Inorganic nanomaterials have attracted substantial research interest due to their unique intrinsic physicochemical properties.We highlighted recent advances in the applications of inorganic nanoparticles regarding their imaging efficacy, focusing on tumor-imaging nanomaterials such as metal-based and carbon-based nanomaterials and quantum dots. Inorganic nanoparticles gain excellent in vivo tumor-imaging functions based on their specific characteristics of strong near-infrared optical absorption and/or X-ray attenuation capability. The specific response signals from these novel nanomaterials can be captured using a series of imaging techniques, i.e., optical coherence tomography(OCT), X-ray computed tomography(CT) imaging, two-photon luminescence(TPL), photoacoustic tomography(PAT), magnetic resonance imaging(MRI), surface-enhanced Raman scattering(SERS) and positron emission tomography(PET). In this review, we summarized the rapid development of inorganic nanomaterial applications using these analysis techniques and discussed the related safety issues of these materials.  相似文献

Delivery and toxicity are critical issues facing nanomedicine research. Currently, there is limited understanding and connection between the physicochemical properties of a nanomaterial and its interactions with a physiological system. As a result, it remains unclear how to optimally synthesize and chemically modify nanomaterials for in vivo applications. It has been suggested that the physicochemical properties of a nanomaterial after synthesis, known as its "synthetic identity", are not what a cell encounters in vivo. Adsorption of blood components and interactions with phagocytes can modify the size, aggregation state, and interfacial composition of a nanomaterial, giving it a distinct "biological identity". Here, we investigate the role of size and surface chemistry in mediating serum protein adsorption to gold nanoparticles and their subsequent uptake by macrophages. Using label-free liquid chromatography tandem mass spectrometry, we find that over 70 different serum proteins are heterogeneously adsorbed to the surface of gold nanoparticles. The relative density of each of these adsorbed proteins depends on nanoparticle size and poly(ethylene glycol) grafting density. Variations in serum protein adsorption correlate with differences in the mechanism and efficiency of nanoparticle uptake by a macrophage cell line. Macrophages contribute to the poor efficiency of nanomaterial delivery into diseased tissues, redistribution of nanomaterials within the body, and potential toxicity. This study establishes principles for the rational design of clinically useful nanomaterials.  相似文献

Nanomaterials with localized surface plasmon resonance (LSPR) locating in the near-infrared region have broad application prospects in the field of biomedicine. However, the biggest problem that limits the biomedical application of such nanomaterials lies in two aspects: First, the potential long-term in vivo toxicity caused by the metabolism of many nanomaterials with LSPR effect; Second, most of current nanomaterials with LSPR effect are difficult to achieve LSPR wavelength tunability in the near-infrared region to adapt to different biomedical applications. Copper selenide nanomaterials are composed of selenium and copper, which are necessary nutrient elements for human life. Because of the active and flexible chemical properties of selenium and copper, copper selenide nanomaterials can not only be effectively degraded and utilized in human body, but also be endowed with various physicochemical properties by chemical modification or doping. Recently, copper selenide nanomaterials have shown unique properties such as LSPR in the near-infrared region, making them attractive for near-infrared thermal ablation, photoacoustic imaging, disease marker detection, multimode imaging, and so on. Currently, to the best of our knowledge, there is no review on the LSPR properties of copper selenide nanomaterials and its biomedical applications. This review first discusses the relationship between the physicochemical properties and the LSPR of copper selenide nanomaterials and then summarizes the latest progress in the application of copper selenide nanomaterials in biological detection, diagnosis, and treatment of diseases. In addition, the advantages, and prospects of copper selenide nanomaterials in biomedicine are also highlighted.  相似文献

