtained by atomic absorption spectrometric method and conducting the necessary statistical evaluation.  相似文献

The solubility of anthracene was measured in pure water and in sodium chloride aqueous solution (salt concentration, m/mol · kg?1 = 0.1006, 0.5056, and 0.6082) at temperatures between (278 and 333) K. Solubility of anthracene in pure water agrees fairly well with values reported in earlier similar studies. Solubility of anthracene in sodium chloride aqueous solutions ranged from (6 · 10?8 to 143 · 10?8) mol · kg?1. Sodium chloride had a salting-out effect on the solubility of anthracene. The salting-out coefficients did not vary significantly with temperature over the range studied. The average salting-out coefficient for anthracene was 0.256 kg · mol?1.The standard molar Gibbs free energies, ΔtrG°, enthalpies, ΔtrH°, and entropies, ΔtrS°, for the transfer of anthracene from pure water to sodium chloride aqueous solutions were also estimated. Most of the estimated ΔtrG° values were positive [(20 to 1230) J · mol?1]. The analysis of the thermodynamic parameters shows that the transfer of anthracene from pure water to sodium chloride aqueous solution is thermodynamically unfavorable, and that this unfavorable condition is caused by a decrease in entropy.  相似文献

The solubility measurements of sodium dicarboxylate salts; sodium oxalate, malonate, succinate, glutarate, and adipate in water at temperatures from (278.15 to 358.15 K) were determined. The molar enthalpies of solution at T = 298.15 K were derived: ΔsolHm (m = 2.11 mol · kg?1) = 13.86 kJ · mol?1 for sodium oxalate; ΔsolHm (m = 3.99 mol · kg?1) = 14.83 kJ · mol?1 for sodium malonate; ΔsolHm (m = 2.45 mol · kg?1) = 14.83 kJ · mol?1 for sodium succinate; ΔsolHm (m = 4.53 mol · kg?1) = 16.55 kJ · mol?1 for sodium glutarate, and ΔsolHm (m = 3.52 mol · kg?1) = 15.70 kJ · mol?1 for sodium adipate. The solubility value exhibits a prominent odd–even effect with respect to terms with odd number of sodium dicarboxylate carbon numbers showing much higher solubility. This odd–even effect may have implications for the relative abundance of these compounds in industrial applications and also in the atmospheric aerosols.  相似文献

A new type of amperometric hydrogen peroxide biosensor was fabricated by entrapping horseradish peroxidase (HRP) in the organic–inorganic hybrid material composed of zirconia–chitosan sol–gel and Au nanoparticles (ZrO2–CS–AuNPs). The sensitivity of the biosensor was enhanced by a flowerlike polymer–copper nanostructure composite (pPA–FCu) which was prepared from co-electrodeposition of CuSO4 solution and 2,6-pyridinediamine solution. Several techniques, including UV–vis absorption spectroscopy, scanning electron microscopy, cyclic voltammetry, chronoamperometry and electrochemical impedance spectroscopy were employed to characterize the assembly process and performance of the biosensor. The results showed that this pPA–FCu nanostructure not only had excellent redox electrochemical activity, but also had good catalytic efficiency for hydrogen peroxide. Also the ZrO2–CS–AuNPs had good film forming ability, high stability and good retention of bioactivity of the immobilized enzyme. The resulting biosensors showed a linear range from 7.80 × 10?7 to 3.7 × 10?3 mol L?1, with a detection limit of 3.2 × 10?7 mol L?1 (S/N = 3) under optimized experimental conditions. The apparent Michaelis–Menten constant was determined to be 0.32 mM, showing good affinity. In addition, the biosensor which exhibits good analytical performance, acceptable stability and good selectivity, has potential for practical applications.  相似文献

This paper describes, for the first time, the development of a simple and highly sensitive chemical sensor based on a new electroactive material, electrogenerated in situ from xanthurenic acid on an electrode modified with MWCNT. The modified electrode shows efficient electrocatalytic oxidation activity towards NADH at an applied potential of 0.1 V vs. Ag/AgCl. The kinetic constant, kkin, for the electrocatalytic oxidation of NADH, evaluated by chronoamperometry and voltammetry using RDE, provided values close to 105 mol?1 L s?1.  相似文献