In the past few decades, brain diseases have taken a heavy toll on human health and social systems. Magnetic resonance imaging (MRI), photoacoustic imaging (PA), computed tomography (CT), and other imaging modes play important roles in disease prevention and treatment. However, the disadvantages of traditional imaging mode, such as long imaging time and large noise, limit the effective diagnosis of diseases, and reduce the precision treatment of diseases. The ever-growing applications of inorganic nanomaterials in biomedicine provide an exciting way to develop novel imaging systems. Moreover, these nanomaterials with special physicochemical characteristics can be modified by surface modification or combined with functional materials to improve targeting in different diseases of the brain to achieve accurate imaging of disease regions. This article reviews the potential applications of different types of inorganic nanomaterials in vivo imaging and in vitro detection of different brain disease models in recent years. In addition, the future trends, opportunities, and disadvantages of inorganic nanomaterials in the application of brain diseases are also discussed. Additionally, recommendations for improving the sensitivity and accuracy of inorganic nanomaterials in screening/diagnosis of brain diseases.  相似文献

The interest in graphene for biomedical applications has grown substantially in the past few years creating a need for biocompatibility testing. Biomedical engineering applications using graphene such as biosensing devices, microbial detection, disease diagnosis, and drug delivery systems are progressing rapidly, perhaps overlooking any possible hazards as graphene nanomaterials may interact with biological materials differently than other graphitic materials such as carbon nanotubes and fullerenes. As a potential application for graphene is drug delivery, the toxicity of graphene was tested against an in vitro model of the blood brain barrier (BBB) by measuring trans-endothelial-electrical resistance (TEER). A new approach in terms of electrical impedance sensing was also utilized to kinetically analyze the cytotoxicity of graphene nanomaterials towards the BBB model's individual components, rat astrocytes (CRL-2006) and mouse endothelial cells (CRL-2583), in real time by measuring the impedimetric response. Graphene showed little or no toxicity toward both individual cell types as the resistance measurements were similar to those of the control and further, graphene did not interrupt the integrity of the BBB model as a whole showing the biocompatibility of graphene and the broad potential of using these new nanomaterials for biomedical applications.  相似文献

Lab-on-a-chip (LOC) platforms have become important tools for sample analysis and treatment with interest for DNA, protein and cells studies or diagnostics due to benefits such as the reduced sample volume, low cost, portability and the possibility to build new analytical devices or be integrated into conventional ones. These platforms have advantages of a wide set of nanomaterials (NM) (i.e. nanoparticles, quantum dots, nanowires, graphene etc.) and offer excellent improvement in properties for many applications (i.e. detectors sensitivity enhancement, biolabelling capability along with other in-chip applications related to the specificities of the variety of nanomaterials with optical, electrical and/or mechanical properties). This review covers the last trends in the use of nanomaterials in microfluidic systems and the related advantages in analytical and bioanalytical applications. In addition to the applications of nanomaterials in LOCs, we also discuss the employment of such devices for the production and characterization of nanomaterials. Both framed platforms, NMs based LOCs and LOCs for NMs production and characterization, represent promising alternatives to generate new nanotechnology tools for point-of-care diagnostics, drug delivery and nanotoxicology applications.  相似文献

Two-dimensional (2D) nanomaterials have drawn tremendous attention due to their unique physicochemical properties and promising applications in the fields of electronics, energy storage, and catalysis. Recently, the biomedicine community has gradually started to recognize the great potential of these nanostructured materials for biomedical applications – in particular those related to cancer therapy. In this review, we provide a brief overview of a few representative 2D nanomaterials, discuss their preparation strategies and physicochemical properties, and highlight their applications in cancer nanomedicine. We expect that this review will shed some light on the new opportunities associated with 2D nanomaterials for biomedical research.  相似文献

Protein assemblies have recently become known as potential molecular scaffolds for applications in materials science and bio‐nanotechnology. Efforts to design protein assemblies for construction of protein‐based hybrid materials with metal ions, metal complexes, nanomaterials and proteins now represent a growing field with a common aim of providing novel functions and mimicking natural functions. However, the important roles of protein assemblies in coordination and biosupramolecular chemistry have not been systematically investigated and characterized. In this personal account, we focus on our recent progress in rational design of protein assemblies using bioinorganic chemistry for (1) exploration of unnatural reactions, (2) construction of functional protein architectures, and (3) in vivo applications.  相似文献

Understanding the interactions of nanomaterials with the immune system is essential for the engineering of new macromolecular systems for in vivo applications. Systematic study of immune activation is challenging due to the complex structure of most macromolecular probes. We present here the use of engineered gold nanoparticles to determine the sole effect of hydrophobicity on the immune response of splenocytes. The gene expression profile of a range of cytokines (immunological reporters) was analyzed against the calculated log P of the nanoparticle headgroups, with an essentially linear increase in immune activity with the increase in hydrophobicity observed in vitro. Consistent behavior was observed with in vivo mouse models, demonstrating the importance of hydrophobicity in immune system activation.  相似文献

Designing of nanomaterials has now become a top-priority research goal with a view to developing specific applications in the biomedical fields. In fact, the recent trends in the literature show that there is a lack of in-depth reviews that specifically highlight the current knowledge based on the design and production of nanomaterials. Considerations of size, shape, surface charge and microstructures are important factors in this regard as they affect the performance of nanoparticles (NPs). These parameters are also found to be dependent on their synthesis methods. The characterisation techniques that have been used for the investigation of these nanomaterials are relatively different in their concepts, sample preparation methods and obtained results. Consequently, this review article aims to carry out an in-depth discussion on the recent trends on nanomaterials for biomedical engineering, with a particular emphasis on the choices of the nanomaterials, preparation methods/instruments and characterisations techniques used for designing of nanomaterials. Key applications of these nanomaterials, such as tissue regeneration, medication delivery and wound healing, are also discussed briefly. Covering this knowledge gap will result in a better understanding of the role of nanomaterial design and subsequent larger-scale applications in terms of both its potential and difficulties.  相似文献

Electrochemiluminescent biosensors are nowadays an established technology in the field of immunosensors and diagnostics. Along with the advent of nanotechnology, the marriage between electrochemiluminescence and nanomaterials results in promising enhancing strategies in many biosensor applications. Among nanomaterials, carbon-based ones are the most used, as (i) scaffolds, (ii) luminophores and (iii) electrode materials of the sensor. In this review, we describe the importance of a rational modification and functionalization of carbon nanomaterials to optimize electrochemiluminescence signal, and we also resume the latest and most relevant applications of electrochemiluminescent biosensors based on carbon nanomaterials.  相似文献

Multifunctional bismuth sulfide (Bi2S3) nanomaterials exhibit significant potential as nanomedicines for the diagnosis and treatment of cancer. These nanomaterials act as excellent photothermal agents and radiation sensitizers for the treatment of tumors, and they can also act as contrast agents for computed tomography (CT) imaging, photoacoustic imaging (PA), and other forms of imaging to provide real-time tumor monitoring and testing guidance. Compared with other nanomaterials, Bi2S3 nanomaterials can readily adapt to different applications by virtue of the fact that they can be easily functionalized. However, these nanomaterials have some limitations that cannot be ignored and need to be addressed, such as poor biocompatibility, toxicity, and low chemical stability. It is widely believed that appropriate functionalization of Bi2S3 nanomaterials could remedy such defects and significantly improve performance. This review summarizes the ways in which Bi2S3 nanomaterials can be functionalized and discusses their applications in cancer theranostics over the last few years, focusing particularly on imaging and therapy. We also discuss issues relating to how Bi2S3 nanomaterials can be analyzed, including how we might be able to use these systems to inhibit and treat tumors and how current limitations might be overcome to improve treatment efficacy. Finally, we hope to provide inspiration and guidance as to how we might create a more optimized multifunctional nano-system for the diagnosis and treatment of tumors.  相似文献