Hydroboration reactions of 4-octene with HBBr2 · SMe2, HBCl2 · SMe2 and H2BBr · SMe2 in CH2Cl2 were studied as function of concentration and temperature and compared with those of 1-octene. On average, hydroboration with dihaloborane proceeded 16 times slower for 4-octene than for 1-octene. In the case of the reactions with the monohaloborane, this factor is halved. This can be explained by the difference in the relative rates of dissociates of Me2S from the dihaloborane and a monohaloborane complex, respectively. The reactions involving H2BBr · SMe2 also exhibited a k?2 value, an indication of the presence of a parallel reaction, most likely a rearrangement process facilitating isomerization by way of a π-complex. The moderate ΔH values accompanied by small ΔS values (94 ± 4 kJ mol?1, ?3 ± 13 J K?1 mol?1 for HBBr2 · SMe2; 93 ± 1 kJ mol?1, ?17 ± 4 J K?1 mol?1 for HBCl2 · SMe2 and in the case of H2BBr · SMe2, 90 ± 13 kJ mol?1, +12 ± 44 J K?1 mol?1 and 83 ± 13 kJ mol?1, ?24 ± 45 J K?1 mol?1, respectively, for the k2 and k?2 processes) imply a process that is dissociatively dominated, with the overall mode of activation being interchange dissociative (Id).  相似文献

MgO polyhedral nanocages and nanocrystals, synthesized by non-catalytic simple thermal evaporation process, were used to fabricate high-sensitive amperometric glucose biosensor which showed a high and reproducible sensitivity of 31.6 μA μM?1 cm?2 with a response time less than 5 s, linear dynamic range from 1.0 to 9.0 μM and correlation coefficient of R = 0.9993. The detection limit of fabricated biosensor (based on S/N ratio = 3) was estimated to be 68.3 ± 0.02 nM. To the best of our knowledge, this is the first report which demonstrates the use of MgO nanostructures for the fabrication of glucose biosensor; hence, this work opens a new way to utilize MgO nanostructures as an efficient electron mediator to fabricate efficient glucose biosensors.  相似文献

A magnesium-based metal organic framework (MOF), also known as Mg-MOF-74, was successfully synthesized, characterized, and evaluated for adsorption equilibria and kinetics of CO2 and CH4. The Mg-MOF-74 crystals were characterized with scanning electron microscopy for crystal structure, powder X-ray diffraction for phase structure, and nitrogen adsorption for pore textural properties. Adsorption equilibrium and kinetics of CO2 and CH4 on the Mg-MOF-74 adsorbent were measured in a volumetric adsorption unit at 278, 298, and 318 K and pressures up to 1 bar. It was found that the Mg-MOF-74 adsorbent prepared in this work has a median pore width of 10.2 Å, a BET specific surface area of 1174 m2/g, CO2 and CH4 adsorption capacities of 8.61 mmol g?1 (37.8 wt.%) and 1.05 mmol g?1 (1.7 wt.%), respectively, at 298 K and 1 bar. Both CO2 and CH4 adsorption capacities are significantly higher than those of zeolite 13X under similar conditions. The pressure-dependent equilibrium selectivity of CO2 over CH4 (qCO2/qCH4) in the Mg-MOF-74 adsorbent showed a trend similar to that of zeolite 13X and the intrinsic selectivity of Mg-MOF-74 at zero adsorption loading is 283 at 298 K. The initial heats of adsorption of CO2 and CH4 on the Mg-MOF-74 adsorbent were found to be 73.0 and 18.5 kJ mol?1, respectively. The adsorption kinetic measurements suggest that the diffusivities of CO2 and CH4 on Mg-MOF-74 were comparable to those on zeolite 13X. CH4 showed relatively faster adsorption kinetics than CO2 in both adsorbents. The diffusion time constants of CO2 and CH4 in the Mg-MOF-74 adsorbent at 298 K were estimated to be 8.11 × 10?3 and 4.05 × 10?2 s?1, respectively, showing a modest kinetic selectivity of about 5 for the separation CH4 from CO2.  相似文献