石墨碳纳米材料因其特殊的光学性质而受到广泛关注。石墨碳纳米材料最引人注目的光学性质之一是其独特的拉曼性质,作为拉曼探针,石墨碳纳米材料在对于复杂生物样品,极端测试条件和定量拉曼检测方面都有很好的应用;除了拉曼性质以外,单壁碳纳米管(SWNTs)独特的近红外二区(NIR-II,1000-1700 nm)荧光性质,具有穿透深度大、分辨率高的荧光成像特点,在生物活体成像领域也得到了很好的应用。除了光致发光特性,石墨碳纳米材料还具有优异的光热转换效应,同时具有比表面积大的特点,被广泛应用在针对肿瘤的热疗及其它疗法协同治疗当中。除此之外,石墨碳纳米材料还是一种高效的信号传导基底,可以猝灭激发态的染料和光敏剂,利用该类性质设计的生物传感器和纳米药物,显现出高灵敏、高选择性的特点。本文主要结合本课题组的工作,总结和探讨了石墨碳纳米材料作为光学探针、光热材料和信号传递基底在生化传感领域的应用。  相似文献

光动力治疗是新兴的非侵入性癌症治疗方法。纳米材料以其独特的结构以及光物理、光化学性质成为可用于光动力治疗的光敏剂。根据纳米材料的不同种类,分别对无机非金属纳米材料、无机金属纳米材料、有机小分子纳米材料以及有机聚合物纳米材料等的构建策略及其在光动力治疗肿瘤中的应用进行综述。展望了纳米材料在未来肿瘤光动力治疗中的挑战和发展方向。为新一代纳米光敏剂的构建提供创新思路,并扩展其在癌症治疗中的潜力。  相似文献

For more than a decade, the exfoliation of graphene and other layered materials has led to a tremendous amount of research in two-dimensional (2D) materials, among which 2D transition metal chalcogenides (TMCs) nanomaterials have attracted much attention in a wide range of applications including photoelectric devices, lithium-ion batteries, catalysis, and energy conversion and storage owing to their unique photoelectric physical properties. With such large specific surface area, strong near-infrared (NIR) absorption and abundant chemical element composition, 2D TMCs nanomaterials have become good candidates in biomedical imaging and cancer treatment. This review systematically summarizes recent progress on 2D TMCs nanomaterials, which includes their synthesis methods and applications in cancer treatment. At the end of this review, we also highlight the future prospects and challenges of 2D TMCs nanomaterials. It is expected that this work can provide the readers with a detailed overview of the synthesis of 2D TMCs and inspire more novel functional biomaterials based on 2D TMCs for cancer treatment in the future.  相似文献

Identifying key factors that govern the in vivo behavior of nanomaterials is critical to the clinical translation of nanomedicines. Overshadowed by size‐, shape‐, and surface‐chemistry effects, the impact of the particle core density on clearance and tumor targeting of inorganic nanoparticles (NPs) remains largely unknown. By utilizing a class of ultrasmall metal NPs with the same size and surface chemistry but different densities, we found that the renal‐clearance efficiency exponentially increased in the early elimination phase while passive tumor targeting linearly decreased with a decrease in particle density. Moreover, lower‐density NPs are more easily distributed in the body and have shorter retention times in highly permeable organs than higher‐density NPs. The density‐dependent in vivo behavior of metal NPs likely results from their distinct margination in laminar blood flow, which opens up a new path for precise control of nanomedicines in vivo.  相似文献

Nanomaterials manipulation using dielectrophoresis (DEP) is one of the major research areas that could potentially benefit the micro/nano science for diverse applications, such as microfluidics, nanomachine, and biosensor. The innovation and development of basic theories, methods or applications will have a huge impact on the entire related field. Specifically, for DEP manipulation of nanomaterials, improvements in comprehensive performance of accuracy, flexibility and scale could promote broader applications in micro/nano science. Therefore, to explore the directions for future research, this paper critically provides an overview on the fundamentals, recent progress, current challenges, and potential applications of DEP manipulation of nanomaterials.