We determined apparent molar volumes V? at 278.15 ? (T/K) ? 368.15 and apparent molar heat capacities Cp,? at 278.15 ? (T/K) ? 393.15 at p = 0.35 MPa for aqueous solutions of tetrahydrofuran at m from (0.016 to 2.5) mol · kg?1, dimethyl sulfoxide at m from (0.02 to 3.0) mol · kg?1, 1,4-dioxane at m from (0.015 to 2.0) mol · kg?1, and 1,2-dimethoxyethane at m from (0.01 to 2.0) mol · kg?1. Values of V? were determined from densities measured with a vibrating-tube densimeter, and values of Cp,? were determined with a twin fixed-cell, differential, temperature-scanning calorimeter. An electropolymerized melatonin modified glassy carbon electrode (EPMT/GCE) was prepared by electrochemically polymerizing of melatonin in a 0.04 mol l?1 perchloric acid solution. In KCl solution, Ca2+ was found to perform a reversible complex reaction on the EPMT/GCE to produce a sharp complex adsorption wave. The configuration of the electropolymerized membrane was characterized by scanning electron microscope and various electrochemical techniques. Based on which, a novel sensitive Ca2+ electrochemical sensor was established. The peak current showed a linear relationship with Ca2+ concentration in range of 6.2 × 10?7–1.0 × 10?4 mol l?1 with detection limit of 4.5 × 10?7 mol l?1. The developed electrochemical sensor has been used for determination of Ca2+ in body fluid.  相似文献

Fast analysis of thiocyanate by ion-pair chromatography using a silica-based monolithic column and direct conductivity detection was carried out. Chromatographic separation was performed on a Chromolith Speed ROD RP-18e using tetrabutylammonium hydroxide (TBA)-phthalic acid-acetonitrile as eluent. The effects of eluent concentration, eluent pH value, column temperature and flow rate on retention time of thiocyanate were investigated. The optimized chromatographic conditions for the determination of thiocyanate were as follows: 0.25 mmol/L TBA-0.18 mmol/L phthalate-7% acetonitrile (pH 5.5) as eluent, column temperature of 30 ℃, and flow rate of 6.0 mL/min. Retention time of thiocyanate was less than 1 min under the conditions. Common anions (Cl^-, NO3 , SO42 and I^- ) did not interfere with the determination of thiocyanate. Detection limit (S/N = 3) of thiocyanate was 0.96 mg/L. Calibration graph between peak area and the concentration of thiocyanate was linear in the range of 2.0- 100.0 mg/L. Relative standard deviation (RSD) of chromatographic peak area was 1.4% (n = 5). This method has been applied to the determination of thiocyanate in ionic liquids. Recoveries of thiocyanate after spiking were 100.5%.  相似文献

Human serum albumin (HSA) was the most abundant protein in human plasma and has significant physiological function. In Tris–HCl buffer solution (pH 7.4), water-soluble semiconductor CdSe quantum dots (QDs) reacted with HSA and the products resulted in a great enhancement of the intensity of resonance Rayleigh scattering (RRS) and second-order scattering (SOS). Based on this, a new method was developed to investigate the interactions between QDs and HSA. The parameters with regard to determination were optimized, and the reaction mechanism was discussed. Under optimal conditions, the increments of scattering intensity (ΔI) were directly proportional to the concentrations of HSA in the range of 0.4–48.0 μmol L?1. The detection limits were 0.10 μmol L?1 for RRS method and 0.25 μmol L?1 for SOS method. The proposed method was sensitive, simple and rapid. It has been successfully applied to the determination of HSA in human urine samples. Analytical results obtained with this novel assay were satisfactory.  相似文献

The heat capacity of polycrystalline germanium disulfide α-GeS2 has been measured by relaxation calorimetry, adiabatic calorimetry, DSC and heat flux calorimetry from T = (2 to 1240) K. Values of the molar heat capacity, standard molar entropy and standard molar enthalpy are 66.191 J · K?1 · mol?1, 87.935 J · K?1 · mol?1 and 12.642 kJ · mol?1. The temperature of fusion and its enthalpy change are 1116 K and 23 kJ · mol?1, respectively. The thermodynamic functions of α-GeS2 were calculated over the range (0 ? T/K ? 1250).  相似文献
